Response Surface Optimization of Phenolic Compounds Extraction From Steam Exploded Oak Wood (Quercus mongolica)

  • Steam explosion was applied to extract phenolic compounds from oak wood (Quercus mongolica). The effects of three independent factors (ethanol concentration, extraction temperature and extraction time) on the total phenolic content, DPPH radical scavenging activity, and antimicrobial activity from the steam exploded oak wood were optimized using response surface methodology (RSM). The independent variables were coded at three levels and their actual values were selected on the basis of preliminary experimental results. The following optimal extraction conditions were selected: ethanol concentration 82.0%, extraction temperature $71.7^{\circ}C$, and extraction time 60.5 min for total phenolic content; ethanol concentration 78.3%, extraction temperature $70.3^{\circ}C$, and extraction time 57.6 min for DPPH radical scavenging activity; ethanol concentration 80.6%, extraction temperature $68.4^{\circ}C$, and extraction time 59.0 min for antimicrobial activity. The experimental values agreed with those were predicted within confidence intervals indicating the suitability of RSM in optimizing the ethanol extraction of phenolic compounds from the steam exploded oak wood. Under the optimized conditions, the experimental value of the total phenolic content was 111.8 mg GAE/g dry steam exploded oak wood, DPPH free radical scavenging activity was 65.7%, and antimicrobial activity was 17.0 mm, and those are reasonably close to the predicted values (109.2 mg GAE/g dry steam exploded oak wood, 62.3% and 15.9 mm, respectively).

Study on Extraction of the Center Point of Steam Generator Tubes (증기발생기 세관의 중심좌표추출에 대한 연구)

  • This paper describes extraction procedure for the center coordinates of steam generator tubes of Youngkwang NPP #6, which are arrayed in triangular patterns. Steam generator tube images taken with wide field-of-view lens and low-light lamp mounted on a ccd camera tend to have low contrast, because steam generator is sealed and poorly illuminated. The extraction procedures consists of two steps. The first step is to process the region with superior contrast in entire image of steam generator tubes and to extract the center points. Using the extracted coordinates in the first step and the geometrical array characteristics of tubes lined up in regular triangle forms, the central points of the rest region with low contrast are estimated. The straight lines from center point of a tube to neighbour points in horizontal and 60, 120$^{\circ}$ degree directions are derived. The intersections of straight line In horizontal direction and slant line in regular triangle direction are selected as the center coordinates of steam generator tubes. The Chi-square interpolation method is used to determine the line's coefficients in horizontal and regular triangle direction.

  • This study was performed to develop extract materials from Elsholtzia splendens by analyzing the functionality and aroma profile by the simultaneous steam distillation extraction. The qualities of extracts such as total yield, total phenolic compound and electron donation ability were affected by extraction temperature than time. The main flavor compounds were analyzed as estragole, thymol and beta-caryophyllene. The response variables had significant with extraction temperature than tim. The established model was suitable to predict calculated value in experimental ranges. The optimum extinction conditions, which were limited of maximum value for dependent variables were $108^{\circ}C$ and 2.1 hr in the simultaneous steam distillation extraction method.

Pretreatments of Softwood Sawdust for Mycelial Growth of Lentinus edodes

  • Mycelial growth of L. edodes by pretreatments of softwood was studied on a sawdust medium. The sawdust used was from the following softwood species : Larix leptolepis, Pinus densiflora and Pinus koraiensis. The pretreatment consisted of cold-water (48 h), hot-water (3 h) and steam extractions (3 h) at a ratio of 500 g : 3,000 mL (sawdust : distilled water). The sawdust medium was a mixture of 76% sawdust, 20% rice bran, 3% glucose, 0.4% potassium nitrate and 0.6% calcium carbonate. Following sawdust pretreatments proved most suitable : L. leptolepis (steam extraction), P. densiflora (hot-water extraction) and P. koraiensis (hot-water extraction). Mycelial growth on P. koraiensis sawdust increased in proportion to an increase in hot-water extraction time. Mycelial growth was optimum on the sawdust extracted for 12 hours, hot-water extraction beyond this period proved unsuitable. With the exception of P. densiflora at 100 ㎍/mL, antifungal activity occurred in every sample. Maximum inhibition of mycelial growth was obtained from following concentration of hot-water extractives : P. densiflora (104 ㎍/mL) and P. koraiensis (104 ㎍/mL). This study has provided useful preliminary information for the cultivation of L. edodes.

