Association of β3-Adrenergic Receptor Polymorphisms and High-Density Lipoprotein Cholesterol (고밀도 지단백 콜레스테롤과 베타 3-아드레날린성 수용체 유전자 변이와의 관련성)

  • The ${\beta}$3-adrenergic receptor (ADRB3) is expressed mainly in visceral adipose tissue and is thought to contribute to lipolysis and the delivery of free fatty acids to the portal vein. This study was aimed at evaluating the association between high-density lipoprotein cholesterol (HDL-C) and ADRB3 genetic polymophisms. A total of 991 healthy examinees who were examined in a university hospital, located in Busan City, between May and December 2006 were enrolled in this study. Height, weight, body mass index, waist circumference, and systolic and diastolic blood pressures of the subjects were examined. Intravenous concentrations of fasting blood glucose, total cholesterol, HDL-C, low-density lipoprotein cholesterol, and triglyceride were also measured. After extracting DNA from the subjects, mutations of the +188T>C (Trp64Arg) of exon 1 and +3893T>C of intron 2 on the ADRB3 gene were genotyped using the single base extension method. We have identified a novel mutation of ADRB3 that is located in intron 2. The frequency of its minor allele was 0.164. Both the +188T>C mutation of exon 1 and +3893T>C mutation of intron 2 were significantly associated with HDL-C. The mean concentration of serum HDL-C was significantly lower in the presence of their minor allele 'C'. These results suggest that both mutations of +188T>C of exon 1 and +3895T>C of intron 2 have significant associations with HDL-C in the Korean population.

Prevalence of Diabetes Mellitus and Associated Diseases in Yeungnam Province Area (영남지방에서의 당뇨병 유병율과 이에 관련돈 질환의 빈도에 관한 연구)

  • To know the prevalence of the diabetes mellitus and associated diseases, we analysed the data of the 3,088 subjects who were examined with the Computed Automated Medi-Screening Test System which consisted of 65 parameters including blood glucose determination fasting and one hour after 100g of oral glucose load. We grouped the subjects by the modified criteria of National Diabetic Data Group. Followings are the results of the various analysis : 1. The prevalence of diabetes mellitus and impaired glucose tolerance is 2.27% and 18.26% respectively. 2. The prevalence of diabetes mellitus is 2.63% In male and 1.66% in female. There is no statistically significant difference between male and female. 3. There is tendency of increasing prevalence of diabetes mellitus as the age increases. From second to eighth decade, the prevalence of diabetes mellitus Increases as 0.0, 0.45, 0.67, 2.28, 3.47, 5.36, 10.00% respectively. 4. There is no statistically significant difference of prevalence of obesity between normal and diabetes: that is, 18.03%, 22.86% respectively.($P{\geq}0.1$) 5. There is no statistically significant difference of prevalence of impaired glucose tolerance and diabetes between non-obese and obses group. ($P{\geq}0.1$) 6. There is statistically significant increases of frequency of proteinuria, azotemia, hypertension as the glucose tolerance decreases. ($P{\leq}0.05$)

Analysis on the perception of body image and awareness of diet among high school students in Zibo city, China (중국 치박시(淄博市) 고등학생의 체형에 대한 인식 및 다이어트에 대한 태도 분석)

  • This study investigated the self-perception of body image and awareness of diet of high school students in Zibo city, China. The data were collected from 260 high school students through the self-administered questionnaires. Frequency analysis, t tests, one-way ANOVA, Duncan's multiple range tests, factor analysis and Cronbach's alpha test were conducted by SPSS Windows V.19.0. The results were as follows. In the self-perception of body image, two fifths of the respondents thought of themselves as standard body image. More female respondents thought of themselves as fat persons compared with male respondents. The level of concern about diet was generally low and the mean for pursuit of skinny figure was highest in the awareness of diet. The percentage of correct answers of knowledge about diet was not that high, and one fifths of respondents had experience of diet. The respondents practiced fasting therapy, exercise therapy, food therapy, sweat therapy, drug/appliance therapy, and therapy using professional organization in order for diet.

