  • In this study, the extraction efficiency and reflection coefficient by a two-dimensional OWC (Oscillating Water Column) WEC (wave energy converter) installed in front of a seawall was investigated for regular/irregular waves. The matched eigenfunction expansion method (MEEM) based on the linear potential theory was applied as an analytical tool. The diffraction problem by the incident wave in the open-chamber and the radiation problem by the oscillating pressure in the closed-chamber were solved to obtain the volume fluxes at the internal free-surface. Applying the volume fluxes into the continuity equation for the airflow in a chamber, we got the oscillating air pressure. The maximum extracted power and corresponding reflection coefficient were determined at the optimal turbine coefficient that maximizes the extracted power. OWC device designed for a high extracted efficiency simultaneously contributes to reduce reflected waves.

  • A solvent extraction-atomic absorption spectrophotometry for determination of trace amount of cadmium, copper, lead and zinc and a flameless atomic absorption spectrophotometry for mercury in sea water were studied. The optimum pH range for solvent extraction was pH 4-7. A better solvent extraction efficiency was obtained with MIBK solvent than nitrobenzene, benzene, isoamylalcohol, n-buthylacetate. DDTC was more advantageous than APDC as chelating agent. The metals, chelated with DDTC and concentrated into MIBK by solvent extraction with a volume of $1\iota$ of sea water for cadmium, copper and lead, and 200m1 for zinc, were determined simultaneously by atomic absorption spectrophotometry. For mercury determination, 500ml of sea water was digested with permanganate-sulfuric acid and mercury( II ) was reduced by stannous chloride and aerated the solution with air pump until the absorbance reached a constant value. The precisions, in standard deviation, of these methods were 0.058ppb for cadmium, 0.084 ppb for copper, 0.44ppb for lead, 2.49ppb for zinc and 0.08 ppb for mercury. The sensitivities, expressed in $ppb/1\%$ absorption, were 0.058 ppb cadmium, 0. 15 ppb copper, 0.6 ppb lead, 1.2 ppb zinc and 0.01 ppb mercury respectively. No significant adsorption on the wall of polyethylene sample bottle occurred during 30 days of storing by acidification to pH 1.5 with nitric acid except zinc. Poor reproducibility was found for zinc with this method.

  • In the present works, the High-speed Cavitation Tunnel (HCT) has been designed and manufactured to have the large test section to conduct various supercavitation experiments. The large amount of air ventilated behind a cavitator produces lots of tiny bubbles, which prevent clear observation of supercavitation at the test section. To collect small bubbles effectively, a bubble collecting section of large volume is equipped upstream of the test section. HCT has the test section dimension of $0.3^H{\times}0.3^W{\times}3.0^L\;m^3$ and provides maximum flow speed of 20.4 m/s at the test section. The blockage and Froude effects on the ventilated supercavitation are investigated successfully at the test section. The basic studies such as the supercavitation evolution, drag measurements and cavity shape extraction with air flow rate are also carried out in HCT.

  • The purpose of this study is to develop a badge-type passive sampler for the measurement of short-term nitrogen dioxide and to evaluate its performance. The principle of the method is a colorimetric reaction of nitrogen dioxide with sulfanilic acid, N-1-naphthylethylendiamine, and phosphoric acid. First, it has been shown that the filter paper should be rinsed with ultrapure water and ultrasound, and then dried in a vacuumed desiccator. The concentration and volume of absorption reagent (triethanolamine) were $20\%$ and 100 ${\mu}L$, respectively. The extraction time was determined as 60 min. Second, duplicate measurements (n= 116) were carried out for evaluating the precision of the passive sampler. The relative error and the correlation coefficient between duplicates are $3.4\pm 3.0\%$ and 0.994, respectively. In addition, the $95\%$ confidence interval of intraclass correlation coefficient and the estimated value are 0.992$\sim$0.996 and 0.994, respectively. Third, a paired t-test was carried out for evaluating the accuracy of the passive sampler (n=40). In the result of the test, the $95\%$ confidence interval of the difference was -1.710 ppb <$\gamma$< 0.788 ppb. Finally, the average concentration of blanks, measurement detection limit, limit of detection, and limit of quantification are $2.4\pm 0.4$ ppb, 104 ppb, 3.8 ppb, and 7.0 ppb, respectively.

