Pop-up satellite archival tag (PSAT) 체외부착에 따른 해산어의 생리학적 반응 (Physiological Responses of Marine Fish to External Attachment of Pop-up Satellite Archival Tag (PSAT))

  • Recently, the pop-up satellite archival tag (PSAT) is being used in studies as a method of using satellites for monitoring organisms. Because PSATs are attached directly on the organism being monitored, it is used mostly on larger species since the health of the organism is an important concern. For this reason, PSAT-based surveys are lacking in Korea with no studies on the physiological responses of organisms with PSAT attached. Accordingly, as a basic biomonitoring study using PSAT, the present study investigated the physiological changes in fish in response to the attachment of PSAT. The present study used red seabream (Pagrus major) and Korean rockfish (Sebastes schlegelii) as the experimental fish. The PSAT was attached to the muscle below the experimental fish's dorsal fin using a mono filament (n = 3). To investigate the changes in physiological responses according to PSAT attachment, blood samples were collected from all experimental fish, including the control (n = 3), at 1, 7, 14 and 21 days after the attachment. Upon blood sample collection, whole blood was used to measure hematocrit and hemoglobin levels. After separating the plasma, the separated plasma was used to measure the GOT, GPT, glucose, total protein, and cholesterol levels. Meanwhile, the plasma cortisol, superoxide dismutase, and catalase levels were measured using the ELISA method. The results showed that attaching the tag did not have any impact on the immunity and stress response of the experimental fish. The findings in this study also demonstrated the possibility of using PSAT for studying relatively smaller species living in the coastal waters of Korea.

Effects of Turbid Water on Fish Ecology in Streams and Dam Reservoirs

  • Turbid water or suspended sediment is associated with negative effects on aquatic organisms; fish, aquatic invertebrate, and periphyton. Effects of turbid water on fish differ depending on their developmental stage and a level of turbidity. Low turbid water may cause feeding and predation rates, reaction distance, and avoidance in fish, and it could make fish to die under high turbidity and long period. Therefore, it is very important to find out how turbid water or suspended sediment can affect fish in domestic watersheds. The objectives of this study were 1) to introduce international case studies and their standards to deal with suspended sediment, 2) to determine acute toxicity in 4 major freshwater fishes, and 3) to determine in relation to adverse effect of macroinvertebrates and fish. Impacts of turbid water on fish can be categorized into direct and indirect effects, and some factors such as duration and frequency of exposure, toxicity, temperature, life stage of fish, size of particle, time of occurrence, availability of and access to refugia, etc, play important role to decide magnitude of effect. A review of turbidity standard in USA, Canada, and Europe indicated that each standard varied with natural condition, and Alaska allowed liberal increase of turbidity over natural conditions in streams. Even though acute toxicity with four different species did not show any fatal effect, it should be considered to conduct a chronic test (long-term) for more detailed assessment. Compared to the control, dominance index of macroinvertebrates was greater in the turbid site, whereas biotic index, species diversity index, species richness index, and ecological score were smaller in the turbid site. According to histopathological analysis with gills of macroinvertebrate and fishes, morphological and physiological modification of gills due to suspended sediments can cause disturbance of respiration, excretion and secretion. In conclusion, in order to maintain good and healthy aquatic ecosystem, it is the best to minimize or prevent impact by occurrence of turbid water in stream and reservoir. We must make every effort to maintain and manage healthy aquatic ecosystem with additional investigation using various assessment tools and periodic biomonitoring of fish.

An EST-based approach for identifying genes expressed in the gills of olive flounder Paralichthys olivaceus

  • Analysis of expressed sequence tags (ESTs) is an efficient approach for gene discovery, expression profiling, and development of resources useful for functional genomics studies. As part of studies on the immune system of olive flounder, a total of 251 EST sequences from gill cDNA library were generated to identify and characterize important genes in the immune machanisms of olive flounder. Of the 251 clones, 126 clones (50.2%) were identified as orthologues of known genes from olive flounder and other organisms. Among the 126 EST clones, 16 clones (12.7%) were representing 9 unique genes identified as homologous to the previously reported olive flounder ESTs, 100 clones (79.4%) representing 103unique genes were identified as orthologs of known genes from other organisms. We also identified several kinds of immune associated proteins, indicating EST as a powerful method for identifying immune related genes of fish as well as identifying novel genes. Further studies using cDNA microarrays are needed to identify the differentially expressed transcripts after disease infection.

