Transition nuclear protein 1 as a novel biomarker in patients with fertilization failure

  • Objective: Although intracytoplasmic sperm injection (ICSI) is a way to deal with in vitro fertilization failure, 3% of couples still experience repeated fertilization failure after attempted ICSI, despite having sperm within normal parameters. These patients are a challenging group whose sperm cannot fertilize the egg during ICSI. Unfortunately, no test can predict the risk of fertilization failure. Phospholipase C zeta (PLCζ) and transition nuclear proteins (TNPs) are essential factors for chromatin packaging during sperm maturation. This study aimed to assess PLCζ1 and TNP1 expression in the sperm of patients with fertilization failure and the correlations among the DNA fragmentation index, PLCζ1 and TNP1 gene and protein expression, and the risk of fertilization failure. Methods: In this study, 12 infertile couples with low fertilization rates (<25%) and complete failure of fertilization in their prior ICSI cycles despite normal sperm parameters were chosen as the case group. Fifteen individuals who underwent ICSI for the first time served as the control group. After sperm analysis and DNA fragmentation assays, quantitative reverse-transcription polymerase chain reaction (qRT-PCR) and Western blot analyses were performed to compare the gene and protein expression of PLCζ and TNP1 in both groups. Results: DNA fragmentation was significantly higher in the fertilization failure group. The qRT-PCR and Western blot results demonstrated significantly lower PLCζ and TNP1 gene and protein expression in these patients than in controls. Conclusion: The present study showed that fertilization failure in normozoospermic men was probably due to deficient DNA packaging and expression of TNP1.

  • 최근 지반공학 분야에서는 깊은기초와 얕은기초에 관한 한계상태설계법에 관한 연구가 선행되고 있으며, 옹벽구조물에 관한 연구는 미미한 실정이다. 본 연구는 철도 옹벽의 한계상태설계법을 도입하기 위한 기초 연구로써 옹벽 표준도의 파괴모드별 신뢰도지수가 목표신뢰도지수를 만족하는지를 평가하였으며, 옹벽 표준도에 대한 하중저항계수 설계를 수행함으로써 한계상태설계법의 적용성을 분석하였다. 철도 옹벽 표준도의 일부는 활동 및 지지력파괴에 대한 신뢰도지수가 목표신뢰도지수에 미달하며, 하중저항계수설계법에 의한 한계상태를 만족하지 못하여 한계상태설계 도입 시 철도 옹벽 표준도의 수정이 필요한 것으로 평가되었다.

토양 손실 평가에 의한 식생매트의 허용 소류력 결정 (Determination of Permissible Shear Stresses on Vegetation Mats by Soil Loss Evaluation)

  • 친환경 하천사업의 활성화로 식생매트의 사용은 증가하고 있으나 수리적 안정성에 대한 평가기법은 제시되지 않은 실정이다. 본 연구는 호안용 식생매트 제품의 객관적인 수리안정성 시험 기법 개발을 위해 수행하였다. 이를 위해서 식생매트 2종에 대한 실규모 실험을 수행하여 수리량을 측정하고 분석하여 작용 소류력을 계산하였다. 작용 소류력에 대한 토양손실평가를 위하여 지상라이다에 의한 측량을 수행하고 실험 전후의 하상고 변화를 평가하여 토양손실지수(CSLI)를 산정하였으며, 작용 소류력과 함께 도시하여 허용 소류력을 정량적으로 평가하였다. 하상고에 대한 정밀 측량 결과 분석에 의해서 식생매트가 안정한 경우에는 하상 변동이 국부적으로 제한되나, 불안정으로 판정되는 경우에는 식생매트 하부에서 비교적 큰 규모의 하상 변동이 발생하며 이는 자연 하상의 거동과 유사함을 확인하였다. 이상의 연구를 통해서 ASTM D 6040에 의한 식생매트의 허용 소류력 평가가 파괴 메카니즘 및 토양손실판정 기준에서 유효함을 확인할 수 있었다.

혼성제 직립 케이슨의 활동에 대한 파괴확률 (Probability of Failure on Sliding of Monolithic Vertical Caisson of Composite Breakwaters)

  • 신뢰성설계법의 적용성을 확대하기 위하여 혼성제 직립 케이슨의 활동에 대한 신뢰성 해석이 수행되었다. 충격쇄파 효과를 고려한 결정론적 설계법에 의하여 단면의 안전성이 자세히 해석되었으며, 특히 신뢰성 해석의 결과를 직접 결정론적 설계법의 결과와 연결시키기 위한 연구가 시도되었다. 해석 결과에 의하면 현재의 결정론적 설계 법에서 적용되고 있는 안전율은 약간 안전 측에 속한다. 또한 동일한 입사조건과 안전율에 대하여 수심이 증가함에 따라 신뢰지수는 감소하는 경향을 보이고 있다. 한편 방파제가 그 기능을 수행하는데 지장이 없을 것으로 예상되는 목표 파괴확률을 설정하고 그 목표 범위내에서 가장 최적의 파괴 확률을 산정하였는데, 입사조건에 따라 약간 씩 다른 최적의 안전율이 추정되었다. 마지막으로 혼성제 케이슨의 단면 결정에 영향을 주는 변수들의 변화에 따른 민감도 분석이 수행되었다. 해석 결과에 의하면 입사각, 주기의 영향이 크게 나타났으며 하상의 경사나 마운드의 두께에 대한 영향의 정도는 상대적으로 낮게 평가되었다.

