Effect of zinc supplementation on insulin resistance and metabolic risk factors in obese Korean women

  • Zinc deficiency is known to be associated with insulin resistance in obese individuals. This study was performed to evaluate the effect of zinc supplementation on insulin resistance and metabolic risk factors in obese Korean women. Forty obese women (body mass index (BMI) ${\geq}25kg/m^2$) aged 19-28 years were recruited for this study. Twenty women of the study group took 30 mg/day of supplemental zinc as zinc gluconate for 8 weeks and 20 women of control group took placebo. Usual dietary zinc intake was estimated from 3-day diet records. Insulin resistances were measured using Homeostasis model assessment (HOMA) indices, and insulin sensitivities Matsuda indices, which were calculated using oral glucose tolerance test data. Metabolic risk factors, such as waist circumference, blood pressure, fasting glucose, triglyceride, high density lipoprotein (HDL) cholesterol, and adipocyte hormones such as leptin, and adiponectin were also measured. At the beginning of study, dietary zinc averaged 7.31 mg/day and serum zinc averaged $12.98{\mu}mol/L$ in the study group. Zinc supplementation increased serum zinc by 15% and urinary zinc by 56% (P < 0.05). HOMA values tended to decrease and insulin sensitivity increased slightly in the study group, but not significantly so. BMI, waist circumference, blood pressure, blood glucose, triglyceride, HDL cholesterol, and adipocyte hormones did not change in either the study or control group. These results suggest that zinc status may not affect insulin resistance and metabolic risk factors in obese Korean women. Further research is required on a larger cohort with a longer follow-up to determine the effects of zinc status on insulin resistance and metabolic variables.

Validation of G-protein beta-3 subunit gene C825T polymorphism as predictor of obesogenic epidemics in overweight/obese Korean children

  • Purpose: We investigated the potential interaction between the G-protein beta-3 subunit gene (GNB3) C825T polymorphism, a risk factor for chronic disease in various ethnicities, and obesogenic environments in overweight/obese Korean children. Methods: The present study was conducted as a cross-sectional study using measures of anthropometry, blood pressure (BP), and fasting blood samples as well as 3-day food records. Subjects were recruited from seven elementary schools in an urban district in Seoul, South Korea, between 2007 and 2008. A total of 1,260 children aged 8-9 years were recruited in the study, including 633 boys (50.3%) and 627 girls (49.7%). Results: The allele frequencies of the GNB3 polymorphism were C allele = 49.7% and T allele = 50.3% in subjects. In general, boys with T allele had higher BMI, systolic BP (SBP), and triglycerides, although their energy intake was not significantly different from boys with C allele. In contrast to boys, girls with T allele had lower BMI but higher SBP and energy intake than those with C allele. The girls with T allele had a significantly lower BMI and waist circumference in both the normal weight group and obese group (OB). T allele carriers in both genders had significantly higher TC than C allele carriers in the OB group. At last, girls with T allele in OB appeared to have significantly lower HOMA-IR than those with C allele. Conclusion: Unlike higher risk for negative health outcomes by the GNB3 polymorphism in various ethnicities, GNB3 polymorphism did not influence obesogenic environments in overweight/obese Korean children.

Effects of Mushroom Supplementation on Blood Glucose Concentration, Lipid Profile, and Antioxidant Enzyme Activities in Patients with Type 2 Diabetes Mellitus (표고버섯 보충이 제 2 형 당뇨병 환자의 혈당, 지질 대사 및 항산화 효소 활성에 미치는 영향)

  • The present study was carried out to evaluate the physiological effects of mushroom supplementation on blood glucose levels, lipid profile, and antioxidant enzyme activities in subjects with type 2 diabetes mellitus. Subjects were randomized into either a control group or mushroom supplementation group. Mushroom supplementation was provided 3 times a day for 4 weeks. We found that total dietary fiber intake was about 2.5 times higher (30.3 g vs. 12.3 g) in subjects receiving mushroom supplementation than in the control group. Two groups maintained the same food intake and amount of activity, exercise during the supplementation. We observed no difference in age, height, weight, BMI (body mass index), blood pressure between the groups. Nutrient intake did not differ appreciably between the two groups, except for fiber intake, during the supplementation. Fasting blood glucose levels and 2-hour postprandial blood glucose levels were significantly lower in those ingesting mushroom than in controls. Furthermore, the concentrations of low-density lipoprotein cholesterol were decreased significantly in the mushroom supplementation group. Small changes were observed in the concentration of total cholesterol, triglyceride, high-density lipoprotein cholesterol of those supplemented with mushroom, but these changes were not statistically significant. Activities of superoxide dismutase and catalase with mushroom supplementation were higher than in controls, but and glutathione peroxidase activity was not affected. The levels of thiobarbituric acid reactive substance of mushroom group were lower than control group, but were not significant. We conclude that addition of mushroom influences glycemic control and may be effective in lowering blood lipids and improving antioxidant enzyme activities. Accordingly, such effects may reduce risk factors for cardiovascular disease in patients with type 2 diabetes. However, to confirm these effects and to make dietary recommendations for patients with type 2 diabetes, further studies are necessary.

