Anti-Diabetic Effect of Pectinase-Processed Ginseng Radix (GINST) in High Fat Diet-Fed ICR Mice

  • In the present study, we investigate anti-diabetic effect of pectinase-processed ginseng radix (GINST) in high fat diet-fed ICR mice. The ICR mice were divided into three groups: regular diet group, high fat diet control group (HFD), and GINSTtreated group. To induce hyperglycemia, mice were fed a high fat diet for 10 weeks, and mice were administered with 300 mg/kg of GINST once a day for 5 weeks. Oral glucose tolerance test revealed that GINST improved glucose tolerance after glucose challenge. Compared to the HFD control group, fasting blood glucose and insulin levels were decreased by 57.8% (p<0.05) and 30.9% (p<0.01) in GINST-treated group, respectively. With decreased plasma glucose and insulin levels, the insulin resistance index of the GINST-treated group was reduced by 68.1% (p<0.01) compared to the HFD control group. Pancreas of GINST-treated mice preserved a morphological integrity of islets and consequently having more insulin contents. In addition, GINST up-regulated the levels of phosphorylated AMP-activated protein kinase (AMPK) and its target molecule, glucose transporter 4 (GLUT4) protein expression in the skeletal muscle. Our results suggest that GINST ameliorates a hyperglycemia through activation of AMPK/GLUT4 signaling pathway, and has a therapeutic potential for type 2 diabetes.

동물실험에 의한 몇가지 해조식품(海藻食品)의 소화흡수율(消化吸收率) (Digestibility of Some Korean Seweeds through an Animal Experiment)

  • Apparent digestibility of three representative seaweeds commonly used in Korea was obtained through an animal experiment and their calorie values were calculated therefrom. 1) Digestion coefficients for the nutrients of laver were crude protein, 42%; crude ash, 9%; carbohydrate, 62% and crude fat, none. 2) Digestion coefficients for the nutrients of tang1e were crude protein, 10%; crude fat, 40%; carbohydrate, 40% and crude ash, none. 3) Digestion coefficients for the nutrients of green laver were crude protein, 38%; crude fat, 59%; carbohydrate, 66% and crude ash, none. 4) Calorie values of seaweeds on the basis of above digestion coefficients were assessed to be only 52% in laver, 32% in tang1e and 59% in green laver as compared with those customarily obtained from proximate composition and Atwater's coefficient.

월동기 민물도요 Calidris alpina 의 지방과 단백질의 변화 (Variation of Fat and Protein Reserves in Wintering Dunlin, Calidris alpina)

  • The changes of pectoral muscle size (as an index of protein reserves) and fat reserves (as an index of lipid) were measured in wintering Dunlin during 1988 -1989 on two estuaries located in the south coast of Korea. The mean of lean mass of February population was greater(11.8%) than that of October population. SMI of October population was greater(4.5%) than that of February population and lipid index of October population also greater(1.6%) than that of February population. This show that the body condition of October population is better than February population. The mean of fat and protein reserves of 22 % of Dunlin arriving on October for their wintering were higher than that of others, this group seems to continue their migration to south further and 11% of the wintering population seems to starve during winter.

Product Characteristics of Comminuted Sausages as Affected by Various Fat and Moisture Combinations

  • Comminuted sausages with reduced fat (25-0%) and increased moisture (19-44%) combinations were manufactured, and their chemical composition, and physico-chemical, textural and sensory properties were determined for the selection of the most palatable sausage treatment. The moisture and fat contents of the comminuted sausages varied from 55-79% and 26.4-2.14%, respectively, whereas, the protein content was relatively constant at 13.5-14.5%. Expressible moisture (EM, %) decreased with increased fat addition, and the high-fat control sausage (~25% fat) had lower EM than those with lower than 15% fat addition. Increased fat level also increased Hunter L values (lightness) of sausage samples taken from the core, and differences in lightness were observed between 15 and 25% fat level at the initial mixture. Sausages without fat addition had different textural characteristics from high-fat control sausages in most texture profile analysis (TPA) values. However, no differences in TPA values were observed among treatments with added fat (<25%) in the formulation. Results of the sensory evaluation showed that the most appropriate fat content of comminuted sausages to have better sensory properties ranged from 15-20% of added fat at the initial mixture. These results also indicated that decreased fat and increased moisture contents produce sausages with higher EM and lower lightness. Comminuted sausages without fat addition had different textural characteristics from the high-fat control.

