Bioactivity of two medicinal plant Xylocarpus granatum Koen. (Meliaceae) and Sarcolobus globosus Wall. (Asclepiadaceae) of Sundarbans mangrove forest

  • Two medicinal plant of Sundarbans mangrove forest has been tested for the evaluation of growth inhibitory and antibacterial activity. The methanol extract of Xylocarpus granatum stem bark showed potent wheat rootlet ($IC_{50}=0.01{\mu}g/ml$) and shoot ($IC_{50}=0.23{\mu}g/ml$) growth inhibitory activity in a concentration related manner. The growth inhibitory activity was markedly decreased in residual methanol extract. The methanol extract showed antibacterial activity (MIC > 3 mg/ml) against Bacillus subtilis, Staphylococcus aureous and Proteus vulgaris. The residual methanol extract did not show any antibacterial activity. The results suggest the bioactive principle(s) of Xylocarpus granatum may be relatively non polar compound(s). The methanol extract and residual methanol extract of Sarcolobus globosus stem showed poor wheat rootlet and shoot growth inhibitory activity and no antibacterial activity.

Morphological Characteristics and Antioxidant Activity Changes in 'Autumn Sense' Hardy Kiwi (Actinidia arguta) as Honey Plant during Fruit Ripening

  • In this study, we studied the changes in antioxidant activity of Actinidia arguta fruit of Autumn Sense cultivar during fruit ripening. The aim of this investigation was to find the knowledge of the changes of physiochemicals associated with fruit quality, antioxidant properties (free-radical scavenging activity and reducing power), total phenolics and vitamin C during fruit ripening. The highest free-radical scavenging activity (at $100{\mu}g/ml$) and reducing power (at $100{\mu}g/ml$) in A. arguta fruit were 78.57% and 0.22, respectively. Total phenolic content and vitamin C content in fruit of 10 days after fruit set were $639.48{\mu}g/g$ and $1052.2{\mu}g/g$, respectively. In general, the antioxidant activity and the related parameters, including total phenolic content and vitamin C content decreased during fruit ripening. These results improve knowledge of the effect of ripening on the antioxidant activity and related compounds contents that could help to establish the optimum A. arguta fruit harvest data for various usages.

Antifungal Activities of Essential Oils from Six Conifers against Aspergillus fumigatus

  • The antifungal activities of the essential oils from Pinus densiflora, Pinus koraiensis, Abies holophylla, Larix kaempferi, Cryptomeria japonica and Chamaecyparis obtusa against Aspergillus fumigatus were evaluated. Essential oils were extracted from each conifer leaves for 8 h using water distillation method. The essential oil from A. holophylla appeared the strongest antifungal activity among the six oils with MIC value 0.125 mg/$m{\ell}$. The composition of the A. holophylla oil was analysed by GC/MS and then MIC was determined for main constituents of the oil. As the results, borneol and ${\alpha}$-bisabolol, which have a hydroxyl group, showed effective antifungal activities against A. fumigatus (0.25 mg/$m{\ell}$). In addition, the synergistic antifungal effects were observed in the combination of borneol and ${\alpha}$-bisabolol.

Fertilization and Tree Density Effects on Cellulose Decomposition in a Larix leptolepis Plantation

  • Cellulose mass loss by cellulose filter papers was measured for 3 time (35 days, 70 days, 105 days) incubation during the growing season (from May to September 2002) with different tree density and after fertilization in a Japanese larch (Larix leptolepis) plantation. Cellulose mass loss rates were significantly different between tree density types and fertilization treatments during the study periods. After 105 day incubation of cellulose filter paper, cellulose mass loss rates were significantly higher in the low tree density (70.1 $\%$) than in the high tree density (49.9$\%$). Cellulose mass loss rates averaged 62.8$\%$ in the fertilization and 58.9% in the unfertilization treatments during the same periods. However, cellulose mass loss was not significantly different between the forest floor and the mineral soil layer except for 35 day incubation. The results indicate that cellulose decomposition rates are a useful index to express differences in organic matter decomposition activity in different tree density and after fertilizer treatments.