상수도관의 개량을 위한 시간에 따른 노후도 및 안전성 분석 (Safety analysis and deterioration evaluation of water pipe for improvement according to service year)

  • 본 논문에서는 부식깊이의 실측 데이터를 통해 부식깊이 예측식을 수립하고 상수관망의 노후도 및 안전도 등을 결정할 수 있는 운영인자를 수립하여 실제 관망에 적용하였다. 신뢰성해석과 운영인자 분석을 통해 노후도를 비교·분석 하였다. 신뢰성해석결과 가덕의 직경 200 mm 관의 경우 시간이 경과함에 따라 4.36%, 6.18%, 8.23%로 증가하는 것으로 나타났고 남이의 직경 200 mm 관의 경우 시간이 경과함에 따라 7.35%, 9.51%, 12.99%로 증가하는 것으로 나타났다. 운영인자를 결정하여 방사형 차트 면적을 통해 노후도를 비교한 결과 가덕은 1.009, 남이는 1.174로 남이 지역이 노후도가 큰 것으로 나타났다. 신뢰성해석과 운영인자를 통해 교체 우선순위를 결정할 수 있었다.

Seismic damage assessment of a large concrete gravity dam

  • In the present work, a new global damage index is proposed for the seismic performance and failure analysis of concrete gravity dams. Unlike the existing indices of concrete structures, this index doesn't need scaling with an ultimate or an upper value. For this purpose, the Beni-Haroun dam in north-eastern Algeria, is considered as a case study, for which an average seismic capacity curve is first evaluated by performing several incremental dynamic analyses. The seismic performance point of the dam is then determined using the N2 method, considering multiple modes and taking into account the stiffness degradation. The seismic demand is obtained from the design spectrum of the Algerian seismic regulations. A series of recorded and artificial accelerograms are used as dynamic loads to evaluate the nonlinear responses of the dam. The nonlinear behaviour of the concrete mass is modelled by using continuum damage mechanics, where material damage is represented by a scalar field damage variable. This modelling, which is suitable for cyclic loading, uses only a single damage parameter to describe the stiffness degradation of the concrete. The hydrodynamic and the sediment pressures are included in the analyses. The obtained results show that the proposed damage index faithfully describes the successive brittle failures of the dam which increase with increasing applied ground accelerations. It is found that minor damage can occur for ground accelerations less than 0.3 g, and complete failure can be caused by accelerations greater than 0.45 g.

The contents are consisted of the possibility for the replacement of a metal valve disk to composite valve disk in butterfly valve based on the diversification. Purpose in this study is to predict failure field by each fly by appling Tsai-Wu Failure Index.

  • For the purpose of developing the method for efficiently calculating the design sensitivity and the reliability for the complicated structure such as ship structure, the probabilistic finite element method is introduced to formulate the deterministic design sensitivity analysis method and incorporated with the second moment reliability methods such as MVFOSM, AFOSM and SORM. Also, the probabilistic design sensitivity analysis needed in the reliability-based design is performed. The reliability analysis is carried out for the initial yielding failure, in which the derivative derived in the deterministic desin sensitivity is used. The present PFEM-based reliability method shows good agreement with Monte Carlo method in terms with the variance of response and the associated probability of failure even at the first or first few iteration steps. The probabilistic design sensitivity analysis evaluates explicitly the contribution of each random variable to probability of failure. Further, the reliability index variation can be easily predicted by the variation of the mean and the variance of the random variables.

Simplified Machine Diagnosis Techniques Using ARMA Model of Absolute Deterioration Factor with Weight

  • In mass production industries such as steel making that have large equipment, sudden stops of production process due to machine failure can cause severe problems. To prevent such situations, machine diagnosis techniques play important roles. Many methods have been developed focusing on this subject. In this paper, we propose a method for the early detection of the failure on rotating machine, which is the most common theme in the machine failure detection field. A simplified method of calculating autocorrelation function is introduced and is utilized for ARMA model identification. Furthermore, an absolute deterioration factor such as Bicoherence is introduced. Machine diagnosis can be executed by this simplified calculation method of system parameter distance with weight. Proposed method proved to be a practical index for machine diagnosis by numerical examples.

  • This paper develops practical and realistic reliability models and methods for the evalusion of system reliability and system reliability-based rating of R.C box-girder bridge superstructures. The precise prediction of reserved carrying capacity of bridge as a system is extremely difficult expecially when the bridges are highly redundant and significantly deteriorated or damaged. This paper proposes a new approach for the evaluation of reserved system carrying capacity of bridges in terms of equivalent system-strength, which may be defined as a bridge system-strength corresponding to the system reliability of the bridge. This can be derived from an inverse process based on the concept of FOSM form of system reliability index. The strength limit state models for R.C box-girder bridges suggested in the paper are based on the basic bending and shear strength. and the system reliability problem of box-girder superstructure is formulated as parallel-series models obtained from the FMA(Failure Mode Approach) based on major failure mechanism or critical failure states of each girder. AFOSM and IST(Importance Sampling Technique) simulation algorithm is used for the reliability analysis of the proposed models.