사료 내 수준별 조단백질 조건에서 보호메티오닌과 보호라이신의 사료첨가 급여가 착유우유생산성 및 유성분에 미치는 영향 : 메타분석 (Effect of protected Methionine and Lysine on Milk yield and Composition in Holstein Dairy Cow under Different Dietary Crude Protein Levels : Meta-analysis)

  • The present study was conducted to investigate effect of dietary protected amino acid on milk yield and composition in dairy cow using meta-analysis. Total 21 research papers were employed in analysis, and mixed model was used for the analysis of effects. Effect of protected methionine (PM) and combination of protected methionine and lysine (PML) were investigated under two different levels of dietary crude protein (CP, <18% and >18%). For performance of dairy cow, milk yield, milk composition including milk fat and protein content and yield and 4% FCM (fat corrected milk) production were used for analysis. In case of milk yield, a trend of increment was found at PM supplementation at low CP (P=0.055). However, the effect of PM at high CP was detected as not significant (P>0.05). In case of milk protein, inclusion of PM at low CP showed significant decrement (P<0.05). However, there was no significant effect of MP on milk protein at high CP (P>0.05). Supplementation of MP at high CP level showed significant increment of milk fat (P<0.05). MP supplementation represented significant increment of 4% FCM production (P<0.05) regardless of dietary CP levels. Effects of PML on milk yield and composition at both of low and high dietary CP were not significant in this study. However, it seem to be that there was a possible positive effect of MPL application at high dietary CP on performance of dairy cow.

Plant Proteins Differently Affect Body Fat Reduction in High-fat Fed Rats

  • This study examined the effects of corn gluten (CG), wheat gluten (WG), and soybean protein isolate (SPI), as well as their hydrolysates, on weight reduction in rats fed a high-fat diet. Eight-month-old male Sprague-Dawley rats (n=70) were fed a high-fat diet (40% of the calories were fat) for 4 weeks. Rats were then randomly divided into seven groups and were fed isocaloric diets with different protein sources for 8 weeks. The protein sources were casein (control group), intact CG (CG group), CG hydrolysate (CGH group), intact WG (WG group), WG hydrolysate (WGH group), intact SPI (SPI group), and SPI hydrolysate (SPIH group). Body weight gain, adipose tissue weights, lipid profiles in plasma and liver; and hepatic activities of carnitine palmitoyl transferase, fatty acid synthase (FAS), malic enzyme, and glucose-6-phosphate dehydrogenase were assessed. The CGH group showed significant weight reduction compared with the other groups. Epididymal fat pad and plasma triglycerides in the CGH group were the lowest and were significantly different than those in the control group. FAS activity in the CGH group was significantly lower than that in the other groups. In conclusion, the CGH diet of these experimental animals demonstrated a weight-reducing effect by lowering the adipose tissue weight and by affecting the activities of hepatic lipogenic enzymes.

Loss of FAT Atypical Cadherin 4 Expression Is Associated with High Pathologic T Stage in Radically Resected Gastric Cancer

  • Purpose: Recent studies have revealed recurrent alterations in the cell adhesion gene FAT4, a candidate tumor suppressor gene, in cancer. FAT atypical cadherin 4 (FAT4) is a transmembrane receptor involved in the Hippo signaling pathway, which is involved in the control of organ size. Here, we investigated the loss of FAT4 expression and its association with clinicopathological risk factors in gastric cancer. Materials and Methods: We assessed the expression of FAT4 by using immunohistochemistry on three tissue microarrays containing samples from 136 gastric cancer cases, radically resected in the Soonchunhyang University Cheonan Hospital between July 2006 and June 2008. Cytoplasmic immunoexpression of FAT4 was semi-quantitatively scored using the H-score system. An H-score of ${\geq}10$ was considered positive for FAT4 expression. Results: Variable cytoplasmic expressions of FAT4 were observed in gastric cancers, with 33 cases (24.3%) showing loss of expression (H-score <10). Loss of FAT4 expression was associated with an increased rate of perineural invasion (H-score <10 vs. ${\geq}10$, 36.4% vs. 16.5%, P=0.015), high pathologic T stage (P=0.015), high tumor-node-metastasis stage (P=0.017), and reduced disease-free survival time (H-score <10 vs. ${\geq}10$, mean survival $62.7{\pm}7.3$ months vs. $79.1{\pm}3.1$ months, P=0.025). However, no association was found between the loss of FAT4 expression and tumor size, gross type, histologic subtype, Lauren classification, lymphovascular invasion, or overall survival. Conclusions: Loss of FAT4 expression appears to be associated with invasiveness in gastric cancer.

