The Effects of Two Inoculants Applied to Forage Sorghum at Ensiling on Silage Characteristics

  • Whole forage sorghum (saccharatum) cultivar FS5 was harvested at the soft dough ($303{\pm}9g\;kg^{-1}$ DM) stage of maturity. The sorghum was chopped into approximately 20 mm pieces and ensiled under laboratory conditions in 1.5 L Weck glass jars. At ensiling, it was treated with two commercial silage inoculants: Pioneer 1188 (Inoculant A) and Eco-corn (Inoculant B). The inoculant A and B was applied at ca $2{\times}10^5$ or $2{\times}10^4$ colony forming units $g^{-1}$ DM., respectively. Silage with no additives served as a control. Three jars per treatment were opened on days 2, 4, 8, 15 and 60 post-ensiling to study fermentation dynamics. After 60 days of ensiling the silages were analyzed and subjected to an aerobic stability test lasting 5 days. Results showed that both inoculants caused a more rapid rate of pH decrease and a higher amount of lactic acid production. All the silages were well preserved and were stable upon exposure to air. Inoculants did not influence (p>0.05) the ash and total N contents, but tended to reduce acetic acid (p<0.05), butyric acid (p<0.01) and propionic acid (p<0.01) contents, and to increase the lactic acid content (p<0.01). The lower DM content of silages treated with Inoculant A agrees with the greater gas loss resulting from the DM loss, which was in good agreement with the higher yeast counts upon aerobic exposure. Silage treated with inoculant B had the highest DM (p<0.05) and lactic acid contents (p<0.01), and the lowest acetic acid content (p<0.05), which agrees with the rapid reduction of pH and smaller gas loss. Inoculant B reduced the ADF (p<0.01), ADL and NDF (p<0.05) contents, which also indicates smaller losses of organic soluble material. The control silages contained the highest levels of volatile fatty acids but no lactic acid, indicating secondary fermentation. It was concluded that both inoculants may improve the fermentation process, since silages from all treatments were stable upon aerobic exposure, noadvantage could be attributed to any of the inoculants used.

Development of Automatic Nutrient-Solution Mixing System Using a Low-Cost and Precise Liquid Metering Device (액제 정밀계량 장치를 이용한 양액 자동조제 시스템 개발)

  • This study was conducted to develop an automatic nutrient-solution mixing system for small-scale sewers. The nutrient-solution mixing system consisted of a low-cost and precise metering device and data acquisition & control system with a personal computer. and, the metering device was composed of three parts those were supply pumps, metering cylinders and venturi tube. The system controlled electric conductivity(EC) and pH of nutrient-solution based on the time-based feedback control method with the information about temperature, EC, and pH of the nutrient-solution. The performance of the nutrient-solution mixing system was evaluated through the control of EC and pH while compared with those of commercial system. Also an experimental cultivation of tomato was conducted to verify and to improve the developed system. Results of this study were as follows. 1. The correlation coefficient of meteing device between the flow rate and operating time was 0.9999, and the linear reuession equation computed was y=21.759x, where y is the discharge($g$) and x is the operating time(s). 2. Calculated errors for the developed metering device and two commercial pump were $\pm$0.3% $\pm$2.45% and $\pm$1.38 % FS error respectively. 3. An automatic nutrient-solution mixing system based on a low-cost and precise metering device was developed. 4. The full scale errors of the developed system in controlling EC and pH at 23$\pm$1$^{\circ}C$ were $\pm$0.05mS/cm and $\pm$0.2, respectively 5. When using the commercial system, the controlled values of EC and pH of the 500 $\ell$ of water were 1.29 mS/cm and 6.1 pH for the setting points of 1.4 mS/cm and 6.0 pH respectively at 23$pm1^{\circ}C$. 6. The developed nutrient-solution control system showed $\pm$0.05 ms/cm of deviation from the setting EC value over the experimental cultivation period. 7. The deviation from the average values of Ca and Mg mass content in the several nutrient-solution were 0.5% and 1.8% respectively.

