  • Changes in protein metabolism were studied in hemolymph and fat body on days 1, 3, 5 and 7 of the fifth-instar silkworm, Bombyx mori, exposed to lethal, sublethal doses and prevailing levels of fluoride in groundwater in Karnataka and Andhra Pradesh States of India. The total protein content indicated a depletion followed by a concomitant increase in accumulation of free amino acids. Concurrently, the activity of protease in both of the tissues was also increased. A steady enhancement in the activities of alanine aminotransferase and aspartate aminotransferase paralleled the elevation of glutamate dehydrogenase activity in the tissues studied. It is presumed, on the basis of these results, that the fluoride toxicity causes major changes in protein metabolism of the silkworms.

  • Animals possess a highly sophisticated mechanism of storing energy in adipose tissue inside their bodies. However, in humans it has been clarified that adipocyte (fat cell), which composes the body fat (adipose) tissues, development and the extent of subsequent fat accumulation are closely associated with the occurrence and advancement of various common diseases (e.g., type-2 diabetes, coronary artery disease, and hypertension) resulting from obesity. Recent exciting progress in clinical and biochemical studies of adipocytes has rapidly clarified the functions of adipocytes and adipose tissue. Interesting findings are the function of white adipocytes as "secreting cells" and the molecular mechanism undelying adipocyte differentiation at the transcriptional level in relation to nuclear receptors. Consequently, the adipose tissue is being targeted for the prevention or treatment of many common diseases. In this review, I will focus on recent information on characteristics of adipocytes and the relationship between obesity and common obesity-related diseases. diseases.

  • Evidence from recent studies in several laboratories indicates a relationship between type or level of fat in the diet and occurance of tumor at specific sites. The essential fatty acids in fat and degree of their unsaturation are important to determine the influence of a dietary fats on carcinogenesis. Alteration of dietary fat can also change carcinogenesis of cell in several tissues. Dietary fats appear to be important in both initiation and promotion stages of carcinogenesis. Several possible mechanisms have been investigated how dietary fat could affect to carcinogenesis at cellular level. One potential mechanism of dietary fat on carcinogenesis is through modulation of protein kinase C activity in the cell.

  • Two separate trials were designed to determine effects of dietary level of whole flaxseed (WFS) on fatty acid composition of serum, and subcutaneous, perirenal, intermuscular, and intramuscular adipose tissues of Korean Hanwoo cattle. Twentyone bulls (trial 1) and 15 cows (trial 2) were assigned to diets containing 0, 10 or 15% WFS. Relative treatment effects were similar between bulls and cows. The proportion of C18:3 in serum and to a lesser extent in adipose tissues were increased by dietary inclusion of WFS, reflecting supplemented lipid composition of WFS that escaped ruminal biohydrogenation. Animals fed WFS had a lower proportion of saturated fatty acids in serum and adipose tissues than animals fed diets without WFS, while the opposite trend was observed in unsaturated fatty acids with little differences between two WFS groups. WFS-fed animals had higher proportions of C18:1, 18:2, 18:3, 20:3, and 22:3 and lower proportions of C12:0, 14:0, 16:0 and 18:0 in intramuscular fat than animals fed diets without WFS. Furthermore, feeding WFS increased proportions of both $\omega$-3 and $\omega$-6 fatty acids but decreased the ratio of $\omega$-6/$\omega$-3 substantially. In conclusion, feeding WFS can be an effective method of increasing absorption of unsaturated fatty acids, and subsequent deposition in adipose tissues.

  • Angiopoietin-like protein 4 (ANGPTL4) is involved in a variety of functions, including lipoprotein metabolism and angiogenesis. To reveal the role of ANGPTL4 in fat metabolism of sheep, ovine ANGPTL4 mRNA expression was analyzed in seven adipose tissues from two breeds with distinct tail types. Forty-eight animals with the gender ratio of 1:1 for both Guangling Large Tailed (GLT) and Small Tailed Han (STH) sheep were slaughtered at 2, 4, 6, 8, 10, and 12 months of age, respectively. Adipose tissues were collected from greater and lesser omental, subcutaneous, retroperitoneal, perirenal, mesenteric, and tail fats. Ontogenetic mRNA expression of ANGPTL4 in these adipose tissues from GTL and STH was studied by quantitative real time polymerase chain reaction. The results showed that ANGPTL4 mRNA expressed in all adipose tissues studied with the highest in subcutaneous and the lowest in mesenteric fat depots. Months of age, tissue and breed are the main factors that significantly influence the mRNA expression. These results provide new insights into ovine ANGPTL4 gene expression and clues for its function mechanism.

