Molecular Analysis of the Salmonella Typhimurium tdc Operon Regulation

  • Efficient expression of the Salmonella Typhimurium tdc ABCDEG operon involved in the degradation of L-serine and L-threonine requires TdcA, the transcriptional activator of the tdc operon. We found that the tdcA gene was transiently activated when the bacterial growth condition was changed from aerobic to anaerobic, but this was not observed if Salmonella was grown anaerobically from the beginning of the culture. Expression kinetics of six tdc genes after anaerobic shock demonstrated by a real-time PCR assay showed that the tdc CDEG genes were not induced in the tdcA mutant but tdcB maintained its inducibility by anaerobic shock even in the absence of tdcA, suggesting that an additional unknown transcriptional regulation may be working for the tdcB expression. We also investigated the effects of nucleoid-associated proteins by primer extension analysis and found that H-NS repressed tdcA under anaerobic shock conditions, and fis mutation delayed the peak expression time of the tdc operon. DNA microarray analysis of genes regulated by TdcA revealed that the genes involved in N-acetylmannosamine, maltose, and propanediol utilization were significantly induced in a tdcA mutant. These findings suggest that Tdc enzymes may playa pivotal role in energy metabolism under a sudden change of oxygen tension.

Discrimination of Korean Native Chicken Lines Using Fifteen Selected Microsatellite Markers

  • In order to evaluate the genetic diversity and discrimination among five Korean native chicken lines, a total of 86 individuals were genotyped using 150 microsatellite (MS) markers, and 15 highly polymorphic MS markers were selected. Based on the highest value of the number of alleles, the expected heterozygosity (He) and polymorphic information content (PIC) for the selected markers ranged from 6 to 12, 0.466 to 0.852, 0.709 to 0.882 and 0.648 to 0.865, respectively. Using these markers, the calculated genetic distance (Fst), the heterozygote deficit among chicken lines (Fit) and the heterozygote deficit within chicken line (Fis) values ranged from 0.0309 to 0.2473, 0.0013 to 0.4513 and -0.1002 to 0.271, respectively. The expected probability of identity values in random individuals (PI), random half-sib ($PI_{half-sibs}$) and random sibs ($PI_{sibs}$) were estimated at $7.98{\times}10^{-29}$, $2.88{\times}10^{-20}$ and $1.25{\times}10^{-08}$, respectively, indicating that these markers can be used for traceability systems in Korean native chickens. The unrooted phylogenetic neighbor-joining (NJ) tree was constructed using 15 MS markers that clearly differentiated among the five native chicken lines. Also, the structure was estimated by the individual clustering with the K value of 5. The selected 15 MS markers were found to be useful for the conservation, breeding plan, and traceability system in Korean native chickens.

OBSERVATIONS OF STAR FORMATION INDUCED BY GALAXY-GALAXY AND GALAXY-INTERGALACTIC MEDIUM INTERACTIONS WITH AKARI

  • Nearby spiral galaxies M101 and M81 are considered to have undergone a galaxy-galaxy interaction. M101 has experienced HI gas infall due to the interaction. With AKARI far-infrared (IR) photometric observations, we found regions with enhanced star forming activity, which are spatially close to regions affected by the interaction. In addition, the relation between the star formation rate (SFR) and the gas content for such regions shows a significant difference from typical spiral arm regions. We discuss possible explanations for star formation processes on a kiloparsec scale and the association with interaction-triggered star formation. We also observed the compact group of galaxies Stephan's Quintet (SQ) with the AKARI Far-infrared Surveyor (FIS). The SQ shows diffuse intergalactic medium (IGM) due to multiple collisions between the member galaxies and the IGM. The intruder galaxy NGC 7318b is currently colliding with the IGM and causes a large-scale shock. The 160 micron image clearly shows the structure along the shock ridge as seen in warm molecular hydrogen line emission and X-ray emission. The far-IR emission from the shocked region comes from the luminous [CII]$158{\mu}m$ line and cold dust (~ 20 K) that coexist with molecular hydrogen gas. Survival of dust grains is indispensable to form molecular hydrogen gas within the collision age (~ 5 Myr). At the stage of the dusty IGM environment, [CII] and $H_2$ lines rather than X-ray emission are powerful cooling channels to release the collision energy.

