Fe65, a neuron-specific adaptor protein, has two phosphotyrosine binding (PTB) domains. The second PTB (PTB2) domain interacts with intracellular domain fragment (AICD) of amyloid beta precursor protein (APP). Recent studies suggested that tile complex is composed of AICD and Fe65 transactivates genes that are responsible for neuronal cell death in Alzheimer's disease (AD). Therefore, a compound inhibiting the interaction between Fe65 and AICD can be a drug candidate to treat AD. However, it remains unclear how Fe65 recognizes AICD at a molecular level. Here, we report high-level production of the PTB2 domain of Fe65 in the baculovirus system. We found that the baculovirus system is an efficient method to obtain the Fe65 PTB2 domain, compared with the bacterial and mammalian expression systems. The purified recombinant protein was used for crystallization to determine its crystal structure helping to understand the molecular mechanism of Fe65-dependent signaling and to design its inhibitors.
Fe65 is characterized as an adaptor precursor (APP) through its PID2 element, as well as with the other members of the APP protein family. With the serum- and glucocorticoid-induced kinase 1 (SGK1) substrate specificity information, we found that the putative site of phosphorylation in Fe65 by SGK1 is present on its $Ser^{566}$ residue in $^{560}CRVRFLSFLA^{569}$(X60469). Thus, we demonstrated that Fe65 and the fluorescein-labeled Fe65 peptide $FITC-^{560}CRVRFLSFLA^{569}$ are phosphorylated in vitro by SGK1. Phosphorylation of the $Ser^{566}$ residue was also demonstrated using a $Ser^{566}$ phospho-specific antibody. The phospho Fe65 was found mainly in the nucleus, while Fe65 S556A mutant was localized primarily to the cytoplasm. Therefore, these data suggest that SGK1 phosphorylates the $Ser^{566}$ residue of Fe65 and that this phosphorylation promotes the migration of Fe65 to the nucleus of the cell.

Glycogen synthase kinase $3{\beta}$ (GSK $3{\beta}$) is a serine/threonine kinase that phosphorylates substrates such as ${\beta}$-catenin and is involved in a variety of biological processes, including embryonic development, metabolism, tumorigenesis, and cell death. Here, we present evidence that human GSK $3{\beta}$ is associated with Fe65, which has the characteristics of an adaptor protein, possessing a WW domain, and two phosphotyrosine interaction domains, PID1 and PID2. The GSK $3{\beta}$ catalytic domain also contains a putative WW domain binding motif ($^{371}PPLA^{374}$), and we observed, using a pull down approach and co-immunoprecipitation, that it interacts physically with Fe65 via this motif. In addition, we detected co-localization of GSK $3{\beta}$ and Fe65 by confocal microscopy, and this co-localization was disrupted by mutation of the putative WW domain binding motif of GSK $3{\beta}$. Finally, in transient transfection assays interaction of GSK $3{\beta}$ (wt) with Fe65 induced substantial cell apoptosis, whereas interaction with the GSK $3{\beta}$ AALA mutant ($^{371}AALA^{374}$) did not, and we noted that phosphorylation of the Tyr 216 residue of the GSK $3{\beta}$ AALA mutant was significantly reduced compared to that of GSK $3{\beta}$ wild type. Thus, our observations indicate that GSK $3{\beta}$ binds to Fe65 through its $^{371}PPLA^{374}$ motif and that this interaction regulates apoptosis and phosphorylation of Tyr 216 of GSK $3{\beta}$.
To investigate the effect of the catalyst synthesis method on the oxidative dehydrogenation (ODH) of nbutenes, $BiFe_{0.65}MoP_{0.1}$ oxide catalysts were prepared with various synthesis methods such as co-precipitation, citric acid method, hydrothermal method, and surfactant templated method. The catalysts were characterized by X-ray Diffraction (XRD), $N_2$ sorption, and $NH_3/1$-butene-temperature programmed desorption ($NH_3/1$-butene-TPD) to correlate with catalytic activity in ODH reaction. Among the catalysts studied here, $BiFe_{0.65}MoP_{0.1}$ oxide catalyst prepared with co-precipitation method marked the highest activity showing 1-butene conversion, 79.5%, butadiene selectivity, 85.1% and yield, 67.7% after reaction for 14 h. From the result of $NH_3$-TPD, the catalytic activity is closely related to the acidity of the $BiFe_{0.65}MoP_{0.1}$-x oxide catalyst and acidity of the $BiFe_{0.65}MoP_{0.1}$ oxde catalyst prepared with co-precipitation method was higher than that of other catalysts. In addition, combined with the 1-butene TPD, the higher catalytic activity is closely related to the amount of weakly adsorbed intermediate (< $200^{\circ}C$) and the desorbing temperature of strongly adsorbed intermediates (> $200^{\circ}C$).
