Characteristics of Plantlets Redifferentiated from F1 Hybrid between Panax ginseng and Panax quinquefolius (고려인삼과 미국삼의 종간잡종으로부터 재분화된 식물체의 특성)

  • The characteristics of plantlets redifferentiated from calli of F1 hybrid between Panax ginseng and Panax quinquefolius were investigated. Growth of plantlets redifferentiated from F1 hybrid was superior to the plants redifferentiated from Korean ginseng. Stem color of plantlets redifferentiated from F1 hybrid was more purple than that from Korean ginseng and leaf color of the former was also greener than that of the latter. Chunpoong, Yunpoong and Seonweon which are belonged to Korean ginseng showed same PCR band(A), while American ginseng showed different PCR band (B) in Internal Transcribed Spacer (ITS) region. F1 hybrid exhibited both A and B PCR band which belonged to Korean ginseng and American ginseng, respectively. F1 hybrid calli and plantlets redifferentiated from F1 hybrid calli showed same PCR band with that of F1 hybrid plant in ITS region. Therefore it was confirmed that piantlets redifferentiated from F1 hybrid exhibited genetic stability in ITS region.

Morphological, Cytological and Molecular Evidence for Intersubgeneric F1 Hybrid between Glycine max x G. tomentella (콩 Glycine max와 G. tomentella의 종간교잡으로부터 얻은 Fl식물체 검증을 위한 형태적 · 세포학적 · 분자유전학적 연구)

  • This study was carried out to demonstrate morphological, cytological and molecular evidence for intersubgeneric $F_1$ hybrid between Glycine tomentella and G. max cv. ‘Baemkong’. Morphological features of $F_1$ plant for pistil and stamen, flower color and growth habit showed intermediate type between G. tomentella and G. max cv. ‘Baemkong’. Chromosome number of $F_1$ plant was 2n=39, which explained the evidence of $F_1$ hybrid between G. tomentella (2n=38) and G. max cv. ‘Baemkong’ (2n=40). Polyacrylamide isoelectric focusing pattern for esterase and peroxidase also showed that the $F_1$ plant was true $F_1$ hybrid between G. tomentella and G. max cv. ‘Baemkong’. From RAPD analysis, we identified that 62 primers showing bands in $F_1$ hybrid had both bands from G. tomentella and G. max cv. ‘Baemkong’, which suggested that this was true $F_1$ hybrid. Based on our results from morphological, cytological and molecular analyses, we suggest that the $F_1$ plant was true intersubgeneric hybrid between G. tomentella and G. max cv. ‘Baemkong’. Our results still remain us further study to recover fertility of $F_1$ hybrids. The occurrence of maternal and/or paternal inheritance in $F_1$ hybrid from intersubgeneric cross between G. tomentella and G. max cv. ‘Baemkong’ need to be explained.

Breeding Hybrid Rice with Genes Resistant to Diseases and Insects Using Marker-Assisted Selection and Evaluation of Biological Assay

  • Developing elite hybrid rice varieties is one important objective of rice breeding programs. Several genes related to male sterilities, restores, and pollinators have been identified through map-based gene cloning within natural variations of rice. These identified genes are good targets for introducing genetic traits in molecular breeding. This study was conducted to breed elite hybrid lines with major genes related to hybrid traits and disease/insect resistance in 240 genetic resources and F1 hybrid combinations of rice. Molecular markers were reset for three major hybrid genes (S5, Rf3, Rf4) and thirteen disease/insect resistant genes (rice bacterial blight resistance genes Xa3, Xa4, xa5, Xa7, xa13, Xa21; blast resistance genes Pita, Pib, Pi5, Pii; brown planthopper resistant genes Bph18(t) and tungro virus resistance gene tsv1). Genotypes were then analyzed using molecular marker-assisted selection (MAS). Biological assay was then performed at the Red River Delta region in Vietnam using eleven F1 hybrid combinations and two control vatieties. Results showed that nine F1 hybrid combinations were highly resistant to rice bacterial blight and blast. Finally, eight F1 hybrid rice varieties with resistance to disease/insect were selected from eleven F1 hybrid combinations. Their characteristics such as agricultural traits and yields were then investigated. These F1 hybrid rice varieties developed with major genes related to hybrid traits and disease/insect resistant genes could be useful for hybrid breeding programs to achieve high yield with biotic and abiotic resistance.

