본 논문에서는 자원 재활용을 위해 시멘트를 전혀 사용하지 않고 잠재 수경성을 지닌 고로슬래그와 알칼리 활성화제를 이용하여 모래를 고결시키는 연구를 수행하였다. 기존 수산화칼슘이나 수산화나트륨과 같은 화학적 알칼리 활성화제뿐 아니라 pH 10 이상에서 생존하는 극한미생물을 화학적 알칼리 활성화제에 혼합한 미생물 알칼리 활성화제를 개발하여 흙의 고결 가능성을 평가하였다. 낙동강모래에 고로슬래그의 함유량을 네 종류(4, 8, 12, 16%)로 달리하면서 화학적 또는 미생물 알칼리 활성화제를 혼합하여 공시체를 제작한 다음 7일 동안 대기중 양생시킨 후 일축압축시험을 실시하였다. 알칼리 활성화제의 종류에 관계없이 고로슬래그의 함유량이 4%에서 16%로 증가함에 따라 건조밀도가 증가하면서 일축압축강도는 평균 178kPa에서 2,435kPa까지 증가하였다. 화학적 알칼리 활성화제를 사용한 경우, 수산화칼슘이 포함된 공시체의 일축압축강도가 수산화나트륨을 사용한 경우보다 5-54% 정도 높게 나타났다. 한편 본 연구에서 개발한 미생물 알칼리 활성화제를 사용한 경우, 수산화칼슘 성분이 포함된 공시체의 경우에는 화학적 알칼리 활성화제보다 일축압축강도가 11-60% 감소하였으나, 수산화나트륨이 포함된 경우에는 일축압축강도가 19-121% 증가하였다. 고결된 공시체에서 C-S-H 화합물이 생성되었으며, SEM분석에서 고로슬래그 함유량이 증가할수록 수화물의 양도 증가하였다.

The Earth contains billions of microbial species, although the vast majority cannot be cultured in laboratories and are thus considered unidentified and uncharacterized. Extremophiles are microorganisms that thrive in extreme conditions, including temperature, salinity, and pH. Extremophilic microorganisms have provided important insights for biological, metabolic, and evolutionary studies. Between 2017 and 2019, as part of a comprehensive investigation to identify bacterial species in Korea, eight bacterial strains were isolated from marine and non-marine environments in Jeju Island. These strains were cultured under extreme salinity or pH conditions. Phylogenetic analysis using 16S ribosomal RNA(rRNA) gene sequencing indicated that all eight strains belonged to the phyla Gammaproteobacteria, Bacilli, and Alphaproteobacteria. Based on their high 16S rRNA gene sequence similarities(>98.7%) and the formation of strong monophyletic clades with their closest related species, all isolated strains were considered as an unrecorded strain, previously unidentified species. Gram stain reaction, culture conditions, colony and cell morphology, biochemical characteristics, isolation source, and National Institute of Biological Resources(NIBR) IDs are described in this article. The characterization of these unrecorded strains provides information on microorganisms living in Korea.
그람 음성이며 황색인 긴 막대 모양의 세균 Spirosoma montaniterrae $DY10^T$는 전라북도 덕유산에서 분리가 되었다. 이 세균의 세포는 감마선에 대해 12 KGy의 $D_{10}$값을 보이며 극단적인 감마선 저항성을 보였다. $DY10^T$ 균주의 완전한 게놈서열은 5,116개의 유전자, 39개의 tRNA 유전자를 포함하는 원형 염색체(5,797,678 bp)로 구성되었다. 유전체 특징은 감마선 및 UVC에 대응하는 주요 효소를 포함하였다.

