Detection of E. coli O157:H7 and Listeria monocytogenes, and Appraisal for Microbiological Qualities in the Commercial Frozen Yogurt Products in Korea (국내 시판용 Frozen Yogurt의 병원성 미생물 검출 및 미생물학적 품질 평가에 관한 연구)

  • Recently the high outbreaks of intestinal disease caused by the consumption of frozen dairy foods containing pathogenic bacteria has generated considerable interest in the causative agent such as Listeria monocytogenes and E. coli O157:H7. This study was carried out to detect the above pathogens and compare the microbiological qualities of three commercial forzen yogurt products. The main results obtained were as follows. L. monocytogenes coliforms and E. coli O157:H7 were not detected in the total of seven frozen yogurt samples. For microbiological qualities the viable lactic counts of products manufactured by FA company were about 2.9$\times$108 -1.6$\times$109cfu/ml 1.7$\times$106 cfu/ml for FB's and 1.2$\times$106 cfu/ml for FC's The PH values of FA's FB's and FC's products was in the range of pH 4.1~5.3 and the values of FA's were 4.1~4.6 compared by the pH 5.2~5.3 of FB's and FC's products. During refrigeration of the test samples the survival rates of L. monocytogenes spiked into thawed frozen yogurt sample(FA's FB,s and FC's) were 0.55% 15.61% and 16.89% respectively. On the other hand E. coli O157:H7 and L. monocytogenes were 12.4% and 25.0% for FA's 10.8% and 20.8% for FB's and 10.26% and 22.7% for FC's under 37$^{\circ}C$ storage, As the results described above each frozen yogurt products were different in microbiological qualities. The survival rates of pathogens spiked into the samples increased with the pH of the products. This indicates that the pH or any other factors pre-sumable supressed the growth of E. coli O157:H7 and L. monocytogenes in frozen yogurt products.

The effects of limestone powder and fly ash as an addition on fresh, elastic, inelastic and strength properties of self-compacting concrete

  • In this study, limestone powder (LS) and fly ash (FA) were used as powder materials in self-compacting concrete (SCC) in increasing quantities in addition to cement, so that the two powders commonly used in the production of SCC could be compared in the same study. Considering the reduction of the maximum aggregate size in SCC, 10 mm or 16 mm was selected as the coarse aggregate size. The properties of fresh concrete were determined by slump flow (including T500 time), V-funnel and J-ring experiments. The experimental results showed that as the amount of both LS and FA increased, the slump flow also increased. The increase in powder material had a negative effect on V-funnel flow times, causing it to increase; however, the increase in FA concretes was smaller compared to LS ones. The increase in the powder content reduced the amount of blockage in the J-ring test for both aggregate sizes. As the hardened concrete properties, the compressive and splitting strengths as well as the modulus of elasticity were determined. Longitudinal and transverse deformations were measured by attaching a special frame to the cylindrical specimens and the values of Poisson's ratio, initiation and critical stresses were obtained. Despite having a similar W/C ratio, all SCC exhibited higher compressive strength than NVC. Compressive strength increased with increasing powder content for both LS and FA; however, the increase of the FA was higher than the LS due to the pozzolanic effect. SCC with a coarse aggregate size of 16 mm showed higher strength than 10 mm for both powders. Similarly, the modulus of elasticity increased with the amount of powder material. Inelastic properties, which are rarely found in the literature for SCC, were determined by measuring the initial and critical stresses. Crack formation in SCC begins under lower stresses (corresponding to lower initial stresses) than in normal concretes, while critical stresses indicate a more brittle behavior by taking higher values.

A Study on Correlation Between Cyclic Drying-Wetting Accelerated Corrosion Test and Long-term Exposure Test (건습반복 부식촉진시험 및 장기폭로시험의 상관성에 대한 연구)

  • There are various method for evaluating the durability life of concrete structures due to salt damage. The best way is to perform a corrosion test for a rebar embedded in concrete specimen was exposure to marine environment. However, this method has the disadvantage that it takes a long period of time. Also, accelerated corrosion test which was complemented complements the time-consuming weakness is limited to apply because it could not reveal a correlation between long-term exposure test. Accordingly, the purpose of this study is to derive a correlation coefficient between cycle drying-wetting accelerated corrosion test and long-term exposure test. Corrosion initiation time was measured in four types of concrete samples, i.e., two samples mixed with fly ash(FA) and blast furnace slag(BS), and the other two samples having two water/cement ratio(W/C = 0.6, 0.35) without admixture(OPC 60 and OPC 35). The accelerated corrosion test was carried out by two case, i.e., one is a cyclic drying-wetting method(case 1), and the other is a artificial seawater ponding test method(case 2). Whether corrosion occurs, it was measures using half-cell potential method. The results indicated that case 1 is to accelerated the corrosion of rebar about 24~36% as compared with case 2, then the corrosion of rebar embedded in concrete occurred according to the order of OPC60, FA, BS, OPC35. Correlation coefficient between accelerated corrosion test and long-term exposure test, case 1 is 4.23 to 5.42, and case 2 is 6.54 to 7.82.

