Dietary maifanite supplementation did not affect the apparent total tract digestibility of calcium and phosphorus in growing pigs

  • Objective: This study was conducted to determine the effects of dietary maifanite supplementation and fecal collection method on the apparent total tract digestibility (ATTD) of calcium (Ca) and phosphorus (P) and blood parameters in growing pigs. Methods: Thirty-six growing barrows (Duroc${\times}$Landrace${\times}$Yorkshire; $27.0{\pm}2.6kg$) were allotted to six dietary treatments with 6 pigs per treatment according to body weight in a completely randomized design. The experimental treatments were: i) Low Ca+cornstarch (2.25%), ii) Low Ca+maifanite (2.25%), iii) Medium Ca+cornstarch (1.42%), iv) Medium Ca+maifanite (1.42%), v) High Ca+cornstarch (0.64%), and vi) High Ca+maifanite (0.64%). Feces were collected by the total collection (TC) and indicator method (IM). At the beginning and the end of the experiment, blood samples were collected from each pig. Results: For the TC method, there were no difference in Ca intake, fecal Ca output, Ca retention and the ATTD of Ca between cornstarch and maifanite diets at the same dietary Ca level. However, urinary Ca excretion was lower (p = 0.01) in pigs fed low Ca diets without maifanite supplementation compared with other dietary treatments. Dietary maifanite supplementation had no effect on the P metabolism in growing pigs. For the IM method, there was no difference in Ca digestibility between cornstarch and maifanite diets at the same dietary Ca level. The ATTD of P was greater (p<0.01) in pigs fed the high Ca diet with maifanite supplementation compared with the high Ca diet with cornstarch treatment. Dietary inclusion of maifanite had no effect on blood parameters in growing pigs. Conclusion: Dietary maifanite supplementation had no effect on the ATTD of Ca and P and serum parameters in growing pigs. The IM resulted in lower digestibility values than the TC method.

A Rapid Technique for Determination of Total Disappearance of Dietary Nitrogen in the Digestive Tract Using Washed Fecal Sample after Freezing and Thawing

  • Three Holstein steers, fitted with ruminal and duodenal cannulas, were used in a replicated $3{\times}3$ Latin square design to determine the digestibility of dietary nitrogen in total digestive tract by three methods, 1) mobile nylon bag (MNB); 2) total fecal collection (TFC); and 3) washed fecal sample after freezing and thawing through a sieve with a pore size of $45{\mu}m$ (WFS). A basal diet of oaten hay-barley was supplemented with one of the following protein sources; soybean meal, fish meal or blood meal. Steers were fed at a level of 2% of body weight. The experimental diets were contained approximately 1.85% nitrogen. There were no differences (p>0.05) among the diets on DM, NDF and nitrogen disappearances, and the diet results were pooled to assess the methods. Total tract disappearances of dry matter and neutral detergent fiber were 61.6, 71.1 and 78.9 and 25.3, 63.2 and 64.6 for MNB, TFC and WFS methods, respectively. The lower digestibility of DM and NDF in the MNB method could be a result of low ruminal incubation time. The TFC method had the lower (p<0.05) determination of nitrogen disappearance in the total digestive tract than the MNB and WFS methods. On the other hand, nitrogen disappearance in the total digestive tract determined by the WFS technique was comparable to that in MNB technique, as there was no significant difference (p>0.05) between the methods. It is shown that the disappearance of dietary nitrogen in the total digestive tract could be estimated in the intact animals by using washed fecal sample prior to freezing and thawing.

