Optimization of biodiesel production via methyl acetate reaction from cerbera odollam

  • Cerbera Odollam (sea mango) is a proven promising feedstock for the production of biodiesel due to its high oil content. Fatty acid methyl esters (FAME) were produced as the final reaction product in the transesterification reflux condensation reaction of sea mango oil and methyl acetate (MA). Potassium methoxide was used as catalyst to study its reacting potential as a homogeneous base catalyst. The initial part of this project studied the optimum conditions to extract crude sea mango oil. It was found that the content of sea mango sea mango oil was 55%. This optimum amount was obtained by using 18 g of grinded sea mango seeds in 250 ml hexane. The extraction was carried out for 24 hours using solvent extraction method. Response surface methodology (RSM) was employed to determine the optimum conditions of the reaction. The three manipulated variables in this reaction were the reaction time, oil to solvent molar ratio, and catalyst wt%. The optimum condition for this reaction determined was 5 hours reaction time, 0.28 wt% of catalyst and 1:35 mol/mol of oil: solvent molar ratio. A series of test were conducted on the final FAME product of this study, namely the FTIR test, GC-FID, calorimeter bomb and viscometer test.

대추의 과육지질(果肉脂質)에 존재(存在)하는 지방산(脂肪酸)의 동정(同定)과 숙성(熟成)에 따른 그 조성(組成)의 변화(變化) (Identification of Fatty Acids in the Pulp Oils of Jujube and Their Compsitional Changes in the Ripening Period)

  • In search for several fatty acid with unusual structure in vegetable oils, we have found that unknown peaks were shown on GLC in the analysis of fatty acids of the lipids from the pulp of ripened jujube (Zizypus jujuba var. inermis) fruits. These fatty acids were identified as a series of cis-monoenoic acids with ${\omega}-5$ double bond system such as $C_{14:1{\omega}5}$, $C_{16:1{\omega}5}$ and $C_{18:1{\omega}5}$, including ${\omega}-7$ fatty acid as $C_{16:1{\omega}7}$ and $C_{18:1{\omega}7}$, by GLC, solid-phase extraction silver ion-column chromatographic, GLC-mass spectrometric and IR techniques. First of all, total fatty acid methyl esters were resolved into saturated and branched fatty acid, monoenoic acid, dienoic acid, and trienoic acid fraction, respectively, with 100% dichloromethane (DCM), DCM/acetone (9:1, v/v) 100% acetone, and acetone/ acetonitrile (97:3, v/v) solvent system. Unknown fatty acids were included in the monoenoic fraction and were confirmed to have cis-configuration by IR. Picolinyl esters of monoenoic fatty acids gave distinct molecular ion peak and dominant diagnostic peaks, for example, m/z 317, 220 and 260 fragment for $cis-C_{14:1{\omega}5}$, m/z 345, m/z 248 and 288 fragment for $cis-C_{16:1{\omega}5}$ and m/z 373, m/z 276 and 316 fragment for $cis-C_{18:1{\omega}5}$. In this way the occurrence of $cis-C_{16:1{\omega}7}$ and $cis-C_{18:1{\omega}7}$ could be deduced from the appearance of prominent fragments as m/z 345, 220 and 260, and m/z 373, 248 and 280. Level of total ${\omega}-5$ fatty acids amounted to about 30% in the fatty acid composition with the predominance of $C_{16:1{\omega}5}$ $ (18.7{\sim}25.0%)$, in the semi-ripened and/or ripened samples collected in September 14 ($C_{16:1{\omega}5}$ ; 18.7%, $C_{14:1{\omega}5}$ ; 3.6% and $C_{18:1{\omega}5}$ ; 3.0%), September 22 ($C_{16:1{\omega}5}$ ; 25.0%, $C_{14:1{\omega}5}$ ; 1.4% and $C_{18:1{\omega}5}$ ; 2.6%), and October $7 (C_{16:1{\omega}5}$ ; 24.7%, $C_{14:1{\omega}5}$ ; 7.7% and $C_{18:1{\omega}5}$ ; 2.5%). However, the lipids extracted from unripened jujube in July and August contain these unusual fatty acids as low as negligible. It could be observed that the level of ${\omega}-5$ fatty acids in the pulps increased sharply with an elapse of ripening time of jujube fruits. Other monoenoic fatty acids with ${\omega}-7$ series, $C_{16:1{\omega}7}$ (palmitoleic acid) and $C_{18:1{\omega}7}$ (cis-vaccenic acid) could be detected. And in the lipids of the kernel and leaf of jujube, none of ${\omega}-5$ fatty acids could be detected.

