Effect of 17β-estradiol on Life History Parameters and Morphological Deformities in Tigriopus japonicus sensu lato: A Two-generation Studies (17β-estradiol에 노출된 저서성 요각류 Tigriopus japonicus s.l.의 세대간 생활사 및 형태학적 변이 연구)

  • To characterize ecotoxicological responses to a natural estrogen, $17{\beta}$-estradiol, we evaluated the life-history of the parental ($F_0$) and first generation ($F_1$) of the harpacticoid copepod, Tigriopus japonicus sensu lato. We evaluated the survival of nauplii and copepodites, the number of days until the emergence of copepodites and adult males, the sex ratio, brooding success, and the first brooding day of adult females. No significant differences in the survival rate were noted in response to treatments with different concentrations of $17{\beta}$-estradiol. However, $17{\beta}$-estradiol induced developmental delay and skewed the sex ratio toward males. Copepod development was delayed significantly in the 0.1 and $1\;{\mu}g\;l^{-1}$ $17{\beta}$-estradiol treatment groups relative to the control group, with a more pronounced delay in the $F_1$ group. Body length and biomass were significantly smaller in the $17{\beta}$-estradiol treated groups than in the controls. The male emergence of T. japonicus s.l. was very high in the 10 and $30\;{\mu}g\;l^{-1}$ $17{\beta}$-estradiol treatment group. Furthermore, exposure to $17{\beta}$-estradiol resulted in morphological deformities such as shrinking and swelling of the urosome, twisted setae of the caudal rami, setal loss of swimming legs, abnormal segmentation of antennules, and dwarfism.

The Biologists' Boon Activation Patterns during the Generation of Scientific Questions on Biological Phenomena (생명현상에 관한 과학적 의문 생성 과정에서 나타나는 생물학자의 두뇌 활성 양상)

  • The purpose of this study was to investigate biologists' brain activation patterns during the generation of scientific questions on biological phenomena. Eight right-handed healthy biologists volunteered to be participants in the present study. The question-generation tasks were presented in a block design. The BOLD signals of the biologists' brain were measured by 3.0T fMRI system and data were analyzed using Statistical Parametric Mapping (SPM2). According to our results, the left inferior and middle frontal gyri, the medial prefrontal cortex, the bilateral hippocampus, the occipito-parietal route, the fusiform gyrus, and the cerebellum were activated significantly during the generation of scientific questions. Therefore, we suggested that generating scientific question is associated with analyzing observed situations, using verbal strategy, retrieving episodic memories for comparisons, and feeling cognitive conflicts.

Adverse Effect of Nonylphenol on the Reproductive System in F2 Male Mice : A Qualitative Change?

  • Previously, we reported negative effects of low-dose nonylphenol (NP) exposure on the reproductive organs of F1 male mice. In the present study was further investigated the endocrine disrupting effect of NP exposure to F2 generation male mice. Mice were divided into 2 groups; (1) CON, control animals and (2) NP-50 ($50{\mu}g/L$), animals were treated with NP via drinking water. NP exposures were continuously conducted from parental pre-mating period until the postnatal day (PND) 55 of F2 offsprings. Mice were sacrificed on PND 55 and the reproductive tissue weights were measured. The initial (at PND 21) and terminal (PND 55) body weights of the NP-50 group animals were not significantly different from those of control group animals. NP exposure fail to induce a significant weight change of the testes, seminal vesicle and prostate except absolute epididymal weight (p<0.05). However, pathohistological studies revealed that NP-treated F2 animals showed evident decrease in seminiferous tubule diameters, reduced luminal area and number of germ cells. Also, sloughing morphologies in the tubules were notable. In the caudal epididymis, fewer mature sperms and swollen epithelial cells were found in the NP-treated group. The present study demonstrated that the subchronic low-dose NP exposure induced pathohistological abnormalities in testis and epididymis of F2 mice, and we assumed that these 'qualitative' changes in reproductive tissues could be derived from the epigenetic modifications such as DNA methylation, histone modification, altered DNA accessibility and chromatin structure. Further studies are needed to achieve a better understanding on the multi- or trans-generational effects of NP on the reproductive health and a human application.

