Inhibitory Effects of Bogigambi-tang on the Obese-mouse Induced by High Fat Diet (보기감비탕(補氣減肥湯)이 고지방사료 식이로 비만이 유발된 생쥐에게 미치는 영향)

  • In order to investigate the effects of Bogigambi-tang(here in after referred to BGGBT) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high fat diet. C57BL/6 mice were divided into three groups(normal, high fat diet with control, high fat diet with BGGBT extract) and fed for 15 weeks. Items of this experimental study are as follows. Body weight change, final inclose of body weight, the weight change of the adipocytes in body, the level change of ALT, AST, total cholesterol, LDL-Cholesterol, triglyceride, glucose, free fatty acid and creatinine, the expression of ${\beta}$3AR and leptin gene in primary adipocytes, the production change of leptin in primary adipocytes, the expression of ${\beta}$3AR and leptin in adipocytes tissue. The following results have been obtained All experimental group have shown that the weight and the final increase of weight have decreased considerably. All experimental group have shown that the amount of the adipocyte in weight has decreased considerably. All experimental group have shown that the amount of leptin has decreased considerably. All experimental group have shown that the revelation of ${\beta}$3AR in primary adipose cell and 3T3-L1 cell has increased considerably, and that the revelation of leptin in primary adipose cell and 3T3-L1 cell has decreased considerably, All experimental group have shown that the size of adipocyte in adipocytes tissue has decreased. The high density group have shown that the adipose vacuoles in liver tissue has decreased considerably, and that the cell nucleuses has similar with normal group.

Effects of Samhwang-tang(Sanhuang-tang) on Obese Rats Induced by High Fat Diet, MSG and Ovariectomy (삼황탕(三黃湯)이 고지방식이, MSG 및 난소적출로 유발된 흰쥐의 비만에 미치는 영향)

  • Objectives : Obesity is a metabolic disease resulted from various causes including nutritional disproportion, hormonal imbalance, and hypothalamic nuclei disorder. Samhwang-tang(Sanhuang-tang, SHT) is composed of 5 herbs (Ephedrae Herba, Scutellariae Radix, Typhae pollen, Acori Rhizoma and Ansu Semen) and is frequently used for obese patients. This study investigated anti-obesity effects of SHT on the rats. Methods : Obesity was induced by high fat diet (HFD), monosodium L-glutamate injection (MSG) and ovariectomy (OVX) in the Sprague-Dawley rats. Water extract of SHT was treated for 2 weeks, then changes of body weight gain and epididymal fat cell size were measured. In OVX animals, c-Fos and neuropeptide Y (NPY) expressions in hypothalamic nuclei were observed with immunohistochemistry. Results : SHT treatment demonstrated significant decrease of body weight gain in obesity induced by HFD and OVX, but not in obesity induced by MSG. SHT treatment demonstrated significant decrease of fat cell size in obesity induced by HFD, but not in obesity induced by MSG. On c-Fos and NPY expressions in hypothalamic nuclei, SHT treatment resulted significant decrease of immuno-reactivity of c-Fos and NPY in obesity induced by OVX. Conclusions : These results suggest that SHT has a weight loss effect against obesity induced by nutritional disproportion of diet intake and hormonal imbalance, especially without central nervous system disorder.