Troglitazone Regulates white Adipose Tissue Metabolism by Activating Genes Involved in Fatty Acid ${\beta}$-Oxidation in High Fat Diet-fed C57BL/6J Mice

  • This study aimed to determine whether troglitazone stimulates genes related to fatty acid ${\beta}$-oxidation, leading to modulation of white adipose tissue (WAT) metabolism in high fat diet-fed mice. Female C57BL/6J mice were randomly divided into two groups (n=10/group). After they received either a high fat diet or the same high fat diet supplemented with troglitazone for 4 weeks, the effects of troglitazone on gene expression and physiology of WAT were measured using Northern, histological and serological analyses. Administration of troglitazone induced the expression of genes involved in mitochondrial and peroxisomal fatty acid ${\beta}$-oxidation in mesenteric WAT. Troglitazone also significantly increased uncoupling protein 2 mRNA levels. The changes in WAT gene expression were accompanied by reductions in circulating levels of free fatty acids and triglycerides as well as glucose and insulin. Histological studies showed that troglitazone treatment decreased the average size of adipocytes in mesenteric WAT. These results suggest that troglitazone-stimulated WAT expression of genes associated with fatty acid ${\beta}$-oxidation regulates WAT metabolism of high fat diet-fed mice, contributing to improvement of insulin sensitivity.

근무시간이 불규칙한 사람들의 식생활태도 및 건강상태에 대한 조사 연구 (Nutritional and Health Status of Nurses, Medical Doctors and Factory Workers)

  • The purpose of this survey was to investigate the nutritional status of special working persons related to health status, nutrient intake and living environment. The survey was conducted on workers in 9 general hospitals and 2 factories from Nov. 24 in 1979 to Jan. 20 in 1980. Nurses, medical doctors on intership or residency, and factory workers were chosen as subjects because of their irregular working hours, i.e., they sometimes work during the day and. sometimes at night. The total number of subjects was 417 for nurses, for medical doctors was 91, and was 216 for factory workers, all together 724 subjects were investigated. The results obtained are summarized as follows: 1) Average nutrient intake of subjects per day. For nurses the caloric intake was 1743 Kcal. This was composed of carbohydrate 251g(57.6%), protein 81g (18.5%), and fat 46g (23.9%). Other nutrient intake, such as iron (16mg), thiamin (0.98mg), riboflavin (1mg) was lower than the recommended dietary allowance. For male medical doctors, the caloric intake was 2064 Kcal, This was composed of carbohydrate 288g (55.8%), protein 102g (19.8%) and fat 56g (24.4%). Other nutrient consumption, such as riboflavin (1.31mg) was lower than the recommended dietary allowance. For female medical doctors, the caloric intake was 1909 Kcal. This was composed of carbohydrate 156g(53.7%), protein 90g (18.9%) and fat 58g (27.4%). For male factory workers, caloric intake was 1699 Kcal. This was composed of carbohydrate 317g(74.5%), protein 64g (15.0%) and fat 20g(10.5%). Other nutrient intake, such as vitamin A (4765IU), ascorbic acid (46mg) and riboflavin (1.11mg) were lower than the recommended dietary allowance. For female factory workers, the caloric intake was 1630 Kcal. This was composed of carbohydrate 287g (70.5%), protein 65g (15.8%) and fat 25g (13.7%). Other nutrient consumption, such as iron (15mg), riboflavin (1.04mg), ascorbic acid (42mg), and vitamin A (4334IU) was lower than the recommended dietary allowance. The percentage of animal protein to total protein was 65.2% in nurses, 67.4% in male medical doctors, 64.9% in female medical doctors, 38.6% in male factory workers, and 45.6% in female factory workers. 2) The average Kaup's value of subjects was 1.9 in nurses, 2.2 in male mcdical doctors, 1.9 in female medical doctors, 2.0 in male factory workers, and 2.1 in female factory workers. 3) The health status of subjects according to their self diagnosed answer was as follows; The percentage of food health was 82. 7% in nurse, 82.7% in nurse, 83.5% in medical doctor, and 56% in factory workers. The remainder answered that they were in poor health.

어린이의 식생활태도가 영양 상태 및 성격에 미치는 영향에 관한 연구 (An Effect of Children's Food Alttitude on Nutritional Status and Personality)

  • This survey of 527 children, aged 4 to 6 years, focused on family and dietary environment, nutritional status and personality. It was conducted from June to October, 1986. The results showed that 68.0%(breakfast)and 55.4% (evening meal ) of the subjects indicated they had meals with the family everyday. Analysis of the nutritional status children showed all nutrient intake except protein an diron was insufficient compared with RDA. Carbohydrate provided 58.3% of total calorie intake ; protein accounted for 15.7% . fat provided 26.0%. There was a positive relation between income and nutrient intake. Children with employed mothers showed lower nutrient intake than children with unemployed mothers. Food attitude was positively related to nutrient intake, nutrient intake of children with good eating behavior was higher than that of children had poor eating behavior . Activity level had a significant correlation with intake of calories, fat, vitamin A, vitamin B$_1$, and Vitamin B$_2$. Mood was correlated with intake of protein, iron, vitamin A, vitamin B$_2$ and vitamin C.