AKARI MID- TO FAR-INFRARED OBSERVATIONS OF DIFFUSE GALACTIC EMISSION

  • We have collected dozens of mid-infrared spectra showing UIR bands from diffuse Galactic emitting regions with the AKARI's Infrared Camera (IRC) onboard AKARI, as part of the ISMGN Mission Program. The datasets cover various directions in the inner Galactic Plane ($|l|$ < 70 deg), in the outer Galactic Plane ($|l|$ > 70 deg), and in the off-Plane ($|b|$ > 2 deg). The variations in the UIR band ratios are examined in terms of the radiation environments judged from the far-infrared ($50-170{\mu}m$) spectral energy distribution (SED) made with AKARI/FIS All Sky Survey data at each slit position where mid-IR spectra were obtained. We have found that the band ratios of $6.2{\mu}m/11.2{\mu}m$ and $7.7{\mu}m/11.2{\mu}m$ toward the inner Galaxy are systematically higher than those toward the outer Galaxy and off the Galactic plane. Likely causes of the variations in properties of UIR bands in diffuse emission on a Galactic scale are discussed in this paper.

Genomic Heterogeneity of Chicken Populations in India

  • A comprehensive genome profiling study was undertaken based on automated genotyping and analysis of 20 microsatellite markers that involved 155 birds representing eight different populations. The distribution of microsatellite markers in each of these breeds helped us to decipher genetic heterogeneity, population genetic structure and evolutionary relationships of the present day chicken populations in India. All the microsatellite loci utilized for the analysis were polymorphic and reasonably informative. A total of 285 alleles were documented at 20 loci with a mean of 14.25 alleles/locus. A total of 103 alleles were found to be population/strain specific of which, only 30 per cent had a frequency of more than 10. The mean PIC values ranged from 0.39 for the locus ADL158 to 0.71 for loci MCW005 or ADL267 across the genomes and 0.55 in Dahlem Red to 0.71 in Desi (non-descript), among the populations. The overall mean expected and observed heterozygosity estimates for our populations were 0.68 and 0.64, respectively. The overall mean inbreeding coefficients (FIS) varied between -0.05 (Babcock) and 0.16 (Rhode Island Red). The pairwise FST estimates ranged from 0.06 between Aseel and Desi (non-descript) to 0.14 between Dahlem Red and Babcock. The Nei's genetic distance varied from 0.30 (WLH-IWD and WLH-IWF) to 0.80 (Dahlem Red and Babcock. Phylogenetic analysis grouped all the populations into two main clusters, representing i) the pure breeds, Dahlem Red and Rhode Island Red, and ii) the remaining six populations/strains. All the chicken populations studied were in the state of mild to moderate inbreeding except for commercial birds. A planned breeding is advised for purebreds to revive their genetic potential. High genetic diversity exists in Desi (non-descript), local birds, which can be exploited to genetically improve the birds suitable for backyard poultry.

기후변화를 고려한 한강유역 저수지의 적응능력 평가 (Adaptation Capability of Reservoirs Considering Climate Change in the Han River Basin, South Korea)

  • 변화할 것으로 예측되고 있는 기후환경에서 현재 수공구조물의 적응능력을 평가하고 지속가능한 시스템을 만들고자 하는 것은 최근의 수자원 관리의 핵심이다. 본 연구에서는 한강수계 5개의 댐을 대상으로 다양한 유입량에 따른 방류량 및 저류량의 변화를 퍼지 추론 시스템을 이용하여 분석하였다. 유입량의 변화에 대한 최대 저류량 및 최소 저류량의 변화를 저수지의 적응능력이라 정의하여 분석한 결과, 저류용량이 상대적으로 작은 광동댐은 유입량의 급격한 증가를 감당하기 어려우며, 소양강댐은 강우량 변화에 대한 적응능력이 가장 뛰어난 것으로 판단되었다. 그러나 퍼지 추론 시스템은 소속함수를 임의로 지정하고, 과거 자료를 이용하여 검증하기가 용이하지 않으므로, 보다 정확하고 효율적인 모의를 위해 소양강댐을 대상으로 적응 신경망-퍼지추론 시스템을 구축하여 적응능력을 평가하였다. 과거 자료의 빈도분석 결과와 기후변화 시나리오를 바탕으로 구축된 9개의 강우 시나리오에 대해 소양강댐의 방류량 및 저류량을 모의한 결과, 유입 시나리오에 따라 매우 상이한 저수지 운영결과를 나타냄을 알 수 있으며, 적응 신경망-퍼지 추론 시스템이 변화하는 강우량과 패턴에도 불구하고 안정적으로 저수지를 운영함을 알 수 있었다.