A Comparative Study of the Shoulder Scoring Systems (견관절 Scoring System의 비교연구)

  • Aim: To evaluate validity and responsiveness of four shoulder scoring systems. Material and Method: Twenty-five cases of shoulder instability(22 traumatic, 3 non-traumatic) and twenty-three cases of rotator cuff tear(12 small or medium, 10 large or massive) treated surgically were evaluated with the Shoulder Function Score of the University of Pennsylvania(Penn FS), Constant Score, UCLA Shoulder Rating Scale and Simple Shouler Test(SST), preoperatively and at final follow-up. The average follow-up was 16.0 months in instability group and 17.5 months in rotator cuff tear group. Using the SPSS program, Pearson linear correlation coefficiency(PLCC) between the scores were calculated. And to assess the construct validity, PLCC between patients' satisfaction and the scores were also calculated. Responsiveness was measured by the standardized response mean(SRM). Result: In instability group, correlation between the scoring systems was low preoperatively except between Constant and SST, but high after operation. Patients' satisfaction with the scores showed low PLCC preoperativley, but high PLCC postoperatively. SRM was high in PENN and UCLA, but when the satisfaction segment of the score was eliminated from UCLA, the SRM was the lowest. In rotator cuff tear group, there was high correlation between the scores not only preoperatively but postoperatively. And the patients' satisfaction matched well with the scores. SRM was particularly high in UCLA and SST. Even when satisfaction segment was eliminated from UCLA, the SRM was still the highest. Conclusion : Evaluation by the 4 scoring systems investigated in the study showed less consistency in instability than rotator cuff tear in terms of correlation and validity. Responsiveness was generally higher in rotator cuff tear group than in instability group except for Pennsylvania Shoulder Function Score. Therefore it is construed that use of any among the four scoring systems doesn't make difference in evaluation of rotator cuff lesions. However in instability group, care is needed because different result may be obtained according to the selection of a scoring system.

The Development of Water Quality Monitoring System and its Application Using Satellite Image Data

  • In this study, we was measured the radiance reflectance by using multi-spectral image of low resolution camera(LRC) which will be loaded in the multi-purpose satellite(KOMPSAT) to use the data in analyzing water pollution. Also we investigated the possibility of extraction of water quality factors in rivers and water body by using high resolution remote sensing data such as Airborne MSS. Especially, we tried to extract the environmental factors related with eutrophication, and also tried to develop the process technique and the radiance feature of reflectance related with eutrophication. The results were summarized as follows: First, the spectrum of sun's rays which reaches the surface of the earth was consistent with visible rays bands of 0.4${\mu}{\textrm}{m}$~0.7${\mu}{\textrm}{m}$ and about 50% of total quantity of radiation were there. And at around 0.5${\mu}{\textrm}{m}$ of green spectral band in visible rays bands, the spectrum was highest. Second, as a result of the radiance reflectance Chlorophyll-a represented high spectral reflectance mainly around 0.52${\mu}{\textrm}{m}$ of green spectral band, and suspended sediments and turbidity represented high spectral reflectance at 0.8${\mu}{\textrm}{m}$ and at 0.57${\mu}{\textrm}{m}$ each. Third, as a result of the water quality analysis by using Airborne MSS, Chlorophyll-a could have a distribution chart when carried out ratio of B3 and BS to B7. And Band 7 was useful for making the distribution chart of suspended sediments. And when we carried out PCA, suspended sediments and turbidity had distributions at PC 1 , PC 4 each similarly to ground truth data. Above results can be changed according to the change of season and time. Therefore, in order to analyze more exactly the environmental factors of water quality by using LRC data, we need to investigate constantly the ground truth data and the radiance feature of reflectance of water body. Afterward in this study, we will constantly analyze the radiance feature of the surface of water in water body by measuring the on-the-spot radiance reflectance and using low resolution satellite image(SeaWiFs). Besides, we will gather the data of water quality analysis in water body and analyze the pattern of water pollution.

