Nonlinear FE modelling and parametric study on flexural performance of ECC beams

  • Engineered Cementitious Composite (ECC) is a special class of the new generation of high performance fiber reinforced cementitious composites (HPFRCC) featuring high ductility with relatively low fiber content. In this research, the mechanical performance of ECC beams will be investigated with respect to the effect of slag and aggregate size and amount, by employing nonlinear finite element method. The validity of the models was verified with the experimental results of the ECC beams under monotonic loading. Based on the numerical analysis method, nonlinear parametric study was then conducted to evaluate the influence of the ECC aggregate content (AC), ECC compressive strength ($f_{ECC}$), maximum aggregate size ($D_{max}$) and slag amount (${\phi}$) parameters on the flexural stress, deflection, load and strain of ECC beams. The simulation results indicated that when increase the slag and aggregate size and content no definite trend in flexural strength is observed and the ductility of ECC is negatively influenced by the increase of slag and aggregate size and content. Also, the ECC beams revealed enhancement in terms of flexural stress, strain, and midspan deflection when compared with the reference beam (microsilica MSC), where, the average improvement percentage of the specimens were 61.55%, 725%, and 879%, respectively. These results are quite similar to that of the experimental results, which provides that the finite element model is in accordance with the desirable flexural behaviour of the ECC beams. Furthermore, the proposed models can be used to predict the flexural behaviour of ECC beams with great accuracy.

Effects of Sire Birth Weight on Calving Difficulty and Maternal Performance of Their Female Progeny

  • Weight records from birth to calving and calving scores of 407 two-year old heifers and weights of their offspring from birth to one year of age were used to study the effects of sire birth weight on maternal traits of their female progeny. The heifers ($G_1$) were Ihe progeny of 81 sires ($G_0$) and were classified into three classes based on their sires' birth weights (High, Medium and Low). The heifers were from three distinct breed-groups and were mated to bulls with medium birth weights within each breed-group to produce the second generation ($G_2$). The data were analyzed using a covariance model. The female progeny of high birth-weight sires were heavier from birth to calving than those sired by medium and low birth-weight bulls. The effect of sire birth weight on calving difficulty scores of their female progeny was not significant. Grand progeny (G2) of low birth-weight sires were lighter at birth than those from high birth-weight sires (p < 0.05) but they did not differ significantly in weaning and yearling weights from the other two Grand progeny groups. The results indicated that using low birth weight sires would not result in an increase in the incidence of dystocia among their female progeny calving at two-year of age and would not have an adverse effect On weaning and yearling weights of their grand progeny.

  • The two-phase flow is observed in power plants, chemical process plants, and refrigeration systems etc., and it is very important to solve the heat transfer mechanism of a boiler, an automic reactor, a condenser and various types of evaporators. Recently, the problem of two phase heat transfer is braught up in many regions with development of energy saving technique. In flow boiling system it is necessary to store data in each condition because the heat transfer characteristics of flow boiling region vary by the change of flow pattern and the magnetude of heat flux to tube length, and be subtly affected by the flow and heating condition. So basic study for knowing flow pattern in heat transfer region and the relation between heat transfer characteristic and flow condition is desired to accumulate data in wide variety of liquid and flow system in the study of heat transfer of two phase flow. In this study R-113 was selected as working fluid whose properties were programmed by least square method, and experiment was conducted in the region of mass flow $1.628{\times}10^6$~$4.884{\times}10^6$/kg/$m^2$hr with inlet subcooling 10~3$0^{\circ}C$, sustaining test section inlet pressure to 1.5kg$_f$/$cm^2$abs.

Mitochondrial DNA Sequence Variation of the Mason Bee, Osmia cornifrons (Hymenoptera: Apidae)

