Bovine Genome-wide Association Study for Genetic Elements to Resist the Infection of Foot-and-mouth Disease in the Field

  • Foot-and-mouth disease (FMD) is a highly contagious disease affecting cloven-hoofed animals and causes severe economic loss and devastating effect on international trade of animal or animal products. Since FMD outbreaks have recently occurred in some Asian countries, it is important to understand the relationship between diverse immunogenomic structures of host animals and the immunity to foot-and-mouth disease virus (FMDV). We performed genome wide association study based on high-density bovine single nucleotide polymorphism (SNP) chip for identifying FMD resistant loci in Holstein cattle. Among 624532 SNP after quality control, we found that 11 SNPs on 3 chromosomes (chr17, 22, and 15) were significantly associated with the trait at the p.adjust <0.05 after PERMORY test. Most significantly associated SNPs were located on chromosome 17, around the genes Myosin XVIIIB and Seizure related 6 homolog (mouse)-like, which were associated with lung cancer. Based on the known function of the genes nearby the significant SNPs, the FMD resistant animals might have ability to improve their innate immune response to FMDV infection.

  • 구제역(Foot-and-Mouth Disease: FMD)이란 소, 돼지, 양, 염소 등의 cloven-hoofed 동물에서 나타나는 바이러스성 질병으로 입, 코, 유두, 발굽 등에 수포가 형성되는 것이 특징이다. 일곱 가지 혈청형(O, A, C, Asia1, SAT1, SAT2 and SAT3)으로 분류되는 구제역바이러스(Foot-and-Mouth Disease Virus: FMDV)는 single stranded positive RNA virus로 nonenveloped capsid virus이다. Viral genome은 8.2 Kb로 하나의 ORF인 polyprotein으로 되어있으며, 크게 capsid protein coding region인 P1, replication related protein coding region인 P2, RNA dependent RNA polymerase coding region인 P3로 구성된다. FMDV는 respiratory tract의 pharynx epithelial cell에 감염되며, lung epithelial cell에서 replication을 한다. 구제역바이러스는 감염율은 높지만 낮은 치사율을 가진다. 2002년 한국에서 구제역이 발병하여 많은 경제적 손실을 입었다. FMDV의 감염을 조절할 수 있는 조절방법이 없는 실정이며, 현재 많은 나라에서는 구제역바이러스의 감염을 막을 수 있는 효과적인 방법을 연구하고 있다. 본 보고서에서는 FMD에 대한 보다 효과적인 예방법인 DNA vaccine, edible vaccine, peptide vaccine에 대해 고찰하였다.

주요 혈청형 구제역바이러스의 발생분포와 분자역학적 분석 (Geographical distribution and molecular epidemiology of the foot-and-mouth disease viruses of major groups)

  • Foot-and-mouth disease (FMD) virus exists in seven serotypes and is known to be a highly contagious disease that is hard to eradicate from the world. The O, A, Asia1 and SAT2 serotypes commonly infected cattle, sheep and goats during 2007~2009 throughout the world. In particular, the outbreak of the Asia1 serotype in China appeared in all areas from 2005 and is still present. Surprisingly, in 2009, Taiwan reported the first outbreak of the type O serotype since 2001. Then type A appeared in China for the first time since the early 1960s. The virus shows a close relationship to the viruses from Southeast Asia suggesting one or more recent introductions into China in the OIE reports. Recently the subtype of A/Iran05 spread to nearby countries exhibiting genomic evolution. The use of molecular epidemiology is an important tool in understanding and consequently controlling the FMD virus. The phylogenetic analysis with VP1 gene was especially useful for molecular epidemiological studies and showed the same pattern which matches with serotype classification. This paper describes basic information about the disease, and the serotype-specific characteristics and evolution to perform molecular epidemiological analysis. Furthermore, we show the importance of the genetic evolution on the FMD serotypes in global surveillance and molecular epidemiology of FMD for outbreak investigation.

국내 사슴에서 구제역 A형 감염증례 (Infection of Foot and Mouth Disease of Serotype A in Farm-raised Deer in Korea)

  • 2010년초 국내 구제역 발생에서 사슴에서 구제역 A형 감염 사례를 확인하였다. 2002년 O형 구제역이 돼지에서 발생된 이후에 8년만인 2010년 1월에 처음 소에서 A형 구제역이 발생된 이후 6농가에서 발생하였고, 같은 해 3월 발생농가 주변 이동제한 해제를 위한 사슴 항체검사에서 2마리의 사슴에서 구제역 항체양성을 확인하였다. 추가적으로 동일농장에 사육중인 12두에 대한 추가 검사에서 구제역바이러스는 검출되지 않았으며, 비교적 높은 구제역A형에 대한 중화항체가 검출되었다. 그 이후 구제역 A형에 대한 추가 발생은 없었다.