Stable Isotope and Fluid Inclusion Studies of Gold-Silver-Bearing Hyarothermal-Vein Deposits, Cheonan-Cheongyang-Nonsan Mining District, Republic of Korea: Cheongyang Area (한반도 천안-청양-논산지역 광화대내 금-은 열수광상의 안정동위원소 및 유체포유물 연구 : 청양지역)

  • Electrum-sulfide mineralization of the Samgwang and Sobo mines of the Cheongyang Au-Ag area was deposited in two stages of quartz and calcite veins that fill fault zones in granite gneiss. Radiometric dating indicates that mineralization is Early Cretaceous age (127 Ma). Fluid inclusion and sulfur isotope data show that ore mineralization was deposited at temperatures between $340^{\circ}$ and $180^{\circ}C$ from fluids with salinities of 1 to 8 wt. % equiv. NaCl and a ${\delta}^{34}S_{{\sum}S}$ value of 2 to 5 per mil. Evidence of fluid boiling (and $CO_2$ effervescence) indicates a range of pressures from < 200 to $\approx$ 700 bars, corresponding to depths of ${\approx}1.5{\pm}0.3\;km$ in a hydrothermal system which alternated from lithostatic toward hydrostatic conditions. Au-Ag deposition was likely a result of boiling coupled with cooling. Meaured and calculated hydrogen and oxygen isotope values of ore-forming fluids indicate a significant meteoric water component, approaching unexchanged paleometeoric water values. Comparison of these values with those of other Korean Au-Ag deposits reveals a relationship among depth, Au/Ag ratio and degree of water-rock interaction. All investigated Korean Jurassic and Cretaceous gold-silver-bearing deposits have fluids which are dominantly evolved meteoric waters, but only deeper systems (${\geq}1.5\;km$) are exclusively gold-rich.

  • The Au-Ag deposit of the Oknam mine occurs as gold-silver-bearing rhodochrosite veins in biotite schist and phyllite of the Precambriam Yulri Group. Five stages of ore deposition are recognized, each showing a definite mineral assemblage. General mineral parageneses in veins (stage III) associated with gold and silver vary inwardly from the vein margin: arsenopyrite + pyrite $\Rightarrow$ sphalerite+chalcopyrite+galena+gold $\Rightarrow$ ga1ena+Ag-bearing minerals. Fluid inclusion data indicate that temperature and salinity of ore fluids overally decreased with time: $345^{\circ}{\sim}240^{\circ}C$ and 3.4~7.8 wt. % NaCl equiv during stage I (quartz vein mineralization), $313^{\circ}{\sim}207^{\circ}C$ and 2.3~8.7 wt.% NaCl equiv during manganese-bearing carbonate stages (II and III), and $328^{\circ}{\sim}213^{\circ}C$ and 3.6-5.4 wt.% NaCl equiv during stage IV (quartz vein mineralization). The ore fluids probably evolved through repeated pulses of boiling and later mixing with cooler and more dilute meteoric waters. Fluid inclusion data and geologic arguments indicate that pressures during the mineralization were in the range of 90 to 340 bars. Gold occurs as silver-rich electrums (21 to 29 atom. % Au) and was deposited at temperatures between $300^{\circ}$ and $240^{\circ}C$. Thermochemical calculations suggest that gold was deposited as a combined result of increase in pH and decreases in temperature, $fs_2$ and $fo_2$.

Diagnostic Mutational Analysis of MECP2 in Korean patients with Rett syndrome

  • Purpose: Rett syndrome (RTT) is an X-linked dominant neurodevelopmental disorder affecting 1 per 10,000~15,000 female births worldwide. The disease-causing gene has been identified as MECP2 (methyl-CpG-binding protein). In this study, we carried out diagnostic mutational analysis of MECP2 gene in RTT patients. Methods: We analyzed four exons and putative promoter of MECP2 gene from the peripheral blood of 43 Korean patients with RTT by PCR-RFLP and direct sequencing. Results: Mutations were detected in MECP2 gene about 60.5% of patients. The mutations consisted of 14 different types including 9 missense mutations, 4 nonsense mutations and 1 frameshift mutation. Of these, three mutations (G161E, T311M, P385fsX409) were newly identified and these were determined as disease-causing mutations by PCR-RFLP and direct sequencing analysis. Most of the mutations were located within MBD (42.3%) and TRD (50%). T158M, R270X, and R306C mutations were identified with high frequency. An intronic SNP (IVS3+23C>G) was newly identified in only three of the patients. It may be a disease-related and Korea-specific SNP with RTT. The L100V and A201V have been reported to be unclassified variant and SNP. However, these mutations were not found in more than 100 normal Korean control samples. These base substitutions seem to be the disease-causing mutations in Korean RTT contrary to previous studies. Conclusion: Disease-causing mutations and polymorphisms would be very important for diagnosing of RTT in Korean. The experimental procedure used in this study might be considered for molecular biologic diagnosis used in clinical field.

