Enhanced production in recycle fed-batch cultivation by Lactic acid bacteria Isolated from Kimchi

  • A process for efficient recycle fed-batch culture was carried out to increase cell mass and spore production by Lactic acid bacteria isolated from Kimchi. A large quantity of cell mass obtained by feeding concentration of sugar in recycle fed-batch culture. When the high density of salt was created that the cell mass was come-down. In this study, cultured in different feeding concentration of sugar conditions. Lactic acid bacteria by recycle fed-batch culture was investigated in 2L working volume of fermenter, obtained the maximum cell mass was 15.17g/L.

Control of Feed Rate Using Neurocontroller Incorporated with Genetic Algorithm in Fed-Batch Cultivation of Scutellaria baicalensis Georgi

  • To enhance the production of flavonoids [baicalin, wogonin-7-Ο-glucuronic acid (GA)], which are secondary metabolites of Scutellaria baicalensis Georgi(G.) plant cells, a multilayer perceptron control system was applied to regulate the substrate feeding in a fed-batch cultivation. The optimal profile for the substrate feeding rate in a fed-batch culture of S. baicalensis G. was determined by simulating a kinetic model using a genetic algorithm. Process variable profiles were then prepared for the construction of a multilayer perceptron controller that included massive parallelism, trainability, and fault tolerance. An error back-propagation algorithm was applied to train the multiplayer perceptron. The experimental results showed that neurocontrol incorporated with a genetic algorithm improved the flavonoid production compared with a simple fuzzy logic control system. Furthermore, the specific production yield and flavonoid productivity also increased.

Maximization of cell growth and polysaccharide production from Agaricus blazei by fed-batch cultivation

  • 본 연구에서는 fed-batch를 통하여 Agaricus blazei의 균체증식과 다당체 생산을 증가시키고자 공급배지와 배지공급속도에 변화를 주어 균체증식과 다당체 생성에 미치는 영향을 비교 ${\cdot}$ 검토하였다. 유가식 배양에서의 최종균체량 및 다당체 생성량은 회분식 배양에 비해 훨씬 높았으며, 초기에 균체증식을 위해 공급배지로 yeast extract를 먼저 공급하고 4일째부터 다당체 생성을 위해서 glucose를 공급할 경우 균체량은 18.2 g/L, 다당체 생성량은 10.4 g/L를 생산하여 배양결과 최대 생산량을 나타내었다.

Combined Age and Segregated Kinetic Model for Industrial-scale Penicillin Fed-batch Cultivation

  • This paper proposes a cell age model for Penicillium chrysogenum fed-batch cultivation to supply a qualitative insight into morphology-associated dynamics. The average ages of the segregated cell populations, such as growing cells, non-growing cells and intact productive cells, were estimated by this model. A combined model was obtained by incorporating the aver-age ages of the cell sub-populations into a known but modified segregated kinetic model from literature. For simulations, no additional effort was needed for parameter identification since the cell age model has no internal parameters. Validation of the combined model was per-formed by 20 charges of industrial-scale penicillin cultivation. Meanwhile, only two charge-dependent parameters were required in the combined model among approximately 20 parameters in total. The model is thus easily transformed into an adaptive model for a further application in on-line state variables prediction and optimal scheduling.

  • 1,2-propanediol (1,2-PD) is a commodity chemical that is currently produced from petrochemical derivatives. Saccharomyces cerevisiae is well characterized and a successful industrial microorganism to enable the improvement of the 1,2-propanediol production by metabolic engineering. A recombinant S. cerevisiae M3G3 was used to produce 1,2-propanediol. S. cerevisiae M3G3 is the diploid strain that contains 3 copies of mgs (methylglyoxal synthase) and gldA (glycerol dehydrogenase). S. cerevisiae M3G3 was cultivated at various culture conditions by changing culture temperature, glucose concentration, and inducer concentration. Also the effect of induction time was studied to optimize the production of 1,2-propanediol. Batch and fed-batch cultivation of S. cerevisiae M3G3 was performed by using a 5 L jar fermenter. The highest concentration of 1,2-propanediol in batch cultivation was 0.86 g/L and it was further improved to 1.33 g/L in fed-batch cultivation.

