Chromosomes of gonadal tissues from Fasciola spp, Eurytrema pancreaticum and Calicophoron calicophorum occurred Korean cattle were egamined using modified air-drying method. To compare their phenotype with three different genotypes among Fasciola spp, the adult and egg si2e were measured since they have been known as important taxonomical characters. The results obtained were as followed; Cattle liver fluke, Fasciola spp were classified into three types based on their chromosomal complements such as individual with 2o chromosome(diploid), 30 chromosome(triploid) and 20/30 mosaic constitution(mixoploid). The propotions of appearance of three types were 40.00%, 54.29% and 5.71%, respectively. The frequency of three types in type I which was regarded as F gigantica were 58.82% for diploid, 35.29% for triploid and 5.88% for mixoploid, but in type II which was regarded as F hepatica were 72.2% for triploid, 22.22% for diploid and 5.56% for mixoploid. Egg length of triploid forms was significantly larger than that of diploid forms and egg size of mixoploid forms was similiar to that of triploid forms. Worm size of triploid forms was larger than that of diploid forms and was more similar to that of mixoploid forms, but the statistical data were not significant. Diploid chromosome consisted of one pair of metacentric chromosome(m), four pairs of submetacentric chromosomes(sm), five pairs of subtelocentric chromosomes(st), while triploid chromosome consisted of one pair of metacentric chromosome, seven pairs of submetacen.tric chromosomes, one pair of subtelocentric chromosome and telocentric chromosome(t), respectively. In mixoploid chromosome, constitution of the chromosomes of diploid or triploid cell was consistent with that of diploid or triploid. Chromosomes of gonadal tissues from pancreatic fluke, Eurytrema pancreaticum consisted of 13 pairs of homologs(2n=26, n=13). The mitotic and meiotic divisions were observed frequently. In the mitotic metaphase, Karyotype consisted of five pairs of metacentric chromosomes, four pairs of submetacentric chromosomes, three pairs of subtelocentric chromosomes and one pair of telocentric chromosome. Chromosomes of gonadal tissues from stomach fluke, Calicvphoron calicophorum consisted of 9 pairs of homologs(2n=18, n=9). The meiotic divisions was frequently observed, but mitotic divisions was rare. In the mitotic metaphase, Karyotype consisted of two pairs of metacentric chromosomes, three pairs of submetacentric chromosomes and four pairs of subtelocentric chromosomes. Karyotype of Calicophoron calicophorum differed from that of Japanese C calicophorum which was similar to that of Paramphistomum cervi of Korean cattle. Though that of Calicophoron calicophorum of Korean cattle was similar to that of Paramphistomum explanatum of Korean cattle, that have been recognized to be a different species of fluke.
In order to obtain the most specific and sensitive antigen from crude antigens of Fasciola hepatica for the immunodiagnosis of bovine fascioliasis by the enzyme linked immunosorbent assay (ELISA), phosphate buffered saline extract of F. hepatica was prepared. The crude extract was fractionated into 7 antigens using sephadex G-100 column chromatography. Seven fractionated antigens were applied to ELISA, precipitation test and intradermal test, respectively. Results obtained are as follows: 1. The specificity (95% confidence interval in negative sera of bovine fascioliasis; Mean+$2{\times}SD$ of absorbence) of the first (MW>150,000) and the second antigens(MW 120,000) were 93.7%, but those of others including crude antigen showed 100%. 2. The sensitivity(positive sera of bovine fascioliasis having higher values with compared to the criterion) of the first, the sixth (MW 16,000) and the seventh antigen (MW<5,000) were 91.6%, 87.5% and 0%, respectively, but those of others showed all 100%. 3. The absorbance by ELISA using the fifth antigen (MW 26,000) was 8. 43-folds higher in the positive sera than that in the negative sera. This could be used as one of the most specific antigens for the immunodiagnosis of bovine fascioliasis. 4. In Ouchterlony test, precipitin lines were not found in the sera naturally infected with F. hepatica, but some were found in the sera of rabbits immunized with the crude antigens. The numbers of precipitin lines in the sera of rabbits were different in the different fractionated antigens. They were 6 in the crude, 2 in the second and the third antigens, 1 in the fourth, the fifth and the sixth antigens and absent in the seventh antigen, respectively. 5. The wheal size for bovine infected with F. hepatica was $2.46{\pm}0.15cm$ in the intradermal test antigen(saline extract of F. hepatica) supplied by the Veterinary Research Institute, Rural Development Administration, Korea. The wheal size of the first, the second and the third antigens were larger than that. of intradermal test antigen, whereas those of the fouth, the fifth, the sixth and the seventh antigens showed smaller than that of the intradermal test antigen. The results suggest that the fifth antigen may be specific antigen for the immunodiagnosis of bovine fascioliasis.
An abattoir survey was carried out on 376 Korean native cattle reared and slaughtered in Soon-chun area to determine the prevalence of Fasciola hepatica infections. The average prevalence of fascioliasis was 44%; the infection rate had a tendency to increase with age. The mean(${\pm}SE$) length and width of the flukes were $24.9{\pm}2.5mm$ and $10.5{\pm}1.7mm$, respectively.
Field investigations were made in 1991 and 1992 in Kwangju and the western and central Chonnam area to determine the distribution of Lymnaea viridis, the intermediate host of Fasciola hepatica, and to study their life history during the different seasons. It had a wide distribution, existing all over the places, and the principal habitats of the snail were rice paddies. The life cycle was determined by making frequent visits to a series of habitats. The snsils began to produce egg masses in April, and egg production was the highest in May and June. Egg laying was continued but decreased in summer. There was sudden decrease in egg production in October when the temperature was below 2$0^{\circ}C$. The number of snails in July. Under the unfavourble conditions the snail either crawle up on the snail to aestivation. At low temperature below 8$^{\circ}C$ the snails hibernated inthe mud of the habitat. It was found in some habitats thao the snails were loving together with L. auricularia and Physa s pp. which indicates the neccessity of differentiation among them.
