보리와 옥수수 잎의 색소, 단백질 함량 및 관계II 활성에 미치는 ${SO}^2$의 영향 (Effects of Sulfur Dioxide on Pigments, Frotein Content and Photosystem II Activity of Barley and Corn Leaves)

  • This investigation was carried out to clarify the changes of pigments and soluble protein, and photosystem II activity in the leaves of barley (${SO}_2$-sensitive) and corn (${SO}_2$-resistant) seedlings induced by the ${SO}_2$ fumigation (10, 50ppm). The pH changes of the leaf extract, the content of sulfite and sulfate, the activities of catalase, peroxidase, and polyphenoloxidase were compared in the leaves of barley and corn seedlings induced by ${SO}_2$ fumigation. The results are summarized as follows: An appreciable effect of pH change of leaf extract by ${SO}_2$ fumigation was observed in barley leaves (pH 6.10 to 5.18), but only a small change occurred in corn leaves (pH 5.66 to 5.50). The same pattern of pH changes was recorded when the solution of 0.2N HCl was added to leaf extract, providing lower buffering capacity of the barley leaves than corn leaves. After 2 hours of exposure to 10 ppm ${SO}_2$, the contents of ${SO}^{2-}_3$ and ${SO}^{2-}_4$ were increased in barley leaves, while only ${SO}^{2-}_4$ increased in corn leaves. After fumigation with 10ppm ${SO}_2$ for 2 hours, barley leaves showed significant decreases in activities of catalase, to 17% peroxidase, to 58%, and polyphenoloxidase, to 88%. Corn leaves showed increases in activities of peroxidase, to 136%, and polyphenoloxidase, to 128%. Absorption spectra of pigments obtained from ${SO}_2$-fumigated leaves were gradually decreased with the fumigation time increases, but the decrease was more significant in barley leaves. Fumigation with 50ppm ${SO}_2$ for 2 hours induced the greatest decomposition in carotenoid, followed by chlorophyll a and then chlorophyll b in barley leaves. The ratio of chlorophyll a/b was decreased from 4.1 to 3.6 in barley leaves, but in corn leaves it was maintained almost a constant level(4.9-4.8). The rate of decomposition of chlorophyll and carotenoid in corn leaves was very slow than those in the barley leaves. Fumigation with 50 ppm ${SO}_2$ for 2 hous, decreased the protein content of barley leaves to 59%, and that of corn leaves to 89%, and the extent of decrease in protein content was greater than that of pigments in barley and corn leaves. The rate of DCIP9dichlorophenol indophenol) photoreduction in ${SO}_2$-fumigated leaves was decreased to 18 and 67% in barley and corn leaves, respectively. However, DCIP photoreduction was considerably recovered about 32 and 92% with the addition of DPC(diphenylcarbazide) as an exogenous electron donor in barley and corn leaves, respectively.

작약 검은뿌리썩음병 발생실태와 방제 (Occurrence and Control of Black Root Rot of Peony (Paeonia lactiflora) on Continuous Cropping)

  • 작약검은뿌리썩음병은 연작을 할 경우 현저히 발생이 증가하였으며, 태백작약 품종에 비해 의성작약 품종에서 좀 더 심한 발병을 보였다. 종구로 사용하는 뇌두를 약제에 침지시 병원균은 검출되지 않았으나 물에만 침지한 경우 병원균이 검출되었다. 연작지의 토양개량시 검은뿌리썩음병의 방제는 효과가 없었으나 다조메입제로 토양을 훈증소독할 경우 토양훈증처리구에서 주근중과 근장이 단순연작구와 비교하여 50% 정도 증가하였으며 검은뿌리썩음병의 발병율은 무처리구의 경우 40.6% 정도 발병되었으나 토양훈증처리구는 4.7% 정도로 발병되어 아주 높은 방제효과를 보였으며 종구소독과 훈증처리를 병행시 발병율은 더욱 낮아졌다.

상족중 회전섶 포르말린훈증 소독효과 (Effect of Formalin Fumigation on Rotatory Cocooning Frame during Mounting Period)

  • 다회육시 회전섶을 소독할 여유없이 재사용하므로서 누에병발생의 가장 큰 오염원이 될 가능성이 크므로 이를 상족중에 포르말린으로 훈증처리하여 소독효과 및 사질에 미치는 영향에 대해 조사 검토하였다. 소독후 회전섶 먼지를 채취하여 도말급상법으로 소독효과를 검토한 결과 훈증소독구에서 첨식 6일째까지 82%의 병잠이 발생하여 실용적 소독효과가 거의 없음을 보였고 또한 훈증처리한 구의 고치 실풀림새율이 41%로 극히 불량하여 사질에도 악영향을 미치는 것으로 나타나 상족중 포르말린 훈증소독방법은 전혀 의미가 없는 것으로 판명되었다.