A Study on Annual Cyclical Rituals and Medicine in the late Joseon Dynasty - With focus on Uibanghappyeon 'Wolhaeng' - (조선후기 세시풍속과 의학 - "의방합편(醫方合編)" 월행(月行)을 중심으로 -)

  • Objective : This study considered the annual cyclical rituals related to medicine in an effort to look into the general medical conditions in the late Joseon Dynasty. Method : This study selected Uibanghappyeon 'Wolhaeng' as its research material in that it reflects the medical conditions at that time as a writing in the late Joseon Dynasty, and it's a record about annual cyclical rituals included in a medical book. The annual cyclical rituals are possible to analyze from various angles, but this study perceives the fundamental elements forming the annual rituals as (1) the time when a ritual was held (2) purpose, (3) method, and analyzed the calendaric rituals by dividing them according to the three elements. Result : There appear 97 sorts of annual rituals period in all according to their contents in Uibanghappyeon 'Wolhaeng.' (1) the case of directly suggesting a specific day like May 5 was found to be the most with 46 cases on the basis of the character of the implemented time, followed by the part (16 cases) with the sexagenary cycle like the Day of the Ox. (2) From the aspect of the purpose, the case of aiming at preventing diseases was found to be the most with 24 cases, and the case of expressing 'having to do like that' without any clear reason, or 'that would be better' was found to be the third-most 19 cases. In a broad sense, should these two be perceived as disease prevention, these statistics accounted for a half of the whole with 43 cases. Meanwhile, the style of rituals whose purpose was disease treatment(22 times) was to make a medicine on a specific day. Among the 22 sorts related to disease treatment, 16 sorts were the custom of making medicine on a designated day. (3) When classified from the aspect of methods, they can be divided into pharmaceutical preparations(23 cases), specific behavior(20cases), taboo(19 cases), taking medicine(15 cases), fasting(11 cases) and external medical treatment(9 cases), etc. This shows a comparatively even distribution rather than the case of dividing the annual rituals into a period or purpose. Conclusion : Medicine is the knowledge for preventing and treating diseases. However, the medical science and medical treatment of a traditional society show not a little difference from what we feel. The research on the annual cyclical rituals related to medicine is expected to be helpful to our perception of such a difference.

Effects of Dietary Supplementation of Magnesium Hydrogen Phosphate (MgHPO4) as an Alternative Phosphorus Source on Growth and Feed Utilization of Juvenile Far Eastern Catfish (Silurus asotus)

  • The present study was conducted to investigate a supplemental effect of magnesium hydrogen phosphate (MHP, $MgHPO_4$) as an alternative phosphorus (P) source on growth and feed utilization of juvenile far eastern catfish (Silurus asotus) in comparison with three conventional P additives (monocalcium phosphate (MCP), dicalcium phosphate (DCP) and tricalcium phosphate [TCP]) as positive controls. A basal diet as a negative control was prepared without P supplementation and four supplemental P sources were added at the level of 2%. Five groups of 450 fish having mean body weight of 11.3 g following 24 h fasting after three week adaptation period were randomly distributed into each of 15 tanks (30 fish/tank). Fish were hand-fed to apparent satiety twice a day for 8 weeks. Fish fed MHP had weight gain (WG), protein efficiency ratio and specific growth rate comparable to those fed MCP. Fish fed MHP and MCP had feed efficiency (FE) significantly higher (p<0.05) than those fed DCP. Fish groups fed control and TCP showed the lower FE than the other groups which was significantly different (p<0.05) from those of fish fed the other diets. Survival rate was not significantly different (p>0.05) among treatments. Fish fed control had the lowest hematocrit, which was significantly different (p<0.05) from that of fish fed MHP. Fish fed MCP and MHP had plasma P higher (p<0.05) than fish fed the other diets. Relative efficiencies of MCP, DCP and TCP to MHP were found to be 100.5 and 101.3%, 92.0 and 91.6%, and 79.1 and 80.9% for WG and FE, respectively. P availability was determined to be 88.1%, 75.2%, 8.7%, and 90.9% for MCP, DCP, TCP, and MHP, respectively. Consequently, MHP recovered from wastewater stream showed that as an alternative P source its performance was comparative with MCP on growth and feed utilization of juvenile far eastern catfish.

