In order to fractionate sardine oil by different solvents for an effective use of fish oil being subjected to the limit of use, an attempt was to investigate the proper solvents, ratios and fractionation time. The results of the study were as follows: 1. The proper solvent of fractionation using ethanol, isopropyl alcohol, acetone, and hexane was ethanol, and its optimum ratio was 2:1 (ethanol: oil, v/w). The proper time of ethanol fractionation by the ratio (2:1) was 4hr at $10^{\circ}C$, 6hr at $5^{\circ}C$, 8hr at $0^{\circ}C$and 8hr at $-5^{\circ}C$, respectively. 2. In the fractionation by stages using the ratio (2:1) at each temperature, the yield of stearine was 8% at $10^{\circ}C$ (Fraction I), 32% at $5^{\circ}C$ (Fraction II), 7% at $0^{\circ}C$ (Fraction III) and 10% at $0^{\circ}C$ (Fraction IV), respectively. When ethanol fractionation was undertaken at $5^{\circ}C$ by stages, the yield of stearine (Fraction II) was high. 3. Iodine value of Fraction II was 96.8. This result indicated that the hydrogenation process would be simplified by fractionation. 4. The percentage of the decrease of polyenoic acids from original sardine oil to Fraction II oil was from 30.5% to 13.5%. The major fatty acids of Fraction II were palmitic and oleic acids and these fatty acids were about 52% of total fatty acids. Therefore, Fraction II, which remained liquid oil at room temperature because solid fat content was 6.9% at $20^{\circ}C$, would be used as frying oil.

To evaluate energy efficiency and partition of nutrients, 32 piglets were weaned at 14 d of age and individually fed diets containing 15% fat from coconut oil (CO, medium chain saturated), high oleate sunflower oil (HOSO, n-9 series), soybean oil (SO, n-6 series), or linseed oil plus fish oil, (LF, n-3 series). After 4 weeks, the subjects were sacrificed to evaluate empty body composition and apparent ileal digestibility with the slaughter method. No statistical effect of dietary fat sources on growth was observed. The digestibility of fat from the coconut oil diet was higher than fats from the diets containing high levels of unsaturated fatty acids. The efficiency of use of metabolizable energy for growth averaged 63% and was not affected by the diet. Dietary fat composition was reflected strongly in backfat. Total body neutral and polar fatty acids were influenced too. For the whole body phospholipid fraction the ratio of n-6 to n-3 and the double bond index were 4.3, 5.8, 7.2, 0.78 and 69, 87, 89, 87 for CO, HOSO, SO, and LF respectively. These results show that for the coconut oil diet the degree of unsaturation of phospholipids in the body was lower and that, in the other diets, it did not differ, but double bond index was maintained with different n-6 to n-3 ratios in carcass fat. On the whole the data on body fat composition indicate that the dietary fat tended to be deposited in similar quantity in the body, whatever was the dietary fatty acid profile.
To develop mil fat fractions with desirable physico-chemical properties, anhydrous milk fat (AMF) was fractionated by one- and two-stage extractions using supercritical $CO_2$(SC-$CO_2$). Tow-stage extraction of AMF was performed by first producing tow fractions, an extract and a residue at 4$0^{\circ}C$/241bar, which were subsequently used as the feed for an extraction at 6$0^{\circ}C$/241bar and 4$0^{\circ}C$/345bar, and separated into five and four fractions, respectively, based one extraction time. These fractions were quantified and analyzed for fatty acids and physico-chemical properties. SHort-chain (C4~C8) fatty acids in extract fractions from an extract were 200~150% compared with those of the original AF. Long-chain (C14~C18) fatty acids in extract fractions from a residue were 118~141%. The ratio of unsaturated fatty acids in the residue fraction was 131%. Melting point ranged from 22 to 43$^{\circ}C$, iodine value 21.8 to 36.9, and saponification value 255 to 221 in the extract and residue fractions. SC-$CO_2$ fractionation of AMF by two-stage extraction offers the possibility of developing ractions with discrete fatty acid compositions and physico-chemical properties such as melting point, iodine value and saponification value.

