Cytotoxicity of the Methanol Extract of Crotalariae sessiliflorae L. (농길리 메탄올 추출물의 세포독성)

  • The cytotoxic activity of Cratalariae sessiliflorae on cultured NIH 3T3 fibroblasts and human oral epithelioid carcinoma cells (KB) were evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) colorimetric method These fractions of methanol extract of Cratalariae sessiliflorae showed inhibitory effect in vitro in the milligram range against KB cell lines. In general, the antitumor activities of these fractions were does-dependent over the milligram range. The comparison of IC50 values of these fractions in tumor cell lines showed that their susceptibility to these fractions decrease in the following order: Fr. 4> Fr. 6> Fr. 10> Fr. 2> Fr. 11> Fr. 3> Fr. 8> Fr. 7> Fr. 9> Fr. 1> Fr. 5 by the MTT assay. These fractions were tested for their cytotoxic effects on NIH 3T3 fibroblasts using MTT assay. They exhibited potent cytotoxic activities in vitro in the milligram range against NIH 3T3 fibroblasts. In general, the cytotoxic activities of these fractions were does-dependent over the milligram range. The comparison of CD50 values of these fractions in NIH 313 fibroblasts shows that their susceptibility to these fractions in decrease the following order: Fr. 10> Fr. 9> Fr. 2 = Fr. 4> Fr. 8> Fr. 11> Fr. 1 = Fr. 7> Fr. 3> Fr. 5 = Fr. 6 by the MTT assay. These results suggests that fraction 5 has the most growth - inhibitory activity against KB cell lines.

Investigation of Food Quality Characterization of Processing By-product (Frame Muscle) from the Sea Rainbow Trout Oncorhynchus mykiss (해수산 무지개송어(Oncorhynchus mykiss) 가공부산물(프레임육)의 식품학적 품질 특성 조사)

  • This study compared the meat quality of sea rainbow trout Oncorhynchus mykiss frame muscle (S-FR-TY) and fillet (S-FI-TY) with the frame muscle of freshwater rainbow trout (F-FR). There was a difference of < 1% in the proximate composition of S-FR-TY vs. S-FI-TY, and of S-FR-TY vs. F-FR. The Hunter a value of F-FR-TY was lower compared with that of F-FR cultured in Jecheon (F-FR-JC), but higher than that of F-FR cultured in Pyeongtaek (F-FR-PT). However, no difference in the Hunter a value of S-FR-TY compared with any other F-FR was observed. The odor intensity of S-FR-TY was lower compared with that of F-FR, while the taste of S-FR-TY was milder than that of S-FI-TY, but stronger than that of F-FR. Although its total amino acid content was lower, the essential amino acid content of S-FR-TY was higher compared with S-FI-TY; the potassium content of S-FR-TY was also significantly greater. S-FR-TY contained high levels of nutritional and functional components, such as anserine, lysine, threonine, and eicosapentaenoic and docosahexaenoic acid. These results suggest that S-FR-TY could be used as a resource of canned food or fish jerky for children.

Antioxidant Effects of Crotalaria sessiliflora L. Leaf Extracts Fractionated from Each Solvent (활나물 잎 추출물의 분획별 항산화 활성)

  • In this study, Crotalaria sessiliflora L., leaf was extracted with a solvent mixture of $CH_2Cl_2$ and $CH_2OH$ (3:1). In order to draw or separate effective components from the leaf extract with high activity, a 1:1 solvent mixture of water and butanol was used after removal of the sugar and fat from the extract. From the decompressed and concentrated butanol layer, 10 small fractions (Fr1, Fr2, Fr3, Fr4, Fr5, Fr6, Fr7, Fr8, Fr9, and Fr10) were obtained using a chromatography column filled with sephadex LH-20. The activity of each fraction was measured by the same method as that used to measure the antioxidant effect of each part(p<0.05). The Fr1, Fr4, and Fr6 fractions showed the highest activity levels(p<0.05). The vital unknown material that vitality exists in the strong butanol fractions of F1, F4 and F6 will be isolated and identified. Currently, we are performing for a single compound from an unknown peak by instrumental analysis.

  • As face recognition (FR) has been well studied over the past decades, FR technology has been applied to many real-world applications such as surveillance and biometric systems. However, in the real-world scenarios, FR performances have been known to be significantly degraded owing to variations in face images, such as the pose, illumination, and low-resolution. Recently, visual intelligence technology has been rapidly growing owing to advances in deep learning, which has also improved the FR performance. Furthermore, the FR performance based on deep learning has been reported to surpass the performance level of human perception. In this article, we discuss deep-learning based high-performance FR technologies in terms of representative deep-learning based FR architectures and recent FR algorithms robust to face image variations (i.e., pose-robust FR, illumination-robust FR, and video FR). In addition, we investigate big face image datasets widely adopted for performance evaluations of the most recent deep-learning based FR algorithms.

