Comparison and validation of rutin and quercetin contents according to the extraction method of tartary Buckwheat (Fagopyrum tataricum Gaertn.) (쓴메밀 종자의 추출방법에 따른 루틴 및 퀘세틴 함량 비교)

  • The stability and accuracy of ultra-performance liquid chromatography (UPLC) used for evaluating the contents of rutin and quercetin in tartary buckwheat (Fagopyrum tataricum Gaertn.) seeds extracted by seven different extraction methods were determined. The seven extraction methods were reflux extraction (RE), ultra-sonification extraction (UE), stirrer extraction (SE), RE after UE (UE+RE), RE after SE (SE+RE), UE after SE (SE+UE), and RE with UE after SE (SE+UE+RE). Among the seven extraction methods used, RE yielded comparatively higher contents of rutin (2,277 mg/ 100 g) and quercetin (158 mg/100 g) than those yielded by other six extraction methods. The intra-day repeatability and inter-day precision of RE was 0.4-3.2% considering relative standard deviation (RSD), while accuracy was 88.8-102.4%. Therefore, RE with UPLC would be a rapid, accurate, and stable method for analyzing rutin and quercetin contents in tartary buckwheat.

  • Ixeris sonchifolia Hance (Godulbaegi), Oenanthe javanica (Dolminari), Fagopyrum esculentum Moench (Buckwheat>, Hizikia fusiforme (Seaweed Fusiforme) and Zingiber ojficinale Roscoe (Ginger) have been used respectively as one of folk remedies as well as food materials. However, reportedly few studies on their immunomodulating effects have been made, although it has been known from other preceding studies that the ex vivo supplementation of each Ish, OJ, Fem, Hf, Zor water extracts tends to enhance the proliferation of splenocyte in comparison to the control group. This study on the combined immunomodulative effect of water extract mixture of these five food materials (Ish + Oj + Fem +Hf+Zor) lasted covering seven or eight weeks. The old mice (balb/c) was fed ad libitum on chow diet, and the water extract of plant mixture was orally administrated every other day for four weeks at two different concentrations (50 and 500 mg/kg B.W) . After preparing the single cell suspension, the proliferation of splenocyte was determined by MTT (3-[4,5-dimethylthiazol-2-y]-2,5-diphenyl terazolium bromide) assay. The production of cytokine ($IL-{\beta}$, IL-6, and $TNF-{\alpha}$) which was secreted by macrophages stimulated with LPS or not was detected by ELISA assay using the cytokine kit. After the 48 hours of incubation with the mitogen (ConA or LPS) stimulation, the proliferation of the mice splenocyt in the experimental group statisticaly increased at both of two different concentrations in comparison to the control group. The cytokines production was more significantly enhanced at the lower supplementation (50 mg/kg B.W.) group than at the higher concentration (500 mg/kg B.W.). The result of this study may suggest that the supplementation of water extract of plant mixture can regulate and enhance the immune function by increasing the splenocyte proliferation and regulating the cytokine production capacity by the activated macrophages in mice.

Growth of Three Sprout Vegetables and Absorption Characteristics of Vanadium according to Jeju Water Concentration (제주 Water 농축에 따른 3종 새싹채소의 생장 및 Vanadium의 흡수 특성)

  • Vanadium is a trace mineral present in dietary sources, most commonly vegetables. The aim of this study was to investigate the growth and vanadium absorption characteristics of Brassica napus L., Fagopyrum esculentum, and Hordeum vulgare L. using different Jeju water concentrations. The growth of three sprout vegetables and absorption characteristics of vanadium were different depending on Jeju water concentration containing vanadium. In the case of B. napus L. and H. vulgare L., germination rate was inhibited by increasing Jeju water concentration, whereas in F. esculentum, the best germination rate (60.7 %) was obtained at 40 times concentration of Jeju water (vanadium, 1.34 mg/L). For the sprout growth, growth of shoots increased with increasing Jeju water concentration, but root growth was gradually inhibited by increasing Jeju water concentration. For the absorption of vanadium by Jeju water concentration, vanadium was not detected in H. vulgare L. The vanadium contents of B. napus L. and F. esculentum were 4.2 mg/kg and 2.9 mg/kg at 10 times concentration of Jeju water (vanadium, 0.46 mg/L), respectively.

