보중익기탕(補中益氣湯) 투여가 장거리 달리기 선수의 에너지 및 전해질 대사에 미치는 영향 (The effects of Bo-joong-ik-gi-tang administration on Metabolic Responses ; changes in the energy and electrolyte metabolism among long distance runners)

  • 스포츠 과학 분야에서는 운동 선수들의 경기력 향상과 운동 피로의 신속한 회복을 도모하기 위하여 한약을 복용하고 그 효과와 유의성에 대하여 연구가 활발히 이루어지고 있다. 격심한 운동을 수행할 때 나타나는 Glucose 등 에너지원의 고갈, Lactate 등 대사 산물의 축적, 전해질의 불균형 등의 여러 제반 증상이 기허(氣虛)로 인하여 나타나는 전신의 증후와 유사하다고 생각하여 보중익기탕(補中益氣湯)을 투여한 후 인체의 대사 변화를 관찰함으로서 운동수행능력에 미치는 영향과 유의성을 평가하고자 하였다. 본 연구는 장거리 달리기 선수를 대상으로 2주간의 보중익기탕(補中益氣湯)의 투여가 에너지 대사 및 전해질 대사에 미치는 영향을 규명하기 위하여 최대심박수의 70% 강도의 운동을 실시한 후 운동전 안정시, 운동 직후, 휴식 후 10분, 휴식 후 30분, 휴식 후 1시간으로 나누어 혈액검사를 하였을 때 나타나는 각종 변인의 반응 양상을 비교 분석하여 유의한 결과를 얻었다. 혈중 에너지 대사에서는 Free fatty acid와 Lactate 반응에서 유의성 있는 차이가 나타났고, 전해질 대사에서도 Na+, Cl-와 K+반응에서 모두 유의성 있는 차이를 나타냈다. 그러므로 보중익기탕(補中益氣湯)은 에너지 및 전해질 대사에서 유의성있는 변화를 나타내었으며 운동수행능력의 향상에 유효하였다.

Free fatty acid-induced histone acetyltransferase activity accelerates lipid accumulation in HepG2 cells

  • BACKGROUND/OBJECTIVES: Non-alcoholic fatty liver disease (NAFLD) is a common metabolic disease triggered by epigenetic alterations, including lysine acetylation at histone or non-histone proteins, affecting the stability or transcription of lipogenic genes. Although various natural dietary compounds have anti-lipogenic effects, their effects on the acetylation status and lipid metabolism in the liver have not been thoroughly investigated. MATERIALS/METHODS: Following oleic-palmitic acid (OPA)-induced lipid accumulation in HepG2 cells, the acetylation status of histone and non-histone proteins, HAT activity, and mRNA expression of representative lipogenic genes, including $PPAR{\gamma}$, SREBP-1c, ACLY, and FASN, were evaluated. Furthermore, correlations between lipid accumulation and HAT activity for 22 representative natural food extracts (NExs) were evaluated. RESULTS: Non-histone protein acetylation increased following OPA treatment and the acetylation of histones H3K9, H4K8, and H4K16 was accelerated, accompanied by an increase in HAT activity. OPA-induced increases in the mRNA expression of lipogenic genes were down-regulated by C-646, a p300/CBP-specific inhibitor. Finally, we detected a positive correlation between HAT activity and lipid accumulation (Pearson's correlation coefficient = 0.604) using 22 NExs. CONCLUSIONS: Our results suggest that NExs have novel applications as nutraceutical agents with HAT inhibitor activity for the prevention and treatment of NAFLD.

