Application of Loop-Mediated Isothermal Amplification (LAMP) Assay to Rapid Detection of Methicillin-Resistant Staphylococcus aureus from Blood Cultures

  • We developed the multiplex LAMP assay using 16S rRNA, femA and mecA genes for direct detection of the methicillin resistance in Staphylococci from positive blood culture. To simultaneously recognize Staphylococci genus, S. aureus and methicillin resistance, three sets of six primers for 16S rRNA, femA and mecA were designed, respectively. The performance of LAMP assay was affirmed using VITEK system for the phenotypic methods of identification and for oxacillin and cefoxitin antimicrobial susceptibility. The optimal condition for LAMP assay was obtained under $64^{\circ}C$ for 50 min. The detection limit was determined to be of 20 copies and CFU/reaction ($10^4CFU/mL$). For clinical application of comparison with phenotypic methods, the sensitivity and specificity of the LAMP with femA gene for detecting S. aureus was 95.31% and 100%, respectively. The sensitivity and specificity of the LAMP with mecA gene for detecting methicillin resistance was 98.46% and 100%, respectively. The multiplex LAMP assay with femA and mecA gene successfully detected all of MRSA (38 isolates) isolates from 103 Staphylococci in blood cultures. The LAMP assay developed in this study is sensitive, specific, and of excellent agreement with the phenotypic methods.

Analyzing the Effects of the Initiation Sequences of the MDS Blasting Method on Rock Fracturing Using SPH-FEM Coupling Technique (SPH-FEM 연계기법을 이용한 MDS 발파법의 기폭패턴별 암석파괴 효과 분석)

  • The conventional bench blasting method uses the bottom initiation in all blast holes in a round, whereas the MDS (mixture detonation system) method applies the bottom and top initiations alternately according to the spatial position or temporal sequence of each blast hole. The former and latter are respectively called the SMDS (spatial MDS) and TMDS (temporal MDS) methods. Another variant called MMDS (modified MDS) is designed for the specific use in the site having a fly-rock problem. This study compares the MDS method to the conventional method in the aspect of rock fracturing effect. The comparison is made by numerical simulations for a two-row bench blasting model in the LS-DYNA. The SPH-FEM coupling method is utilized for constructing the blasting model. The SPH elements are used for the rock in the near-field region of the blast holes, and the FEM elements for that in the far-field region. The RHT material model is used for the rock. As a result of the simulations, it was found that up to 0.4 m deeper damaged zone was appeared in the SMDS method than in the conventional method for the case of the burden 1.6 m and bench height 3.0 m. In addition, the fly-rock velocity to the normal direction of the bench slope was appeared about 2.0 m/s lower in the MMDS method compared to the other methods.

  • This paper describes the loss analysis based on load conditions of the air conditioning compressor motors using variable dc-link voltage. The losses of PMSM (Permanent Magnet Synchronous Motor) should be analyzed by the PWM (Pulse Width Modulation) output of inverter. The harmonic loss by the PWM cannot consider that using the current source analysis of the inverter. In addition, when the voltage of dc-link is variable with the condition of variable speed and load conditions in motor, the losses of motor are also changeable, however it is hard to analyze those losses by only electromagnetic finite element method (FEM). Therefore, this paper proposes the analysis method considering the carrier frequency of the inverter and the varying state of the dc-link voltage through the FEM-control coupled analysis. Using proposed analysis method, additional core loss and eddy current loss of permanent magnet caused by PWM could be analyzed. Finally, the validity of the proposed analysis method is verified through the comparison the result of coupled analysis with experiment.

  • This paper deals with friction-induced vibration of disc brake system under constact friction coefficient. A linear, finite element model to represent the floating caliper disc brake system is proposed. The complex eigenvalues are used to investigate the dynamic stability and in order to verify simulations which are based on the FEM model, The comparison of experimental and analytical results shows a good agreement and the analysis indicates that mode coupling due to friction force and geometric instability is responsible fur disc brake squeal. And the Front brake system reduced the squeal noise using design of experiment method(DOE). This helped to validate the FEM model and establish confidence in the simulation results. Also they may be useful during real disk brake model.

Free Vibration Analysis of Two Identical Rectangular Plates Coupled with Fluid (유체로 연성된 동일한 두 직사각 평판의 고유진동 해석)

  • In order to investigate the vibration characteristics of fluid-structure interaction problem, we modeled two identical rectangular plates coupled with fluid. A commercial computer code, ANSYS was used to perform finite element analysis and FEM solutions were compared with the experimental results to verify the finite element model. As a result, comparison of FEM and experiment showed good agreement, and the transverse vibration modes, in-phase and out-of-Phase, were observed alternately in the fluid-coupled system. The effect of fluid gap size on the fluid-coupled natural frequency were investigated. It was shown that the mode numbers increased, the normalized natural frequencies monotonically increased. And it was also found that an increase of the fluid gap reduced the coupled natural frequencies for the in-phase modes but increased the coupled natural frequencies for the out-of phase modes, and eventually converged to the results of an infinite fluid gap.