Landslide Analysis of River Bank Affected by Water Level Fluctuation I (저수위 변동에 영향을 받는 강기슭의 산사태 해석 I)

  • The change of water level in reservoirs is an important factor causing failure of bank slopes, i.e. landslide. The water level of Three Gorges reservoir in China fluctuate between 145 m and 175 m, as a matter of flood control. During its normal operational state, the rate of water level fluctuation is supposed to range from 0.67 m/d to 3.0 m/d. Majiagou slope is located on the left bank of Zhaxi River, 2.1 km up from the outlet. Zhaxi River is a tributary of the Yangtze River within the Three Gorges area, of which the water level changes with the reservoir. At the back of Majiagou slope, a 20 m long and 3~10 cm wide fissure developed just after the reservoir water level rose from 95 m to 135 m in 2003. This big fissure was a full suggestion of potential failure of this slope. In this study, unsaturated-saturated seepage analyses were carried out to simulate the change of pore-water pressures in the bank slope subjected to the reservoir water level change. The obtained pore-water pressures were then used to evaluate the change in factor of safety (FS) with reservoir water level. It was found that the phreatic line showed a delayed response with respect to the change of the reservoir water level, because the seepage through soil layer was generally slower than water flows itself. During the rising and drawdown process, the phreatic lines take the shapes of concave and convex, respectively. And the fluctuation of reservoir water level just affected the front part of the bank slope, but had little influence on the back of the slope.

  • Homologue and isomer patterns of polychlorinated dibenzo-p-dioxins(PCDDs) and polychlorinated dibenzofurans(PCDFs) congeners formed from phenols in the gas-phase at $600{\sim}700^{\circ}C$ and via particle-mediated reactions at $400^{\circ}C$ were studied in an isothermal flow reactor. A mixture of 20 phenols in relative concentrations found in a municipal waste incinerator(MWI) stack gas was used for this study. PCDDs and PCDFs homologue and isomer patterns obtained from the phenol. From the phenol experiments, gas-phase formation at $600{\sim}700^{\circ}C$ favors PCDFs formation whereas particle-mediated formation at $400^{\circ}C$ favors PCDDs formation. DD and DF were most abundant homologue groups, PCDDs and PCDFs homologue fraction decreased with increasing number of chlorine substituents. PCDDs and PCDFs homologue and isomer fractions were almost constant from gas-phase formation and particle-mediated formation. Unsubstituted phenol, which was present in high concentration, played a significant role in the formation of PCDD/Fs congeners under both sets of experimental conditions.

A Study on the Development of the Repair Standards for Underground Pipelines Carrying Natural Gas (도시가스 매설배관 보수기준 개발에 관한 연구)

  • Grinding, weld deposition, type A sleeve, type B sleeve, composite sleeve, hot tapping and clamp are used as the method to repair the buried pipelines in the United States, UK and Europe. In the event of defect to the pipeline, they have repaired the pipeline through the fitness-for-service assessments. In addition, they have guidelines for the possible repair methods to apply to each type of damage, which is occurred due to the 3rd party construction or corrosion. According to the KGS FS551, Safety Validation in Detail including ECDA(External Corrosion Direct Assessment) as one method of integrity management should be carried out for the old pipeline which supply natural gas as the middle pressure in Korea. Where a defect on the pipelines is found, on the result of Safety Validation in Detail, the pipelines should be repaired or replaced by new piping. However, there are no guidelines or regulations regarding the repair and reinforcement of pipeline, so that, cutting the damaged pipeline and replacing it as a segment of new pipe is the only way in Korea until now. We have suggested pipeline repair methods including type A, B sleeve, composite sleeve, after the survey of foreign repair method and standards including the method of United States and the United Kingdom, and after analysis of the results on pipeline repair test including type A, type B sleeve and composite sleeve.