새덕이 잎 정유의 성분분석 및 항산화활성 평가 (Chemical Compositions and Antioxidant Activities of Essential Oil Extracted from Neolitsea aciculata (Blume) Koidz Leaves)

  • 본 연구는 hydrodistillation 방법으로 추출한 새덕이 잎 정유의 화학적 조성과 항산화활성을 조사하였다. 새덕이 잎 정유의 화학적 조성은 GC/MS로 분석하였다. 새덕이 잎 정유의 주요 구성성분은 cis-ocimene (11.00%), trans-ocimene (9.65%), elemol (9.15%), ${\beta}$-elemene (8.75%), germacrene-D (7.55%), trans-caryophyllene (5.90%), ${\gamma}$-elemene (5.40%), ${\tau}$-muurolol (4.95%)인 것으로 나타났다. 새덕이 잎 정유의 항산화 활성은 총 페놀함량 분석, DPPH radical 소거활성, ABTS radical 소거활성, ferric reducing antioxidant power (FRAP) 분석을 통해 평가하였다. 정유의 총 페놀 함량은 $136.7{\pm}0.13\;mg\;GAE/g$으로 분석되었으며, 50% DPPH radical을 소거하는데 필요한 정유의 효과 농도($EC_{50}$ value)는 $639.33\;{\mu}g/m{\ell}$로 나타났다. 또한, ABTS radical 소거활성은 농도 의존적으로 나타났으며, FRAP value는 $31.21{\pm}0.12{\mu}M$ $FeSO_4{\cdot}7H_2O/g$이었다. 이러한 결과는 새덕이 잎 정유가 건강에 대해 중요한 이점과 기능을 가질 수 있는 항산화 활성을 가지고 있다는 것을 제안한다.

Inhibition of Osteoclast Differentiation and Promotion of Osteogenic Formation by Wolfiporia extensa Mycelium

  • Osteoporosis, Greek for "porous bone," is a bone disease characterized by a decrease in bone strength, microarchitectural changes in the bone tissues, and an increased risk of fracture. An imbalance of bone resorption and bone formation may lead to chronic metabolic diseases such as osteoporosis. Wolfiporia extensa, known as "Bokryung" in Korea, is a fungus belonging to the family Polyporaceae and has been used as a therapeutic food against various diseases. Medicinal mushrooms, mycelium and fungi, possess approximately 130 medicinal functions, including antitumor, immunomodulating, antibacterial, hepatoprotective, and antidiabetic effects, and are therefore used to improve human health. In this study, we used osteoclast and osteoblast cell cultures treated with Wolfiporia extensa mycelium water extract (WEMWE) and investigated the effect of the fungus on bone homeostasis. Subsequently, we assessed its capacity to modulate both osteoblast and osteoclast differentiation by performing osteogenic and anti-osteoclastogenic activity assays. We observed that WEMWE increased BMP-2-stimulated osteogenesis by inducing Smad-Runx2 signal pathway axis. In addition, we found that WEMWE decreased RANKL-induced osteoclastogenesis by blocking c-Fos/NFATc1 via the inhibition of ERK and JNK phosphorylation. Our results show that WEMWE can prevent and treat bone metabolic diseases, including osteoporosis, by a biphasic activity that sustains bone homeostasis. Therefore, we suggest that WEMWE can be used as a preventive and therapeutic drug.

Physiological and psychological effects of nature-based outdoor activities on firefighters in South Korea