  • The effects of dietary levels of lysine and energy on growth performance, the content of DNA, RNA and protein in liver, thigh muscle composition and nutrient utilization in broiler chicks were investigated in an experiment involvies with 2 levels of dietary energy : 3,200 (2900) 2,900 (2700) kcal ME/kg) and 6 levels of lysine : 0.6(0.5), 0.8(0.7), 1.0(0.9), 1.2(1.1), 1.4(1.3), and 1.6(1.5)% was carried out. A total number of 384 male broiler chicks was used for a period of 7 weeks. Body weight gain of 1.0(0.9)% lysine level group was significantly (p < 0.01) higher than that of any other groups. Interaction between lysine and energy in the feed intake was observed (p < 0.05). Present data indicate that the content of DNA in liver tissues was significantly (p < 0.05) different by the levels of lysine, namely, 1.0(0.9)% or 1.2(1.1)% lysine level groups showed higher content than other groups (p < 0.01). Dietary levels of 1.2(1.1)% or 1.6(1.5)% lysine groups showed the highest protein content in thigh muscle tissues than that of any other groups (p < 0.05). Interaction between energy and lysine in the content of protein of thigh muscle tissues was shown (p < 0.01). The level of 0.6% lysine group showed the highest fat content in thigh muscle tissues than any other groups. Interaction between lysine and energy in the content of crude ash and crude fat of thigh muscle tissues was observed (p < 0.01). Apparent amino acid availability of arginine, glycine and threonine (p < 0.01), phenylalanine (p < 0.05) were significantly affected by the levels of lysine and interaction between lysine and energy was found only in arginine (p < 0.01).

  • In order to evaluate the effect of Chekambohyulansin-tang (CST) which was a traditional poly-herbal formula has been used for treatment of obesity in Korea, on the obesity in ovariectomized rats, the changes of body weight, peri-kidney, abdominal and perineal subcutaneous fat weight and their histological changes were measured with the detection of number and/or size of adipocytes after an oral administration of CBT extracts (125, 250 and 500 mg/kg, respectively). CBT significantly attenuated elevations in the body weight, the absolute and relative weight of peri-kidney, abdominal and perineal fat tissues and decrease the severe hypertrophy, the number of adipocytes, and the diameter of adipocytes of peri-kidney, abdominal and perineal fat tissues in ovariectomized rats in a dose dependent manner. These results provide a story evidence that CBT can playa role in the treatment of obesity induced by ovariectomy. Determination of the specific mechanisms involred toxicological dosage of CBT on obesity will require addition study.

  • The influence of baculovirus Bombyx mori Nuclear Polyhedrosis virus (BmNPV) infection on intermediary metabolic pathways in silkworm Bombyx mori L. was investigated. Studies revealed that NADP-linked malate dehydrogenase activity in hemolymph of infected silkworms at 96 hrs post infection (p.i.) with visible symptoms of infection was enhanced in comparison to healthy larvae of the same age. Also, NADP-dependent MDH activity was significantly lower in fat body cytosol of infected larvae at 96 hrs p.i. when compared to healthy larvae. Similarly, some biometabolic parameters like growth, protein content and cholesterol titer were observed to be influenced by baculovirus infection. While the growth of infected larvae was significantly retardedi protein content was also drastically reduced in both hemolymph and fat body tissues. Cholesterol titers however, was enhanced in infected larvae. The results observed herein point to a significant change in the normal biochemical and biometabolic pathways required for growth and development following BmNPV infection.

  • This study was to detect the expression of heart fatty acid-binding protein (H-FABP) and adipocyte fatty acid-binding protein (A-FABP) gene mRNA in different tissues of Rugao and Luyuan chickens at 56 d and 120 d by real-time fluorescence quantitative reverse transcription polymerase-chain reaction (FQ-RT-PCR). The primers were designed according to the sequences of HFABP, A-FABP and GAPDH genes in Gallus gallus, which were used as target genes and internal reference gene, respectively. The levels of H-FABP and A-FABP gene expression were detected by SYBR Green I FQ-RT-PCR. The relative H-FABP and A-FABP gene mRNA expression level was calculated with 2-$^{{\Delta}Ct}$. Melting curve analysis showed a single peak of three genes. Intramuscular fat (IMF) content in breast muscle and leg muscle of the two chicken breeds at 120 d was higher than at 56 d. IMF content in breast muscle and leg muscle at 56 d and 120 d in Luyuan was significantly higher than in Rugao, however, abdominal fat of Luyuan was significantly lower than that of Rugao. The relative H-FABP gene mRNA expression level in cardiac muscle was the highest in both chicken breeds. The relative H-FABP and A-FABP gene expression of different tissues in Luyuan was higher than in Rugao. H-FABP gene mRNA expression had a negative effect on IMF of leg and breast muscles, and was significantly negatively correlated with IMF content. The relative A-FABP gene mRNA level in abdominal fat was higher than in liver. The A-FABP gene mRNA was not expressed in leg, breast and cardiac muscles. A-FABP gene mRNA expression level was significantly positively correlated with abdominal fat and had a significant effect on abdominal fat but not IMF content.