  • Objective : In order to investigate the effects of Soongijeseub-bang(here in after referred to SJB) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high fat diet Methods : C57BL/6 mice were divided into three groups(normal, high fat diet with control, high fat diet with SJB extract) and fed for 15 weeks. And measured body weight change, the weight change of the adipocytes in body, the levels of ALT, AST, creatinine, total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, NEFA(non-esterified fat acid), glucose in serum and the expression of ${\beta}3AR$, leptin, and TNF- ${\alpha}$ gene in primary adipocytes and adipocytes tissue. Results : The following results were obtained in this study. 1. SJB 500 mg/kg extract group showed significant decrease in final weight. 2. SJB 500 mg/kg extract group showed significant decrease in the amount of adipocyte in weight. 3. All experimental group showed significant decrease in ALT and AST levels. 4. All experimetal group did not show significant decrease in glucose and creatinine levels. 5. All experimental group showed significant decrease in total cholesterol, LDL-cholesterol, triglyceride and NEFA levels. HDL-cholesterol were increase significantly in SJB 500 mg/kg extract group. 6. SJB 500 mg/kg extract group showed significant decrease in leptin levels. 7. All experimental group showed significantly increased expressions of fJ 3AR in primary adipose cell and 3T3-Ll cell, and those of leptin in primary adipose cell and 3T3-Ll cell were decreased significantly. SJB $100{\mu}l/ml$ extract group showed significant decrease in the expression of TNF-${\alpha}$. 8. SJB 500 mg/kg extract group showed decrease in the size of adipocyte in adipocytes tissue. 9. All experimental group showed significant decrease in adipose vacuoles in liver tissue.

  • In Oriental medicine, there has been a theory that the deficiency of the Qi(氣) and the Phlegm(濕)-Damp(痰) bring Obesity. And so a clinically representative herb-medicine of the obesity treatments is Chegameyiin-tang . We observed the effects of Chegameyiin-tang on the fat tissues and what the function of Chegameyiin-tang is. These experimental studies were designed to investigate the effects of Chegameyiin-tang on the weight and lipid metabolism of obesity rats induced by high fat diet. And what is changed in the blood and how the leptin and uncoupled protein is affected. The measurement has been performed on (1) the weight of obese rats fed high fat diet, (2) the average size and number of epididymal fat cells, (3) the total cholesterol, triglyceride, glucose. and free fatty acids in the blood. and (4) the leptin and uncoupled protein in the blood are observed. The results are as follows; 1. In the sample group, the weight decrease occured significantly throughout the whole research period than that of control group. 2. In the sample group, epididymal fat weight showed significantly decrease in the 8th and 14th weeks than that of control group.3. In the sample group, epididymal fat cell size was decreased significantly in the 8th and 14th weeks than that of control group. 4. In the sample group, total cholesterol. triglyceride and glucose increased rather than control group in 8 weeks, those decreased significantly in 14 weeks. 5. In the sample group, free fatty acids and insulin increased rather than control group in 8 weeks. those showed some decrease in 14 weeks . 6. In the sample group, leptin decreased significantly than control group in 8, 14 weeks. Uncoupled protein showed some decrease in 8 weeks. that decreased significantly in 14 weeks.

Long Term Feeding with Soy Isoflavone and L-Carnitine Synergistically Suppresses Body Weight Gain and Adiposity in High-Fat Diet Induced Obese Mice

  • Objective: We investigated the efficacy of a 12-week supplementation of soy isoflavone with L-carnitine on the development of obesity in high fat-induced obese C57BL/6J mice, which are known as a good model of diet-induced obesity. Methods: We measured body weights, adipose tissue mass, serum/liver lipid profiles and fat cell size/number in C57BL/6J mice fed diets containing either low fat (4%) or high fat (35%), or high fat supplemented with soy isoflavone powder containing 10% isoflavone and L-camitine for 12 weeks. Results: Body weight gain, abdominal adipose tissue and liver weight were lower by 31% 78% and 31.4% respectively, in mice on high fat diet containing soy isoflavone+L-carnitine (SC mixture) compared with high fat diet group. Also, SC mixture improved serum lipid profiles such as total cholesterol (TC), triglycerides (TG), and liver lipid profiles such as total lipids and TG. As subsequent results, this SC mixture prevented high-fat diet from accumulating TG in the liver. The size of fat cell was also significantly decreased in SC mixture fed mice. At the end point of this experiment, our results showed that feeding with soy isoflavone for 12 weeks finally increased camitine palmitoyltransferase 1 (CPT 1) activity through elevating the level of CPT1 expression. Conclusions: This study suggests that long-tenn supplementation with dietary soy isoflavone and L-carnitine is more synergistically beneficial for the suppression of high-fat diet induced obesity by inhibiting liver TG accumulation and the gain in abdominal adipose tissue weight than that with soy isoflavone. The antiobesity effects of SC mixture might be attributed, at least in part, to the induction of fatty acid catabolism by soy isoflavone, genistein.