  • This study investigates the physiological and psychological effects of a nature-based outdoor activity program in an environment reflecting the characteristics of forest and coastal areas on 30 firefighters (average age: 40.4 ± 9.8 years) who are frequently exposed to dangerous situations. Blood pressure, pulse pressure, and heart rate variability were used as physiological measurement indicators, and the Korean versions of PANAS (positive affect and negative affect schedule), WEMWBS (Warwick-Edinburgh mental well-being scale), and PRS (perceived restorativeness scale) were used as psychological measurement indicators. For four days and three nights, the participants experienced programs at Hallyeohaesang Nature Center and the surrounding mountains, seas, and islands, utilizing forest resources such as trekking on forest trails, walking barefoot, taking aromatic footbaths, meditating in forest oxygen domes, and lying on relaxation chairs, and programs utilizing marine resources such as taking a boat to an island, walking on forest trails with seascape views, and sailing on a yacht. Participants' systolic blood pressure and pulse pressure decreased, and participants' positive emotions increased and negative emotions decreased after the program. There was a statistically significant increase in mental well-being and perceived restorative environment. Through this study, it was found that nature-based outdoor activity programs based on forest and marine resources are effective in physiological and psychological stability of firefighters. It is hoped that the results of this study will be applied to other high-risk workers for PTSD, who have high stress levels, by combining forest healing and marine healing, and expanding the scope and diversity of programs in more diverse environments and conditions.

추출 방법에 따른 삼나무 잎 추출물의 항산화 활성 평가 (Antioxidant Activities of Cryptomeria japonica Leaves Extracts by Extraction Methods)

  • 본 연구는 삼나무 잎 추출물의 항산화 활성을 평가하여 잠재적인 천연 항산화제로의 적용 가능성 구명에 관한 연구이다. 삼나무 잎 추출물은 세 가지 추출방법에 의해 정유, 메탄올 추출물, 열수 추출물로 획득하여 항산화 활성을 평가 하였고, 대조구와 비교하여 DPPH radical 소거활성, FRAP 활성, zanthin oxidase 저해활성, 철이온 chelate 활성 평가를 실시하였다. 그 결과, 메탄올 추출물과 열수 추출물의 항산화 활성이 정유의 항산화 활성보다 우수했다. 정유가 함유하고 있는 테르펜 화합물은 항산화 활성이 비교적 낮은 성분으로 사료된다. 활성이 우수했던 메탄올추출물과 열수 추출물은 다양한 유기용매(n-hexane, chloroform, ethyl acetate, n-butanol, methanol, ethanol, acetone)와 물을 사용하여 분획을 실시하였고, 메탄올 추출물의 acetone 및 물 분획물, 그리고 열수 추출물의 에탄올 분획물이 천연 항산화 성분으로 알려진 ${\alpha}$-tocopherol과 유사한 활성을 나타내거나, 보다 우수한 항산화 활성을 나타냈다. 이들은 공통적으로 수산기 및 당류를 가지는 성분을 함유하고 높은 페놀성 화합물 함량을 나타냈기 때문에 삼나무 잎 추출물은 수산기 및 당류를 가지는 페놀성 화합물에 의해 항산화 활성을 나타내는 것이라 판단되었다.

서양측백나무 잎으로부터 식물정유 추출 및 생리활성 (Extraction and Biological Activities of Essential Oil from Thuja occidental Leaves)

  • 서양측백나무의 잎으로부터 정유를 steam distillation법으로 추출하고, GC-MS로 정유 조성성분을 분석하였으며, 28종의 미생물종을 대상으로 항균활성을, DPPH법으로 항산화 활성을, 집모기에 대한 살충효과를 검정하였다. 서양측백나무 정유 주성분은 monoterpene류였으며, d-isothujone, l-fenchone, ${\alpha}-thujone$, camphor, terpin-4-ol 순으로 나타났다. 정유의 박테리아에 대한 항균력은 박테리아 중 Streptococcus pyogenes에서, 그리고 효모 및 곰팡이 중 Candida albicans에서 가장 높게 나타났다. 특히 서양측백나무 정유는 그램 양성균의 박테리아에 대해 가장 장한 항균력을 보였다. 정유는 DPPH radical 소거능을 보유하고 있었으며, $EC_{50}$ 값은 $36\;{\mu}{\ell}/m{\ell}$이었다. 집모기에 대한 살충 효과는 분무, 훈증, 자연증발 등 처리방법에 따라 다르게 나타났으며, 그 중 분무처리가 가장 효과적인 것으로 나타났다. 이상의 결과는 환경 친화적인 항균 및 살충소재로 이용될 수 있을 것이다.