  • In order to understand geographic genetic variation and relationship among populations of the mason bee (Osmia cornifrons Radoszkowsky), which is used as pollinator for apple tree, we sequenced a portion of mitochondrial (mt) COI gene, which corresponds to "DNA Barcode" region (658 bp) from 81 O. cornifrons individuals collected over eight localities in Korea. The sequence data revealed overall moderate to low genetic diversity within species, with a maximum sequence divergence of 0.76%. Geographically, two haplotypes (BAROC01 and BAROC02) were widespread with a frequency of 82.7%, whereas several haplotypes were found in a locality as a single individual, suggesting that haplotype distribution can be summarized as coexistence of a few widespread haplotypes and several regionally restricted haplotypes. Overall, high rate of per generation female migration (Nm=$1.1{\sim}$infinite) and low level of geographic subdivision ($F_{ST}=0{\sim}0.315$) among localities were characteristic. Although two populations (p < 0.026) were genetically subdivided from the remaining localities, no clear polarity was observed. Taken together, the nature of genetic divergence of the mason bee populations is characterized as one that possessing moderate to low genetic diversity, high gene flow, and wide spread haplotypes with ahigh frequency, concordant with the capability of dispersal in connection with the lack of historical biogeographic barriers.

  • Four programs, i.e. TRMM, ADEOS2, ASTER, and ALOS are going on in Japanese Earth Observation programs. TRMM and ASTER are operating well, and TRMM operation will be continued to 2009. ADEOS2 was failed, but AMSR-E on Aqua is operating. ALOS (Advanced Land Observing Satellite) was successfully launched on $24^{th}$ Jan. 2006. ALOS carries three instruments, i.e., PRISM (Panchromatic Remote Sensing Instrument for Stereo Mapping), AVNIR-2 (Advanced Visible and Near Infrared Radiometer), and PALSAR (Phased Array L band Synthetic Aperture Radar). PRISM is a 3 line panchromatic push broom scanner with 2.5m IFOV. AVNIR-2 is a 4 channel multi spectral scanner with 10m IFOV. PALSAR is a full polarimetric active phased array SAR. PALSAR has many observation modes including full polarimetric mode and scan SAR mode. After the unfortunate accident of ADEOS2, JAXA still have plans of Earth observation programs. Next generation satellites will be launched in 2008-2012 timeframe. They are GOSAT (Greenhouse Gas Observation Satellite), GCOM-W and GCOM-C (ADEOS-2 follow on), and GPM (Global Precipitation Mission) core satellite. GOSAT will carry 2 instruments, i.e. a green house gas sensor and a cloud/aerosol imager. The main sensor is a Fourier transform spectrometer (FTS) and covers 0.76 to 15 ${\mu}m$ region with 0.2 to 0.5 $cm^{-1}$ resolution. GPM is a joint project with NASA and will carry two instruments. JAXA will develop DPR (Dual frequency Precipitation Radar) which is a follow on of PR on TRMM. Another project is EarthCare. It is a joint project with ESA and JAXA is going to provide CPR (Cloud Profiling Radar). Discussions on future Earth Observation programs have been started including discussions on ALOS F/O.

Allozyme Variation and Population Structure of Carex okamotoi (Cyperaceae), a Korean Endemic Species (한국 내 국부적으로 분포하는 지리사초의 알로자임 변이와 집단구조)

  • The genetic diversity and population structures of fourteen Carex okamotoi (Cyperaceae) populations in Korea were determined using genetic variation at 25 allozyme loci. The Carex okamotoi species is native to Korea. It is endemic to three mountains (Mt. Taeback, Mt. Sobak, and Mt. Noreong) where it is found at 700~1,500 m above sea level. The percentage of polymorphic loci was 40.0%. Genetic diversity at the species level and at the population level was low ($H_{ES}$=0.106; $H_{EP}$=0.094), and the extent of the population divergence was relatively low ($G_{ST}$=0.082). Measurement of deviation from random mating ($F_{IS}$) within the 14 populations was 0.238. An indirect estimate of the number of migrants per generation was 2.78 (Nm=2.78). Analysis of fixation indices revealed a substantial heterozygosity deficiency in some populations and at some loci. Mean genetic identity between populations was 0.986.