조피볼락 치어 및 성어에서 분 수집 방법에 따른 영양소 소화율 (Evaluation of the Nutrient Digestibilities by Different Fecal Collection Methods in Juvenile and Adult Korean Rockfish (Sebastes schlegeli))

  • 분 수집 방법에 따른 건물, 단백질, 지질, 에너지, 탄수화물 및 아미노산의 소화율을 어체 크기별로 측정하기 위해 산화크롬 $(Cr_{2}O_{3})$을 지표물질로 사용하여 표준사료와 단백질 무 첨가 사료를 제조하였다. 조피볼락 성어(평균체중: 300g)와 치어 (평균체중: 30g)를 대상으로 위, 장, 항문을 각각 절개한 방법, 항문 부위를 눌러서 분을 짜내는 방법과 배설된 분을 분 수집 통으로 수집한 방법을 사용하여 각 방법별로 각각 3 반복으로 실험하였다. 건물, 단백질, 지질 및 에너지의 소화율은 분 수집 방법에 따라 유의한 차이 (P<0.0001)를 보인 반면 어체 크기에 대해서는 영향을 받지 않았다. 성어 및 치어 모두 위에서는 건물, 단백질, 지질 및 에너지가 전혀 소화 또는 흡수되지 않았고, 장, 항문 부위로 갈수록 유의하게 (P<0.01) 소화율이 높아지는 경향을 보였다. 성어에서 건물과 단백질의 소화율은 stripping한 것이 항문 부위에서 절개하여 채취한 분보다 유의하게 높은 값을 보인 반면 (P<0.01), 지질과 에너지의 소화율은 유의차가 없었다. 건물 소화율을 제외 한 단백질, 지질과 에너지의 소화율은 분 수집 장치에서 측정한 값이 항문 부위에서 stripping한 것보다 높았지만 유의한 차이는 없었다 (P>0.01). 분 수집 장치를 이용하여 측정된 표준사료의 소화율은 건물이 성어 $61\%$, 치어 $58\%$, 단백질이 성어 $94\%$, 치어 $93\%$, 지질이 성어 $96\%$, 치어 $94\%$, 에너지가 성어 $80\%$, 치어 $79\%$ 및 NFE (nitrogen-free extract)가 성어 $29\%$, 치어 $32\%$로 나타났다. 단백질 무 첨가 사료 (단백질 함량: $1.57\%$)를 먹인 실험어의 분에 단백질 함량이 성어는 $1.35\~1.41\%$, 치어는 $1.16\~1.33\%$ 함유되어 있어 사료의 단백질함량과 거의 비슷하거나 낮게 나타났다. 성어 및 치어 모두 아미노산 종류별로 소화율이 유의한 차이 (P<0.01)를 보였는데, 성어의 경우는 Met이 가장 낮았고, 다음으로 Cys과 Val이 낮은 값을 보였다. 그 외 아미노산의 소화율은 서로 유의한 차이없이 $93\%\~97\%$로 나타났다. 치어의 경우도 Met, Cys, His과 Val의 소화율이 다른 아미노산에 비해 낮은 값을 보였다. 본 실험의 결과로부터, 조피볼락의 외견상 소화율 측정은 stripping하거나 분 수집 통을 이용하는 것이 바람직할 것으로 생각된다.

$^{125}I-triolein$$^{51}Cr_2O_3$의 동시투여(同時投與)에 의(依)한 지방흡수시험법(脂肪吸收試驗法)의 개발(開發)에 관(關)한 임상연구(臨床硏究) (A Clinical Study on the Development of a Simplified Fat Absorption Test by Simultaneous Administration of $^{125}I$-triolein and Chromic Oxide($^{51}Cr_2O_3$))

  • The conventional triolen absorption test has its defect in that the stool collection was cumbersome, time and energy-wasting. In the present study, the triolen absorption test was carried out using double tracer technique with $^{125}I-triolen\;and\;^{51}Cr_2O_3$ to determine if it can overcome the defect of the conventional method also with satisfactory results. Following were the results: 1. The clinical significance of this double tracer method was essentially the same with that previously done by radioactive. triolen alone. With the fractional fecal samples, the equation, y=0.626x+2.010 was substantiated, hence, this method appears to be clinically valuable if the appropriate correction is applied. With the mixed fecal samples, the equation y=0.642x+1.468 was substantiated (p<0.005) which appears to be also clinically valuable. When these two data were compared, the equation y=0.975x+0.090 (P<0.05) was substantiated, hence, $x{\fallingdotseq}y$. 2. The normal ranges of the fecal triolen excretion rate in this double tracer method were $3.46{\pm}1.69%$, namely, less than 6.9%. 3. The samplings were done from the first to third defecation in cases of clinically normal, and from the first to second defecation in cases of diarrhea or malabsorption. 4. The intestinal malabsorption of triolen was not observed in whom the triolen absorption was supposed to be clinically normal, however. a good number of suspicious malabsorptive cases showed the normal values.