Anti-cancer Mechanism of Docosahexaenoic Acid in Pancreatic Carcinogenesis: A Mini-review

  • Pancreatic cancer is a highly aggressive malignant tumor of the digestive system and radical resection, which is available to very few patients, might be the only possibility for cure. Since therapeutic choices are limited at the advanced stage, prevention is more important for reducing incidence in high-risk individuals with family history of pancreatic cancer. Epidemiological studies have shown that a high consumption of fish oil or ${\omega}3-polyunsaturated$ fatty acids reduces the risk of pancreatic cancers. Dietary fish oil supplementation has shown to suppress pancreatic cancer development in animal models. Previous experimental studies revealed that several hallmarks of cancer involved in the pathogenesis of pancreatic cancer, such as the resistance to apoptosis, hyper-proliferation with abnormal $Wnt/{\beta}-catenin$ signaling, expression of pro-angiogenic growth factors, and invasion. Docosahexaenoic acid (DHA) is a ${\omega}3-polyunsaturated$ fatty acid and rich in cold oceanic fish oil. DHA shows anti-cancer activity by inducing oxidative stress and apoptosis, inhibiting $Wnt/{\beta}-catenin$ signaling, and decreasing extracellular matrix degradation and expression of pro-angiogenic factors in pancreatic cancer cells. This review will summarize anti-cancer mechanism of DHA in pancreatic carcinogenesis based on the recent studies.

어류군집 특성과 하안형태복잡도와의 관계 (Relationship between fish assemblages community and Streamline complexity)

  • Numerous studies suggested that fish assemblage structure reflects the status of stream ecosystems. The status of streams integrity, including various trophic levels, water quality and habitat degradation, can be assessed by fish assemblages. In this study, we investigated the relationships between fish assemblages and streamline geometry of streams. Previous studies suggested that geomorphologic parameter can be a critical factor of permeability between adjacent two systems. From a landscape ecological perspective, edges may partially control the flow rate of energy between two adjacent systems. Thus, the Streamline geometry can be a geomorphologic parameter that exhibits the integrity of stream. We selected the Nakdong river for study areas, which is one of major rivers and the longest (525 km) River in South Korea. We used the revised IBI representing overall ecological characteristics of Korean fish assemblages and eight sub-assessment criteria of IBI, collected from 82 sampling sites in the Nakdong River. For calculating the Streamline geometry, we measured fractal dimension index that generally used in biology, ecology and landscape ecology. We used the digital land-use/land-cover map and generated a 1-km buffer for each sampling site and refined the shape of the Streamlines. Pearson correlation analyses were performed between Streamline geometry and IBI and sub-assessment criteria of IBI. The results show that IBI and eight sub-assessments of fish are significantly correlated with geometry of Streamline. The fractal dimension of Streamline geometry were related with IBI (r = 0.48) and six sub-assessments of IBI, including total number of native fish and native species, the number of riffle benthic species, sensitive species, tolerant species and native insectivore. Especially, the number of tolerant species(r = -0.52) and native insectivore(r = 0.52) show strong correlation with geometry of Streamline. These results indicate that lower Streamline geometry can result in poor fish assemblages, while higher geometry of Streamline can enhance fish assemblages by potentially supplying insects and better habitat conditions. We expect the results of our study to be useful for stream restoration and management. However, we see the necessity of study investigating the mechanisms how Streamline geometry affect fish assemblages.

황복의 마취에 미치는 수온 및 연령과 MS-222 농도의 영향 (Effect of Water Temperature, Fish Age, and MS-222 Concentration on the Anesthetization of River Pufferfish, Takifugu obscurus)

  • 황복(Takifugu obscurus)은 양식 어종 중 하나이며, 유전체 연구에서도 의미 있는 종이다. 양식 어종의 경우 어류 마취제를 사용하면 양식 과정에 있어서 어류를 좀 더 쉽게 다룰 수 있다. 그러나, 지금까지 황복의 효과적인 마취와 회복을 위해 필요한 적절한 조건에 대한 연구는 없었다. 본 연구는 가장 일반적인 어류 마취제인 MS222를 이용하여 황복을 마취하는 데 필요한 연령, 수온 및 마취 농도 등 최적의 조건을 파악하는 데 그 목적이 있다. 실험에서는 세 가지 다른 수온(20℃, 24℃, 28℃) 및 마취 농도(125 mg/L, 150 mg/L, 175 mg/L)와 두 가지 다른 어류 연령(1년생과 2년생)을 테스트하였다. 실험 결과 황복의 적절한 마취 조건은 24℃와 28℃에서 MS-222 150 mg/L에서 175 mg/L까지의 범위였다. 2년생 어류의 경우도 28℃에서 MS-222 150 mg/L에서 175 mg/L의 범위를 보였다. 부작용과 위험을 최소화하기 위한 효과적인 마취 농도는 1년생 어류의 경우 24℃에서 MS-222 150 mg/L, 2년생 어류의 경우 28℃에서 MS-222 175 mg/L가 권장된다.

