The fruit and seeds of Butea monosperma (Lam) Kuntze (Fabaceae) are useful in piles, anthelmintic, eye diseases, and inflammation in the Indian system of medicine. Hence, we have evaluated the anti-inflammatory activity of the fixed oil, mixed fatty acids, and unsaponifiable matter of B. monosperma against carrageenan-induced paw oedema and cotton pellet-induced granuloma in rats. The fixed oil, mixed fatty acids, and unsaponifiable matter of the oil exhibited significant anti-inflammatory activity on the tested experimental animal models. The unsaponifiable matter of the oil produced higher protection compared to fixed oil and mixed fatty acids. Phytochemical analysis of the fixed oil revealed the presence of steroids and terpenoids while unsaponifiable matter of the oil showed the presence of ${\beta}-sitosterol$. Also, four fatty acids were identified in the fixed oil by gas liquid chromatography. The anti-inflammatory activity of the fixed oil may be due to unsaponifiable matter or combination of unsaponifiable matter and mixed fatty acids.
Studies were made to evaluate the specific and combined effects of different fatty acids on the in vitro development of 8-cell rat embryo in culture media with and without carbohydrate substrate. Palmitic, oleic, linoleic and arachidonic acids were added singly and in combination to media which contained fatty acid-free BSA. Cell numbers in blastocysts cultured in the media were counted and compared with cell numbers in blastocysts at the corresponding stage collected from the uterus. Oleic, linoleic and arachidonic acids promoted the rat embryo development from 8-cell to the blastocysts. especially in the absence of carbohydrate substrates. Among these three, oleic acid was the most effective but embryo development was not accelerated by the addition of palmitic acid in either the presence or the absence of carbohydrate substrates. Addition of the mixture of four fatty acids was more effective for rat embryo development than single treatment with any of fatty acids tested. Cell numbers per blastocyst in the presence and absence of carbohydrate substrate were similar, and did not differ from those for blastocysts obtained from the uterus.
This study was conducted longitudinally following the total lipid and cholesterol content, fatty acids composition and Vitamin E content in human milk from 2-5 days to 12 weeks postpartum of 45 lactating Korean mothers. All samples were from well-defined subjects and uniform collection procedures were used. Total lipid concentration increased from 1.98g/dl at 2-5 days to 3.09g/dl at 12 weeks postpartum. However, there was a large variation in the concentration of total lipids. Total cholesterol concentration tended to decrease from 20.68mg/dl at 2-5days to 17.19mg/dl at 12 weeks. In comparison with transitional and mature milk, human colostrum was characterized by a lower percentage of saturated fatty acids including medium chain length fatty acids and a higher percentage of monounsatureated fatty acids and polyunsaturated fatty acids. Therefore, the P/S and M/S ratios of human milk lipids decreased significanlty during lactation. P/M/S ratio of colostrum changed from 0.38 : 0.84 : 1.00 at 2-5 days to 0.29 : 0.61 : 1.00 at 12 weeks of mature milk. Total tocopherol concentration decreased significantly from 833 ug/dl at 2-5 days to 300 ug/dl at 12 weeks.
This study was aimed at evaluating the lipid status of 122 pregnant women, who were attending peripheral community clinics in low-income areas and were vulnerable in terms of public health, by anthropometric measurements, estimating dietary intakes and analyzing serum lipids content and fatty acids composition. Regarding the skinfold thickness, it has increased more significantly in suprailliac than in triceps, biceps, subscapular, which means body fat has been accumulated in central parts more than in distal parts of the pregnants. It is found that intakes of energy, protein, lipid and carbohydrate are desirable while calcium, zinc and copper are low and sodium is excessive. Regarding dietary fat intake, the energy $\%$ of fat was within the same range as RDA(20%) for adults, but energy $\%$ intake from each fatty acid was usually low, especially energy % of monoun-saturated fatty acid intake was below RDA for adult women. And intake ratio of $\omega$6$\omega$3 fatty acids was desirable. Accordingly, it is thought that dietary fat intake was balanced in quality. Serum lipids concentrations were increased with the length of pregnancy and were higher as compared with non-pregnant women. Polyunsaturated fatty acids, especially $\omega$6 fatty acids, have gradually decreased with the length of pregnancy. Serum lipids content and fatty acids composition were not greatly influenced by dietary fat but serum free fatty acid level was negatively related to energy, carbohydrate, fatty acids and cholesterol intake. On the basis of the above results, it can be predicted that lipid metabolism for mother and fetus is changed by pregnant status and multilateral research on maternal nutrition in terms of areas and income levels will have to be made. Hydroxy fatty acids have gained important attentions because of their special properties such as higher viscosity and reactivity compared to non-hydroxy fatty acids. The new bacterial isolate Pseudomonas aeruginosa (PR3) had been well studied to produce mono-, di-, and tri-hydroxy fatty acids from different unsaturated fatty acids. Of those hydroxy fatty acids, 7,10-dihydroxy-8(E)-octadecenoic acid (DOD) was produced with high yield from oleic acid by P. aeruginosa PR3. Mostly, the substrates used for the production of hydroxy fatty acid by microorganisms were free fatty acids or synthetic lipids. However, it is highly plausible to use vegetable oils containing oleic acid such as olive oil for DOD production by P. aeruginosa PR3. As a first step to address the utilization of olive oil as a substrate for DOD production, we tried to determine the effect of vessel type on DOD production from olive oil by P. aeruginosa PR3. Of two different flask types of normal flask and baffled-flask, baffled-flask was highly effective for DOD production with all the media tested. Maximum increase of productivity by baffled-flask represented 221% with the medium supplemented with whey powder instead of lactose. Results from this study demonstrated that vessel type and medium composition could be both significant factors for DOD production from olive oil by P. aeruginosa PR3.