SAF 고정변에서 람블편모충(Giardia lamblia)시스트 검출을 위한 검사방법의 평가 (Evaluation of Giarydia lumblia Detection Method in Stool Specimens Fixed with SAF Solution)

  • The present study has been designed as a basic study on laboratory diagnosis of giardiasis and to demonstrate a more effective method for the detection of Giardia lamblia cyst with the inherent advantages of minimizing both the number of stool examinations required and the interval of stool collections for estimating the real state of prevalence in the shortest time possible. There were 3 subject groups of 75 children each currently residing in an orphanage in Gunsan city, Jeonbuk province from which stool specimens were collected every day, every other day, and every 3 days. The procedure is as follows: 1) resuspend the InKed sample after fixation with SAF solution 2) centrifuge the sediment for 1 min. at 2, 000 rpm after straining through gauze into a tube 3) divide the sediment into 3 parts and use them for direct fecal smear, formalin-ether concentration (MGL) and zinc sulfate ($ZnSO_4$) floatation techniques. The results are summarized as follows: 1. Overall infection rate after 10 trials showed a 60% positive indication. The positive rate among children under 4 years old was significantly higher than the rate in children over 4 years old. No significant difference in rate by sex was observed. 2. The results of eBaminations by direct fecal smear and MGL techniques appeared more accurate than that obtained by $ZnSO_4$ ftoatation method as indicated by a higher positive rate. Of all three methods concerned, combinations of two demonstrated a higher positive rate than that shown by any one alone. 3. In three consecutive examinations under varying conditions such as different days, the cyst detection rate by MGL technique indicated 83%. In 5 examinations under the same varying conditions, the indicated rate was 94%. 4. The interval of stool collection proved to be insignificant for the cyst detection rate. In conclusion, both MGL method and modified fecal direct smear can provide a good cyst detection rate of G. lamblia provided that more than 3 consecutive examinations of stool under varying conditions are carried out.

Casein Supplementation Does Not Affect the Estimates of True Total Tract Digestibility of Phosphorus in Soybean Meal for Growing Pigs Determined by the Regression Method

  • Forty-eight barrows with an average initial body weight of $25.5{\pm}0.3kg$ were assigned to 6 dietary treatments arranged in a $3{\times}2$ factorial of 3 graded levels of P at 1.42, 2.07, or 2.72 g/kg, and 2 levels of casein at 0 or 50 g/kg to compare the estimates of true total tract digestibility (TTTD) of P in soybean meal (SBM) for pigs fed diets with or without casein supplementation. The SBM is the only source of P in diets without casein, and in the diet with added casein, 1.0 to 2.4 g/kg of total dietary P was supplied by SBM as dietary level of SBM increased. The experiment consisted of a 5-d adjustment period and a 5-d total collection period with ferric oxide as a maker to indicate the initiation and termination of fecal collection. There were interactive effects of casein supplementation and total dietary P level on the apparent total tract digestibility (ATTD) and retention of P (p<0.05). Dietary P intake, fecal P output, digested P and retained P were increased linearly with graded increasing levels of SBM in diets regardless of casein addition (p<0.01). Compared with diets without casein, there was a reduction in fecal P in the casein-supplemented diets, which led to increases in digested P, retained P, ATTD, and retention of P (p<0.01). Digested N, ATTD of N, retained N, and N retention were affected by the interaction of casein supplementation and dietary P level (p<0.05). Fecal N output, urinary N output, digested N, and retained N increased linearly with graded increasing levels of SBM for each type of diet (p<0.01). The estimates of TTTD of P in SBM, derived from the regression of daily digested P against daily P intake, for pigs fed diets without casein and with casein were calculated to be 37.3% and 38.6%, respectively. Regressing daily digested N against daily N intake, the TTTD of N in SBM were determined at 94.3% and 94.4% for diets without casein and with added casein, respectively. There was no difference in determined values of TTTD of P or N in SBM for pigs fed diets with or without casein (p>0.05). In summary, our results demonstrate that the estimates of TTTD of P in SBM for pigs were not affected by constant casein inclusion in the basal diets.