Glucoamylase Production in Batch and Fed-Batch Solid State Fermentation: Effect of Maltose or Starch Addition

  • Maltose and soluble starch were used as secondary sources of carbon for glucoamylase production by Aspergillus awamori in solid state fermentation. During batch cultivation, maltose above 2.5%(w/w) repressed glucoamylase production, but, by adding either 2.5% (w/w) maltose or 1.25% (w/w) soluble starch to fed-batch cultivations, glucoamylase activity was increased by 15% and 170% over standard medium, respectively. The data showed that maltose is a weak inducer of glucoamylase production in solid stat fermentation.

Application of a Fed-Batch Bioprocess for the Heterologous Production of hSCOMT in Escherichia coli

  • In this paper, a fed-batch cultivation process in recombinant Escherichia coli BL21(DE3) bacteria, for the production of human soluble catechol-O-methyltransferase (hSCOMT), is presented. For the first time, a straightforward model is applied in a recombinant hSCOMT expression system and distinguishes an initial cell growth phase from a protein production phase upon induction. Specifically, the kinetic model predicts biomass, substrate, and product concentrations in the culture over time and was identified from a series of fed-batch experiments designed by testing several feed profiles. The main advantage of this model is that its parameters can be identified more reliably from distinct fed-batch strategies, such as glycerol pulses and exponential followed by constant substrate additions. Interestingly, with the limited amount of data available, the proposed model accomplishes satisfactorily the experimental results obtained for the three state variables, and no exhaustive process knowledge is required. The comparison of the measurement data obtained in a validation experiment with the model predictions showed the great extrapolation capability of the model presented, which could provide new complementary information for the COMT production system.

Cultivation of the Hyperthermophilic Archaeon Sulfolobus solfataricus in Low-Salt Media

  • Two low-salt complex media, bactopeptone and desalted yeast extract, were used for high density cultivation of the hyperthermophilic archaeon Sulfolobus solfataricus (DSM 1617). Bactopeptone, which has low mineral ion content among various complex media, was good for cell growth in batch cultures; the maximal cell density in bactopeptone was comparable to that in yeast extract. However, cell growth was rather poor when bactopeptone was added by the fed-batch procedure. Since several vitamins are deficient in abctopeptone, the effect of vitamins on cell growth was examined. Among the vitamins tested, pyridoxine was found to improve the growth rate of S. solfataricus. To reduce the growth inhibition caused by mineral ions, yeast extract was dialyzed against distilled water and then fed-batch cultures were carried out using a fed medium containing desalted yeast extract. Although the concentrations of mineral ions in yeast extract were significantly lowered by the dialysis whether low molecular weight solutes in yest extract are crucial for cell growth, we investigated the effect of trehalose, a most abundant compatible solute in yeast extract, on the growth pattern. Cell densities were increased and the length of the lag phase was markedly shortened by the presence of trehalose, indicating that trehalose plays an important role in the growth of S. solfataricus.

Enhancement of BDNF Production by Co-cultivation of Human Neuroblastoma and Fibroblast Cells

  • It has been proved that co-cultivation of human neroblastoma cells and human fibroblast cells can enhance nerve cell growth and the production of BDNF in perfusion cultivation. In batch co-cultivation, maximum cell density was increased up to 1.76${\times}$106 viable cells/mL from 9${\times}$105 viable cells/mL of only neuroblastoma cell culture. The growth of neuroblastoma cells was greatly improved by culturing both nerve and fibroblast cells in a perfusion process, maintaining 1.5${\times}$106 viable cells/mL, which was much higher than that form fed-batch cultivation. The nerve cell growth was greatly enhance in both fed-batch and perfusion cultivations while the growth of fibroblast cells was not. It strongly implies that the factors secreted from human fibrobast cells and/or the environments of co-culture system can enhance both cell growth and BDNF secretion. Specific BDNF production rate was not enhanced in co-cultures; however, the production period was increased as the cell growth was lengthened in the co-culture case. Competitive growth between nerve cells and fibroblast cells was not observed in all cases, showing no changes of fibroblast cell growth and only enhancement of the neuroblastoma cell growth and overall BDNF production. It was also found that the perfusion cultivation was the most appropriate process for cultivating two cell lines simultaneously in a bioreactor.