  • Nitroarenes are ubiquitous environmental pollutants displaying potent mutagenicity in bacteria and carcinogenicity in mammal. In this study, the concentration of nitroarenes in coarse and fine particles and mutagenicity of POC$\_$N/ fraction was investigated in suspended particulates at the Shinchon and Bulkwang area of Seoul. The suspended particulates were collected bimonthly by a high volume cascade impactor air sampler from July 1987 to May 1988. Extractable organic matter was obtained by ultrasonic extraction on diethly ether/cyclohexane (8/2, v/v). Neutral fraction was obtained by liquid-liquid extraction. Polar neutral organic compounds (POC$\_$N/) was fractionated by thin-layer chromatography. Finally, the concentrations of nitroarenes in POC$\_$N/ fraction were measured and determined by capillary gas chromatography. Direct and indirect mutagenicity of POC$\_$N/ fraction were measured using Salmonella typhimurium TA 98. The result were as follows: 1) Major nitroarenes at the Shinchon area was 1-nitropyrene and at the Bulkwang area it was 2,7-dinitro-9-fluorenone during the year. 2) Average concentration of total nitroarenes measured was 67.26 ng/m$^3$in fine particles which was 1,3 folds higher that in coarse particle (52.30 ng/m$^3$). 3) Annual pattern of nitroarenes concentrations revealed that concentration during heating season (Feb., Jan., Mar.) was 2.2 folds higher than that in non heating season (May, Jul., Sep.). Concentration of each season has 157.68 ng/m$^3$and 80.39 ng/m$^3$. 4) The mutagenic activity of POC$\_$N/ fraction from fine particles was higher compared to that of coarse particles and was increased when metabolically activated, with 59 mixture. Mutagenicities, Metabolically activated, were significantly different between Shinchon and Bulkwang area, 322.8 rev/250 $\mu\textrm{g}$/plate and 286.8 rev/250 $\mu\textrm{g}$/plate, respectively. 5) Annual pattern of mutagenicity of POC$\_$N/ fraction revealed that mutagenicity during the heating season was 1.7 folds higher at Shinchon area and 1.2 folds higher at Bulkwang than during the non heating season. The variable contents and levels of nitroarenes in suspended particulates may affect human health significantly. Further studies such as risk assessment should be conducted on the basis of these kind of studies.

  • Transgenic animals production tools have been valuable for research and purpose. The current methods of gene transfer, microinjection and nuclear transfer, which are widely used in transgenic animal production, but all most methods has only had limited success in production of larger species. Here, we report the possibility of a sperm-mediated gene transfer method in porcine embryos. Oocytes were collected from ovaries harvested at a local slaughterhouse were matured in 500${mu}ell$ drops of TCM-199 under mineral oil at 38.5$^{\circ}C$ in a humidified atmosphere of 5%CO2 in air. After 42-43h of in vitro maturation oocytes were denuded. for sperm injection into the cytoplasm of the porcine oocytes, sperm suspension in NIM medium are subjected extraction with TritonX-100 before mixing with a green fluorescent gene (GFP). Sperm with Tritonx-100 were prepared by adding TritonX-100 to a final volume of 0.05% in the sperm suspension and mixing by trituration for 60s before two wishes in NIM medium at 2$^{\circ}C$. A(ter wishing, sperm were mixed with TritonX-100 at $25^{\circ}C$ followed by washes at 2$^{\circ}C$. Sperm were resuspended in ice cold NIM to a final volume of 400${mu}ell$ and 2-20ng/${mu}ell$ DNA were triturated on ice for 60s. All microinjection was performed in HEPES-buffered CZB medium at room temperature within 2h. After culture in NCSU-23 for 72h, percent of porcine embryos transfected GFP gene are 20.7%(6/29) in 20ng/${mu}ell$ sperm-DNA mixed group and other groups were 3.7 %(2/54)and 4.7%(3/67). These data suggests that sperm-mediated gene transfer method should be used to the production tool of transgenic pig efficiently.

  • In recent years, packed column has been widely used in separation processes, such as absorption, desorption, distillation, and extraction, in the petrochemical, fine chemistry, and environmental industries. Packed column is used as a contacting facility for gas-liquid and liquid-liquid systems filled with random packed materials in the column. Packed column has various advantages such as low pressure drop, economical efficiency, thermally sensitive liquids, easy repairing restoration, and noxious gas treatment. The performance of a packed column is highly dependent on the maintenance of good gas and liquid distribution throughout a packed bed; thus, this is an important consideration in a design of packed column. In this study, hydraulic pressure drop, hold-up as a function of liquid load, and mass transfer in the air, air/water, and air-NH3/water systems were studied to find the geometrical characteristic for raschig super-ring experiment dry pressure drop. Based on the results, design factors and operating conditions to handle noxious gases were obtained. The dry pressure drop of the random packing raschig super-ring was linearly increased as a function of gas capacity factor with various liquid loads in the Air/Water system. This result is lower than that of 35 mm Pall-ring, which is most commonly used in the industrial field. Also, it can be found that the hydraulic pressure drop of raschig super-ring is consistently increased by gas capacity factor with various liquid loads. When gas capacity factor with various liquid loads is increased from 1.855 to 2.323 kg-1/2 m-1/2 S-1, hydraulic pressure drop increases around 17%. Finally, the liquid hold-up related to packing volume, which is a parameter of specific liquid load depending on gas capacity factor, shows consistent increase by around 3.84 kg-1/2 m-1/2 S-1 of the gas capacity factor. However, liquid hold-up significantly increases above it.