Effects of Gamma Irradiation and Methyl Bromide Fumigation on the Qualities of Fresh Chestnuts during Storage

  • Effects of irradiation and fumigation on disinfestation and quality attributes of chestnuts during storage were studied. Fresh chestnuts were exposed to gamma irradiation (0-10 kGy) and methyl bromide (MeBr) under commercial conditions and stored at different temperatures for 6 months. Pest of quarantine importance for chestnuts, Curculio sikkimensis Heller showed 100% mortality by MeBr on 3rd day after fumigation and by irradiation at 0.5 kGy in about 3 weeks at $23{\pm}0.5^{\circ}C$. Respiration rates of samples one day after treatments increased in proportion to irradiation dose. Respiration pattern of MeBr group was equal to that of 10 kGy-group. Both MeBr and irradiation at 0.25 kGy or higher showed 100% inhibition of sprouting during storage at $5^{\circ}C$ for 6 months. Flesh firmness was significantly reduced by MeBr or irradiation over 5 kGy (p<0.05) 1 day after treatments. MeBr fumigation resulted in appreciable decrease in flesh weight, reducing sugar and ascorbic acid contents (p<0.05), as compared to irradiated samples. Irradiation at 0.5 kGy was effective as alternative to MeBr in controlling pests while maintaining overall quality of fresh chestnuts during storage.

Effect of Gamma Irradiation and Fumigation on the Biological Qualities of Green, Black, and Oolong Teas

  • The biological qualities of green, black, and oolong teas were monitored by observing their microbial decontamination and insect disinfestation following gamma irradiation (0-10 kGy) and fumigation (MeBr or $PH_3$) during 6-month storage at room temperature. Plodia interpunctella Hubner was found as an important quarantine pest in teas used. In a comparative study, both treatments were found to be effective in disinfecting the stored samples. An irradiation dose of 5 kGy was sufficient to control all microorganisms related to the quality of teas, while fumigation with methyl bromide and phosphine showed no appreciable decontamination effect on the microorganisms. As a result, irradiation was found an effective alternative to fumigants for the improvement of biological tea qualities during storage.

Effect of Thymol and Linalool Fumigation on Postharvest Diseases of Table Grapes

  • Several postharvest diseases of table grapes (Vitis vinifera) occur during storage, and gray mold rot is a particularly severe disease because the causal agent, Botrytis cinerea, grows at temperatures as low as $0^{\circ}C$. Other postharvest diseases, such as those caused by Penicillium spp. and Aspergillus spp., also often lead to deterioration in the quality of table grapes after harvest. The use of plant essential oils such as thymol and linalool, to reduce postharvest diseases in several kinds of fruits, including table grapes and oranges, has received much attention in European countries. However, to the best of our knowledge there has been no report of the use of thymol fumigation to control gray mold in table grapes in Korea. Thymol ($30{\mu}g/mL$) and linalool ($120{\mu}g/mL$) significantly inhibited mycelial growth and conidia germination of B. cinerea. The occurrence rate of gray mold rot of B. cinerea and other unknown fungi was significantly reduced by fumigation with $30{\mu}g/mL$ thymol in several table grape cultivars, such as Campbell early, Muscat Bailey A, Sheridan, and Geobong. In this study, fumigation with $30{\mu}g/mL$ thymol, had no influence on the sugar content and hardness of grapes, but reduced fungal infection significantly. This suggests that $30{\mu}g/mL$ thymol could be utilized to reduce deterioration of grapes due to gray mold and other fungal infections during long-term storage.

감마선과 Methyl Bromide 처리가 도토리 종실의 해충사멸과 이화학적 품질에 미치는 영향 (Comparative Effects of Gamma Irradiation and Methyl Bromide Fumigation on Disinfestation and Physicochemical Quality of Acorn)

  • 전분 추출용으로 대부분 사용되고 있는 도토리 종실의 가해해충 사멸방법을 연구할 목적으로 현행 검역해충 사멸방법인 methyl bromide (MBr) 훈증처리와 감마선 조사법에 대하여 살충효과와 이들이 몇 가지 이화학적 품질에 미치는 영향을 비교 검토하였다. 국내산 도토리의 가해해충은 도토리밤바구미 (Curculio drntipes Roelofs)와 복숭아명나방 (Dichocrocis punctiferalis Guenee)으로 확인되었다. 이들 해충의 사멸시험에서 상업적 조건의 MBr 훈증은 처리 직후부터 살충효과가 완전하였고, 감마선의 경우에는 조사 3일째에는 1-3 kGy, 조사후 1개월 경에는 0.5-1 kGy의 조사선량에서 해충의 치사율이 100%에 달하였다. 훈증처리된 도토리는 처리 직후 명도 (L값)의 감소와 황색도 (b값) 및 갈색도의 증가현상이 유의적이었으나 0.5-2 kGy 조사구에서는 대조구와 큰 차이가 없었다. 시료의 총페놀 성분과 환원당에 있어서도 훈증처리구는 대조구와 조사구에 비해 함량의 감소가 크게 나타났다. 시료를 통기포장하여 5-1$0^{\circ}C$의 조건에서 6개월간 저장 중 훈증처리구는 부패현상이 심하여 대체방안이 요구되었으며, 3 kGy 미만의 감마선 조사는 적용가능성이 인정되었다.