STUDIES ON THE DIMORPHISM OF THE PERSICARIA SENTICOSA NAKAI (Persicaria senticosa Nakai의 Dimorphism에 관한 연구)

Bot. 3(1) 16-25 During his researches regarding the morphological and physiological properties of Polygonecenae, the author has found that the species, Persicaria senticosa, aiso, besides the heterostylous plants of Polygonum family, Fagopyrum esculentum, and Persicaria japonica which was recently verified by the author as dimorphic, shows the typical floral structure of heterostylism, the description of which is not found in taxonomical works. Further research on this species have revealed that this plant, despite pressessing characteristic structural dimorphism, does not exhibit even the slight signs of heterostlylous properties physiologically. This is a deviation from the usual behavior of normal heterostylous plants. What is interesting is the fact that the physiological behavior of this species is quite contrary to that of P. japonica which is considered to be the most highly specialized dimorphic plant. Thus it is assurred that if some species of this family had taken a heterostylic form in its course of development from autogamy to allogamy, P. seoticosa would be the least differentiated type of dimorphic forms among the three heterostylous plants, including buckwheat, of this family. The results obtained in this experiment are summarized as follows: 1) P. senticosa has two forms of flower, one, long style-short stamened; the other, short style-long stamened. Not only conspicuous is this primary difference, but the secondary difference, such as pollen grain size, is noticeable between long and short styled individuals, thus expressing structurally the definite trait of a dimorphic plant. 2) Structural alteration of floral parts towards dimorphism has preceeded far less in comparison with those of the P. japonica and F. esculentum. 3) Elaborate studies on fertility reveal that this species does not show the slightest sign of the physiological characteristics of dimorphic plants. In other words, regardless of the modes of combinations, legitimate and illegitimate, fertilization and fruit setting flourish unimpaired. 4) Growth of pollen tubes apparently parallels the results in the fertility, tubes reaching ovary approximately 30 minutes after pollination both in legitimate and illegitimate combinations. Pollen tube penetration appears to be comparatively rapid. 5) A slight difference in the growth of pollen tube seems to exist between legitimate and illegitimate combinations, legitimate union giving slightly faster tube penetration. 6) In the present experiment it was clarified that P. senticosa, known to possess one form of flower in taxonomy, is in reality dimorphic plant having two of flowers. Although this species is definitely heterostylous in floral structure, physiological evidence and pollen tube behavior show that the differentiation of this plant toward the dimorphism has apparently proceeded slightly except for some parts of floral organ. In ordinary heterostylous plants it is a matter of common occurrence that when illegitimately cmbined, there is poor or no fertility, Contrary to the universal property of heterostylous plants, no difference is observed in the fertility and pollen tube growth between the legitimate and illegitimate combinations in the case of P. senticosa. Compared to the P. japonica and F. esculentum, which are supposed to have undergone high degree of dimorphic differentiation, it is an unavoidable conclusion that P. senticosa has not yet developed as a heterostyle plant except for some of its floral parts. If P. japonica is assumed to be the most differentiated type of hetenostylous plant, thenthe P. senticosa would be regarded as the primitive, retaining still the self-fertile nature so common to the Polygonum genus. In nature, however, this plant has a better chance to be pollinated legitimately owing to the two forms of flowers than to be pollinated illegitimately. Studies on Heterostylism, Fertility, and Embryological Characteristics in Buckwheat. Fagopyrum esculentum (메밀의 이형예현상과 수정력 발생학적 특징에 관한 연구)

  • Fifty-five local collections of buck wheat, Fagopyrum esculentum, were investigated their ratios of long-styled (LS) and short-styled (SS) flowers, fertility, meiosis of megaspore and microspore mother cell, female and male gametogenesis, and egg apparatus in accordance with the sowing seasons (spring, summer), altitudes (20m, 50-100m, 300m), and parent style types (L, S). Also they were embryologically investigated the fertility, fertilizing phenomenon and proembryogenesis by the legitimate and illegitimate pollination. There were no differences in the ratios of long-styled and short-5tyled flowers along with altitudes, but more irregularness was observed in plain area than that in the mountaineous or coastal area. LS versus SS ratios by sowing seasons were significantly separated into 1 : 1 in the summer sowing (P 0.1), but they were irregularly separated in the spring sowing. The segregating ratios by parent style types showed more number of short-styled flower in the spring sowing, and were statistically seperated into 1 : 1 in the summer sowing (P 0.25), regardless to parent style types. In the artificial legitimate union, the seed setting rates of the summer sowing (59-61%) were much higher than those of the spring sowing (about 30%), but in the artificial illegitimate union the seed setting rates were only fructified about 0.8-1.8% in the spring sowing. The seed setting rates in accordance with flowering stages were larger in turn early, middle, late, in the summer sowing. The grain number and grain weight per plant of short-styled flower were more than those of long-styled one regardless to style types. The 1,000 grain weight of long-styled flower was heavier than that of short-styled one in large grain, but it was lighter than that of short-styled flower in small or medium grain. The percentage of normal female and male gametogenesis in the summer sowing were higher than those in the spring sowing. The ovule was atropous and two polar nuclei were a synkarion before flowering. The pollens germinated at 30 minuts after pollination and the pollen tube grew continually and penetrated into micropyle at 1.5-2 hours and the two male nuclei fertilized with egg nucleus at 3 -5 hours after pollination. Flertilizing times in summer were shorter than in autumn. The fertilized egg was divided in a small apical cell toward the interior of the embryo sac and a large basal cell toward the micropyle cell at 15-24 hours after pollination, and division times in summer were shorter than in autumn. The proembryo began the embryogenesis at 7-8 days and formed itself into the perfect embryo at 15 days after pollination.