Effects of β-glucan with vitamin E supplementation on the growth performance, blood profiles, immune response, fecal microbiota, fecal score, and nutrient digestibility in weaning pigs

  • Objective: This study was conducted to evaluate effects of β-glucan with vitamin E supplementation on the growth performance, blood profiles, immune response, fecal microbiota, fecal score, and nutrient digestibility in weaning pigs. Methods: A total of 200 weaning pigs with an average body weight (BW) of 7.64±0.741 kg were allotted to five treatment groups and were divided based on sex and initial BW in four replicates with ten pigs per pen in a randomized complete block design. The experimental diets included a corn-soybean meal-based basal diet with or without 0.1% or 0.2% β-glucan and 0.02% vitamin E. The pigs were fed the diets for 6 weeks. A total of 15 barrows were used to evaluate the nutrient digestibility by the total collection method. The BW and feed intake were measured at the end of each phase. Blood samples were collected at the end of each phase, and fecal samples were collected at the end of the experiment. Results: The addition of β-glucan with vitamin E to weaning pig feed increased BW, average daily gain, and average daily feed intake. A significant decrease in yeast and mold and Proteobacteria and a tendency for Lactobacillus to increase compared to the control was shown when 0.1% β-glucan and 0.02% vitamin E were added. The fecal score in weaning pigs was lower in the treatments supplemented with 0.1% or 0.2% β-glucan and 0.02% vitamin E compared to the control. In addition, vitamin E was better supplied to weaning pigs by increasing the concentration of α-tocopherol in the blood of weaning pigs when 0.02% vitamin E was supplemented. However, there was no significant difference in either the immune response or nutrient digestibility. Conclusion: Inclusion of 0.1% β-Glucan with 0.02% vitamin E in a weaning pig's diet were beneficial to the growth performance of weaning pigs by improving intestinal microbiota and reducing the incidence of diarrhea.

Effects of Dietary Cellulose on the Basal Endogenous Loss of Phosphorus in Growing Pigs

  • An experiment was conducted to determine the effect of cellulose concentration in diets containing no phosphorus (P) on the basal endogenous loss (BEL) of P in growing pigs. Twelve barrows (an initial mean body weight = $49.6{\pm}3.2kg$) were individually housed in metabolism crates. Pigs were allotted to 4 experimental diets according to a cross-over design with 12 animals and 2 periods. Four P-free diets were mainly based on corn starch, sucrose, and gelatin, and were formulated to contain 0%, 4%, 8%, or 12% cellulose. Each period consisted of a 5-d adaptation and a 5-d collection period. The marker-to-marker method was used for fecal collection. The feed intake (p<0.05, linear and quadratic) and dry feces output (p<0.01, linear and quadratic) were increased with increasing dietary cellulose concentration. However, P concentration in the feces was decreased (p<0.01, linear and quadratic) with increasing dietary cellulose concentration. There was no significant difference in total P output and the BEL of P as mg per kg DMI (ranging from 157 to 214 mg/kg of dry matter intake) among experimental diets. However, values for the apparent total tract digestibility of energy, dry matter, organic matter, crude protein, and calcium were linearly decreased (p<0.01) with increasing cellulose concentration in the diet. In conclusion, dietary cellulose affected the amount of feces and digestibility of energy and nutrients, but did not affect the endogenous loss of P.