Hypotriglyceridemic effects of brown seaweed consumption via regulation of bile acid excretion and hepatic lipogenesis in high fat diet-induced obese mice

  • BACKGROUND/OBJECTIVES: The present study aimed to further investigate the potential health beneficial effects of long-term seaweed supplementation on lipid metabolism and hepatic functions in DIO mice. MATERIALS/METHODS: Four brown seaweeds (Undaria pinnatifida [UP], Laminaria japonica [LJ], Sargassum fulvellum [SF], or Hizikia fusiforme [HF]) were added to a high fat diet (HFD) at a 5% ratio and supplemented to C57BL/6N mice for 16 weeks. Triglycerides (TGs) and total cholesterol (TC) in the liver, feces, and plasma were measured. Fecal bile acid (BA) levels in feces were monitored. Hepatic insulin signaling- and lipogenesis-related proteins were evaluated by Western blot analysis. RESULTS: Fasting blood glucose levels were significantly reduced in the LJ, SF, and HF groups compared to the HFD group by the end of 16-week feeding period. Plasma TG levels and hepatic lipid accumulation were significantly reduced in all 4 seaweed supplemented groups, whereas plasma TC levels were only suppressed in the UP and HF groups compared to the HFD group. Fecal BA levels were significantly elevated by UP, LJ, and SF supplementation compared to HFD feeding only. Lastly, regarding hepatic insulin signaling-related proteins, phosphorylation of 5'-AMP-activated protein kinase was significantly up-regulated by all 4 types of seaweed, whereas phosphorylation of protein kinase B was up-regulated only in the SF and HF groups. Lipogenesis-related proteins in the liver were effectively down-regulated by HF supplementation in DIO mice. CONCLUSIONS: Brown seaweed consumption showed hypotriglyceridemic effects in the prolonged DIO mouse model. Specifically, combinatory regulation of BA excretion and lipogenesis-related proteins in the liver by seaweed supplementation contributed to the reduction of plasma and hepatic TG levels, which inhibited hyperglycemia in DIO mice. Thus, the discrepant and species-specific functions of brown seaweeds provide novel insights for the selection of future targets for therapeutic agents.

Effects of a Rice-Based Diet on Body Weight and Serum Lipid Levels in Mice (밥 중심 식사가 마우스의 체중 및 혈중 지질 함량에 미치는 영향)

  • This study was designed to evaluate the effect of a rice-based diet on body weight and serum lipid levels. Thirty male C57BL/6 mice were randomly divided into three groups and fed a high-fat diet (HFD), rice-based diet (RD), or bread-based diet (BD) for five weeks. Our results showed that the RD group had a significantly lower final body weight, although there was no significant difference in daily food intake among the groups. There was a reduction in body weight gains, adipose tissues weight, and adipocyte size of epididymal adipose tissue in the RD group (p<0.05). Also, serum triglycerides and total cholesterol in the RD group were significantly decreased compared to the BD group. RD also reduced fasting glucose and insulin levels. These results indicate that the intake of RD attenuates increases in body weight and serum lipid concentrations in mice, suggesting its benefits for the dietary control of obesity and hyperlipidemia.

Expression and Significance of ER, PR, VEGF, CA15-3, CA125 and CEA in Judging the Prognosis of Breast Cancer

  • Objective: To explore the expression and significance of estrogen receptor (ER), progestrone receptor (PR), vascular endothelial growth factor (VEGF), CA15-3, CA125 and carcinoma embryonic antigen (CEA) expression in judging the prognosis of breast cancer. Materials and Methods: Sixty-five patients with breast cancer undergoing operations in the general surgery department were considered as the observation group, and 50 healthy outpatients of our hospital as the control group. Cubital venous blood was drawn in the morning from fasting patients in the two groups and chemiluminescence immunoassays were used to detect the levels of CA15-3, CA125 and CEA in serum. The follow-up duration was from 4 months to 2 years, and change in levels of the indicators was detected by dynamically drawing blood. After surgery, cancer tissue samples of patients in observation group remained on file (the non-recurrent patients were biopsied). Immunohistochemistry was applied to determine the expression of ER, PR and VEGF in tissue. Results: The effective rate of 12 patients with negative ER and PR expression was 33.3% in the observation group, being associated with prognosis to varying extents. Serum CA15-3, CA125 and CEA in the observation group were all significantly higher than in control group (p<0.01). With increase in pathological staging, levels of serum CA15-3, CA125 and CEA gradually increased (p<0.01). Levels in patients with lymph node metastasis were markedly higher than in those without (p<0.01). In addition, values with distal lymph node metastasis were notably higher than with adjacent lymph node metastasis (p<0.01). The postoperative follow-up results revealed that positive VEGF and levels of serum VEGF, CA15-3, CA125 and CEA in recurrence group were obviously higher than in non-recurrence group (p<0.01). Conclusions: Joint detection of ER and PR expression as well as levels of serum VEGF, CA15-3, CA125 and CEA is meaningful and can guide the diagnosis and treatment for breast cancer.