Characteristics of Hybrids between Jakyungjong and Hwangsukjong in Korean Ginseng (Panax ginseng C. A. Meyer)

  • A large number of individual ginseng plants have been selected in the farmer's fields to develop new ginseng varieties with desirable traits since 1970s. Among them, Hwangsukjong with green stem and yellow berry was selected as a ginseng germplasm. The phenotype of Hwangsukjong is quite different from Jakyungjong that has violet stem and red berry and has been cultivated in most of ginseng fields. Therefore, Hwangsukjong was crossed with Jakyungjong to clarify the inheritance of stem color and then the characteristics of $F_1\;and\;F_2$ hybrids were investigated. $F_1$ hybrid plants were similar to Jakyungjong in most of aerial part characters and showed hybrid vigor in fresh weight of root and weight of 100 seeds. In $F_2$ generation, the stem color was segregated in a ratio of 3 violet to 1 green. From this result, it was elucidated that violet color was controlled by single dominant gene. In another experiment, DNA was extracted from parents (Jakyungjong and Hwangsukjong) and $F_1$ hybrid. For each primer evaluated, multiple band profile was produced comprising from one to five major bands plus a varying number of minor bands and amplified bands were detected among most primers. In case of UBC primer number 13, 17, 30, 31, and 43, band patterns of parents and $F_1$ hybrid were very similar, but the others were not. Especially, in {\sharp}1$, {\sharp}4$, and {\sharp}33$, specific band was produced in Hwangsukjong and $F_1$ hybrid while in {\sharp}6$, another specific band was produced in Jakyungjong and $F_1$ hybrid. Therefore, $F_1$ hybrid had all specific bands at these primers. So, these selective markers could be used for identification of characteristics of $F_2$ hybrids

Studies on Incompatibility in Interspecific Hybrid Between Panax ginseng C. A. Meyer and Panax quinquefolium L. (고려인삼과 미국삼의 종간 잡종식물체 불화합성에 관한 연구)

  • This study was carried out to clarify the cause of incompatibility in interspecific hybrid plant between Panax ginseng and p. quinquefolium. The floral structure of F,(p.g. x p.q.) hybrid was normal because the redundant anther was 0.2 mm longer than pistil in Fl hybrid and the size and structure of redundant carpel in F, hybrid were similar to P. ginseng and p. quiquefolium Pollens of $F_1$ hybrid did not germinate on stigma of P-quinquefolium but germinated well on stigma of P. ginseng. Pollen tube was able to penetrate styles completely and seed harvest rate was 16.8% in field. However on stigma of $F_1$ hybrid, Pollen did not germinate when P. ginseng was used as male Parent. In addition, the growth of pollen tube was halted on style and seed was not set when P qlfinquefoEi2a was used as male Parent. These suggest that the inhibitor of pollen germination present on stigma caused $F_1$ hybrid sterility. It took 5 hours for pollen grains to germinate, 12 hours to arrive at in trance of ovule, 16 hours to penetrate micropyles in Panax ginseng.

  • choChorion(egg-shell) morphology of the F1 hybrid between Bombyx mori and Bombyx mandarina has been observed by scanning electrom microscope and chorion protein was analyzed by electrophoresis. The chorion surface structure of F1 hybrids in the lateral(flat) region was similar to that of maternal line. The F1 hybrids chorion was found to have basically a three layer structure. The middle and inner layer were very much like those of the Bombyx mandarina and Bombyx mori. There were many conic pillar structures in the outer layer of the F1 hybrid, which was similar to Bombyx mandarina. This conic pillar structure had a thin cover layer was more clear in the dorsal and ventral side of the F1 hybrid chorion. The conic pillar structure of Bombyx mandarina was found to be dominant in F1 hybrid chorion irrespective of their maternal line. Major components of chorion protein were analyzed by two-dimensional electrophoresis and found to have isoelectric points in the range of pH 4.0-6.5 and molecular weight 10 to 50 kd. F1 hybrid chorion protein components related directly to those of the maternal line. The conic pillar structure was dominat characteristic and it was present in all F1 hybrid.

  • This study was carried out to develop the efficient methods for the subsequent progeny of intersectional hybrid between Oriental and Asiatic lily hybrids. The pollen fertility and germination ability of 3 different allotetraploids (OAOA) after somatic chromosome doubling was ranged from 0 to 80 percent on artificial pollen germination medium. The number of BC$_1$ progeny using allotetraploid of F$_1$ OA-hybrid as male and female parent was different. The efficiency of BC$_1$ progeny production was increased when F$_1$OA-hybrids was used as male rather than as female parent. And in back crosses of F$_1$ OA-hybrids with the Asiatic and Oriental hybrids, Asiatic hybrids showed higher efficiency on BC$_1$ progeny production. The ploidy level between 2x or 4x Asiatic hybrid and allotetraploid F$_1$ OA-hybrid was determined and showed higher progeny production in 2x-4x crosses rather than 4x-4x.

