Rheumatoid Fibroblast-like Synoviocytes Downregulate Foxp3 Expression by Regulatory T Cells Via GITRL/GITR Interaction

  • Fibroblast-like synoviocytes (FLS) colocalize with leukocyte infiltrates in rheumatoid synovia. Proinflammatory leukocytes are known to amplify inflammation by signaling to FLS, but crosstalk between FLS and regulatory T cells (Tregs) remains uncharacterized. To address this possibility, we cocultured FLS lines derived from arthritic mice with Tregs. FLS that expressed the ligand for glucocorticoid-induced TNF receptor family-related gene (GITR) decreased expression of Foxp3 and GITR in Tregs in a contact-dependent manner. This effect was abolished by blocking antibody to GITR. On the other hand, the Tregs caused the FLS to increase IL-6 production. These results demonstrate that inflamed FLS license Tregs to downregulate Foxp3 expression via the GITRL/GITR interaction while the Tregs induce the FLS to increase their production of IL-6. Our findings suggest that the interaction between FLS and Tregs dampens the anti-inflammatory activity of Tregs and amplifies the proinflammatory activity of FLS, thereby exacerbating inflammatory arthritis.

재골절예방 서비스 코디네이터 경험 (Experiences of Fracture Liaison Service Coordinators)

  • Purpose: This study aimed to explore the experience of Fracture Liaison Service (FLS) coordinator nurses in a hospital setting. Methods: An in-depth interview was done using a semi-structured questionnaire on 4 FLS coordinator nurses between August 2022 and May 2023. A qualitative content analysis method was used for the data analysis. Results: Three themes of the experience of FLS coordinator were "The absence of identity of FLS coordinator," "struggling with the approach of creating something out of nothing," and "improvement of training and system to establish FLS coordinator role." Conclusion: This study identified three themes about the experience of FLS coordinator nurses in hospital setting. This finding is expected to be useful in developing education programs for FLS coordinator nurses.

  • An efficient algorithm for finding the inverse and the group inverse of the FLS $\gamma-circulant$ matrix is presented by Euclidean algorithm. Extension is made to compute the inverse of the FLS $\gamma-retrocirculant$ matrix by using the relationship between an FLS $\gamma-circulant$ matrix and an FLS $\gamma-retrocirculant$ matrix. Finally, some examples are given.

  • 3D FLS(Foward Looking Sonar)는 상선등의 앞쪽에서 바다내부와 해저를 시각적으로 표현하는 능동 소나이다. 3D FLS에서 수면파에 의한 수중기포의 영향을 감쇄시키기 위한 방법을 논의한다. 실험으로부터 수중기포 감쇄 필터를 사용하여 수중기포의 영향을 감쇄시킨다.

Flow limitation이 일어나는 기도내 위치의 실험적 측정 (Experimental Localization of flow Limiting Segment)

  • A new experimental technique is proposed to localize the flow limiting segment(FLS) during forced expiration. The present technique is based on the pressure drip across FLS and a consequent change in airway resistance, which can provide an accurate and objective location of FLS. During forced expiratory maneuver artificially induced by a strong negative pressure (-100mmHg) applied at the trachea in an anesthetized open chest dog, airway resistance( R) was calculated from air flow and airway pres- sure signals at various airway locations and lung volumes, At the lung volumes above 10 % VC, FLS located in the trachea 6cm lower from the larynx. With the lung volume decreased below 8% VC, FLS jumped upstream to End-3rd generation of the airway. These results were similar with the previous reports from excised dog lungs, which demonstrated the validity of the present technique. Since the present technique provides a more objective measure of FLS location, it would be useful in future studies of expiratory flow limitation.

Synovial Cell Migration is Associated with B Cell Activating Factor Expression Increased by TNFα or Decreased by KR33426

  • Fibroblast-like synoviocytes (FLS) play a crucial role in initiating rheumatoid arthritis. B-cell activating factor (BAFF) plays a role in FLS survival as well as in B cell maturation and maintenance. Here, we investigated whether tumor necrosis factor (TNF)-α-induced BAFF expression controls FLS migration and whether BAFF expression in FLS could be regulated by KR33426 which is the inhibitor of BAFF binding to BAFF receptors (BAFF-R) by using MH7A synovial cells transfected with the SV40 T antigen. More TNF-α-treated cells migrated compared to the control. TNF-α increased BAFF expression in FLS, significantly. FLS migration was inhibited by the transfection with BAFF-siRNA. KR33426 also inhibited BAFF expression increased by TNF-α treatment in FLS as judged by western blotting, PCR, and transcriptional activity assay. Kinases including JNK, p38 and Erk were activated by TNF-α treatment. While JNK and p38 were inhibited by KR33426 treatment, no changes in Erk were observed. Transcription factors including p65, c-Fos, CREB and SP1 were enhanced by TNF-α treatment. Among them, c-Fos was inhibited by KR33426 treatment. Small interference(si)-RNA of c-fos decreased BAFF transcriptional activity. FLS migration induced by TNF-α was inhibited by the transfection with BAFF-siRNA. KR33426 increased Twist, Snail, Cadherin-11 and N-Cadherin. In contrast, KR33426 decreased E-cadherin and TNF-α-enhanced CCL2. Taken together, our results demonstrate that synovial cell migration via CCL2 expression could be regulated by BAFF expression which is decreased by KR33426 and c-Fos-siRNA. It suggests for the first time that the role of BAFF-siRNA on FLS migration might be matched in the effect of KR33426 on BAFF expression.