Evaluation of Amino Acid and Energy Utilization in Feedstuff for Swine and Poultry Diets

  • An accurate feed formulation is essential for optimizing feed efficiency and minimizing feed cost for swine and poultry production. Because energy and amino acid (AA) account for the major cost of swine and poultry diets, a precise determination of the availability of energy and AA in feedstuffs is essential for accurate diet formulations. Therefore, the methodology for determining the availability of energy and AA should be carefully selected. The total collection and index methods are 2 major procedures for estimating the availability of energy and AA in feedstuffs for swine and poultry diets. The total collection method is based on the laborious production of quantitative records of feed intake and output, whereas the index method can avoid the laborious work, but greatly relies on accurate chemical analysis of index compound. The direct method, in which the test feedstuff in a diet is the sole source of the component of interest, is widely used to determine the digestibility of nutritional components in feedstuffs. In some cases, however, it may be necessary to formulate a basal diet and a test diet in which a portion of the basal diet is replaced by the feed ingredient to be tested because of poor palatability and low level of the interested component in the test ingredients. For the digestibility of AA, due to the confounding effect on AA composition of protein in feces by microorganisms in the hind gut, ileal digestibility rather than fecal digestibility has been preferred as the reliable method for estimating AA digestibility. Depending on the contribution of ileal endogenous AA losses in the ileal digestibility calculation, ileal digestibility estimates can be expressed as apparent, standardized, and true ileal digestibility, and are usually determined using the ileal cannulation method for pigs and the slaughter method for poultry. Among these digestibility estimates, the standardized ileal AA digestibility that corrects apparent ileal digestibility for basal endogenous AA losses, provides appropriate information for the formulation of swine and poultry diets. The total quantity of energy in feedstuffs can be partitioned into different components including gross energy (GE), digestible energy (DE), metabolizable energy (ME), and net energy based on the consideration of sequential energy losses during digestion and metabolism from GE in feeds. For swine, the total collection method is suggested for determining DE and ME in feedstuffs whereas for poultry the classical ME assay and the precision-fed method are applicable. Further investigation for the utilization of ME may be conducted by measuring either heat production or energy retention using indirect calorimetry or comparative slaughter method, respectively. This review provides information on the methodology used to determine accurate estimates of AA and energy availability for formulating swine and poultry diets.

Influences of Enzyme Complex Supplementation on Growth, Ileal and Apparent Fecal Digestibility and Morphology of Small Intestine in Pigs

  • A total of 140 weaning pigs were used to determine the effects of digestive enzyme supplementation to corn-soybean meal diets on growth performance, physiological changes of small intestine, microorganisms and pH in the gastrointestinal tract. Two kinds of enzyme complex (A, B) were used in this experiment. Pigs were allotted in a completely random design (CRD) to five replicates with four pigs per pen. Diets and water were provided for ad libitum consumption. Treatments included 1) Control: without enzyme supplementation, 2) Enzyme A 0.05%, 3) Enzyme A 0.10%, 4) Enzyme A 0.15%, 5) Enzyme B 0.05%, 6) Enzyme B 0.10%, 7) Enzyme B 0.15% in the diets. A total of 24 crossbred barrows 25.78${\pm}$0.55 kg BW fitted with simple ileal T-cannulas were used to evaluate the effect the enzyme addition on the nutrient digstibility. Pigs were allotted 4 treatments (No enzyme, enzyme A 0.05%, enzyme A 0.1%, enzyme A 0.15%), 6 replicates according to a completely random design (CRD). Another digestibility trial was followed for enzyme complex B. Twenty pigs, average 31.92${\pm}$0.37 kg BW, fitted with simple ileal T-cannulas for digestibility trial. Neither enzyme A nor enzyme B affected on fecal or ileal digestibility of dry matter, gross energy, crude protein, crude fat and crude ash (p>0.05). The apparent fecal digestibilities of all the nutrients were higher in total feces collection method than in indirect method. At the end of feeding trial, 21 pigs were slaughtered for examining the morphological changes of small intestine and the concentration of microorganisms in the ileum and the colon. Growth performance, intestinal morphology and pH of ileum and colon were not affected by the either enzyme complex supplementation (p>0.05). These results suggested that enzyme complex A and enzyme complex B were of no benefit to early-weaned pigs when corn-soybean meal based diet was provided.