시공간 클러스터링 분석을 이용한 2010~2011 국내 발생 구제역 전파양상 (Temporospatial clustering analysis of foot-and-mouth disease transmission in South Korea, 2010~2011)

  • To investigate the transmission pattern of geographical area and temporal trends of the 2010~2011 foot-and-mouth disease (FMD) outbreaks in Korea, and to explore temporal intervals at which spatial clustering of FMD cases space-time analysis based on georeferenced database of 3,575 burial sites, from 30 November 2010 to 23 February 2011, was performed. The cases represent approximately 98.1% of all infected farms (n = 3,644) during the same period. Descriptive maps of spatial patterns of the outbreaks were generated by ArcGIS. Spatial Scan Statistics, using SaTScan software, was applied to investigate geographical clusters of FMD cases across the country. Overall, spatial heterogeneity was identified, and the transmission pattern was different by province. Cattle have more clusters in number but smaller in size, as compared to the swine population. In addition, spatiotemporal analysis and the comparison of clustering patterns between the first 7 days and days 8 to 14 of the outbreak revealed that the strongest spatial clustering was identified at the 7-day interval, although clustering over longer intervals (8~14 days) was also observed. We further discussed the importance of time period elapsed between FMD-suspected notice and the date of confirmation, and emphasized the necessity of region-specific and species-specific control measures.

Mitigating effect of dietary bromelain on inflammation at the injection site of food-and-mouth disease vaccine

  • The vaccination for foot-and-mouth disease (FMD) is an effective way to control FMD. However, the injection of FMD vaccine causes abnormalities in pork meat by the incidence of lesions at the injection site. This study was conducted to investigate the inhibition effects of dietary bromelain, a natural protease derived from pineapple stems, on the incidence of lesions at the vaccination site on pigs. A total of 335 pigs (LYD [Landrace ${\times}$ Yorkshire ${\times}$ Duroc]; 7-week-old) were randomly allotted to two dietary treatments: control (basic diet) and bromelain treatment (diet supplemented with bromelain 1 kg/ton). The injection of FMD vaccine was conducted on 56- and 84-day-old pigs. Pigs with the bromelain treatment were fed a diet supplemented with bromelain for 14 days from 5 days before the vaccine injection. After slaughtering the pigs, the number of carcasses that had abnormal meat at the injection site of the vaccine and the amount of abnormal meat, discarded meat, and trimmings were recorded. Pork from the bromelain treated pigs had a lower incidence of abnormal meat caused by vaccine injection as well as a lower amount of abnormal meat, discarded meat, and trimmings than those of the control (p < 0.05). Our result suggests that dietary bromelain could improve the quality of pork meat by inhibiting incidence of lesions at the vaccine injection site.

3D프린팅을 활용한 주얼리 디자인 연구 - 곡선 형태를 중심으로 (A Study on Jewelry Design Using 3D-Printing - Focusing on Curved Form)

  • 본 논문은 산업전반에 빠르게 도입되어가고 있는 3D프린팅 기술을 이용하여 주얼리 디자인에 적용하기 위해 시작되었다. 선행연구를 살펴본 결과 디자인 분야에서는 3D출력 방식중 FMD방식이 가장 많이 사용되고 있었다. 하지만 주얼리 디자인에서는 3D프린팅은 제작과정 중 주조 과정에 사용되고 있어 출력방식이 FMD가 아닌 DLP방식을 사용하고 있다. 따라서 연구자는 DLP방식의 3D프린팅을 활용하여 주얼리 디자인에 응용함으로서 주얼리 디자인의 재료적 기능과, 활용 가능성에 대해 연구 작품을 통해 알아보았다. 그 결과 3D프린팅 출력 후 주조과정 없이 주얼리 디자인에 응용하고 에나멜, 정은 등을 함께 활용하여 브로치를 완성함으로서 가볍고 단단하며 다양한 색상의 완성품을 제작하여 적용 가능한 재료임을 알 수 있었다. 또한 기하학 구조가 아닌 주얼리 디자인에서 주로 사용되는 곡선적 디자인을 Rhino CAD로 크기와 형태를 정확히 예측할 수 있어 향후 주얼리 디자인 조형 형태 전개에 쉽게 활용할 수 있는 다양한 발전 가능성을 엿 볼 수 있었다.