Experimental Study on Evaluation of Abrasion Resistance of Concrete Irrigation Facilities (콘크리트 수리구조물의 수중마모저항성 평가기술에 관한 실험적 연구)

  • The purpose of this paper is to propose an experimental method to evaluate the resistance of abrasion about 24 MPa, 27 MPa, and 30 MPa compressive strength. These strength are used in the design and construction of concrete hydraulic structures in Korea. The mixing ratios of the ready mixed concrete strengths were investigated countrywide and set the representative mixture proportion ratios of the nine mixed types of OPC, FA and BFS. After making and curing the test specimens, the underwater abrasion test was performed. ASTM C 1138 International Standard was used to fabricate the test equipment, and the surface abrasion resistance of the specimen was tested using the test equipment. In the case of OPC, the 30% abrasion resistance improvement effect was observed at 72 hours as the water-binder ratio decreased. That was reason the coated cement bond strength of the specimen was strong. In the case of BFS and FA, it was improved by 9.9% and 3.8%, respectively, at 72 hours as the water-binder ratio decreased. It was due to the characteristics of the latent hydraulic and pozzolanic reactions. Generally, the relative abrasion resistance of concrete can be evaluated at 24 hours. However, in case of low strength (under 24 MPa), the surface mortar layer wears much faster at the first 12 hours, so it can be considered to evaluate the relative abrasion loss rate at this point.

Isolation and immunohistochemical diagnosis of Haemophilus parasuis from naturally occurring polyserositis in pigs (자연발생한 돼지 다발성 장막염 예로부터 Haemophilus parasuis의 분리와 면역조직화학적 진단)

  • From Jan. 1996 to Oct. 1997, 29 pigs with 40-70 days old showing dyspnea inappetite and polyserositis were collected and carried out necropsy, bacterial culture, histopathology, avidin biotin complex(ABC) stain, fluorescent antibody(FA) test, and electron microscopy. In the study, 4 strains from 3 pigs were isolated from meninges, pleura and synovial fluid and also were identified as Haemophilus parasuis serovar 5. Main histopathological lesions of 29 pigs with polyserositis were frequently composed of fibrinous peritonitis(27), pleurisy(22), interstitial pneumonia(21), fibrinous epicarditis(20), fibrinopurulent meningitis(8) and synovitis(4). By ABC stain, 11/29(37.9%) pigs with polyserositis were confirmed to have H parasuis serovar 5 in the cytoplasm of macrophages and neutrophils in cerebral meninges, epicardium, pleura surface of lung or serosa of spleen. ABC stain(20.8~40.0%) to detect H parasuis serovar 5 in tissues was more sensitive than bacterial culture(10.3%), but less sensitive than FA test(62.5%) using frozen tissues even though the result of 8 cases. By electron microscopy, a bacterium was also detected in the cytoplasm of macrophages in purulent exudate of cerebral meninges. Consequently, we confirmed that H parasuis serovar 5 has been involving to cause pigs with polyserositis and can be detected by FA and ABC stain as reliable diagnostic tools.

Analysis of Acetaldehyde and Formaldehyde Migrated from Polyethylene Terephthalate Bottles into Mineral Water (페트병 생수 중 아세트알데하이드와 포름알데하이드의 이행 분석)

  • The objective of this study was to determine the extent to which acetaldehyde (AA) and formaldehyde (FA) migrated from polyethylene terephthalate (PET) bottles into mineral water during storage at 25 and 35$^{\circ}C$ for 240 days. Generally, AA and FA were observed to migrate more at 35$^{\circ}C$ than at 25$^{\circ}C$. The amount of FA in the samples irradiated under UV lamps was higher than in those stored in the dark over the storage period and vice versa for AA. Using a triangular test, the sensory panels distinguished a difference between one sample and the other two mineral water samples that had been exposed to UV lamps after 60 days of storage at 35$^{\circ}C$ (p<0.05). At this time, FA and AA concentrations were measured at 267.4 and 515.3 g/L, respectively. After 120 days, an independent mineral water sample from the other two samples in all treatments could be distinguished by the panel members at a significance level of p<0.001.