Effect of Herbal Chokong Pill on the Microstructure of Liver Cell in Rats Fed High Fat Diet (고지방식이 섭식 횐쥐의 간조직에 미치는 한방초콩환의 영향)

  • The purpose of this study is to investigate the effects of the herbal Chokong pill (hereafter HCKP) on the liver cell and enzyme activities of serum in rat. HCKP were mixed with pickled black soybeans and five different kinds of medicinal herbs (Rhynchosia nulubilis, Glycyrrhiza uralensis, Zizyphus vulgaris, Atractylodes macrocephala K., Astragalus membranaceus and Cornus officinalis). Four groups of male Sprague-Dawley rats were fed by different diets for 9 weeks: normal diet (Nor), high-fat diet (HF), high-fat diet supplemented with 1% (T1) and 5% (T5) HCKP powder, respectively. Depending on the presence of HCKP in high fat diet, the activities of the blood serum GOT and GPT were decreased. GOT and GPT activities of T1 and T5 were decreased 6.1%, 17.8% and 25.4%, 32.4% compared with HF. On microstructure observing through the transmission electron microscope (TEM) of liver cell, in normal group, a normal large and clear nucleus, rough endoplasmic reticulum (RER) and mitochondria possessing well-defined double outer-limiting membranes were found. However, in HF, it was hard to observe the microstructures in cytoplasm, because of too many fat granules. It showed severely damaged cell, pyknotic nucleus, swollen disintegrating RER and mitochontria loosing the cristae. In T1, there were more repaired liver cells and less fat granules than HF. In T5, there were much less numbers and smaller size of the fat granules than T1, and the morphology was similar to normal cell.

Effects of Rhizoma Arisaematis on Lipid and Glucose Metabolism and Histochemical Changes of Rats Fed High Fat Diet (천남성이 고지방식이로 유발된 비만 흰쥐의 당지질대사 및 조직화학적 변화에 미치는 효과)

  • Objectives : This study was performed to investigate the effects of Rhizoma Arisaematis water extract on lipid and glucose metabolism and histochemical change of obese rats. Methods and materials : 10 rats were divided into normal, control and RA (Rhizoma Arisaematis) groups. We fed the control group a high-fat diet and administered normal saline for 8 weeks. We fed the experimental group of rats a high-fat diet and administered an extract of Rhizoma Arisaematis for 8 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. The groups were examined for effects on blood serum lipids, blood sugar, blood insulin concentration and epididymal fat cells. Results : 1. Serum total cholesterol, triglyceride, HDL-cholesterol and glucose of the RA group decreased compared with those of the control group. These decreased rates were significant(P<0.05). 2. Serum LDL-cholesterol, total lipid, free fatty acid and the average size of epididymal fat cells of the RA group decreased compared with those of the control group. These decreased rates were significant(p<0.01). Conclusions : These results suggest that RA may be used to prevent or cure the obesity iniduced by a high-fat diet.