Most of microalgae shift their metabolic pathways toward the fatty acid biosynthesis following nitrogen deprivation. Recent studies on Nannochloropsis species, oleaginous microalgae, have been performed to investigate the regulation of contents and compositions of fatty acids under stressful condition. The objective of this experiment is to identify the effect of nitrogen on cell growth and fatty acids production in Nannochloropsis oculata K-1281 and compare fatty acid composition response to nitrogen deficiency between N. oculata LB2164 and K-1281. The fatty acids content in N. oculata K-1281 was increased up to 210%, while the growth rate was decreased under nitrogen deficient condition. The contents of C16:0 and C16:1 increased dramatically in both N. oculata K-1281 and LB2164, while the contents of C20:4 and C20:5 increased in N. oculata LB2164. The fatty acids content and composition in N. oculata K-1281 returned following addition of nitrogen after nitrogen starvation. These results demonstrated that fatty acid contents and compositions under nitrogen deficiency will provide the understanding of fatty acid synthesis in microalgae.
The purpose of this study was to investigate dietary fat and individual fatty acids intake pattern of 174 college women living in Seoul and Gyong-gi province through internet nutritional assessment system. Each of the subjects was required to input their own food intake for three days, which included two days during the week and one day of the weekend, on the web program directly and all of the data collected were used for statistical analysis. The mean daily caloric intake of the subjects was 1,500.9 kcal which was at 71.5% of Estimated Energy Requirement (EER). Dietary fat contributed 27.6% of the total caloric intake which was slightly higher than the recommended limit of 25%. Daily cholesterol intake was 310.0 mg, which was also high to some degree. Mean daily N6 and N3 fatty acid intake was 6.1 g and 0.9 g, respectively, and calory % calculated from each were 3.63% and 0.53%. This result showed the intake of N3 fatty acid fell in Acceptable Macronutrient Distribution Ranges (AMR) $0.5\sim1.0%$ but that of N6 fatty acid was somewhat lower than the AMDR $4\sim8%$. N6/N3 ratio 8.5/l, however, was within the desirable range $4\sim10/1$. Considering overall dietary fatty acids intake, oleic acid was the most abundant, followed by linoleic and palmitic acid. And among polyunsaturated fatty acids intake, linoleic acid was exclusively high, accounting for 97.4% of total N6 fatty acid intake. On the contrary, three fatty acids, linolenic (67.3%), DHA (21.1%) and EPA (10.0%), together supplied 98.4% of total N3 fatty acid intake. Mean P/M/S was 0.9/l.1/1.0. The subjects' intake of fat, many fatty acids and cholesterol came from diverse food groups including meats, fats and oils, milk and milk products, eggs, fish, and soybean products. Nevertheless, the subjects tended to show unfavorable fat and fatty acids intake pattern in terms of quantity and quality. Based on these results, it is important to monitor dietary fat intake pattern of the general population continuously and an internet program such as the one used for this study would be valuable, especially for assessing dietary patterns in the younger generation.
In order to determine the position and the content of fatty acids attached to glycerides and the migration degree of fatty acids in the migration reaction, hydrolysis of fish oil was carried out with lipolase-100T derived from Aspergillus oryzae. The content of fatty acids in the glyceride mixture was analyzed and compared with that of fish oil. The amounts of fatty acid in 2-position and the migration degree of the fatty acid in 2,3-DG (diglyceride) and 2-MG (monoglyceride) were calculated. The results showed that approximately 95% (w/w) of DHA (docosahexaenoic acid) and 65% of EPA (eicosapentaenoic acid) was attached to the 2-position of glycerides in the fish oil. Approximately 87% (w/w) of DHA and 75% of EPA remained in 2,3-DG and 88% of DHA and 65% of EPA in 2-MG were not involved in the migration reaction.
This study deals with investigation of the ethanol tolerance of yeast strains with respect to fatty acid composition and intracelluar ethanol concentration during alcohol fermentation. The cell viabilities and fermentation abilities of Saccharomyces cerevisiae and Kluyveromyces fragilis were improved by aeration and addition of unsaturated fatty acids into growth medium. Aeration decreases the accumulation of ethanol, while increases unsaturated fatty acid contents inside yeast cells. Thus it was found that oxygen and unsaturated fatty acids play decisive roles in the increase of ethanol tolerance of yeasts.
In order to know the antifungal characteristics of saturated fatty acids having 6 to 12 carbons, their minimum inhibitory concentrations (MICs) and minimum fungicidal concentrations (MFCs) were estimated against Saccharomyces cerevisiae. Fatty acids from C6 to C11 exhibited increasing activity with chain length, but C12 fatty acid did not show activity at all. In relation to antifungal modes of actions, fatty acids investigated showed on inhibitory activity toward the plasma membrane H+-ATPase of Saccharomyces cerevisiae. Their inhibitions to the glucose-induced acidification and ATP hydrolysis caused by the proton pump were found to be in common wiht antifungal activities. At the test concentration of 1mM, hexanoic acid (C6) showed the lowest inhibition of about 30%, while undecanoic acid(C11) showed the strongest inhibition of over 90%. In addition, as seen with antifungal activity, the inhibitory activity of dodecanoic acid (C12) was suddenly reduced to less than 50%.