Isolation and Purification of Protein-bound Polysaccharides from Mycelia of Flammulina velutipes Grown on Sawdust Medium (팽나무버섯 수확후의 톱밥배지로부터 단백다당류의 분리 및 정제)

  • Protein-bound polysaccahrides(PBP) were isolated, purified, and characterized from the sawdust media after harvesting the fruit body of Flammulina velutipes. The yield of the crude PBP(Fr.CA) extracted from the sawdust media, was 0.367% relative to the original sawdust media. The total sugar and protein contents of Fr.CA were 19.8% and 23.8% respectively. Using the membrane filtration, the fraction of which the molecular weight is over 300 kDa(Fr.A) was isolated from the Fr.CA and the yield was 44.6% relative to the Fr.CA. This result indicates that high molecular PBP is the dominant components of the Fr.CA. The Fr.A was separated into three fractions (Fr.A-1, Fr.A-2 and Fr.A-3) whose yields are 5.8%, 8.5% and 13.2% respectively. These fractions were further purified using gel filtration, obtaining a single peak in each fraction that considered as pure PBP Among them, the yield of Fr.A-1-${\alpha}$ was 20.9% relative to the Fr.A-1, and the molecular weight was 800 kDa. Monosaccharide components such as glucose, galactose, mannose, xylose and fucose could be detected all fractions by a HPLC analysis. Especially in the Fr.A-1-${\alpha}$ fraction, the content of glucose and galactose appeared to be high.

Free Radical Scavenging Activities and Inhibitory Effects on Xanthine Oxidase by Ethanol Extract from Capsella bursa-pastoris (냉이(Capsella bursa-pastoris)에탄을 추출물의 유리라디칼 소거 및 Xathine Oxidase 저해활성)

  • To examine the characteristics of antioxidative compounds from Capsella bursa-pastoris, ethanol extracts were separated into five organic solvent fractions; hexane(Fr.H), diethyl ether (Fr.E), ethyl acetate(Fr.EA), butanol (Fr.B), and water(Fr.D) fractions. Fr.B showed the greatest electron donating ability and inhibitory effect on lipid peroxidation. Whereas Fr.E had the most excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. The inhibitory effect of each fraction on xanthine oxidase was also measured. Fr.E had the strongest inhibitory effect on xanthine oxidase and $IC_{50}$ was $5.65\;{\mu}g$. The results indicate that the superoxide radical scavenging activity of Fr.E is caused by the inhibitory effect on radical generating system of xanthine oxidase. Also the order of inhibitory effect on xanthine oxidase was Fr.B

Anti-platelet and Anti-thrombotic Effects of Gastrodia elata (천마(Gastrodia elata Blume)의 항혈소판, 항혈전활성)

  • MeOH extract of Gastrodia elata was fractionated to five solvent fractions, hexane fr (fr I), 90% MeOH fr (fr II), EtOAc fr (fr III), BuOH fr (fr IV) and $H_2O$ fr (fr V). Among the five fractions, fr II, III and IV showed platelet anti-aggregating effects against ADP or collagen induced rat platelet aggregation in vitro. Fr II , III and IV were also tested in vivo, in the mouse and rat models of thrombosis. Oral administration of fr II, III or IV enhanced the recovery from the thrombotic shock in the mouse model of thrombosis to 32-40% from 17% of recovery with the control group of mice. Treatment with fr II, III or IV also attenuated the sudden reduction in the blood platelet count following intravenous collagen injection to rat. The above results were indicative of the presence of anti-platelet and anti-thrombotic components in this plant.

  • More than 300 specimens of higher fungi were collected in Mt. Jiri National Park areas from June, 1984 to September, 1985. These higher fungi were identified. As the resulting, Clitocybe flaccida (Fr.) Kummer, Tricoloma sejuctum (Fr.) $Qu\acute{e}l.$, Melanoleuca melaleuca (Fr.) Murr., Amanita rubrovolvata Imai, Rhodophyllus lazulinus (Fr.) ,$Qu\acute{e}l.$ Paxillus involutus (Fr.) Fr., and Lactarius campholatus Fr. were new to be Korea.

Antioxidant Properties and Protective Effects of Inula britannica var. chinensis Regel on Oxidative Stress-induced Neuronal Cell Damage (금불초 추출물의 항산화 효과 및 산화 스트레스에 대한 신경세포 보호작용)

  • The antioxidant properties and protective effects of Inula britannica on ${H_2}{O_2}$-induced SH-SY5Y neuroblastoma cell damage were investigated. A series of solvent fractions, including hexane(Fr.H), petroleum ether, chloroform, ethyl acetate(Fr.EA), and water fraction(Fr.W), were prepared from the 70% methanol extracts of Inula britannica. Fr.W had the highest total contents of phenolics and flavonoids, followed by Fr.EA. The antioxidant properties of the fractions were also evaluated by analyzing their scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radicals, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals, and nitric oxide. Fr.W showed the strongest activities in all assays. The concentrations of Fr.W that resulted in 50% reductions of the DPPH and ABTS radicals were 20.7 ${\mu}g$/mL and 39.4 ${\mu}g$/mL, respectively. Fr.W showed the weakest cytotoxic activities on the SH-SY5Y cells, whereas it effectively protected ${H_2}{O_2}$-induced cell death, increasing cell survival by 35.0-77.0% at a concentration range of 62.5-250 ${\mu}g$/mL. In this range, Fr.W also significantly decreased intracellular ROS levels by 34-39%. Overall, the antioxidant properties of Inula britannica can contribute to rescuring neuronal cells from oxidative stress-induced cell injury.