Both sitagliptin analogue & pioglitazone preserve the β-cell proportion in the islets with different mechanism in non-obese and obese diabetic mice

  • In this study, the effects of sitagliptin analogue (SITA) or pioglitazone (PIO) treatment on glucose homeostasis and ${\beta}$-cell dynamics in animal models of type 2 diabetes-Akita and db/db mice were evaluated. After 4-6 weeks of treatment, both SITA and PIO were shown to lower non-fasting glucose levels and reduced glycemic excursion in the intraperitoneal glucose tolerance test. In addition, both drugs preserved normal islet structure and the proportion of ${\beta}$-cells in the islets. Compared to the controls, SITA treatment induced a higher ${\beta}$-cell proliferation rate in Akita mice and a lower rate of apoptosis in db/db mice, whereas PIO treatment induced a lower rate of apoptosis in db/db mice and reduced proliferation rates in Akita mice. In conclusion, both SITA and PIO appear to exert some beneficial effects on the islet structure in addition to glycemic control via different mechanisms that involve ${\beta}$-cell dynamics in Akita and db/db mice.

Association between Dietary Acid Load and Insulin Resistance: Tehran Lipid and Glucose Study

  • In the current study, we investigated the longitudinal association between dietary acid load and the risk of insulin resistance (IR) in the Tehranian adult population. This longitudinal study was conducted on 925 participants, aged 22~80 years old, in the framework of the third (2006~2008) and fourth (2009~2011) phases of the Tehran Lipid and Glucose Study. At baseline, the dietary intake of subjects was assessed using a validated semi-quantitative food frequency questionnaire, and the potential renal acid load (PRAL) and net endogenous acid production (NEAP) scores were calculated at baseline. Fasting serum insulin and glucose were measured at baseline and again after a 3-year of follow-up; IR was defined according to optimal cut-off values. Multiple logistic regression models were used to estimate the risk of IR according to the PRAL and NEAP quartile categories. Mean age and body mass index of the participants were 40.3 years old of $26.4kg/m^2$, respectively. Mean PRAL and NEAP scores were -11.2 and 35.6 mEq/d, respectively. After adjustment for potential confounders, compared to the lowest quartile of PRAL and NEAP, the highest quartile was accompanied with increased risk of IR [odds ratio (OR)=2.81, 95% confidence interval (CI)=1.32~5.97 and OR=2.18, 95% CI=1.03~4.61, respectively]. Our findings suggest that higher acidic dietary acid-base load, defined by higher PRAL and NEAP scores, may be a risk factor for the development of IR and related metabolic disorders.

An Analysis on Risk Factors of Colon Polyps with Health Examination Examinees (건강검진 대상자에서 대장용종의 위험요인 분석)

  • This study aimed to analyze the occurrence of colon polyps and also risk factors of colon polyps. Among health examination examinees, 508 people were included who underwent colonoscopy and abdominal ultrasonography at the same time. Physical measurements also performed such as height, weight, blood pressure, waist measurement and BMI. General characteristics including age, sex, smoking, drinking and exercise as a risk factors were checked. At the same time, we analyzed various blood tests and fasting blood sugar through blood-gathering. In the results, regarding risk factors of colon polyps, AST, ALT, TC, fatty liver, obesity and smoking were shown significantly high. As a dependent variable with colon polyps, the highest relation was fatty liver(3.4 times), and then T-C(2.3 times). Smoking factor showed 2.3 times higher relation than non-smokers.