A PCR Method for Rapid Detection of Buckwheat Ingredients in Food (식품에서 메밀 성분의 검출을 위한 PCR 방법)

  • Buckwheat often causes severe allergic reactions in sensitive people. One of the major allergenic proteins in common buckwheat (Fagopyrum esculentum) has been found to be a BW10KD protein. In this study, we developed a PCR method to detect buckwheat ingredients in food using primers corresponding to the allergenic BW10KD gene. Five pairs of oligonucleotide primers successfully enabled PCR amplification of the specific regions of the genomic BW10KD DNA from buckwheat, but no amplification from seven other cereals and beans (barley, wheat, German millet, African millet, soybean, red bean, and black bean). The proposed PCR method was applied to analyze 12 processed foods (buckwheat flour, buckwheat noodle, buckwheat jelly, wheat noodle, instant noodle, black sesame gruels, sunsik, cookie, misutkaru, and three kinds of cereal); among them, only three samples including buckwheat flour, buckwheat noodle and buckwheat jelly showed a positive reaction to the detection. This PCR method was able to detect as little as 1 ng of common buckwheat DNA. This rapid and specific PCR method would be applicable to detect allergenic buckwheat ingredients in food.

Quality Characteristics of Buckwheat (Fagopyrum esculentum) Soksungjang (메밀을 이용한 속성장의 품질특성)

  • This study was conducted to provide information for improving the quality characteristics of Buckwheat Soksungjang (BWS). We determined aminotype nitrogen content, total microbial flora counts, the population of Bacillus cereus, presence of volatile compounds, fibrinolytic activity, antioxidant activity, ACE inhibition rate, and a sensory evaluation. The aminotype nitrogen increased gradually during fermentation. We found a decreasing population of B. cereus during fermentation, thus, the edible period for BWS was more than 30 days after fermentation. Acetaldehyde, butanol, and pyrazine were detected as volatile compounds after fermentation. The fibrinolytic activities of a 10% BWS water extract were high at 120.8 units compared to the control (71.6 units). In a sensory evaluation, Soksungjang with 60% added BW showed a significantly higher score (p < 0.001) for color, taste, smell, texture, and overall. The results suggest that a new type of shortened fermented soybean paste had good safety, bioactivities, and sensory characteristics within 4 weeks.

Effects of Added Buckwheat(Fagopyrum esculentum) Extract on Starch Hydrolysis In Vitro and Glucose Responses in Healthy Subjects (메밀추출물 첨가가 In Vitro 전분가수분해율 및 정상성인의 혈당반응에 미치는 영향)