  • Docosahexaenoic acid (DHA, 22:6n-3) and ecosapentaenoic acid (20:5n-3) have attracted increasing attention since the first epidemiological report on the importance of n-3 essential fatty acids. It is thought that DHA has important functions in brain and retinal tissues. Thraustochytrids, a group of marine protists, are capable of heterotrophic growth, and are potential omega-3 producers for industrial use, especially the members of the Schizochytrium and Thraustochytrium genera. The aims of this work were to isolate, identify and screen thraustochytrids from 17 different locations. Twenty-three isolates were screened for biomass, total fatty acid (TFA) and DHA content. Analysis of the fatty acid methyl esters revealed four distinct clusters biomass ranged from $8.68-9.36gL^{-1}$, and lipid and DHA contents ranged from $3.11-4.10gL^{-1}$ and $1.05-1.93gL^{-1}$ biomass, respectively. B-12 isolates were screened for biomass ($9.36gL^{-1}$), TFA ($4.10gL^{-1}$) and DHA (47.01%, w/w) content. C-6 isolates were also screened for biomass ($8.92gL^{-1}$), TFA ($3.30gL^{-1}$) and DHA (49.41%, w/w) content. The 18S rRNA gene sequencing results identified Schizochytrium mangrovei as B-12 and Crypthecodium cohnii as C-6.

Production of Methyl Ester from Coconut Oil using Microwave: Kinetic of Transesterification Reaction using Heterogeneous CaO Catalyst

  • Methyl ester derived from coconut oil is very interesting to study since it contains free-fatty acid with chemical structure of medium carbon chain ($C_{12}-C_{14}$), so the methyl ester obtained from its part can be a biodiesel and another partially into biokerosene. The use of heterogeneous catalysts in the production of methyl ester requires severe conditions (high pressure and high temperature), while at low temperature and atmospheric conditions, yield of methyl ester is relatively very low. By using microwave irradiation trans-esterification reaction with heterogeneous catalysts, it is expected to be much faster and can give higher yields. Therefore, we studied the production of methyl ester from coconut oil using CaO catalyst assisted by microwave. Our aim was to find a kinetic model of methyl ester production through a transesterification process from coconut oil assisted by microwave using heterogeneous CaO catalyst. The experimental apparatus consisted of a batch reactor placed in a microwave oven equipped with a condenser, stirrer and temperature controllers. Batch process was conducted at atmospheric pressure with a variation of CaO catalyst concentration (0.5; 1.0; 1.5; 2.0, 2.5%) and microwave power (100, 264 and 400 W). In general, the production process of methyl esters by heterogeneous catalyst will obtain three layers, wherein the first layer is the product of methyl ester, the second layer is glycerol and the third layer is the catalyst. The experimental results show that the yield of methyl ester increases along with the increase of microwave power, catalyst concentration and reaction time. Kinetic model of methyl ester production can be represented by the following equation: $-r_{TG}=1.7{\cdot}10^6{_e}{\frac{-43.86}{RT}}C_{TG}$.

지역별 소규모 농가 생산 전통 고추장의 휘발성 성분에 관한 연구 (Volatile Components of Traditional Gochujang Produced from Small Farms according to Each Cultivation Region)

  • The purpose of this study is to investigate the volatile compounds of Korean traditional gochujang from various districts. The volatiles from each traditional gochujang are being extracted by simultaneous steam distillation extraction (SDE), and analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). Twenty compounds are identified as major volatile components which include 8 esters, 4 alcohols and 4 acids. The most traditional gochujang possesses more volatile components rather than commercial gochujang products. Most acids come from fatty acids and the alcohols derive from the oxidative degradation of linolenic acid. The most abundant volatile compounds for both traditional and commercial gochujang include 10 compounds such as 2-methyl-1-propanol, hexanal, 2-methyl-1-butanol, octanoic acid ethyl ester, as well as the various type of acids and esters. They represent most of the total GC peak areas, respectively. From the results, the characteristics of the flavors for traditional gochujang from each district are not clear but have shown various components than the commercial products.

김치유래 젖산균의 균체지방산 분석을 이용한 분류학적 연구

  • Two hundreds and thirty lactic acid bacteria, mostly isolated from Kimchi, including type strains were used for analysis of cellular fatty acids. The 230 test strains were recoverd in 7 major and 1 single clusters defined at Euclidian distance of 17.5. These aggregate taxa were equivalent to the genus Leuconostoc (aggregate group A, B, C and D), the genus Lactobacillus (aggregate group F), the genera Lactobacillus and Pediococcus (aggregate group E) and the genera Leuconostoc and Lactobacillus (aggregate group G). It is concluded as evident that FAMEs (Fatty Acid Methyl Esters) profile of cell can be used as a criterion in classification of lactic acid bacteria from kimchi. Additional comparative taxonomic studies need to be carried out on well chosen representative strains to determine the most appropriate methods of value.

Fatty acid composition and docosahexaenoic acid (DHA) content of the heterotrophic dinoflagellate Oxyrrhis marina fed on dried yeast: compared with algal prey