Comparison of F1 and F2 hybrid on authorized silkworm variety (누에 장려품종간 교잡누에의 실용형질 비교)

  • This study was conducted to consider practical use of F2 hybrid and intervariety crossing for the improvement of silkworm eggs export and silkworm eggs manufacture. Now Kumokjam and Baegokjam come into wide use to sericulture farmers but comparison test of F1, F2 hybrid and intervariety have not been done. We weighed against the productivity among economical character. While larval period of F2 among them is most long time, pupation percentage and cocoon yield per 10,000 3rd molted larvae are lower than intervariety. With comparing F2 and intervariety, single cocoon weight of F2 is lower 9 ~ 21% than intervariety, and cocoon shell weight of F2 was lower 10 ~ 18% than intervariety. By number of cocoons per liter, F2 hybrid was lower and more irregular size than intervariety. In coparison with number of egg laid, F2 hybrid Baegokjam was 655 ea (14%) and Kumokjam 625 ea (40%) more than F1 hybrid. Also, F2 hybrid for filament grade went down than F1 hybrid. The advantage of egg production in F2 hybrid can reduce costs because there is no necessity for sex discrimination and germination control. But besides increase of number of egg laid, the productivity including economical characters was greatly lowered. Except for being a lot of laying eggs, cocoon productivity of the F2 was greatly reduced by degeneration of practical traits. Therefore the further studies are needed in order to increase utilization value of the F2 hybrid silkworm.

Fruit Quality Characteristics in Second Generation (F2) Hybrid Cultivar of Minipaprika (Capsicum annuum L.)

  • A field experiment was conducted to examine the fruit quality characters in second generation ($F_2$) hybrid cultivar and to compare the fruit characters with original $F_1$ hybrid cultivar of minipaprika (yellow and orange type) at the Research Farm, Hwacheon in July, 2010. Fruit characters varied within $F_2$ population of each minipaprika type. In minipaprika yellow, fruit weight varied from 12.2 g to 50.8 g (average 28.5 g) and fruit length/width varied from 1.4 to 2.8 (average, 2.0). Pericarp thickness ranged from 1.8 mm to 4.1 mm (average, 2.9 mm). Total soluble solid (TSS) varied from $6.2^{\circ}Brix$ to $13.5^{\circ}Brix$ with an average of $8.7^{\circ}Brix$. Fruit volume varied from 10.3 cc to 46.7 cc with an average of 24.4 cc. In minipaprika orange type, fruit weight ranged from 19.7 g to 42.4 g (average, 29.0 g) and fruit length/width varied from 1.5 to 2.6 (average, 2.0). Pericarp thickness varied from 2.1 mm to 4.1 mm with an average of 3.0 mm. TSS varied from $5.0^{\circ}Brix$ to $12.2^{\circ}Brix$ (average, $7.9^{\circ}Brix$) and average fruit volume was 24.6 cc ranging from 10.7 cc to 35.0 cc. The average fruit quality characters in $F_2$ population in both yellow and orange minipaprika did not differ from their $F_1$ hybrid parent and $F_2$ seed can be an additional way to supply high yielding hybrid cultivars at lower cost to the minipaprika growers.

Biochemical Performance and Quantitative Assessment of F1 Hybrid of Two Ecoraces of Tropical Tasar Silkworm Antheraea Mylitta Drury (Lepidoptera: Saturniidae)

  • Antheraea mylitta Drury is basically a crossbreeding species, as such it seems to be potentially a good material for the exploitation of heterosis. In the present study F1 hybrid of wild ecorace Laria (L) and semi-domestic Daba (D) was raised and evaluated for various quantitative traits and biochemical parameters during larval stage. Improved fecundity ($+18{\pm}1.8%$ and higher egg hatching rate ($+10.96{\pm}1.3%$) was recorded in the F1hybrid ($L{\times}D$). Biochemical parameters studied in the hemolymph, midgut and fatbody of the larva showed significantly higher (P<0.05) total proteins and carbohydrate concentration besides digestive enzyme activity. Correspondingly SDS-PAGE revealed more number of protein bands in the hemolymph sample of F1s, ranging between 29 kDa to 66 kDa compared to parental lines. The present study demonstrates the positive heterosis effect in the F1 hybrid of Laria ${\times}$ Daba. Biochemical analysis indicates that, there is possibilities of exploitation of hybrids with specific parents targeted for desirable commercial traits (silk yield and fecundity). Moreover, most of these biochemical parameters can be used as markers to analyze the genetic improvement in the tasar silkworms.