  • This study determined the effects of added buckwheat extract on the rate of corn starch hydrolysis in vitro as well as blood glucose responses through its supplementation in healthy subjects. The rate of corn starch hydrolysis in the presence or absence of various buckwheat extracts was determined in an in vitro enzyme/dialysis system for 2 hr. The buckwheat was extracted by water, ethanol(40%, 70%, 100%) and methanol(40%, 70%, 100%), respectively. Twenty percent(w/w) additions of the ethanol, methanol and water buckwheat extract to corn starch solution significantly reduced the starch hydrolysis at every minute for 2 hr(p<0.05). The calculated hydrolysis indices of the buckwheat extracts were in the order of 100% ethanol extract(50), 100% methanol(54), 40% ethanol(58), 40% methanol(62), 70% methanol(64), 70% ethanol(68), water (82). For the blood glucose response study, groups of 12 volunteers were given 50 g of boiled rice with or without buckwheat extract(10% and 20% of starch weight) using the 100%, 70%, and 40% ethanol extracts, respectively. The addition of each buckwheat ethanol extract significantly reduced blood glucose concentrations at three or more points during 2 hr and also reduced the mean peak rise and area under the blood glucose curve(p<0.05). The calculated glycemic index(GI) values for all ethanol buckwheat extract groups were significantly decreased compared to the control(rice). At the concentrations of 20%, the buckwheat 100% ethanol extracts lowered the GI by 68%. The 100% ethanol extract was more effective than the 70% and 40% extracts for reducing GI. Therefore, the 100% ethanol buckwheat extract would be the most therapeutically useful in modifying postprandial hyperglycemia.

A Study of the Planting Characteristics of Street Trees and Herbaceous Plants in Gangwon-do (강원도 내 가로수와 가로녹지대 초화류의 식재 특성에 관한 연구)

  • This study surveyed planting areas along streets in Gangwon-do to find out how to improve the planting and use of street trees and herbaceous plants. There were 301,491 trees of 41 species on the streets of Gangwon-do in 2004. The predominant species of street trees were Ginkgo biloba ($40\%$), Prunus spp. (Prunus yedoensis and Prunus sargentii) ($25\%$), Platanus occidentalis ($5\%$), followed by Betula platyphylla var. japonica, Zelkova serrata, Prunus armeniaca var. ansu, Acer palmatum, and Pinus thunbergii. Eighty-four herbaceous plant species were found in the Youngseo district (the southern area of Gangwon-do); the ratio of native species to exotic was 51:33. The predominant species were Cosmos bipinnatus, Petunia hybrida, Tagetes spp., Aster koraiensis, and Fagopyrum esculentum. Eighty-nine herbaceous plant species were found in the Youngdong district (the eastern area of Gangwon-do); the ratio of native species to exotic was 55:33. The predominant herbaceous plants were Aster koraiensis, Tagetes spp., Petunia hybrida, Rudbeckia bicolor, Cosmos bipinnatus, Salvia splendens, Brassica oleraceae var. acephala, Aquilegia buergeriana var. oxysepala, Coreopsis drummondii, Viola tricolor, and Dianthus superbus var. longicalycinus. Appropriate pruning adds to the aesthetic value of trees and prolongs their useful life; it also maintains good health and thereby reduces the need to control insects and diseases. Street trees had not been properly pruned due to the presence of power lines and a shortage of pruning information. The pruning was controlled by Korea Electric Power Company, which has no pruning information. Pruning must be maintained by a professional landscape company in order to maintain good shape, such as that which is done for bonsai. In order to improve the planting, use and maintenance of landscape plants in Gangwon-do, the following recommendations are made: street tree species should be diversified, suitable street trees should be selected for each space, native species should generally be used, trees should be appropriately pruned and properly fertilized, pests and diseases should be controlled, plantings should be done in multiple layers, spatial arrangements should be improved, larger trees should be planted, and drainage and underground electric wires should be considered when planting.

Immuno-enhancing Effect of Seed Extracts on a RAW 264.7 Macrophage Cell Line (종자 추출물의 RAW 264.7 세포에 대한 면역증강 효과)

  • In this study, the immuno-enhancing activity of seed extracts were studied on the macrophage cell lines. We examined the effect of nine seed extracts on nitric oxide (NO) production in RAW 264.7 cells and selected four highly-effective seed candidates (Fagopyrum esculentum, Taraxacum platycarpum, Impatiens balsamina, Helianthus annuus) for further immune-related studies. The effects of the four seed extracts on the production of immune-related cytokines in the RAW 264.7 macrophage cell line and proliferation of Molt-4 as a T cell line were investigated. The secretion of NO from the RAW 264.7 cells was increased up to 39 ${\mu}M$ by adding the seed extracts (25 ${\mu}g/mL$) compared to the control. Also, the secretion of tumor necrosis factor-alpha (TNF-${\alpha}$) was also increased up to 32 times by adding the seed extracts (25 ${\mu}g/mL$). Secretion of cytokines such as interleukin-1 beta (IL-$1{\beta}$), interleukin-6 (IL-6), and interleukin-10 (IL-10) was also increased and induced the proliferation of T cells compared to the control. In conclusion, these results suggest that four seed extracts provide beneficial immuno-enhancing effects for human health.