Antioxidative Effects of Scutellariae Radix Aaquaacupuncture Solution on Lipid Peroxidation Induced by Free Radicals (자유기에 의한 지질과산화 반응에 대한 황금 약침액의 항산화 효능)

  • Scutellariae radix, has been used as a natural drug for fever, inflammation, cataract, and liver disease in traditional medicine. This study was performed in order to investigate the antioxidative effects of Scutellariae radix aqua-acupuncture solution (SRAS) on lipid peroxidation by free radicals. Lipid peroxidation levels were determined by TBA method during the autoxidation of linoleic acid. In this linoleic acid autoxidation system, SRAS markedly exhibited antioxidant activity, which inhibited 89% of linoleic acid peroxidation. SRAS showed scavenging effects on ${\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl$(DPPH) radical, inhibited superoxide generation in xanthine-xathine oxidase system, and also inhibited lipid peroxidation of rat liver tissue by hydroxyl radical derived from $H_2O_2-FE^{+2}$ system. These effects were similar to those of $dl-{\alpha}-tocopherol$, BHA and BHT. In addition, SRAS protected the cell death induced by ter-butyl hydroperoxide (t-BHP) and significantly increased cell viability in the normal rat liver cell (Ac2F). On the basis of these results, it is suggested that SRAS might play a protective role in lipid peroxidation by free radicals.

Formation and Regeneration of Penicillium verruculosum Protoplasts (Penicillium verruculosum의 원형질체 형성과 재생)

  • Optimal conditions for the formation and regeneration of protoplasts of the cellulolytic fungus Penicillium verruculosum were investigated. Among the various commercial cell wall lytic enzymes tested, 0.5%(w/ v) Novozym 234 was the most effective for protoplast formation. The highest yield of protoplast exceeding 4.5$\times$10$^6$/m$\ell$ obtained when 400mg of 20 hr-old mycelia was incubated with 0.5%(w/v) Novozym 234 at 3$0^{\circ}C$ for 1 hr. The best osmotic stabilizer for the isolation and re-generation of protoplasts was 0.7M sorbital (pH 5.6) and 0.6M MgSO$_4$(pH 5.6), respectively. When 0.6M MgSO$_4$was added as osmotic stabilizer to the complete medium, the maximum regeneration frequency obtained was 4.6-27.8%. Micromorphological change of giant protoplasts into hyphae was observed during incubation in the regeneration liquid medium.

  • In this study, the movable flame hardening process of 12Cr steel for a uniform hardness and desirable residual stress have been investigated. For this, the temperature cycles have been controlled accurately as a function of the three processing variables, the flame intensity $I_f$, the scanning velocity $V_s$, and the initial flame holding time $t_h$, where the standard surface temperature $T_{s,\;max}$, was maintained at $960^{\circ}C$. The optimized conditions were $V_s=0.68mn/s\;and\;t_h=67sec$ for the $C_3H_8:O_2\;=\;5:20l/min,\;V_s=0.80mm/s$ and $t_h=56sec$ for the $C_3H_8:O_2=6:24l/min,\;V_s=1.01mm/s\;and\;t_h=48sec$ for the $C_3H_8:O_2=7:28l/min,\;and\;V_s=1.15mm/s$ and $t_h=39sec$ for the $C_3H_8:O_2$=8:32 l/min. The optimally flame-hardened surface exhibited uniform distributions of the hardness and residual compressive stress over the treated area with moderate levels of $470{\sim}490HV_{0.2}$in hardness and $-300{\sim}-450MPa$ in residual stress, which were acceptable on the basis of the acceptance criteria of Siemens AG-KWU and GE Power Generation Engineering.

Green Fluorescent Protein-reporter Mammalian One-hybrid System for Identifying Novel Transcriptional Modulators for Human $p14^{ARF}$ Tumor Suppressor Gene

  • To improve conventional yeast one-hybrid screening, we have developed an efficient mammalian one-hybrid system that allows rapid isolation of com-plementary DNAs which are able to induce human p14$^{ARF}$. tumor suppressor gene. A 1.5 kb promoter region of p14$^{ARF}$ was fused to EGFP to generate ARF promoter-EGFP reporter vector. This reporter plasmid was stably trans-fected into NIH3T3 cells for generation of reporter cell line. When the reporter cell line was infected with E2F-1 together with excess amounts of empty vector, the cells that received the positive modulator were readily identifiable by green fluorescence using FACS. The GFP-positive cells were cloned directly from the cultured cells and expanded in bulk culture. The genomic DNAs from GFP-positive cells were prepared and the CDNA insert in integrated retroviral genome was recovered by PCR using primers annealing to the retroviral vector sequences flanking the insert-cloning site. This system should be useful for efficient screening of expression CDNA libraries in mammalian cells to identify novel upstream regulators for spe-cific genes by one-hybrid interaction.ion.
