Management Plan for the Production of Safe and High Quality Drinking Water From the Paldang Lake (고 품질 수돗물 생산을 위한 팔당 수질관리 방안)

  • It is necessary to implement the management plan for the production of safe and high quality drinking water from lake Paldang. To set up the plan, the water quality items such as BOD, T-P, SS and coliform were monitored for ten years, 1997~2006, and the influence of raw water quality on the drinking water treatment process and the treated water quality was also evaluated from 2004 to 2006. In conclusion, water quality items such as turbidity(SS), T-P(eutrophication), pathogens(fecal coliforms, enterovirus, reovirus, giardia, cryptosporidium), DOC(precursor of disinfection by-products), and micro-pollutants(phthalates, VOCs, heavy metals) are should be managed to get safe and high quality drinking water from lake Paldang.

Study on the Sanitary Indicative Bacteria of Sea Water in Fish Preservatory Tanks during the Mid Summer (여름철 활어조해수(活魚槽海水)의 세균학적(細菌學的)인 연구(硏究))

  • The purpose of the present study is to obtain some fundamental data for hygienic control of sanitary indicative bacteria of sea water in fish preservatory tanks at popular swimming beaches in Busan including Songdo, Kwanganri, Haeundae and Songjeong from July to August in 1982. The geometric mean values of coliform group and fecal coliform MPN (most probable number) of sea water of Kwanganri were 27,300/100ml and 8,700/100ml respectively, which showed the highest population of the study regions. MPN's of the sea water of beaches and fish preservatory tanks showed 21,000/100ml and 8,800/100ml respectively at Kwanganri, which were also the highest numbers. The degree of the hygienic bacteriological populations of both the seawater and that of the fish tanks at a same region showed similar trend. The pollution caused by coliform group generally increased as viable cell counts increased. The types of coliform organisms isolated from seawater in fish presorvatory tanks by IMViC reaction were 42% Escherichia coli group, 2% Citrobacter freundii group, 44% Klebsiella aerogenes group and 12% others.

Evaluation of Influent Water Quality Using Indicator Microorganisms in Lake Shiwha (지표미생물을 이용한 시화호 유입수의 수질평가)

  • Lake Shiwha, an artificial lake located near metropolitan Seoul, offers a unique water environment and has been suspected to have high levels of chemical and microbiological contaminations. Lake Shiwha was originally connected to the sea but currently has four major surface water inputs from agricultural, municipal, industrial areas and in addition an occasional inflow from the sea. The objectives of this study are to investigate the relative contribution of microbial contaminants from each of the inflowing surface waters and to identify appropriate microbial indicator organisms in this unique water environment. We measured the levels of microbial contaminations in the four inflowing surface waters. A number of microbial indicator organisms including total coliform (TC), fecal coliform (FC), E. coli, Enterococci, somatic and male-specific coliphages were analyzed. Bacterial indicator microorganisms were detected and quantified by the $Colilert^{(R)},\;Enterolert^{(R)}$ kit. Surface water (50 l) was sampled by $ViroCap^{TM}\;5"$ cartridge filters and analyzed by the single agar layer method for detecting coliphages. The concentrations of TC, FC, E. coli, and Enterococci were 1543 CFU/100 ml${\sim}1.99{\times}10^6$ CFU/100 ml, 0 CFU/100 ml${\sim}202$ CFU/100ml, 0 CFU/100 ml${\sim}1.80{\sim}10^5$ CFU/100ml, 74 CFU/100 ml${\sim}3408$ CFU/100 ml, respectively. The male-specific and somatic coliphages were detected in three different inflowing surface waters. Isolated E. coli and Enterococci strains were further analyzed by 16s rDNA amplification and subsequent phylogenetic analysis from Jungwang-chun, Ansan-chun, Banwol-chun and penstock of inflowing surface water. Our results indicated that the concentrations of different fecal indicator microorganisms might not be highly correlated with each other. Multiple microbial indicator organisms should be used for monitoring microbial contamination and microbial source tracking methods.