한국인 수유부에 어유의 보충 급여 효과에 관한 연구 -1. 수유부의 지방산 섭취, 혈장 지질 농도 및 혈장과 적혈구 인지질의 지방산 조성에 미친 영향- (Effects of Fish Oil Supplementation to Korean Lactating Women -I. The Effects on Fatty Acid Composition, Plasma Lipid Concentration and Fatty Acid Composition of Plasma Phospholipids and Erythrocyte of Lactating Women-)

  • We studied the effects of fish oil supplementation with low does on the lipid concentration and fatty acid of plasma and the fatty acid composition of plasma phospholipid and erythrocyte of lactating women. The subjects, 18 lactating women, who were exclusively breast-fed their babies were classifed into a control group and 2 fish oil groups according to dose; the subjects of fish oil groups were supplemented with 1.96g/d or 3.92g/d of fish oil, respectively for 2 weeks from 10 to 12 weeks postpartum. All subjects consumed their usual diet at home. Blood sample were collected at the final day of experiment. The plasma HDL-cholesterol level increased significantly by fish oil supplementation. The concentrations of DHA (docesahexaenoic acid) and EPA (eicosapentaenoic acid) in the plasma PC(phosphatidylcholine) and PE (phosphatidylethanolamine)of fish oil groups tended to increase, but not significant. However, the concentrations of DHA and EPA of PC and PE in erythrocyte were not affected by fish oil supplementation. These results demonstrate that fish oil supplementation with low dose does not change the concentration of plasma lipid as well as fatty acid composition in plasma PC and PE and red blood cell obviously. However the increase of plasma HDL-cholesterol level, the reduction of atherogenic index(AI) and the tendency of increase of DHA and EPA concentrations in plasma PC and PE indicate that there may be some beneficial effects on maternal lipid metabolism if fish oil intakes were increased.

Study on the fatty acid profile of phospholipid and neutral lipid in Hanwoo beef and their relationship to genetic variation

  • Maize which has very high omega-6 fatty acid content has been used as a main feed grain for Hanwoo beef production to increase marbling, and thus omega-6 to omega-3 fatty acids ratio in Hanwoo beef is expected to be biased. To elucidate the current status of omega fatty acids ratio in Hanwoo beef, fatty acid profiles of neutral lipid and phospholipid fraction were analyzed separately using 55 Hanwoo steers' longissimus dorsi muscle slaughtered at Pyeongchang, Korea from Oct. to Nov. 2015. In addition, an association study was conducted to evaluate associations between single nucleotide polymorphism (SNP) markers from references and omega fatty acid profiles in phospholipid of Hanwoo beef samples using analysis of variance (ANOVA). In neutral lipid fraction, composition of saturated and monounsaturated fatty acids was higher and polyunsaturated fatty acids was lower compared to those in phospholipid fraction. The mean n-6/n-3 ratios of Hanwoo were $56.059{\pm}16.180$ and $26.811{\pm}6.668$ in phospholipid and neutral lipid, respectively. There were three SNPs showing statistically significant associations with omega fatty acid content. GA type of rs41919985 in fatty acid synthase (FASN) was significantly associated with the highest amount of C20:5 n-3 (p = 0.031). CC type of rs41729173 in fatty acid-binding protein 4 (FABP4) was significantly associated with the lowest amount of C22:2n-6 (p = 0.047). AG type of rs42187261 in FADS1 was significantly linked to the lowest concentration of C20:4 n-6 (p = 0.044). The total n-6/n-3 ratio of the steer which has all four SNP types in above loci (27.905) was much lower than the mean value of the total n-6/n-3 ratio in phospholipid of the 55 Hanwoo steers ($56.059{\pm}16.180$). It was found that phospholipid and neutral lipid of Hanwoo have very high n-6/n-3 ratios compared to the reported data from different cow breeds. Four SNPs in genes related with fatty acid metabolism showed significant associations with the fatty acid profile of phospholipid and may have potential as SNP markers to select Hanwoo steers in terms of n-6/n-3 balance in the future.