[ $F_2$] Production of Gynogenetic Diploid in Olive Flounder, Paralichthys olivaceus (자성발생성 2배체 넙치의 제 2세대 생산)

  • [ $F_2$ ] generation of gynogenetic diploids were produced in olive flounder, Paralichthys olivaceus, by brother-sister mating with full-sibling gynogenetic diploids ($F_1$). The induced ovulations and spawnings were conducted by using intraperitoneal injections of HCG (2,000 IU/kg body weight) with photoperiod controls. Floating rates of artificially ovulated eggs were ranged from 22.9 to $65.7\%$. Fertilization and hatching rates were ranged from 69.0 to $86.2\%$ and 36.8 to $85.8\%$, respectively. All of those three rates between two experimental groups were not significantly different (P>0.05). Average survival rates of $F_2$ generation of 40-day-old 효nogenetic diploid larvae were slightly lower than those of controls (P<0.05), however growth rates were much higher than those of their diploid controls.

A Study on Dimension of the Characteristics of New Young Generation and Information Search for Buying Inner Wear (신세대특성(新世代特性)과 속옷 구매시(購買時) 정보탐색(情報探索)에 관(關)한 연구(硏究))

  • The purpose of this study was to identify the variables which could characteristics the New Young Generation (NYG), to investigate the relationship between level of NYG characteristics and information searching related to underwear purchasing, and the differences level of the NYG characteristics according to demographic variables (sex, grade, major), and examine the casual relationships of demographic variables (sex, place of growing background, parent's education) and level of the NYG characteristics on information searching related to underwear purchasing. The subjects were 723 college students (female = 324, male=398) living in Seoul and Kyuggi were participated in this study. The NYG is limited to person who born after 1970's. The result of this study were as follows; 1. Five factors of New Young Generation oriented characteristics were identified; F.1 'Info-telecommunication(Info-Telecom)'; F.2 'Family and Marriage'; F.3 'Consumption pattern'; FA 'Self-centered (Individualism)'; F.5 Work/Career oriented. 2. The relationship between the NYG characteristics and information search in underwear purchasing were significanted. 3. In demographic variables, gender, grade, and major were partially significant differences. 4. In male students, the result of the casual relationship between demographic variables with NYG characteristics and the level of the NYG in underwear purchasing showed that grade, background of grownplace, level of parent's education were significanted the high level of info-telecom factor associated with higher grade, city-grown, higher level of parent's education. In conclusion, the result showed that the level of NYG characteristics was significant to male students and specially for grade older students. The background of parent's education level was the another important variables to predicting the NYG characteristics. Finally the higher the NYG characteristics, the more information search in underwear purchasing.

Accumulation of triple recessive alleles for three antinutritional proteins in soybean with black seed coat and green cotyledon

  • The black seed coat of soybeans contain anthocyanins which promote health. However, mature soybean seeds contain anti-nutritional factors like lipoxygenase, lectin and Kunitz Trypsin Inhibitor (KTI) proteins. Furthermore, these seeds can be used only after the genetic elimination of these proteins. Therefore, the objective of this study was to develop novel soybean genotypes with black seed coat and triple recessive alleles (lx1lx1lx2lx2lx3lx3, titilele) for lipoxygenase, lectin, and KTI proteins. From a cross of parent1 (lx1lx2lx3/lx1lx2lx3, ti/ti, Le/Le) and parent2 (lx1lx2lx3/lx1lx2lx3, Ti/Ti, le/le), 132 F2 seeds were obtained. A 3:1 segregation ratio was observed during F2 seed generation for the inheritance of lectin and KTI proteins. Between a cross of the Le and Ti genes, the observed independent inheritance ratio in the F2 seed generation was 9: 3 : 3 : 1 (69 Le_Ti_: 32 leleTi_: 22 Le_titi: 9 leletiti) (χ2=2.87, P=0.5 - 0.1). From nine F2 seeds with triple recessive alleles (lx1lx1lx2lx2lx3lx3, titilele genotype), one novel strain posessing black seed coat, and free of lipoxygenase, lectin and KTI proteins, was selected. The seed coat color of the new strain was black and the cotyledon color of the mature seed was green. The weight of 100 seeds belonging to the new strain was 35.4 g. This black soybean strain with lx1lx1lx2lx2lx3lx3, titilele genotype is a novel strain free of lipoxygenase, lectin, and KTI proteins.