This paper presents a micro-mechanical model of ductile fracture for the API X65 steel using the Gurson-Tvergaard-Needleman (GTN) model. Experimental tests and FE damage simulations using the GTN model are performed for smooth and notched tensile bars, from which the parameters in the GTN model are calibrated. As application, the developed GTN model is applied to simulate small-sized, single-edge-cracked tensile and bend bars, via three-dimensional FE damage analyses. Comparison of FE damage analysis results with experimental test data shows overall good agreements.
Ferromagnetic ${\tau}-phase$$Mn_{54}Al_{46}C_{2.44}$ particles were synthesized, and its composites with commercial $Sm_2Fe_{17}N_3$ and synthesized $Fe_{65}Co_{35}$ powders were fabricated. Smaller grain size than the single domain size of the $Mn_{54}Al_{46}C_{2.44}$ without obvious grain boundaries and secondary phases is the origin for the low intrinsic coercivity. It was confirmed that the magnetic properties of the $Mn_{54}Al_{46}C_{2.44}$ can be enhanced by magnetic exchange coupling with the hard magnetic $Sm_2Fe_{17}N_3$ and soft magnetic $Fe_{65}Co_{35}$. The high degrees of the exchange coupling were verified by calculating first derivative curves. Thermo-magnetic stabilities of the composites from 100 to 400 K were measured and compared. It was demonstrated that the $Mn_{54}Al_{46}C_{2.44}$ based composites containing $Sm_2Fe_{17}N_3$ and $Fe_{65}Co_{35}$ could be promising candidates for future permanent magnetic materials with the proper control of purity, magnetic properties, etc.
$Ni_{0.65}Zn_{0.35}Cu_{0.1}Fe{1.9}O_4$ has been studied with Mossbauer spectroscopy and X-ray diffraction. The crystal structure is found to be a cubic spinel with the lattice constant $a_0=8.390{\AA}$. Mossbauer spectra of $Ni_{0.65}Zn_{0.35}Cu_{0.1}Fe{1.9}O_4$ has been taken at various temperatures ranging from 12 K to 705 K. The isomer shift indicates that iron ions are ferric at tetrahedral [A] and octahedral sites [B], respectively. The Neel temperature is determined to be $T_N=705\;K$. As the temperature increases toward $T_N$ a systematic line broadening effect in the Mossbauer spectrum is observed and interpreted to originate from different temperature dependencies of the magnetic hyperfine fields at various iron sites. The quadrupole splitting just on $T_N$ is 0.41 mm/s whereas the quadrupole shift below $T_N$ vanishes. This implies that the orientation of the magnetic hyperfine field with respect to be principal axes of the electric field gradient is random.
$PbTiO_3$/P(VDF/TrFE) thin film for pyroelectric infrared sensor's sensing materials have been fabricated by spin coating technique. 65 wt% VDF and 35 wt% TrFE were for P(VDF/TrFE) powder. $PbTiO_3$ powder was used for a ceramic - polymer composites materials. Surface of composite thin film by ceramic fraction factor was observed by SEM. The $PbTiO_3$/P(VDF/TrFE) thin film capacitancy, dielectric constant and dielectric loss measured by impedence analyzer(HP4192A) and pyroelectric coefficient was measured by semiconductor parameter analyzer(HP4145B).
The influence of phosphorous precursors, $NH_4H_2PO_4$, $(NH_4)_2HPO_4$, $H_3PO_4$, $(C_2H_5)_3PO_4$, and $P_2O_5$, on the catalytic performance of the $BiFe_{0.65}MoP_{0.1}$ catalysts in the oxidative dehydrogenation of 1-butene to 1,3-butadiene was studied. The catalysts were characterized by XRD, $N_2$-sorption, ICP, SEM and TPRO analyses. It was not observed big difference on the physical properties of catalysts in accordance with used different phosphorous precursors, however, the catalytic performance was largely depended on the nature of the phosphorous precursors. Of various precursors, the $BiFe_{0.65}MoP_{0.1}$ oxide catalyst, which was prepared from a phosphoric acid precursor, showed the best catalytic performance. Conversion and yield to butadiene of the catalyst showed 79.5% and 67.7%, respectively, after 14 h on stream. The cation of phosphorous precursors was speculated to affect the lattice structure of the catalysts during catalyst preparation and this difference was influenced on the re-oxidation ability of the catalysts. Based on the results of TPRO, it was proposed that the catalytic performance could be correlated with re-oxidation ability of the catalysts.