  • The extraction efficiency depending on the extracting methods and the conditions of extraction was investigated. The common steam extraction was compared to the distillation extraction method. The effects of the samples size and the extraction time on the extract yield were also investigated by using UV-Vis spectrophotometer. One of the functional components of pine needle extract as the natural phenol base components were detected by the UV-VIS at around 235 nm wavelength range. The absorbance intensity at around 235 nm wavelength of the pine needle extract was used as the indicator of the extraction efficiency in this experiment. The distillation extraction showed the higher extract yield than the steam extraction. The grinding treatment of pine needles also resulted in the better extract performance, but the severe grinding showed a little decrease in the extract yield especially in case of the distillation extraction method. More than half of the extract was collected at the first stage of the extraction, that was the first 15 minutes in the total 60 minutes extraction.

  • Steam defiberated pulp and steam exploded wood(birch chip) were extracted with solvents (hot-water, 1% NaOH, MeOH, hot water, 1% NaOH). The properties of residual fiber were examined for the utilization as ruminants feed. The digestibility is 38% in steam defiberated pulp(10kg /$cm^2$-15min) and 62-77% in exploded wood(17-18kg/$cm^2$-2~10min), respectively. The more steam pressure and time increase, the more the digestibility increase. The sugars obtained from extractives is amount from 7% to 13% in asplund pulp and from 7% to 10% in exploded pulp. The sugars was mainly composed of 70-80% xylose. The digestibility of residual fiber which is extracted with solvents is low than these of original fibers. Considering the yield and digestibility as ruminant feed, exploded pulp under 17kg /$cm^2$ for 10min has the best efficiency. The exploded wood gives 75.3% on yield(O. D. chip) and 48% on the digestibility.

A Two-stage Process for Increasing the Yield of Prebiotic-rich Extract from Pinus densiflora

  • The importance of polysaccharides is increasing globally due to their role as a significant source of dietary prebiotics in the human diet. In the present study, in order to maximize the yield of crude polysaccharides from Pinus densiflora, response surface methodology (RSM) was used to optimize a two-stage extraction process consisting of steam explosion and water extraction. Three independent main variables, namely, the severity factor (Ro) for the steam explosion process, the water extraction temperature ($^{\circ}C$), and the ratio of water to raw material (v/w), were studied with respect to prebiotic sugar content. A Box-Behnken design was created on the basis of the results of these single-factor tests. The experimental data were fitted to a second-order polynomial equation for multiple regression analysis and examined using the appropriate statistical methods. The data showed that both the severity factor (Ro) and the ratio of water to material (v/w) had significant effects on the prebiotic sugar content. The optimal conditions for the two-stage process were as follows: a severity factor (Ro) of 3.86, a water extraction temperature of $89.66^{\circ}C$, and a ratio of water to material (v/w) of 39.20. Under these conditions, the prebiotic sugar content in the extract was 332.45 mg/g.

Comparison of Essential Oil Composition Extracted from Agastache rugosa by Steam Distillation and Supercritical Fluid Extraction (수증기 증류법과 초임계유체 추출법으로 분리한 배초향의 정유성분 조성 비교)

  • In order to compare the extraction patterns of main components from the raw material between the extraction methods, the aerial parts (dried stem, leaves, and flowers) of Agastache rugosa were extracted by SDE simultaneous steam distillation & extraction) and SFE (supercritical fluid extraction). Volatile components of essential oil and extract were identified by GC and GC-MSD. The contents of essential oil extracted by SDE were 0.49% in aerial part of Agastache rugosa on dry basis. Major components were methyl chavicol(27.2%), isomenthone(24.6%), hexadecanoic acid(13.0%). menthone (5.5%) among 32 kinds of components confirmed in essential oil. On the other hand, the contents of SFE extracts revealed 3.21% on dry basis, 6 times higher than those of SDE. Major components were isomenthone(15.3%), hexadecanoic acid(13.7%), methyl chavicol(12.6%), benzoic acid(3.8%) among 33 kinds of components identified in extract.

  • The essential oil of Artemisia apiaceae Hence was extracted by simultanous distillation-extraction and steam distillation and analyzed by GC and GC-MS. The oil content of Artemisia apiaceae Hance was 0.23% when extracted by steam distillation and 0.37% in case of simultaneous distillation-extraction and sensory analysis of the oil indicated camphorous and herbal characteristic notes. the experimental results confirmed the presence 34 volatile components, the major components were camphene, camphor borneol and caryophyllene. 5 fraction have a good aroma character among 11 fraction were seperated by using silicagel column chromatography. This can is used for the pharmaceutical industry because of amedical action.