  • The fat bodies of cabbage worm (Pieris rapae) and silk worm (Bomyx mori) during metamorphosis was comparatively studied by electron microscope. 1. Cell oranelles: Golgi apparatus were not observed in both species. It is observed that RER of cabbage worms initiate to degenerate in prepupa stage with complete degeneration at adult stage, while that of silk worms shows similar degenerative pattern. However, mitochondria of cabbage worms are transformed into autophagic vacuole from prepupa stage until adult stage whereas those of silk worm shows a decrease in number in prepupa stage but maintains a certain level until adult stage. 2. Storage substance in cell: Lipid droplets in cabbage worms were observed to increase in numbers during larval stage but afterward decrease in number with an enlargement in size. However immediately after their pupal stage, they almost disappear. On the contrary lipid droplets in silk worms show rather increase in number until adult stage. Protein storage granules in bothspecies were arised from autophagic vacuoles(lysosome) . Fat cells of cabbage worm in adult stage turn out to be residual bodies which last until final stage, but those of silk worm rapidly decrease. Glycogen particles in both species reach maximum at last larval instar and thee gradually decrease thereafter. 3. Fat body sheath: The average width of fat body sheath was measured to be $0.2{\mu}m$ and $0.6{\mu}m$ and surface of fat cells adjacent to fat body sheath in silk worm is heavily infolded.

Effects of Isoprothiolane administration on fat necrosis of Hanwoo(Korean Cattle) sire (한우 종모우의 지방괴사증에 대한 Isoprothiolane 투여효과)

  • The effect of isoprothiolane(di-isopropyl-1,3-dithiolan-2-ylidenemalonate) aganist fat necrosis in Hanwoo(Korean Cattle) sire was evaluated. The 10 heads of Hanwoo sire suffering from fat necrosis were given 50mg/kg body weight of isoprothiolane(0.2g/kg of Fujix, Japan) orally once a day for 8 weeks. In 30% of these, the size of the necrotic fat masses had decreased significantly 7 months after the adminstration. Isoprothiolane did not affect on live body weight and semen characteristics. However the sire affected with fat necrosis had higher MCHC(Mean corpuscular hemoglobin concentration) than normal sire in hematologic values 10 weeks after administration. Number of RBC(red blood cell) and PCV(packed cell volume) 10 weeks after administration had been increased than those before administration(p < 0.05). The serum concentrations of creatinine, triglyceride, and total cholesterol 10 weeks after administration were higher than those before administration while the concentration of glucose was vice versa. The isoprothiolane may reduce the oxidation of glucose, increase the glucose transfer to lipids, and increase blood supply to necrotic masses. These results indicate that isoprothiolane may be useful as the therapeutic agent aganist fat necrosis.

Effects of an aqueous extract of purple sweet potato on nonalcoholic fatty liver in high fat/cholesterol-fed mice (고지방/고콜레스테롤 식이를 섭취한 마우스에서 자색고구마 열수추출물 보충이 지방간 저항성에 미치는 영향)

  • Purpose: Anthocyanins from purple sweet potato (PSP) have been investigated in vitro and in animals and found to have a protective effect against oxidative hepatic damage. In this study, we investigated that aqueous extract of PSP can ameliorate the dysfunction of lipid metabolism in mice fed a high fat/cholesterol diet. Methods: Forty C57BL/6J mice were randomly divided into 5 groups (n = 8) and fed one of the following diets for 8 weeks; normal fat (NF) diet; high fat/cholesterol (HFC) diet; HFC with 1.25% PSP (HFPL) diet; HFC with 2.5% PSP (HFPM) diet; HFC with 5% PSP (HFPH) diet. Results: Non-alcoholic fatty liver was manifested in the HFC group by showing increased levels in plasma alanine aminotransferase (ALT) activity, total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C), increased level of TC and presence of many large lipid droplets in the liver, and increased fat cell size in the HFC group compared with the NF group. However, administration of HFC induced a significant decrease in food intake, resulting in decrease in fat mass. Co-administration of PSP did not lead to reversal of body weight changes, ALT activity, and lipid levels in plasma and the liver, but suppressed excess enlargement of the fat cell size through increasing carnitine palmitoyltransferase-1 (CPT-1) gene expression in the liver. Accordingly, the number of fat droplets in the liver was reduced in PSP administered groups. Conclusion: Taken together, these results suggest that PSP may have a protective effect on the dysfunction of lipid metabolism. Conduct of further studies on the coordinated regulation of PSP for lipid metabolic homeostasis at the liver-adipose tissue axis is needed.