  • In order to study the vibration characteristics of fluid-structure interaction problem, two rectangular plates coupled with bounded fluid were investigated. Experimental modal analyses were carried out to extract the modal parameters of the system. Additionally. finite element modal analyses performed using a commercial computer code, ANSYS. The FEM solutions were compared with the experimental solutions to verify the finite element model. As a result, the comparison between the experiment and FEM results showed excellent agreement. The transverse vibration modes, in-phase and out-of-phase, were observed alternately in the fluid-coupled system. The thickness effect of the plates on the fluid-coupled natural frequencies and mode shapes was investigated for two different cases with the identical thickness and the unequal thickness. It was found that the coupled natural frequencies increase with the thickness for the identical plates regardless of the mode phase. The experimental and the finite element analysis results showed that the out-of-phase mode shapes were deviated from the symmetrical mode shapes in the plate transverse direction fur the unequal plate thickness case.

Effect of Water Level on the Hydroelastic Vibration of Two Rectangular Plates Coupled with Water (물로연성된 두 직사각평판의 접수진동에 대한 수위의 영향)

  • The effect of water level on the free vibration of a partially water-filled two rectangular plates structure was investigated by experimental modal analysis and finite element analysis using ANSYS computer program. Modal parameters of two rectangular plates coupled with water were obtained by means of experiment and the FEM solutions were compared with the experimental solutions to verify the finite element model. As a result, the comparison between the experiment and FEM results showed excellent agreement. The transverse vibration modes, in-phase and out-of-phase, were observed alternately in the fluid-coupled system. The effect of water level and water gap size on the fluid-coupled natural frequency were investigated. It was found that the natural frequency of the partially water-filled two rectangular plates are not proportional to the water level, but depend on mode number of plates.

The comparison of the fatigue crack initiation life in a notch (노치부의 피로균열발생 수명 비교)

  • For the life evaluating of notched members, it is the best way that performing the real fatigue test of structure containing notch. But this method required generally much times and costs to evaluate fatigue life. So, generally we use the modified S-N curve or several methods to predict fatigue life. In this study, crack initiation life was evaluated by fatigue testing the SAE keyhole specimen and smooth specimen made of Al 7075-T6 alloys using the constant load then obtained S-N curve of smooth specimen and P-N curve of SAE keyhole specimen. And, fatigue lives of keyhole specimen are predicted using some life prediction methods (Nominal range I method, Nominal range II method, FEM analysis) for investigating experimented results, and that were compared with experimental data. Predicted fatigue lives by FEM analysis were corresponded with experimental data between 1/3times and 3times on the whole, and predicted fatigue lives using modified S-N curve (Nominal range I method, Nominal range II method) were nonconservative compared with that of FEM analysis.

  • An identification method of the structural damping matrix for a steel bar by the genetic algorithm is proposed. Supposing the damping matrix were in proportion to the stiffness matrix, the proportional factors can be identified from the curve fitting of the experimental frequency response function(FRF) by the genetic algorithm. Applying the identified damping matrix to FEM of a beam model, the values of the objective function could be reduced to about 1/60 in comparison with conventional FEM model without damping. The damping matrices of some sub-structures which have large damping partly could be identified by the algorithm, and they could be used as some parts of the FEM model for a whole structure.

  • The mold transformers have been widely used in underground substations in large building and have some advantages in comparison to oil-transformer, that is low fire risk, excellent environmental compatibility, compact size and high reliability. In addition, the application of mold transformer for outdoor is possible due to development of epoxy resin. The mold transformer generally has cooling duct between low voltage coil and high voltage coil. A mold transformer made by one body molding method has been developed for small size and low loss. The life of transformer is significantly dependent on the thermal behavior in windings. To analyse winding temperature rise, many transformer designer have calculated temperature distribution and hot spot point by finite element method(FEM). Recently, numerical analyses of transformer are studied for optimum design, that is electric field analysis, magnetic field, potential vibration, thermal distribution and thermal stress. In this paper, the temperature distribution of 50 kVA pole mold transformer for power distribution are investigated by FEM program and the temperature rise test of designed mold transformer carried out and test result is analyzed compare to simulation data. In this result, the designed mold transformer is satisfied to limit value of temperature and the other property is good such as voltage ratio, winding resistance, no-load loss, load loss, impedance voltage and percent regulation.