Paleoparasitology is a discipline that applies existing conventional and molecular techniques to study parasites found in ancient ruins. This review focuses on the history of the discovery of parasites (mostly helminth eggs and larvae) in archaeological soil samples and mummies in Korea from the Three Kingdoms Period to the Joseon Dynasty (100 BCE-1910 CE). We also briefly review important milestones in global paleoparasitology. The helminth species reported so far in Korea included Ascaris lumbricoides, Trichuris trichiura, Strongyloides stercoralis (larva), Trichostrongylus sp. (larva), Paracapillaria philippinensis (syn. Capillaria philippinensis), Enterobius vermicularis, Fasciola hepatica, dicrocoeliids, Paragonimus westermani, Clonorchis sinensis, Metagonimus yokogawai, Pygidiopsis summa, Gymnophalloides seoi, Isthmiophora hortensis, Dibothriocephalus nihonkaiensis (syn. Diphyllobothrium nihonkaiense), and Taenia spp. tapeworms. The findings obtained by Korean paleoparasitologists/archaeologists have brought about deep insight into the status of helminthic infections in Korea's past populations. Continued paleoparasitological research is essential for further understanding of ancient parasites and parasitic diseases in Korea.
The cattle liver flukes were collected at daughter house and classified with gross morphology. The hepatica type was only 14 flukes (4.7%) and the gigantica type was 284 flukes (95.3%) among total 298 flukes collected. In order to determinate the intermediate host of the flukes, artificial infection with the miracidia to snail hosts, Radix auricularia and Austropelea ollula, were successfully carried out, but the development in the later was terminated at the sporocyst stage. Intermediate host of the cattle flukes used to hibernate in deep part of the stubbles of rice pady. The number of A. ollula that has been collected from a rice stubble was ranged 0 to 19. Their mean number was 7 snails per the stubble. In artificial infection with miracidia, the reasonable number of them were 1 to 9 and the snail size was 2 to 5mm in length. The productivility of metacercaria in the intermediate host was observed. The metacercarial production per snail was ranged 5.5 to 48.0 per one miracidium.
This study was undertaken to characterize the properties of a 100 kDa somatic antigen from Metagonimus yokogawai. Monoclonal antibodies (mAbs) were produced against this 100 kDa antigen, and their immunoreactivity was assessed by western blot analysis with patients' sera. The mAbs against the 100 kDa antigen commonly reacted with various kinds of trematode antigens, including intestinal (Gymnophalloides seoi), lung (Paragonimus westermani), and liver flukes (Clonorchis sinensis and Fasciola hepatica). However, this mAb showed no cross-reactions with other helminth parasites, including nematodes and cestodes. To determine the topographic distribution of the 100 kDa antigen in worm sections, indirect immunoperoxidase staining was performed. A strong positive reaction was observed in the tegumental and subtegumental layers of adult M. yokogawai and C. sinensis. The results showed that the 100 kDa somatic protein of M. yokogawai is a common antigen which recognizes a target epitope present over the tegumental layer of different trematode species.
A 17 kDa protein from Clonorchis sinensis adults was purified by a procedure including Sephacryl S-200 HR gel filtration and Q-Sepharose anion exchange chromatography. The protein was proved to be a cysteine protease as it showed hydrolytic activity toward Cbz-Phe-Arg-AMC in the presence of dithiothreitol and was inhibited by specific inhibitors such as iodoacetic acid or trans epoxy-succinly-L-leucyl-amido(4 guanidino) butane. The polyclonal antibody raised against the protein reacted to 17 kDa proteins of trematodes such as Paragonimuf westermani, Fasciola hepatica, Opisthorchis viverrini, Gymnophalloides seoi, and Metagonimus yokogawai. The antibody recognized the 17 kDa and 16 kDa cysteine proteases purified from C. sinensis, P. westemani, and G. seoi as well. These results suggest that the 17 kDa protein may be a cysteine protease commonly present in trematodes.
In order to monitor the parasites, 303 fecal samples were taken from Korean native goats (n=279) and sheep(n=24) in Chonbuk area. The identification of the parasites were determimed by the fecal examination using the floatation or sedimentation methods and microscopical examination, respectively. The obtained results were summarized as follows ; 1. The detection rate of the parasites from Korean native goats and sheep was 98.2% and 83.3%, respectively. 2. In Korean native goat, mixed infection rate was single 7.5%(21 heads), double 47.3%(132 heads), triple 40.8%(114 heads), quadraple 5.0%(14 heads) and quintuple 2.5%(7 heads). 3. In sheep, mixed infection rate was single 12.5%(3 heads), double 45.8%(13 heads) and triple 16.6%(4 heads). 4. The parasites isolated were identified as Eimeria spp. from 283 heads, Oesophagostomum spp. from 96 heads, Ostertagia spp. from 97 heads, Strongylodes papillus from 97 heads, Heamonchs spp. from 73 heads, Trichostrongylus spp. from 42 heads, Moniezia expenza from 26 heads, Cooperia spp. from 16 heads, Nematodirus spp. from 9 heads, Bonostomum spp. from 9 heads, Paraphistomum spp. from 7 heads, and Fasciola hepatica and Capillaria spp. In 2 heads, in order.