IT2TSK 퍼지논리 기반 전력부하 예측 모델 설계에 관한 연구 (Design of Electric Power Load Forecasting Model based on IT2TSK FLS)

  • In most cases, the use of electric power is associated with the economic scale of a nation closely. Thus, the electric power load forecasting plays an important role for the national economic plan. This paper deals with the design method for the electric power load forecasting system. In this paper, RCR-MA data processing, which can make the complex properties of the original data form simple, is proposed. Next, IT2TSK FLS, which can reflect the uncertainty of data more than T1TSK FLS, is applied. Consequently, the structural advantage of the proposed system can improve the forecasting accuracy, and is verified by using two types of electric power data.

  • Due to the limitations of onshore wind power, the wind power industry is currently transitioning to offshore wind power. There has been active research on the development of a floating LiDAR system (FLS) that is easy to install at a low cost. The Carbon Trust published a commercialization roadmap for FLS in 2013, and an updated version was released in 2018, taking into account industry experience. The roadmap divides the development maturity of FLS into three stages: Stage 1 (prototype), Stage 2 (pre-commercialization), and Stage 3 (commercialization), each of which requires availability and accuracy assessment. The results must meet the requirements of the Key Performance Index (KPI) for each stage. Therefore, when developing FLS, the motion compensation algorithm of the FLS is essential because the LiDAR can produce incorrect measurements of wind speed and direction due to the six degrees of freedom in motion. In this study, we implemented the FLS motion compensation algorithm developed by Nassif, F.B. et al. and validated it using data provided by Fraunhofer. In conclusion, the results showed that the determination coefficients of wind speed and wind direction were improved compared to those obtained from the met mast.

자유학기제 운영에 대한 가정과 교사의 인식 분석 (A Study on Home Economics Teachers' Perception in Free Learning Semester)

  • 본 연구는 자유학기제가 전면 시행된 시점에 중학교 가정과 교사들의 자유학기제에 대한 인식과 이해 정도를 알아봄으로써 향후 자유학기제가 학교교육과정에서 효율적으로 운영될 수 있는 방안 및 가정과 교육이 나아가야할 방향을 탐색해보고자 실시하였다. 본 조사대상 중학교 가정과교사들은 자유학기제가 운영되는 목적으로 학생의 진로탐색 및 1차적 진로 의사결정이라고 인식하고 있었으며, 학생 참여 중심 수업방법을 핵심과제로 하여 꿈과 끼를 탐색하는 교육 방향으로 운영되어야 한다고 생각하고 있었다. 자유학기제의 도입취지나 목적에 대한 이해도도 높은 편이었다. 그러나 자유학기제를 운영하기 위한 자료나 정보를 취득하는 데 있어 어려움을 느끼는 것으로 조사되었으며 지역사회 기관과의 연계활동도 충분하지 못한 것으로 나타났다. 연구학교 교사들은 자유학기제 운영에 대한 준비가 필요하다는 것을 인식하고 있었으나, 일반학교 교사들의 경우 자유학기제 관련 교사연수의 경험이 부족한 것으로 조사되었다. 연수 경험이 없는 연구학교와 일반학교 교사 모두 자유학기제 관련 내용의 연수의 필요성을 느끼고 있었다. 자유학기제에 대한 가정과교사의 역할로는 생애설계적 관점에서 진로교육을 지도하고 학생들이 자신의 생애를 자기 주도적으로 이끌어 나갈 수 있는 역량을 길러주어야 하는 것으로 인식하고 있었다. 중학교 시기는 자아, 소질, 기회 등을 탐색하며 여러 가지 실질적인 경험을 통해 자신의 적성이 무엇인지 탐색해볼 수 있는 매우 중요한 시기이다. 따라서 자유학기를 통해 학생들이 자신의 꿈과 끼를 찾을 수 있고, 여러 가지 다양한 길에 대하여 생각해 볼 수 있도록, 앞으로 가정 교과를 통해 생애설계적인 진로계획을 수립하여 자기 주도적으로 인생을 이끌어 나갈 수 있는 역량을 길러주어야 할 것이다. 또한, 교사들은 자유학기가 한 학기의 이벤트성 운영으로 끝나지 않기 위해서 일반학기에도 교과를 통한 진로교육 수업으로 연계성을 이어나갈 수 있도록 하는 노력이 필요하다.

MicroRNA-27a Inhibits Cell Migration and Invasion of Fibroblast-Like Synoviocytes by Targeting Follistatin-Like Protein 1 in Rheumatoid Arthritis

  • Fibroblast-like synoviocytes (FLS) with aberrant expression of microRNA (miRNA) are critical pathogenic regulators in rheumatoid arthritis (RA). Previous studies have found that overexpression or silencing of miRNA can contribute to the development of miRNA-based therapeutics in arthritis models. In this study, we explored the effects of miR-27a on cell migration and invasion in cultured FLS from RA patients. We found that miR-27a was markedly downregulated in the serum, synovial tissue, and FLS of RA patients. Meanwhile, the expression of follistatin-like protein 1 (FSTL1) was upregulated, which suggests that FSTL1 plays a key role in RA development. The results of a Transwell assay showed that miR-27a inhibited FLS migration and invasion. However, miR-27a inhibition promoted the migration and invasion of FLS. In addition, the down-regulated expression of matrix metalloproteinases (MMP2, MMP9, and MMP13) and Rho family proteins (Rac1, Cdc42, and RhoA) was detected after treatment with miR-27a in RA-FLS by quantitative reverse transcription-PCR and western blot analysis. Then, a luciferase reporter assay validated that miR-27a targeted the 3-untranslated region (3'-UTR) of FSTL1. Moreover, miR-27a caused a significant decrease of FSTL1. In addition, the expression of TLR4 and $NF{\kappa}B$ was inhibited by miR-27a but increased by FSTL1 overexpression. In conclusion, we found that miR-27a inhibited cell migration and invasion of RA-FLS by targeting FSTL1 and restraining the $TLR4/NF{\kappa}B$ pathway.