Incubation Routine and Sex Role of Streaked Shearwaters Calonectris leucomelas at Sasudo Island, South Korea (사수도에서 번식하는 슴새의 포란 일정 및 성 역할)

  • Incubation routine and sex role of Streaked Shearwaters Calonectris leucomelas at Sasudo Island, in Jeju, South Korea, were studied during the incubation period, June to August in 2002. Incubation routine in Procellariiformes represents a sequence of alternating shifts taken in turn by female and male in a species-specific pattern. Hence, coordination of individual incubation rhythms between partners is crucial for successful breeding attempt. In Streaked Shearwaters, incubation routine represents a sequence of alternating shifts taken in turn by male and female. The first incubation shift was made by male after female had laid the egg. The mean incubation period was 50.8 days until hatching. Males had spent on average 26.5 days incubating and females 24.3 days accordingly. The mean duration of incubation shifts decreased progressively from 6th and 7th shift to hatching. Overall, males had spent more time incubating than females during the incubation period, but the mean duration of the incubation shift 5.6 days for males and 5.7 days for females did not differ between males and females. There were no effect of the body size of the breeding pair on incubation performance. For males the mean of body weight decreased during the incubation, whereas for females it remained approximately stable. In Streaked Shearwaters, the duration of incubation shift and subsequent foraging trip are related to loss of body weight during the period of fasting. In addition, coordination of individual incubation rhythms affects their incubation behaviour.

The Relationship between Serum Cholesterol Level and Dietary Intake in Obese Children (비만 아동의 식이 섭취 양상과 혈청 콜레스테롤 수준과의 상관성)

  • To assess the relationship between serum cholesterol level and dietary intake in obese children, 50 subjects were selected as an obese group and 50 subjects as a normal group depending upon body mass index. Fasting blood samples were obtained and analyzed for serum total cholesterol(TC), high density lipoprotein cholesterol(HDLC), low density lipoprotein cholesterol(LDLC) and triglyceride(TG). The dietary intake and information on dietary behavior and family environment were obtained by questionaire using 24 hour dietary recall method for 2-consecutive days. The concentrations of TC, LDLC, HDLC and TG of obese group were estimated to be 193.3mg/dl, 122.5mg/dl, 53.7mg/dl and 86.0mg/dl, respectively. The obese group showed higher TC, LDLC, TG level(p<0.001) and lower HDLC level(p<0.001) than normal group. Food intakes in obese group were similar to those of normal group except meats & fishes(P<0.01), milk & dairy products, fruits(p<0.05), and green & yellow vegetables(p<0.001). Nutrient intakes in obese group exceeded Korean recommended dietary allowance for the corresponding ages. Saturated fatty acid(p<0.05) and cholesterol(p<0.001) intakes in obese group were higher than those of normal group. In correlation analysis, meats & fishes, eggs and milk & dairy products intakes were positively related to serum TC, LDLC and TG ; on the contrary, green & yellow vegetables and fruits intakes were negatively related to serum TC, LDLC and TG ; but positively to HDLC. Obesity index, serum cholesterol level, cholesterol and saturated fatty acid intake were positively correlated with parent's obesity index, frequency of snack per day, eating rate and late snack.

  • Mean values for the fasting serum levels of the individual amino acids are given for 34 adult males and 35 adult females, ranging in age from 20 to 39 years. Threonine plus glutamine, alanine and lysine accounted for about 40% of total amino acids, and total essential amino acids was significantly lower than total nonessential amino acids. There were highly significant differences between men and women in the mean serum amino acid levels. Men had significantly .higher levels than women for most of the amino acids except lysine and histidine. Total essential amino acids, and the molar ratios of total essential to total nonessential amino acids and total amino acids showed significantly higher levels in men than in women. The molar ratios of each essential amino acid to total essential amino acids were also examined.

Plasma Gastrin Concentraion after Ingestion of Sesame Gruel in Normal Human Subjects (깨죽이 정상 성인의 혈장 Gastrin 농도에 미치는 영향)

  • This study was conducted to investigate the effect of ingestion of sesame (Sesamum indicum) gruel as a nourishing meal upon the plasma gastrin concentration in normal Korean. Sixteen normal persons with no history of gastrointestinal diseases, including male and female were studied. After an overnight(about 15 hrs) fast, eight persons(mean age: 26.6, range: $20{\sim}40$ years) of them ingested a 350 ml sesame gruel corresponding to 12 g protein, 13 g fat and 99 g carbohydrate, and the remaining 8 subjects(mean age: 21.3, range: $20{\sim}24$ years) ingested a 350 ml glutinous rice gruel(control meal) corresponding to 8 g protein, 1 g fat and 115 g carbohydrate. The venous blood samples were drawn before and after the ingestion of the test meal for the measurement of gastrin by means of radioimmunoassay. 1) Plasma gastrin concentration in response to the ingestion of sesame gruel or glutinous rice gruel increased significantly compared with the concentration in fasting state. 2) Mean increment or percent increment in postprandial plasma gastrin concentration after the ingestion of sesame gruel was not significantly different from that after the control meal, i.e. the glutinous rice gruel. It is inferred from the above results that the ingestion of sesame contained in sesame gruel may have no significant influence on gastrin release in normal human subjects.