  • The heterotrophic dinoflagellate Oxyrrhis marina is known to produce high levels of docosahexaenoic acid (DHA) when fed on diverse algal prey. However, large-scale culturing of algal prey species is not easy and requires a large amount of budget, and thus more easily cultivable and low-cost prey is required. Dried yeast was selected as a strong candidate for an alternative prey in our preliminary tests. Thus, we explored the fatty acid composition and DHA production of O. marina fed on dried yeast and compared these results to those of O. marina fed on two algal prey species: the phototrophic dinoflagellate Amphidinium carterae and chlorophyte Chlorella sp. powder. O. marina fed on dried yeast, which does not contain DHA, produced the same high level of DHA as those fed on DHA-containing A. carterae. This indicates that O. marina is likely to produce DHA by itself regardless of prey items. Furthermore, the DHA content (and portion of total fatty acid methyl esters) of O. marina satiated with dried yeast, 52.40 pg per cell(and 25.9%), was considerably greater than that of O. marina fed on A. carterae (26.91 pg per cell; 15.7%) or powder of Chlorella sp. powder (21.24 pg per cell; 16.7%). The cost of dried yeast (approximately 10 US dollars for 1 kg dried yeast) was much lower than that of obtaining the algal prey (approximately 160 US dollars for 1 kg A. carterae). Therefore, compared to conventional algal prey, dried yeast is a more easily obtainable and lower-cost prey for use in the production of DHA by O. marina.

유화전이에스테르화에 의한 대두유의 biodiesel화

  • 대두유의 유화전이에스테르화에 의한 바이오디젤의 생산기술을 연구개발함에 있어서 지방산메틸에스테르의 수율에 미치는 메탄올 몰비, 알칼리 촉매량, 유화제 첨가량 및 반응시간의 영향을 규명하고 유화전이에스테르화 공정의 반응조건을 최적화한 결과, 반응온도 $45^{\circ}C$, 대두유에 대한 에탄올 몰비 1:6, 대두유에 대한 유화제의 첨가량 l(v/v)%에서 0.8wt% 수산화캄륨과 1.2wt% 소디움메톡시드로 대두유를 각각 30분과 50분간 전이에스테르화 하여 $95{\sim}96%$수준의 총괄수율을 얻었다.

Mixotrophic Cultivation of Marine Alga Tetraselmis sp. Using Glycerol and Its Effects on the Characteristics of Produced Biodiesel

  • As a possible feedstock for biodiesel, the marine green alga Tetraselmis sp. was cultivated under different conditions of phototrophic, mixotrophic and heterotrophic cultures. Glycerol, a byproduct from biodiesel production process, was used as the carbon source of mixotrophic and heterotrophic culture. The effects of glycerol supply and nitrate-repletion were compared for different trophic conditions. Mixotrophic cultivation exhibited higher biomass productivity than that of phototrophic and heterotrophic cultivation. Maximum lipid productivity of 55.5 mg L-1 d-1 was obtained in the mixotrophic culture with 5 g L-1 of glycerol and 8.8 mM of nitrate due to the enhancement of both biomass and lipid accumulation. The major fatty acid methyl esters (FAME) in the produced biodiesel were palmitic acid (C16:0), oleic acid (C18:1), linoleic acid (C18:2), and linolenic acid (C18:3). The degree of unsaturation was affected by different culture conditions. The biodiesel properties predicted by correlation equations based on the FAME profiles mostly complied with the specifications from the US, Europe and Korea, with the exception of the cold-filter plugging point (CFPP) criterion of Korea.

미역 추출물로부터 충치 원인균, Streptococcus mutans에 대한 항균물질의 분리 및 동정 (Isolation and Identification of an Antibacterial Substance from Sea Mustard, Undaria pinnatifida, for Streptococcus mutans)

  • 건조 미역 10.4 kg을 acetone으로 추출, 여과한 다음 액-액 분배, 각종 크로마토그라피를 통하여 충치원인균, Streptococcus mutans에 대한 항균물질의 한 성분을 분리, 정제하고(160 mg, 수율 $1.5\times10^{-3}$%) 물질을 동정한 결과는 다음과 같다. 건조 미역 30 g 상당량의 추출액을 기준으로 용매 획분별 항균활성을 조사하였을 때, $CHCl_3$층이 73.2%로 가장 강하였고, hexane층이 62.0%이었으며, BuOH층이나 물층은 거의 없었다. 알루미나 칼럼에서는 산성 획분인 1% $NH_4OH:MeOH(1:1)$용매 획분이 81.0%의 항균활성을 나타내었으며, 실리카 칼럼에서는 $CHCl_3:MeOH(95:5)$용매에서 가장 높은 95.5%의 항균성을 나타내었고, ODS칼럼에서는 85% MeOH에서 96.4%의 항균성을 나타내었다. 최종적으로 ODS칼럼에서 95% MeOH를 이동상으로 하여 3개의 물질 S1(10 mg), S2(90 mg), S3(60 mg)을 분리 정제하였다. TLC에서 각 성분은 동일한 Rf값 0.42를 나타내어 동일한 물질로 추정되었으며, 이들을 메칠 유도체화한 성분들은 Rf값이 0.95로 바뀌어 이들 물질이 carboxyl기를 가지는 지방산으로 추정되었다. GC분석에서 표품 지방산과 비교한 결과, 이들은 $C_{18:4,n-3}$ 지방산과 retention time이 일치하였다. 또한, 메칠 유도체의 mass spectrum 분석 결과, m/z 290에 분자 이온 peak가 관측되어 $C_{18:4,n-3}$ 지방산의 methyl 유도체의 분자량과 일치하여, 이 물질을 3,6,9,12-octadecatetraenoic acid(stearidonic acid, $C_{18:4,n-3}$) 지방산으로 동정하였다.
