Physico-Chemical Properties of Ginseng Leaf Tea As Affected by Decontamination Treatment (인삼엽록차의 살균처리에 따른 이화학적 특성 변화)

  • Physico-chemical properties were investigated on ginseng leaf tea which was decontaminated by the current ethylene oxide fumigation and gamma irradiation, respectively. Chemical components such as soluble matter, saponin and polyphenols were found to be stable to both treatments, while ascorbic acid and chlorophylls were relatively labile to fumigation and irradiation more than 5 kGy. Instrumental analysis has shown that EO fumigation causes apparent changes in pH and color of extracts and volatile profile of the sample, as compared to negligible changes in the irradiated sample at a decontaminating dose, 5 kGy.

  • Grapes(Vitis labruscana B) were fumigated with acetic acid at 20mg/liter to reduce storage decay and packaged with polyethylene film(0.03mm), then stored for 90 days at 2$^{\circ}C$. Modified atmosphere packaging reduced slightly weight loss and soluble solids content during storage. Acetic acid fumigation decreased effectively berry shattering and achieved remarkable control of rotting for storage. Two grape cultivars, 'Campbell Early' and 'Sheridan', fumigated with acetic acid had only 0.7~2.9 berries rot compared with 8.3~27.6 berries rot of cluster for grapes that were not fumigated.

Biological Quality and Storage Characteristics of Gamma-Irradiated Whilte Ginseng (감마선 조사된 백삼의 생물학적 품질 및 저장특성)

  • Microbiological quality of commercial white ginseng was examined, together with investigation on its water absorption pattern and radiosensitivity of molds isolated from the samples. Comparative effects of phosphine fumigation and gamma irradiation on microbial control and disinfestation of the samples packed in a commercial laminated film and stored for six months at ambient(20oC, 70% RH) and accelerated(40oC, 90% RH) conditions were studied. Commercial white ginseng was contaminated with coliforms by 103~104CFU/g and molds by 102~104CFU/g and thus decontamination process was required for hygienic quality and storage stability. Phosphine fumigation showed no sterilizing effects on microbes contaminated, while gamma irradiation at around 5kGy was effective for decontaminating microorganisms, showing D10 values of 0.48~0.60kGy for isolated molds. Even though the storage insects, Plodia interpuctella Hubner and Lasioderma serricorne(cigarette beetle) were easily destroyed by phosphine fumigation, gamma irradiation less than 5kGy was found effective for both purposes to improve biological quality of stored white ginseng, thereby extending the storage life of packed samples resulting from increased critical moisture content by about 1%(Aw 0.76).

Comparative Effect of Gamma Irradiation and Phosphine Fumiagion on Color and Organoleptic Quality of White Ginseng (감마선과 Phosphine 처리가 백삼의 색도 및 관능적 품질에 미치는 영향)

  • Comparative effects of phosphine fumigation and gamma irradiation were evaluated on the qualities of white ginseng prepacked in a commercial laminated film in terms of water absorption, Hunter color parameters(L, a, b) and organoleptic properties(overall color/appearance, flavor) during 6 months of storage at ambient(20oC, 70% RH) and accelerated(40oC, 90% RH) conditions. Moisture content of packed white ginseng(initial 8.44%) linearly increased with storage period, thereby reaching to about 10% (r=0.9966) in ambient and more than 15%(r=0.9886) in accelerated conditions, respectively, at the 6th month of storage. The storage at severe conditions resulted in remarkable changes in Hunter's color values, decrease in whiteness(L) and increase in redness(a) and yellowness(b), while both treatments, phosphine and irradiation less than 5kGy, did not cause any significant changes in color parameters. Phosphine fumigation was shown detrimental to overall flavor of white ginseng(p<0.01), however or ganoleptic qualities of stored samples were effectively maintained by irradiation less than 5kGy(p< 0.05). As the index on the guality of white ginseng stored at severe conditions its organoleptic quality was highly correlated with moisture content(r= 0.9777) and Hunter color values(over 0.9), suggesting the critical values of 15.5% moisture and 72.56, 7.15, 21.45 in Hunter's L, a, b values, respectively.

Effect of Calcium Cyanamide Soil Fumigation on Sterilization of Rhizoctonia solani, Pythium sp., Soil Microbes and Plant Seed (석회질소 토양훈증의 라이족토니아 소라니, 피시움, 토양미생물과 씨앗의 사멸효과)

  • The effect of calcium cyanamide (China-made) soil fumigation on the growth of the ginseng pathogen Rhizoctonia, Pythium), soil microbes, and seed germination of lettuce and radish was investigated. At twice the recommended level (2S0-ppm $CaCN_2$), the growth of Rhizoctonia and Pythium, and the seed germination, were not inhibited. Rather, the effective level was 10,000 ppm. The powder form was more effective than the granular form in inhibiting pathogen growth and seed germination. The lettuce seed was also more sensitive than the radish seed. Calcium cyanamide appearedto decrease the fungi population and to increase Actinomycetes in the soil.