[Purpose] Blood glucose and insulin resistance were lower following hypoxic exposure in previous studies. However, the effect of hypoxia as therapy in obese model has not been unknown. [Methods] Six-week-old mice were randomly divided into chow diet (n=10) and high-fat diet (HFD) groups (n=20). The chow diet group received a non-purified commercial diet (65 % carbohydrate, 21 % protein, and 14 % fat) and water ad libitum. The HFD group was fed an HFD (Research Diet, #D12492; 60% kcal from fat, 5.24 kcal/g). Both groups consumed their respective diet for 7 weeks. Subsequently, HFD-induced mice (12-weeks-old) were randomly divided into two treatment groups : HFD-Normoxia (HFD; n=10) and HFD-Hypoxia (HYP; n=10, fraction of inspired=14.6%). After treatment for 4 weeks, serum glucose, insulin and oral glucose tolerance tests (OGTT) were performed. [Results] Homeostatic model assessment values for insulin resistance (HOMA-IR) of the HYP group tended to be lower than the HFD group. Regarding the OGTT, the area under the curve was 13% lower for the HYP group than the HFD group. [Conclusion] Insulin resistance tended to be lower and glucose uptake capacity was significantly augmented under hypoxia. From a clinical perspective, exposure to hypoxia may be a practical method of treating obesity.
Purpose: To investigate and compensate the effects of respiration-induced B0 variations on fat quantification of the bone marrow in the lumbar spine. Materials and Methods: Multi-echo gradient echo images with navigator echoes were obtained from eight healthy volunteers at 3T clinical scanner. Using navigator echo data, respiration-induced B0 variations were measured and compensated. Fat fraction maps were estimated using $T2^*$-IDEAL algorithm from the uncompensated and compensated images. For manually drawn bone marrow regions, the estimated B0 variations and the calculated fat fractions (before and after compensations) were analyzed. Results: An increase of temporal B0 variations from inferior level to superior levels was observed for all subjects. After compensation using navigator echo data, the effects of the B0 variations were reduced in gradient echo images. The calculated fat fractions show significant differences (P < 0.05) in L1 and L3 between the uncompensated and the compensated. Conclusion: The results of this study raise the need for considering respiration-induced B0 variations for accurate fat quantification using gradient echo images in the lumbar spine. The use of navigator echo data can be an effective way for the reduction of the effects of respiratory motion on the quantification.

Rats were adapted to diets containing 10% cellulose,10% sodium alginate and fiber-free diet for 5 weeks. Following a 14 hour fasting, rats were fed 5g of a test meal that provided 50% energy from fat, then killed at 4 hour postprandially. Plasma and lipoprotein fraction-cholesterol levels were lower in sodium alginate-fed animals than in rats fed other diets. Plasma TG did not differ among diet treatments. Increase in TG content of HDL fraction occurred in dietary fiber groups. Intestinal apolipoprotein B level and lipase activity were lower in sodium alginate-fed group than in other dietary groups. These results suggest that chronic consumption of sodium alginate affects plasma cholesterol level as in the case of fiber supplemetation, but is less likely to modify the acute Plasma TG response to high fat meal than if a fiber supplement is incorporated into the meal.
The abnormal content of blood lipids often results in metabolic diseases, such as hyperlipidemia and obesity. Many agents, including natural sources from traditional food, have been developed to regulate the blood lipid contents. In this study, we examined the anti-hyperlipidemic activity of Rhynchosia nulubilis seeds pickled with brown rice vinegar (RNSpBRV), a Korean traditional pickled soybean food. Since RNSpBRV is made of R. nulubilis seeds (RNS) soaked in brown rice vinegar (BRV), we compared the anti-adipogenic activity between RNS, BRV and solid fraction of RNSpBRV (SF-RNSpBRV), liquid fraction of RNSpBRV (LF-RNSpBRV). For this, the inhibitory effect of lipid accumulation in 3T3-L1 adipocyte was checked by adding methanol extracts of mixed RNS and BRV, LF-RNSpBRV, and SF-RNSpBRV. The addition of each methanol extract up to 1 mg/ml showed no cytotoxicity on 3T3-L1 adipocyte, and approximately 20% of the lipid droplet formation was suppressed with the methanol extract of BRL or SF-RNSpBRV. The highest suppression (42.1%) was achieved with LF-RNSpBRV. In addition, mice fed a high fat diet (HFD) supplemented with 5% RNSpBRV powder led to increased high density lipoprotein (HDL) cholesterol and lower blood glucose, triglyceride, and total cholesterol compared to mice fed with a HFD diet only. Interestingly, the size of the epididymis cells gradually decreased in HFD + 1% RNSpBRV and HFD + 5% RNSpBRV-fed mice if compared those of HFD-fed mice. Taken together, these results provide evidence that RNSpBRV has a regulatory role in lipid metabolism that is related to hyperlipidemia.