To investigate the mode of bactericidal action for antimicrobial peptide, pediocin, synthetic and mutant pediocins were prepared by direct chemical synthesis. Native pediocin was purified from Pedio-coccus acidilactici M and its conformational structure and bactericidal functions were analyzed and compared to synthetic pediocin. Schematic mode of pediocin actions, how pediocin binds on the target cell membrane, penetrates and makes tunnel are proposed. For these purposes, primary and secondary structures of pediocin was analyzed and disulfide bond assignment was also done. The pediocin purified from P. acidilactici M had high effective bactericidal ability against gram positive bacteria, especially Listeria monocytogenes and was very stable at extreme pHs and even at high temperatures such as autoclaving temperature (121$^{\circ}C$). Pediocin was consisted of 44 amino acids with four cysteines. Novel synthetic peptides were achieved by solid phase peptide synthesis(SPPS) method. To explain the function of cysteine in C-terminal region, mutant pediocin, Ped[C24A+C44A], was synthesized and their structural and biological functions were analyzed. Second mutant pediocin, Ped[KllE], was prepared to explain the function of lysine at 11 of N-terminal part of pediocin, especially loop of $\beta$-sheet, and to predict the initial binding site of pediocin. The native and synthetic pediocins was showed random coil conformation by spectropolarimetry in moderate conditions. This conformation was observed in extreme conditions such as high temperature and low and high pHs, also. Circular dichroism(CD) data also showed the existence of $\beta$-turn structure in N-terminal part both native and synthetic pediocins. A structural model for pediocin predicts that 18 amino acids in the N-terminal part of the peptide assume a three-strand $\beta$-sheet conformation. This random coil in C-terminal part of pediocin was converted to folding structure, helix structure, in nonpolar solvents such as alcohol and TFE. The disulfide bond between $^{9}$ Cys and $^{14}$ Cys was concrete and inevitable, however, evidences of disulfide bond between $^{24}$ Cys and $^{44}$ Cys was not. Data of Ped[C24A+C44A], pediocin mutant showed that $^{44}$ Cys was required during killing the target cells but not inevitable, since Ped[C24A+C44A] still have bactericidal activity but much less than native pediocin. Another pediocin mutant, Ped[KllE], had still bactericidal activity, was controversial to propose that positive charge like as $^{11}$ Lys in loop or hinge in bacteriocin bound or helped to binding to microorganism with electrostatic interaction between cell membrane especially teichoic acid and positive amino acid nonspecifically. The conformation of pediocin among native, synthetic and mutant pediocins did not show big difference. The conformations between oxidized and reduced pediocin were almost similar regardless of native or synthetic.
Literatures on microorganisms of traditional Korean nuruk published since 1945 were reviewed in this paper. Traditional Korean nuruk consists of raw barley and various grains. Traditional Korean nuruk consists of unbolied raw barely and various grains. They are ground to paste and moistened, and then naturally inoculated by airborne microorganisms. Therefore, many kinds of microorganisms such as fungi, yeast, and bacteria grwo in nuruk. Since 1945, new 14 species of Aspergillus and 9 species of Penicillium have been identified from traditional Korean nuruk. Total number of fungal species identified so far is now up to 38 species among 12 different genus. Among newly isolated fungal species, Aspergillus penicilloides and Penicillium, expansum showed not only high production rate of acid and amylase but also extreme stability of the enzyme at room temperature for 3 months. As examples of newly isolated yeast species, there are 5 species of Candida, 4 species of Hansenula, 1 species of Pichia and 1 species of Schizosaccharomyces. Total number of yeast species isolated so far is up to 18 species from different 8 genus. Newly isolated bacteria, were Bacillus pumilus, Lactobacillus casei and Leuconostoc mesenteroides.
The structural components of microorganism are quite related to the toxin and environmental conditions. The vegetative and dormant cells are quite affected by the physical and chemical environments to survive and they will be dormant when they are in the extreme environment. The mechanism to activate the microorganisms however, is not well defined yet in the area of activation state and sporulation state through the analysis of EPS. Other than that even the main mechanism of prior to acquisition of analysis values is not well understood. Therefore, what kind of specific environment to affect the activation and sporulation will be discussed through the analysis of the extracellular polymeric substances(EPS). EPS are a high molecular weight mixture of polymers presenting both outside of cells and interior of microbial aggregates. They are a major complex materials in microbial aggregation for sustaining them together in a three dimensional matrix. Three commonly used extraction methods were applied to this study their effectiveness and quantification in extracting EPS from several Bacillus microorganisms and activated sludge. Three extraction methods used for this study are regular centrifugation with formaldehyde (RCF), Steaming, and EDTA extraction. The results of EPS contents such as the quantitative proteins, carbohydrates and the ratio of protein versus carbohydrate from the several species with the several specific methods showed in this research. This study aims to get comparable results of the quantitative production of EPS and the effectiveness of sedimentation for Bacillus microorganisms and activated sludge from several wastewater treatment plans. The results revealed that the protein amount extracted was the highest by the Steaming method of three extraction methods before sporulation and the carbohydrate amount extracted was the highest by the RCA method of three extraction methods after sporulation. The higher amount of protein compared with carbohydrate from Bacillus microorganisms affected higher sedimentation efficiency, however it could not be found the relation between the EPS production and sedimentation efficiency for the activated sludge.