Evaluation of Chloride Diffusion Characteristics in Concrete with Fly Ash Cured for 2 Years (2년 양생된 Fly Ash 콘크리트의 염화물 확산 특성 평가)

  • When RC(Reinforced Concrete) structures are exposed to harsh environment, deterioration phenomenon occurs, and the corrosion in rebar due to chloride intrusion is known as representative deterioration, so called chloride attack. In this paper, chloride resistance performance of 2 years aged concrete is evaluated considering 3 levels of water to binder ratio(0.37, 0.42, and 0.47) and 2 levels of substitution ratio of fly ash(0% and 30%). Accelerated chloride diffusion coefficient tests referred to Tang's method, total passed charge tests referred to ASTM C 1202, and compressive strength tests referred to KS F 2405 are performed. With adaptation of the previous test results and the results from this study, time-dependent chloride diffusion characteristics are analyzed for each concrete. The FA(Fly Ash) concrete has higher chloride resistance performance than OPC(Ordinary Portland Cement) concrete. According to the evaluation standard of ASTM C 1202, the FA concrete has "Moderate" grade after 49 days while OPC concrete does "Moderate" grade after 365 days. As the results of time-parameter for chloride diffusion, OPC concrete and FA concrete show the decreasing behavior of time-parameters with increasing water to binder ratio. Also, FA concrete has 1.57~2.74 times of time-parameter than OPC concrete. That's cause is thought that the time-parameter indicates the gradient of decreasing of diffusion coefficient. FA concrete has higher time-parameters than OPC concrete by pozzolanic reaction of FA.

  • Monoclonal antibodies(MAbs) against field isolates of the bovine rotavirus A strain(G6), V strain(G10) and reference I-801 strain(G8) were produced and characterized. Six MAbs(4C2, 4D9, 5E1, 5E7, 5D5, 3E4) against A strain had neutralizing activity and reacted only with the G6 bovine rotaviruses determined by fluorescence focus neutralization (FFN) test. Otherwise, five neutralizing MAbs(1G2, 2G6, 5E2, 5E12, 5H7) against I-801 strain neutralized the G6 and G8 bovine rotaviruses. Five non-neutralizing MAbs(5F12, 7F12, 5E11, 2A11, 2B12) were VP6-specific and cross-reacted with all bovine and porcine rotaviruses examined by fluorescence antibody(FA) test. None of the MAbs reacted with bovie viral diarrhea virus(BVDV) and bovine coronavirus(BCV) determined by FA and FFN test.

Quality Analysis of Fly Ash Through Correlation between Density by Hydrometer and Test Report (Hydrometer법을 이용한 밀도 측정값과 시험 성적서간 상관분석을 통한 플라이애시의 품질특성 분석)

  • To evaluate the reliability of fly ash quality supplied to ready-mixed concrete plant using mass cylinder and hydrometer, in this research, the correlationship between the fly ash properties provided from certification and density measurement with suspension was evaluated. As a result, the reliability of the certification, except fineness and loss on ignition, all properties had a discord. Additionally, in the case of density, fineness, and L.O.I, the relation with the density measured using hydrometer showed high correlation, especially fineness was strongly related with the density measured using hydrometer. Furthermore, according to the comparative analysis with previous research, the fly ash used in this research was similar measurement with raw powder without any refining process, it is considered that the constant error of blaine test or using raw ash sample as a fly ash. In current standard regarding fly ash, the fineness range of class 2 can be changed from $3,000-4,500cm^2/g$ to $3,500-4,500cm^2/g$ for improved quality of fly ash in fineness aspect.

Evaluation of Shrinkage Cracking Characteristics and Degree of Restraint for Ultra-High-Strength Concrete (초고강도 콘크리트의 수축 균열 특성 및 구속도 평가)

  • The concrete cracking from the restrained stress caused by the shrinkage may play significant cause of deterioration of concrete structures by allowing the permeation of sulphate and chloride ions which in turn triggers corrosion of steel reinforcement. In particular, the cracking becomes more critical as water binder ratio (W/B) is reduced and concrete strength increases. Therefore, it needs to evaluate correctly the comprehensive shrinkage behavior of concrete with high strength: high-strength concrete (HSC), ultra-highstrength concrete (UHSC). The unrestrained shrinkage tests, however, cannot estimate the net shrinkage effectively which affects cracking after full development of strength and stiffness because it does not consider the degree of restraint, strength development, stress relaxation, and so on. Therefore, in this study, both free and restrained shrinkage tests with variables of W/B (W/B of 30, 25 and 16%) and admixtures (fly ash (FA) and granulated blast-furnace slag (BFS)) for HSC, very-high-strength concrete (VHSC) and UHSC were performed. The test results indicated that the autogenous shrinkage and total shrinkage at drying condition were reduced as W/B increased and FA, BFS were added, and the cracking behavior was suppressed as W/B increased and FA was added.