The effects of dietary n-3 fatty acids, ${\alpha}$-linolenic acid (18:3), eicosapentaenoic acid (EPA, 20:5), and docosahexaenoic acid (DHA, 22:6), on brain phospholipid content and fatty acid composition were compared in rats fed with a diet containing constant ratios of saturated fatty acid/monounsaturated fatty acid/polyunsaturated fatty acid (PUFA) and n-3/n-6. The dietary fat in each diet was added at the level of 10%. In each diet, n-3 PUFA comprised two-thirds of the PUFA and the remaining one-third was linoleic acid (18:2). Dietary fat containing linoleic acid as the sole source of PUFA was also given to the control group. The content of brain phospholipid in the three n-3 PUFA groups was significantly lower than that of the linoleic acid group. This reduction was greater in the EPA and DHA groups than in the ${\alpha}$-linolenic acid group. The decrease in phospholipid content in rats fed n-3 fatty acid-rich diets was largely due to the decrease in the phosphatidylethanolamine fraction. Each dietary n-3 PUFA was found to affect the fatty acid composition of brain phospholipids; the most pronounced alteration was observed in phosphatidylethanolamine fraction. Furthermore, the proportion of DHA in the phosphatidylethanolamine fraction tended to be higher in the DHA group than in other PUFA groups. In conclusion, dietary ${\alpha}$-linolenic acid, EPA and DHA can influence the phospholipid content, phospholipid subclass, and fatty acid composition in rat brain.
Objectives: Schizandrae fructus (Schizandra chinensis) is one of very common herbs, it is known as natural antioxidants, anti-inflammatory agent. Also some reports show that its extract works to regulate of dyslipidemia. This study was designed to investigate the effects of Schizandrae fructus chloroform fraction (SFCF) on serum lipid levels in dyslipidemic mice. Methods: The levels of total cholesterol, high density lipoprotein-cholesterol, triglyceride, aspartate aminotransferase (AST), alanine aminotransferase (ALT), fasting blood glucose in serum were measured. Histopathological and gene expression changes in liver tissue were also observed. Results: Oral administration of SFCF lowered levels of total cholesterol and triglyceride, which were elevated by high-fat diet. But SFCF did not affect on weight changes and serum AST, ALT levels in dyslipidemic mice. After carrying out gene ontological analysis, large numbers of genes in high-fat diet group were up-(347) or down-regulated (235). In SFCF treated mice, some changed expression of the genes was restored to normal levels, with a recovery rate of 17%. And it seems that fatty acid biosynthesis pathway was one of important key pathways to recovery. Conclusions: SFCF has beneficial effect on dyslipidemia, and could be used to prevent and treat cardiovascular disease.
The present study was carried out to investigate the antioxidative effect of ginger extracts on fish oil. The changes of sardine oil with and without ginger extract were estimated by periodically measuring peroxide value (POV), thiobarbituric acid (TBA) value, weighing method, acid value (AV) and fatty acid composition. The results obtained are summarized as follow : The POV of sardine oil by $80\%$ ethanol extract and fat soluble fraction obtained from ginger during storage was rapidly increased after 10 days, while water soluble fraction was slowly increased during storage for 25 days. TBA value of sardine oil by water soluble fraction was appeared to increase slowly until 10 days, but that of $80\%$ ethanol extract and fat soluble fraction was remarkably increased in early stages of storage. The weighing change of sardine oil by $80\%$ ethanol extract and fat soluble fraction were shown $3.5\%\;and\;1.7\%$ for 15 days, but by water soluble fraction was marked $0.5\%$ of weight gain. Docosahexaenoic acid (DHA) in polyunsaturated fatty acid of sardine oil during storage markedly decreased, but by the addition of each fraction of ginger extracts, the oxidative degradation of DHA was effectively inhibited, of which water soluble fraction was most effective.