Effects of Dietary Probiotic Mixture on Caecal Microflora, Immune Response, Egg Quality and Production of Layerunder Heat Stress (혼합 생균제가 열 스트레스에 노출된 산란계의 생산성, 계란품질, 면역반응 및 맹장 미생물에 미치는 효과)

  • This study was conducted to investigate the effects of probiotic mixture on fecal ammonia, caecal microorganism, immune response, egg quality and production in layer under heat stress (HS).A total of four hundred 50 week olds Hy-Line Brown layers were randomly divided into four groups of 100 heads each: C (control, room temperature $25^{\circ}C$), HS (heat stress $33^{\circ}C$), PM (HS plus probiotic mixture 500, 750 mg/kg of diets). Egg production, egg quality, spleen weight, blood IgG and lymphocyte concentrations were increased in the PM group compared to the HS group, while mortality, the heterophil:lymphocyte (H:L) ratio, and corticosterone levelswere significantly decreased. Lactobacillus was increased in the PM group compared to the HS group, but E. coli, coliform bacteria and aerobic bacteria were significantly reduced. Fecal ammonia production was significantly increased in the HS group compared to the PM group. In conclusion, the results of this study that these mixed probiotics can reduce the heat damage of the summer laying hens and can be an effective nutritional strategy to reduce odor generation from feces, and to improve egg quality and laying production through immune response and caecal microbial balance.

The Effects of Inland Pollution Sources around the Port of Jeokyang and Jangpo after Rainfall Events on Bacteriological Water Quality in the Changseon Area, Korea (강우에 따른 적양항과 장포항 육상오염원이 창선해역에 미치는 세균학적 영향평가)

  • The influences of inland pollution sources nearJeokyang and Jangpoports following rainfall events on the bacteriological water quality and safety of commercial shellfish were investigated in the Changseon area, Korea. Stream flow rates exhibited 1.9- to 5.7-fold increases after rainfall events and then fell to 68~81% of that level after 24 h. The calculated impact area of inland pollution sources was 0.47 $km^2$ in the Jeokyangport area and 0.27 $km^2$ in the Jangpoport area at 24 h following 11 mm of rainfall. When the flow rate of inland pollution increased, the level of male-specific bacteriophage (MSB) decreased, and no MSB could be detected in seawater samples, whereas 30 PFU/100 g was detected in shellfish samples. Fecal coliform levels in seawater and shellfish samples did not exceed their respective tolerance levels (4.5 MPN/100 mL) and 130 MPN/100 g, respectively and thus complied with the standard for approved shellfish growing area. The sanitary conditions of areas adjacent to Jeokyang and Jangpoports under conditions of rainfall below 29 mm met the criteria for approved growing area under the United States National Shellfish Sanitation Program and the EC Regulations.

Temporal and Spatial Evaluation of Water Pollution Characteristics in Gohyeon Stream and Its Tributaries (고현천 및 유입지류의 수질오염 특성의 시·공간적 평가)