  • This study is to develop new wheat breeding material through wide hybridization with wild species harboring useful characteristics such as salt, heat, and drought tolerance. Leymus mollis, wild rye was used to improve wheat genetic quality. L. mollis, is a perennial plant harboring tolerance against salt, heat, and drought because L. mollis distributes on the seaside. The F1 hybrids were produced by crossing between common wheat (Triticum aestivum L., Chinese Spring) and L. mollis. Genomic in situ hybridization revealed that the F1 hybrids have L. mollis genome. For the evaluation of salt and drought tolerance, seeds from the F2 were used. Under 2% NaCl solution, the F3 wheat-Leymus addition plants with salt tolerance showed more tillering and longer roots than other F3 plants without salt tolerance. Also, the F3 plants with salt tolerance showed better shallow-rooted than other F3 plants without salt tolerance. Finally, the F3 plants with salt tolerance made seed-setting under 2% NaCl condition, but other F3 plants without salt tolerance were not. Under drought conditions, the F3 plants with drought tolerance showed longer culm and spike length than other F3 plants without drought tolerance and even those of Chinese Spring under well-water conditions. We evaluated and selected the F3 plants with salt or drought tolerance for generation advancement.

Growth Regulation in IGF-1 Receptor Transgenic Mice

  • To study the signaling effect of insulin-like growth factor-I(IGF-1), transgenic mice containing IGF-1 Receptor (IGF-1R) cDNA fused to metallothionein promoter were produced by DNA microinjection into the pronucleus of mouse zygote. Three founders were produced with transgenic mice containing IGF-1R gene. Transgenic mice lines contained approximately $4{\sim}20$ copies of transgenes per cell and transmission of this gene into the progeny with Mendelian manner were determined. The founder mice were mated with normal mice to produce $F_1$ mice and then $F_2$ mice. Transmission rates of IGF-1R transgene in the progeny mice were $25{\sim}60%$ in $F_1$ generation and $40{\sim}50%$ in $F_2$ generation. The mRNA expression of IGF-1R transgene in liver was analyzed using RT-PCR for IGF-1R gene in liver. When body weights of transgenic pups were measured during 4, 10 and 14 weeks after birth, IGF-1R transgenic mice grew faster than non transgenic littermates. This study indicated that growth regulation by IGF-1 signaling through IGF-1R can be elucidated using IGF-1R transgenic mice.

Inheritance of Resistance to Potato Virus Y Vein- necrosis Strain of N. africana (N. africana의 감자바이러스Y 엽맥괴저 계통에 대한 저항성의 유전)

  • A program was initiated to transfer potato virus Y vein-necrosis strain resistance from N. africana to N. tabacum The Fl plants between the above species were self-sterile, but all amphidiploid plants from the Fl plants and backcrossed flowers, that is, the N. tabacum flowers crossed with amphidiploid were self-fertile. The parent, amphidiploid plants of Fl, F2 population of the amphidiploid and the backcrossed generation were screened for a resistance of potato virus Y vein-necrosis strain isolated in Korea. The Chi-square values for the F2 population of the amphidiploid and the backcrossed generation fitted 35: 1 and 5 : 1 ratios of resistant to susceptible for the potato virus Y vein-necrosis strain, respectively. Therefore, it was found that the resistance of N. tabacum for the potato virus Y vein-necrosis strain was controlled by a single dominant gene.

Enhanced Seed Development in the Progeny from the Interspecific Backcross (Fagopyrum esculentum ${\times}$ F. homotropicum) ${\times}$ F. esculentum

  • To facilitate the introgression of F. esculentum into the traits of F. homotropicum, several accessions of the hybrids between these two species were pollinated with F. esculentum as the recurrent parent. The embryo in vitro rescue was performed to increase the recovery of backcross progenies. The $F_{2}$ generation was more amenable than $F_{1}$ hybrids to produce backcross progenies. The $F_{1}$ hybrids were backcrossed twice with common buckwheat (pin-type F. esculentum) (recurrent backcrossing). Also, alternate backcrosses with common buckwheat and F. homotropicum (congruity backcrossing) were carried out. Pollen tube growth of BC$F_{1}$ ${\times}$ F. esculentum (thrum) and F. homotropicum ${\times}$ BC$F_{1}$ was the disturbed penetration exceeded for all initial interspecific hybrids, and its requirement was proportionally lower when the common buckwheat was used as the recurrent parent and as the last parent of congruity hybrids. Effects of both common buckwheat and F. homotropicum on seed success rate for hybridization were observed. Growth of hybrid embryos before rescue, regeneration of mature hybrids all increased recurrent and congruity backcrosses and inter-crosses between $F_{1}$ plants and selected fertile plants of the second congruity backcrosses.