Genome wide association study of fatty acid composition in Duroc swine

  • Objective: Genome wide association study was conducted to identify and validate candidate genes associated with fatty acid composition of pork. Methods: A total of 480 purebreed Duroc pigs were genotyped using IlluminaPorcine60k bead chips while the association test was implemented following genome-wide rapid association using Mixed Model and Regression-Genomic Control (GRAMMAR-GC) approach. Results: A total of 25, 29, and 16 single nucleotide polymorphisms (SNPs) were significantly associated with stearic (18:0), oleic (18:1) and saturated fatty acids (SFA), respectively. Genome wide significant variants were located on the same region of swine chromosome 14 (SSC14) that spanned from 120 to 124 Mb. Top SNP ALGA008191 was located at 5 kb near the stearoyl-CoA desaturase (SCD) gene. This gene is directly involved in desaturation of stearic acid into oleic acid. General relationship of significant SNPs showed high linkage disequilibrium thus genome-wide signals was attributed to SCD gene. However, understanding the role of other genes like elongation of very long chain fatty acids-3 (ELOVL3) located on this chromosomal segment might help in further understanding of metabolism and biosynthesis of fatty acids. Conclusion: Overall, this study provides evidence that validates SCD gene as strong candidate gene associated with fatty acid composition in Duroc pigs. Moreover, this study confirms significant SNPs near ELOVL3 gene.

도시 저속득층 지역의 모자 영양 및 섭식에 관한 생태학적 연구 -I. 임산부의 인체 계측, 식이 섭취 및 혈청 지질/지방산 조성 - (Ecological Studies of Maternal-Infant Nutrition and Feeding in Urban Low Income Areas)

  • This study was aimed at evaluating the lipid status of 122 pregnant women, who were attending peripheral community clinics in low-income areas and were vulnerable in terms of public health, by anthropometric measurements, estimating dietary intakes and analyzing serum lipids content and fatty acids composition. Regarding the skinfold thickness, it has increased more significantly in suprailliac than in triceps, biceps, subscapular, which means body fat has been accumulated in central parts more than in distal parts of the pregnants. It is found that intakes of energy, protein, lipid and carbohydrate are desirable while calcium, zinc and copper are low and sodium is excessive. Regarding dietary fat intake, the energy $\%$ of fat was within the same range as RDA(20%) for adults, but energy $\%$ intake from each fatty acid was usually low, especially energy % of monoun-saturated fatty acid intake was below RDA for adult women. And intake ratio of $\omega$6$\omega$3 fatty acids was desirable. Accordingly, it is thought that dietary fat intake was balanced in quality. Serum lipids concentrations were increased with the length of pregnancy and were higher as compared with non-pregnant women. Polyunsaturated fatty acids, especially $\omega$6 fatty acids, have gradually decreased with the length of pregnancy. Serum lipids content and fatty acids composition were not greatly influenced by dietary fat but serum free fatty acid level was negatively related to energy, carbohydrate, fatty acids and cholesterol intake. On the basis of the above results, it can be predicted that lipid metabolism for mother and fetus is changed by pregnant status and multilateral research on maternal nutrition in terms of areas and income levels will have to be made. Effects of a Pre-Exercise Meal on Plasma Growth Hormone Response and Fat Oxidation during Walking

  • The purpose of this study was to determine the effects of a pre-exercise meal on the plasma human growth hormone (hGH) response and fat oxidation during walking. Subjects (n=8) were randomly provided with either 1 g/kg body weight of glucose in 200 mL water (CHO) or 200 mL water alone (CON) 30 min prior to exercise and subsequently walked on a treadmill at 50% of VO2max for 60 min. Plasma hGH concentrations were significantly higher in subjects who received CHO compared to those who received CON at 15 and 30 min. The fat oxidation rate in the CHO was significantly lower than the CON while walking for 5~15, 25~35 and 45~55 min. Plasma FFA levels were also significantly lower in the CHO compared to the CON at 30, 45 and 60 min. Plasma glucose levels in the CHO were significantly lower while plasma insulin levels were significantly higher than in the CON at 15 and 30 min. Therefore, the results of this study suggest that the elevation of plasma hGH levels due to the intake of a pre-exercise meal may not be strongly related to fat oxidation and plasma free fatty acid (FFA) levels during low-intensity exercise.