Photosynthetic Inhibition in Leaves of Ailanthus altissima under O3 Fumigation

  • We investigated the enect of $O_3$ on the photosynthetic characteristics of tree of heaven (Ailanthus altissima) that is naturalized plant and used as restoration plant for contaminated area. Two-year-old seedlings were planted to pots and transferred into closed $O_3$ chamber. Photosynthetic pigments contents and photosynthetic characteristics were measured every three weeks under 100 pub $O_3$ fumigation. There was no visible foliar injury by $O_3$ exposure and contents of photosynthetic pigments did not show significant differences between control and $O_3$-treated seedlings. Also there were no significant differences in stomatal conductance, and water use efficiency. But photosynthetic rate and apparent quantum yield (AQY) of $O_3$ treated seedlings were reduced after nine weeks of ozone fumigation. In addition, the reduction of carboxylation efficiency and photorespiration were observed in the leave of $O_3$ treated seedlings after six weeks. In accordance with our result, carbon fixation system of A. altissima was most sensitive to $O_3$ stress to evaluate physiological damage induced by $O_3$.

  • Rice (Oryza sativa L.) plants were cultivated to examine changes in antioxidative defence mechanism induced by elevated ozone levels. Catalase activities in tolerant Jinpumbyeo and susceptible Chucheongbyeo under ozone fumigation were reduced at 5 hrs and 3 hrs after ozone fumigation, respectively. With the increased ozone supply, peroxidase activity in Jinpumbyeo was steadily enhanced whereas in Chucheongbyeo it was not changed. Four SOD-isozymes were detected by NBT staining of native-PAGE. Two isozymes of them were obviously induced by ozone supply, particularly in Jinpumbyeo. The continuous ozone fumigation increased remarkably putrescine levels in leaves whereas it did not affect the levels of spermidine and spermine. In this study, it was implied that ozone in cell inhibits strongly diamine oxidase and thus promotes ethylene biosynthesis which will cause the senescence in rice plants.

Development of a New Disinfestation Method Using Methyl Bromide and PH3 in Cut Rose Fumigation (절화장미 훈증처리시 Methyl Bromide와 PH3를 이용한 새로운 살충방법의 개발)

  • Fumigation by combination of methyl bromide/phosphide ($MB/PH_3$) was much more effective in disinfestating the rose insect pests (Aphis gossypii, Tetranychus urticae, and Frankliniella occidentalis) than that by individual MB or $PH_3$. Fumigation for 2 hours in the chamber using ;$MB/PH_3$ at the concentration of $10/3g{\cdot}m^{-3}$ brought about 91.3% and 100% mortality in aphids and thrips, respectively. Fumigation for 3 hrs using ;$MB/PH_3$ at the concentration of 5/3 or $7/3g{\cdot}m^{-3}$ showed over 96% mortalities in mites and thrips as well as no phytotoxicity symptoms, e.g. tip burn or bent neck, in cut roses, while mortalities of mites and thrips were not influenced by increasing $CO_2$ concerntration from 0.03% to 10% or decreasing $O_2$ concentration up to 15% in fumigation chamber.

The 2nd Stability Appraisement on Cultural Property Material with the Replacing Fumigation Gas of Methyl Bromide II (Methyl Bromide를 대체하는 훈증 가스의 문화재 재질 안정성 평가 II)

  • In this study, the substitution fumigation gases (15% Ethylene Oxide + 85% HFC 134a, 20% Ethylene Oxide + 80% $CO_2$, 99% Sulfuryl Fluoride + 1% Inert Gas) were applied on the metal, pigment, fabric and paper specimen. The result of the fumigation treatment with 15% Ethylene Oxide + 85% HFC 134a (200g/$m^3$, 48hours) is the color changes (${\Delta}E$) of 1st and 2nd Cu specimens showed significant difference as 3.40, 4.17. On the other hand, other specimens except for Cu showed less than 3.0 in chrominance values. The result with 20% Ethylene Oxide + 80% $CO_2$ (150g/$m^3$, 48hours) is that the color changes (${\Delta}E$) of 1st and 2nd specimens were overall less than 3.0 so that color differences were subtle and hardly recognized with naked eyes. So it is proved that the fumigation treatment with Ethylene Oxide 20% + $CO_2$ 80% is relatively stable on materials in this study. In the case of 99% Sulfuryl Fluoride + 1% Inert Gas (50g/$m^3$, 48hours), the color difference of Lead red ($PbO_4$) of pigment was more than 3.0 that was compared with contrast specimens. But chrominance values in the other specimens were less than 3.0 on average.

A Study on Gravity Penetration of Fumigants in the Jumbo Silo (대형싸이로에 있어서 훈증제의 수직적 침투력)

  • This experiment was carried out to investigate the vertical penetration and diffusion of fumigants for soybean disinfection in jumbo silo. The results were as follows, 1. Using the methylbromide independently as a soybean fumigant, penetration and diffusing velocity of methylbromide gas which moved from the top to the bottom through the vertica1 silo was too rapidly, it was possible to find out more 60mg/l of methylbromide gas concentration at bottom of silo within 4 hours after beginning the fumigation. And it showed the tendency of reducing methylbromide gas concentration gradually over the 10 hours from tile beginning the fumigation. 2. In case of added $CO_2$ gas to the methylbromide as a carrier is much more rapid velocity of penetration and diffusion of methylbromide gas than that of methylbromide gas independently. Therefore methylbromide gas concentration at bottom of silo was detected over the 70mg/l within 1.5 hours after beginning the fumigation.. 3. On the other hand, hence the phostoxin as a soybean fumigant was less velocity of Penetration and diffusion of the gas through the vertical silo compare to methylbromide gas, the phostoxin gas concentration couldn't detect over the 10mg/1 during the whole fumigation period at the bottomside of silo. 4. Test insects (rice weevil; sitophilus oryzae. L.) inserted at bottom of silo for examine the fumigation effect were killed completly by using the methylbromide independently and added $CO_2$ gas to methylbromide, while using the phostoxin the test insects were most alive.