  • BACKGROUND: Gohyeon Stream is the municipal eco-stream of 7.1km in total length which flows through the downtown area of Gohyeon in Geoje city, rising from the watershed of Mundong Water Fall. Gohyeon district in Geoje city has been a rapid growing area centering in Geoje city and then experienced an rapid increase in population. Large amounts of sewage pollutants have been spewed into Gohyeon Stream from its tributaries, due to the lack of sewer system. Gohyeon Stream is laced with unhealthy levels of fecal coliform (FC). Restoration of water quality in Gohyeon Stream is no less inevitable in behalf of its ecosystem and the citizen. In this study, the water quality of Gohyeon Stream and its tributaries was examined temporally and spatially, and their relationships were comparatively analyzed to give useful basic data applying to a restoration project of the water quality of Gohyeon Stream. METHODS AND RESULTS: The samples ware taken at 20 points in Gohyeon Stream and 19 points in its tributaries during the rainy and dry seasons, respectively, and examined on the parameters of pH, temperature, salinity, dissolved oxygen (DO), suspended solid (SS), biochemical oxygen demand (BOD), chemical oxygen demand (COD), dissolved inorganic nitrogen (DIN; $NH_3$-N, $NO_3$-N, $NO_2$-N), disolved inorganic phosphorus (DIP; $PO_4$-P) and FC. The data were analyzed using a comparative analysis and Pearson's correlation analysis among the parameters. During the rainy season, the concentration of SS was high in the upper region of Gohyeon Stream, and the concentrations of COD, DIN and DIP were low in the upper region and high in the middle and lower regions. During the dry season, the concentration of SS was low and the concentrations of COD, DIN, DIP and FC were high in all regions. The Pearson's correlation analyses showed that the relationships between DO and FC, COD and DIP, and DIN and FC during the rainy season as well as between DO and DIN, SS and FC, COD and DIP, and DIN and DIP during the dry season were significant. CONCLUSION: During the rainy season, the upper region of Gohyeon Stream flowed the high level of SS, the middle region the high level of nutrients due to an agricultural run-off, and the lower region the high level of nutrients due to a sewage inflow. During the dry season, the water quality of Gohyeon Stream was directly and sensitively influenced on the inflow of sewage from the tributaries.

Cleaning Effects of Environmental-Friendly Washing Detergent on Milking Installation in Dairy Farms (젖소농가의 착유시설에 따른 친환경세제의 세척에 미치는 영향)

  • This study was conducted to determine cleaning effects of environmental-friendly washing detergent on milking installation in dairy farms. Milking machine, cooling machine and milking liner were washed by alkaline detergent and acid detergent for first three weeks and alkaline detergent and environmental~friendly detergent for next three weeks and the results get through microorganism and physico-chemistry analysis at the five district dairy farms different from environment. E-coli, coliform, fecal coliform, staphylococcus aureus, fecal streptococcus, prsudomonas aeruginosa and yersinia were not found in all of the dairy farms, and total colony counts were no difference compared with chemical detergent. The water for washing exceeded the determining acceptable level of nitrate nitrogen in the Anseong, Onyang and Cheonan and the remaining substance of washed water was also high levels. The cause of the result was that the level of nitrate nitrogen of water is basically high. Therefore, this study indicated that washing effect is no differences between environmental-friendly detergent and chemical detergent by washing for milking machine and cooling machine. Above all, producing high quality milk is to manage the water quality as well as using detergent.

Bacteriological Contamination of Water in Han River basin (한강유역의 수중미생물 오염도 조사)

  • In order to investigate the bacteriological contamination of water in Han river, the survey was carried out in eight reservoirs of Seoul water supply during the period from January to December in 1985. 1. The counts by means of total bacteria in eight reservoirs by standard plate count method were as follows: $7.7\times10^2$ per ml in Paldang reservior, $9.6\times10^3$ per ml in Gueiri, $8.4\times10^4$ per ml in Doogdo, $1.6\times10^6$ per ml in Bogwang, $2.5\times10^6$ per ml in Noryangjin, $2.2\times10^6$per ml in Seon yoo, $5.9\times10^6$ per ml in Yungdeungpo and $1.9\times10^7$per ml in Gayang. 2. The average counts of total coliform in eight reservoirs by MPN method were as follows : $2.4\times10$ per 100 ml in Paldang, $5.6\times10^2$ per 100 ml in Gueiri, $2.3\times10^3$ per 100 ml in Doogdo, $5.1\times10^4$ per 100 ml in Noryang-jin, $1.2\times10^5$ per 100 ml in Bogwang, $6.2\times10^4$ per 100 ml in Seonyoo, $1.1\times10^5$ per 100 ml in Yungdeungpo and $2.8\times10^5$ per 100 ml Gayang. 3. The counts by means of fecal coliform in eight reservoirs by MPN method were as follows : non detection per 100 ml in Paldang, 5.2 per 100 ml in Gueiri, $1.2\times10^2$ per 100 ml in Doogdo, $1.6\times10^3$ per 100ml in Bogwang, $2.0\times10^3$per 100ml in Noryangjin, $6.6\times10^2$ per 100ml in Seonyoo, $1.2\times10^3$ per 100 ml in Yungdeungpo and $2.5\times10^3$per 100 ml in Gayang. 4. The counts by means of fecal streptococci in eight reservoirs by MPN method were as follows: non detection per 100 ml in Paldang and Gueiri, $6.9\times10$ per 100 ml in Doogdo, $3.2\times10^2$ 102 per 100 ml in Bogwang, $2.9\times10^2$ per 100 ml in Noryangjin, $3.0\times10^2$ per 100 ml in Seonyoo, $4.0\times10^2$ per 100 ml in Yungdeungpo and $14\times10^3$ per 100 ml in Gayang. 5. The counts means of pseudomonas aeruginosa in eight reservoirs by MPN method were as follows; non detection per 100 ml in Paldang, 2.4 per 100 ml in Gueiri, $1.5\times10$ per 100 ml in Doogdo, $2.0\times10$ per 100ml in Bogwang,$6.2\times10$ per 100mI in Noryangjin, $2.1\times10$ per 100ml in Seonyoo, $6.4\times10$ per 100mI in Yungdeungpo and $7.1\times10$ per 100ml in Gaynag.