  • Essential fatty acids (EFA) are fatty acids that must be obtained from the diet because they can not be biosynthesized by human or animals. Gamma fatty acids contain gamma-linolenic acid (GLA, 18:3n-6) and dihomo-gamma-linolenic acid (DHGLA, 20:3n-6) as intermediate metabolites of linoleic acid (LA, 18:2n-6), which is an EFA found in vegetable oils. GLA is an important essential fatty acid that is required by human and animals to function normally. Recently, studies have indicated that GLA may be an essential component of the cell membrane, as well as an active component of dietary supplements and medicine. GLA must beadministered through the diet because it is converted into DHGLA in the body quickly and completely. DHGLA is a key material involved in the metabolism of LA. GLA is biosysthesized by the rate limiting step of ${\Deltac}^6$-desaturase, which is an enzyme that desaturates LA, there by allowing it to be converted into DHGLA via chain elongation. In addition, DHGLA exerts bioactive effects via action as a precursor of eicosanoid series 1. Breast milk contains an abundant amount of GLA; however, GLA is also available directly in evening primrose oil, black currant seed oil, borage oil and hemp seed oil. In addition, GLA enriched animal and plant can be produced using biotechnology, and highly pure GLA can be extracted using supercritical fluids, such as supercritical carbon dioxide, which will allow economically feasible production of GLA for use in medicines.

A Role of Unsaturated Fatty Acid in Animal Reproductive Cells and Biology

  • As a one of unsaturated fatty acid, polyunsaturated fatty acids (PUFAs) have multiple actions: as precursor of prostaglandins (PGs), steroid hormone synthesis and energy production in animal reproduction. PUFAs, which include omega-3 (n-3) and omega-6 (n-6), are derived from the diet and changed by diet, species, breed and season. The plasma membrane of spermatozoa in mammals contain various PUFAs. These composition of PUFAs regulate the membrane fluidity and cause lipid peroxidation via generation of reactive oxygen species (ROS). Induced lipid peroxidation by ROS decreased viability and motility of spermatozoa, and it is reduced by addition of antioxidant and low concentration of PUFAs. Because oocytes of animal have a high lipid components, process of oocyte maturation and embryo development are influenced by PUFAs. In in vitro study, oocyte maturation, embryo development, intracellular cAMP and MAPK activity were increased by treatment of n-3 ${\alpha}$-linolenic acid (ALA) during maturation, whereas n-6 linoleic acid (LA) negatively influenced. Also, inhibition of fatty acid metabolism in oocyte influenced blastocyst formation of cattle. PGs are synthesized from PUFAs and various PUFAs influence PGs via regulation of PG-endoperoxide synthase (PTGS). Steroid hormone synthesis from cholesterol is regulated by expression of steroid acute regulator (StAR) protein and mRNA. Exogenous n-3 and n-6 PUFAs altered sex hormone in animal through stimulate or inhibit StAR activity. Because PUFAs altered PG and steroid hormone synthesis, follicular development was influenced by PUFAs. This effect of unsaturated fatty acid could provide information for improvement of reproductive ability in animals.

  • 신미종(辛味種)고추의 추숙(追熟)에 따른 지질(脂質)의 변화(變化)를 중성지질(中性脂質)과 극성지질(極性脂質)로 분획(分劃)하여 실험(實驗)한 결과(結果)는 다음과 같다. 1. 중성지질(中性脂質)의 함량(含量)은 cli. stage에서 최저치(最低値)가되고 극성지질(極性脂質)의 함량(含量)은 post-cli. stage에서 격감(激減)하고있다. 2. 중성지질(中性脂質)의 지방산(脂肪酸)으로서 주(主)가 되는 것은 linoleic acid이고 이것은 추숙(追熟)에 따라 점감(漸減)하고 있다. 3. 중성지질(中性脂質) fatty acid의 saturates/unsaturates치(値)가 cli. stage에서 최저(最低)가 되는데, 이것을 중성지질함량(中性脂質含量)도 cli.stage에서 최저(最低)를 가리키는 것과 아울러 생각할때 climacteric rise가중성지질(中性脂質)의 대사(代謝)와 관계(關係)가 있는것 같다. 4. 극성지질(極性脂質)의 지방산(脂肪酸) 조성(組成)은 추숙(追熟)에 따라 linoleic acid가 줄곧 감소(減少)하고 linoleic acid가 증가(增加)하여 cli. stage에서 역전(逆轉)하고 있는데, 이것은 고추의 climacteric rise에 따라 일어나는 막성분(膜成分)의 변화(變化)를 가리키는 한 지표(指標)가 될 수 있을 것 같다.