수출입 식물검역업체 근로자의 공기 중 Methyl Bromide 노출에 관한 연구 (Workers' Exposure to Airborne Methyl Bromide in the Exporting/Importing Plants and Products Quarantine Company)

  • Methyl bromide has been used as a representative fumigant for quarantine, and several poisoning cases have occurred recently by this chemical in Korea. The purpose of this study is to evaluate workers' exposures to airborne methyl bromide in the importing and exporting plant products quarantine companies. Air samples were collected 400/200 mg Anasorb 747TM and were analyzed by gas chromatograph /flame ionization detector according to the Occupational Safety and Health Agency (OSHA) Method PV2040. Geometric mean (GM) and arithmetic mean (AM) of total 27 workers' exposure concentrations to airborne methyl bromide were 1.12 ppm and 0.24 ppm, respectively. Two exposures(12.1 ppm and 12 ppm as 8hr-TWA) of total 27 workers' exposures exceeded the Korean standard (5 ppm) of Ministry Labor, while 4 exposures (15%) exceeded the Threshold Limit Value (TLV) (1 ppm) of American Conference of Governmental Industrial Hygienists (ACGIH). Seven samples (11%) of total 63 short-term air samples exceeded the OSHA Permissible Exposure Limit (PEL) 20 ppm (Ceiling). The opening (management) task in wood fumigation by tent showed the highest short-term exposure concentrations (AM: 18.6 ppm, GM: 0.58 ppm, maximum: 340.7 ppm). The maximum level in treatment task of the same process was 2.01 ppm. Methyl bromide concentrations in opening operation was significantly higher than that in treatment operation (p<0.05). In conclusion, the GM of workers' 8hr-TWA exposures to airborne methyl chloride in the importing/exporting plant quarantine industry was estimated below the ACGIH TLV (1 ppm). However, opening task in the fumigation of wood being covered with tent or fumigation of pant products in container showed the levels exceeding ACGIH TLV (1 ppm), and opening task in the fumigation of wood being covered with tent showed the level exceeding the Korean standard of Ministry of Labor (5 ppm).

The Major Postharvest Disease of Onion and Its Control with Thymol Fumigation During Low-Temperature Storage

  • Onion (Allium cepa L.) is one of the major vegetable crops in Korea that are damaged and lost by pathogenic fungal infection during storage due to a lack of proper storage conditions. The aim of this study was to determine an appropriate control measure using thymol to increase the shelf life of onions. To control fungal infections that occur during low-temperature storage, it is necessary to identify the predominant fungal pathogens that appear in low-temperature storage houses. Botrytis aclada was found to be the most predominant fungal pathogen during low-temperature storage. The antifungal activity of the plant essential oil thymol was tested and compared to that of the existing sulfur treatments. B. aclada growth was significantly inhibited up to 16 weeks with spray treatments using a thymol solution. To identify an appropriate method for treating onions in a low-temperature storage house, thymol was delivered by two fumigation treatment methods, either by heating it in the granule form or as a solution at low-temperature storage conditions (in vivo). We confirmed that the disease severity was reduced up to 96% by fumigating thymol solution compared to the untreated control. The efficacy of the fumigation of thymol solution was validated by testing onions in a low-temperature storage house in Muan, Jeollanam-do. Based on these results, the present study suggests that fumigation of the thymol solution as a natural preservative and fungicide can be used as an eco-friendly substitute for existing methods to control postharvest disease in long-term storage crops on a commercial scale.

Reciprocal effect of ethyl formate and phosphine gas on two quarantine pests, Tetranychus urticae(Acari: Tetranychidae) and Myzus persicae(Hemiptera: Aphididae)

  • Fumigation of fruits and vegetables during quarantine and pre-shipment(QPS) treatment should be effective with a shorter fumigation time to minimize phytotoxicity. In this research study, a shorter fumigation time, 2 hours exposure which is shorter than that of the current commercial fumigation procedures using a lower dose of ethyl formate (EF) mixed with phosphine (PH3) on strawberry was investigated. The reciprocal effect between EF and PH3 against nymphs and adult Myzus persicae (Sulzer) and Tetranychus urticae (Koch) was evaluated. In addition, L(Ct)50 and L(Ct)99 of EF only and EF mixed with PH3 were analyzed at 5℃ and 20℃. The synergistic ratio (SR) of L(Ct)50 and L(Ct)99 for the nymph and adult stages of M. persicae were >1.0, which indicated a synergistic effect between EF and PH3. However, the SR values of L(Ct)50 and L(Ct)99 of the nymph and adult stages of T. urticae were ≤1.0 indicating that there was no synergistic effect between the two fumigants against T. urticae. Our results showed that the reciprocal effect between EF and PH3 has different effects on M. persicae and T. urticae. This could be attributed to the biological and physical differences between the class Arachnida and Insecta. The synergistic effect between EF and PH3 against M. persicae within a shorter exposure period and without phytotoxicity on fruits and vegetables will significantly benefit the horticultural industry.