Effect of a probiotic mixture on egg quality and egg production in laying hens (복합생균제가 산란계의 생산성, 계란품질 및 악취저감에 미치는 영향)

  • The objective of the present study was to evaluate effects of probiotic mixture (PM) including Bacillus subtillus, Streptomyces galilaeus and Sphingobacteriaceae on egg production, egg quality, and eliminating odor in laying hens. A total of 240 Hy-Line Brown laying hens (50 wks of age) were divided into four treatment groups (60 replicates of birds, each) in completely randomized design. Groups were assigned to four experimental diets: CON (a basal diet of no PM additive as control), basal diet supplemented with either CP3 (0.3% commercial probiotics), PM3 (0.3% PM), or PM5 (0.5% PM 0.5). Data of egg production and egg quality were obtained during 6 weeks of experimental period. Egg production, Haugh unit, shell thickness, breaking strength, yolk color, and albumin high were significantly increased in CP3 and PM3 compared to those in CON and CP3. Ammonia levels and numbers of E. coli, total aerobic bacteria, Coliform, and Salmonella in feces were significantly increased in order of CON>CP3>PM3>PM5. The present study indicates that addition of 0.3% probiotic mixture including B. subtillus, S. galilaeus and Sphingobacteriaceae to diets may improve egg production, egg quality, and eliminate fecal ammonia of laying hens.

A Study on Hand Hygiene Practices: A Comparison of Food Handlers with General Population (일반인과 식품취급자의 손 위생관리에 관한 비교)

  • Objectives: This study was performed to investigate hygienic behavior of food handlers and general population focusing on awareness of hand-washing and the microbial load of their hands. Methods: A questionnaire survey and microbiological analysis were carried out for sixty-four people each. Samples for microbiological analysis were collected through the glove-juice method from the hands, and were analyzed for the presence of aerobic plate counts, total coliform, fecal coliform, Escherichia coli, Staphylococcus aureus, and Salmonella spp. according to the Food Code of Korea. Results: In the survey, significant differences between the food handlers and general population (p < 0.01) were found in hand-washing frequency, duration, use of hand-washing agents, washing parts of hands, hand-drying method, and method of turning off water. In eight different situations among the ten particular situations in their daily life, more food handlers responded to wash their hands than general population (p < 0.05). Bacterial load on hands with general population was consistently higher than with food handlers (p < 0.05), however, percentages of positive hands of S. aureus and Salmonella spp. were not. Conclusions: Poor hand hygiene practices were indicated by the positive results for E. coli, S. aureus, and Salmonella spp. on the hands of some respondents in both groups. This study reveals that there is the need for programs or campaigns to increase hand-washing practices of both groups.