사료 내 해조류(톳, 감태) 첨가가 돌돔(Oplegnathus fasciatus) 치어의 비특이적 면역반응에 미치는 영향 (Effect of Dietary Supplementation with Alga (Hizikia fusiformis and Ecklonia cava) on the Non-specific Immune Responses of Parrot Fish Oplegnathus fasciatus)

  • Two feeding trials were conducted to determine the effects of dietary supplementation with Hizikia fusiformis or Ecklonia cava on the non-specific immune responses of parrot fish Oplegnathus fasciatus. Fish were fed experimental diets to which H. fusiformis or E. cava powder were added to final concentrations of 0, 2, 4 and 6%, respectively. After feeding for two weeks, phagocytic activity was significantly higher in fish fed diets containing H. fusiformis, but not E. cava, than in fish fed the basal diet. Lysozyme activity was significantly increased in the fish fed diets containing 6% H. fusiformis and E. cava. Myeloperoxidase activity was also significantly higher in fish fed diets containing 2 and 4% H. fusiformis, as compared to the basal diet, but not in those fish fed E. cava. These two studies indicate that dietary supplementation with H. fusiformis or E. cava could enhance the innate immune responses of parrot fish during their growth stage.

Biodynamic understanding of mercury accumulation in marine and freshwater fish

  • Mercury (Hg) is a global environmental pollutant that has been the cause of many public concerns. One particular concern about Hg in aquatic systems is its trophic transfer and biomagnification in food chains. For example, the Hg concentration increases with the increase of food chain level. Fish at the top of food chain can accumulate high concentrations of Hg (especially the toxic form, methylmercury, MeHg), which is then transferred to humans through seafood consumption. Various biological and physiochemical conditions can significantly affect the bioaccumulation of Hg-including both its inorganic (Hg(II)) and organic (MeHg) forms-in fish. There have been numerous measurements of Hg concentrations in marine and freshwater fish worldwide. Many of these studies have attempted to identify the processes leading to variations of Hg concentrations in fish species from different habitats. The development of a biokinetic model over the past decade has helped improve our understanding of the mechanisms underlying the bioaccumulation processes of Hg in aquatic animals. In this review, I will discuss how the biokinetic modeling approach can be used to reveal the interesting biodynamics of Hg in fish, such as the trophic transfer and exposure route of Hg(II) and MeHg, as well as growth enrichment (the increases in Hg concentration with fish size) and biomass dilution (the decreases in Hg concentration with increasing phytoplankton biomass). I will also discuss the relevance of studying the subcellular fates of Hg to predict the Hg bioaccessibility and detoxification in fish. Future challenges will be to understand the inter- and intra-species differences in Hg accumulation and the management/mitigation of Hg pollution in both marine and freshwater fish based on our knowledge of Hg biodynamics.

Typical Patterns of Ecological Engineering in Southern China

  • With the emergence and development of ecological engineering in China, and also the research and demonstration of Chinese Ecological Agriculture (CEA), in recent 10 years many types of ecological engineering have been appeared in Southern China, they could be classified into 4 typical patterns. These typical patterns are; 1. Ecological Building (EB), such as ecological public toilet, ecological garden, ecological integrated building, farmers' ecological household, the combine of ecological building and CEA; 2. Ecological Engineering in Paddy Field (EEPF), like rice-fish system, water caltrop-wild ricestem-fish system, mulberry-silkworm-pig-fish system; 3. Ecological Engineering of Agroforestry Systems (EEAF), as tea-food crops intercropping system, rubber-tea-chicken system, bamboo-chicken system, fruit tree-chicken system, metasequoia-fish-duck system; 4. Ecological Engineering Linked with Biogas (EELB), such as animal (pig)-biogas-fish system, animal (pig)-biogas-fruit tree system, animal-biogas-food crops system. Some case studies were presented in the paper. It is showed that these typical patterns have better ecological, social and economic benefits, now they are deeply been extending in southern China.

어류에 미치는 인위적인 수중소음 영향과 피해기준에 대한 고찰 (Overview of anthropogenic underwater sound effects and sound exposure criteria on fishes)

  • A scientific and objective sound exposure criterion for underwater sound damage on fish has been required since there has been many disputes between an underwater sound maker and a fish damage receiver. The existing criteria are still incomplete scientifically owing to a degree of variability of underwater sounds, diversity of fish hearing sensitivity and damage types, etc. This study reviews existing studies on a hearing mechanism of fish species, manmade underwater sound characteristics and sound exposure assessment parameters, and recent sound exposure criteria. A governing equation for damage coverage estimation and damage coverage dependency on sound source level, ambient noise and transmission loss are also reviewed and interpreted based on sound exposure environments. The foreign and Korean (National Environmental Dispute Medication Commission) criteria are reviewed and compared based on scientific aspects. In addition, the deficit and limit of Korean criteria are presented. The objective of this study is to give a direction for related researches and legislation of sound exposure criteria on fish.