슬러지의 침전성에 영향을 미치는 인자는 F/M비, 유입수의 성분과 조성, 처리수온, 포기조의 pH, 슬러지 일령, 사상성세균의 증식 등으로 알려져 있으며, 유기산의 잔류량이 EPS의 분비에 영향을 미쳐 슬러지의 침강성에 영향을 주게 된다. 바실러스를 이용한 공법에서 생물 반응조는 특히 바실러스 종의 우점화를 위한 포자화의 진행을 이루는데 중요한 것으로 알려져 있으나 이에 대한 기작 설명은 아직 미흡한 실정이다. 바실러스를 이용한 B3 공법과 RABC 공법으로 운전 중인 처리장의 생물반응조는 점감포기를 하고 있으며 침전조에서 포자를 형성한 바실러스가 생물반응조로 반송되면 1.6mg/L로 높아진 DO농도와 유기물질 유입으로 바실러스가 발아를 하고 활성도가 높아지며, 0.5mg/L로 낮아진 DO농도로 인하여 포자를 형성하면 EPS 추출물질 중 Protein의 함량이 109.95mgEPS/gSS에서 131.77 mg EPS/gSS로 증가하고 SVI는 85mL/g에서 96mL/g사이로 양호함에 따라 DO농도가 EPS 추출물질 중 Protein의 생성에 영향을 미치며 Protein의 함량이 침전에 영향을 미치는 것으로 판단된다. 또한 Protein/Carbohydrate 비율에 따라 침강성은 영향을 받는 것으로 나타났다. 특히 RABC 공법에서는 Protein/Carbohydrate의 비가 침전에 영향을 미치는 것으로 나타난 반면, B3 공법에서는 Protein/carbohydrate의 비에 영향을 크게 받지 않는 것으로 나타났다.
시판 샐러드제품의 구입부터 가정까지의 이동 및 소비직전까지 적정하지 않은 온도관리를 예상하여 단기간의 온도 abuse 상황을 설정하고 미생물적 유통기한을 도출하였다. 보다 효율적인 유통기한 설정을 위해 예측미생물학의 3단계 모델인 PMP 7.0을 활용하여 그 활용가능성을 조사하였다. 부적절한 온도에서의 abuse 시간이 증가할수록 미생물은 빠르게 증식하여 샐러드 제품의 유통기한 시점으로 판단되는 log 7 CFU/mL에 도달하는 시간이 짧아졌다. 온도가 증가할수록 0.020에서 1.083까지 grow rate도 증가했으며, 이 모델의 적합도를 나타내는 $r^2$의 값은 전 실험구에서 0.9 이상을 나타내었다. PMP 7.0으로 예측된 미생물의 증식양상은 온도에 따라 0.017~0.235 CFU/mL/hr로 나타났으며, 모든 구에서 0.994~1.000까지 높은 수준의 $r^2$을 나타내었다. 또 PMP를 활용하여 도출한 유통기한의 경우도 온도가 증가함에 따라 감소하였다. 실측된 값을 바탕으로 한 샐러드 제품의 유통기한은 유통매장에 도착하기까지 48시간 소요될 것으로 예상할 경우 유통기한은 109.1~63.0시간까지로 추정되며 PMP로 도출된 유통기한(24.1~626.5시간)에 비해 짧게 나타났다. 이는 온도 abuse에 의한 영향 및 fail safe에 해당하는 결과로 안전성 측면에서는 유리하나 관리적 측면에서 과도한 기준의 설정 등을 통해 관리비용의 증가 등의 단점이 발생할 수 있는 것으로 판단된다. 즉, 예측미생물학을 활용하여, 유통기한 설정 및 품질관리를 위한 초기 미생물 기준 설정시 특정 식품에 적용하는 것은 효율적인 시도가 될 것이나, 이를 전반적인 기준으로 설정하는 것은, 통계적, 실제적 오류 발생이 가능할 것으로 오히려 관련 효율을 저해할 수 있을 것이다.