Determination of plasma ketone body following oximation-trimethylsily| derivatization using gas chromatography-mass spectrometry selected ion monitoring (혈장 중 케톤체의 옥심-TMS 유도체화 후 GC-MS/SIM을 이용한 분석)

  • A ketone body (acetoacetic acid, β-hydroxybutyric acid, and acetone) increases from blood or urine when bio-energy dependence pays more fatty acid than glucose. However, in case oxidation of fat is greater than the capacity of the citric acid cycle the fatty acid oxidation is made from acetoacetyl CoA to acetoacetate then, again form β-hydroxyburytic acid to acetone, the diffusion take place into the blood. Enzymes that oxidize ketone body in the brain and nerve tissue blood ketone dody is increased during prolonged fasting, brain used it as energy. In this study, we developed the rapid two step derivatization method for sensitive detection of the ketone body by GC-MS/SIM. The plasma was deproteinized and then the hydroxy and carboxyl groups of ketone body are subjected to extraction and drying then, keto-group were derivatized with hydoxylamine at 60℃ for 30 min for oximation. Then it was trimetyl-silylated with BSTFA at 80℃ for 30 min and analyzed using a GC-MS. The linear ranges were in between 0.001 μg/mL and 250 μg/mL for β-hydroxy butyrate, and acetoacetate. The method detection limits were below 0.1 pg over each target compound determined. The mean recoveries (%) of target compounds were ranged from 88.2 % to 92.3 % at 1 µg/mL, from 89.5 % to 94.8 % at 10 μg/mL, with RSD of 6.3-9.4 %. This method could be applied to quantification of ketone bodies which are seen in the keto-acidosis in children and adults from a variety of diseases that cause ketones in the blood and urine.

Garlic powder intake and cardiovascular risk factors: a meta-analysis of randomized controlled clinical trials

  • BACKGROUND/OBJECTIVES: Although preclinical studies suggest that garlic has potential preventive effects on cardiovascular disease (CVD) risk factors, clinical trials and reports from systematic reviews or meta-analyses present inconsistent results. The contradiction might be attributed to variations in the manufacturing process that can markedly influence the composition of garlic products. To investigate this issue further, we performed a meta-analysis of the effects of garlic powder on CVD risk factors. MATERIALS/METHODS: We searched PubMed, Cochrane, Science Direct and EMBASE through May 2014. A random-effects meta-analysis was performed on 22 trials reporting total cholesterol (TC), 17 trials reporting LDL cholesterol (LDL-C), 18 trials reporting HDL cholesterol (HDL-C), 4 trials reporting fasting blood glucose (FBG), 9 trials reporting systolic blood pressure (SBP) and 10 trials reporting diastolic blood pressure (DBP). RESULTS: The overall garlic powder intake significantly reduced blood TC and LDL-C by -0.41 mmol/L (95% confidence interval [CI], -0.69, -0.12) (-15.83 mg/dL [95% CI, -26.64, -4.63]) and -0.21 mmol/L (95% CI, -0.40, -0.03) (-8.11 mg/dL [95% CI, -15.44, -1.16]), respectively. The mean difference in the reduction of FBG levels was -0.96 mmol/L (95% CI, -1.91, -0.01) (-17.30 mg/dL [95% CI, -34.41, -0.18]). Evidence for SBP and DBP reduction in the garlic supplementation group was also demonstrated by decreases of -4.34 mmHg (95% CI, -8.38, -0.29) and -2.36 mmHg (95% CI, -4.56, -0.15), respectively. CONCLUSIONS: This meta-analysis provides consistent evidence that garlic powder intake reduces the CVD risk factors of TC, LDL-C, FBG and BP.
