Angiogenesis is a essential part for bone formation and bone fracture healing. Vascular endothelial growth factor (VEGF), one of the most important molecules among many angiogenic factors, is a specific mitogen for vascular endothelial cells. VEGF-mediated angiogenesis is required for bone formation and repair. However, the effect of VEGF on osteoblastic cells during osteogenesis is still controversial. In recent days, substantial progress have been made toward developing tissue-engineered alternatives to autologous bone grafting for maxillofacial bony defects. Periosteum has received considerable interest as a better source of adult stem cells. Periosteum has the advantage of easy harvest and contains various cell types and progenitor cells that are able to differentiate into a several mesenchymal lineages, including bone. Several studies have reported the bone formation potential of periosteal cells, however, the correlation between VEGF signaling and cultured human periosteal cell-derived osteogenesis has not been fully investigated yet. The purpose of this study was to examine the correlation between VEGF signaling and cultured human periosteal-derived cells osteogenesis. Periosteal tissues of $5\;{\times}\;20\;mm$ were obtained from mandible during surgical extraction of lower impacted third molar from 3 patients. Periosteal-derived cells were introduced into the cell culture and were subcultured once they reached confluence. After passage 3, the periosteal-derived cells were further cultured for 42 days in an osteogenic inductive culture medium containing dexamethasone, ascorbic acid, and ${\beta}-glycerophosphate$. We evaluated the alkaline phosphatase (ALP) activity, the expression of Runx2 and VEGF, alizarin red S staining, and the quantification of osteocalcin and VEGF secretion in the periosteal-derived cells. The ALP activity increased rapidly up to day 14, followed by decrease in activity to day 35. Runx2 was expressed strongly at day 7, followed by decreased expression at day 14, and its expression was not observed thereafter. Both VEGF 165 and VEGF 121 were expressed strongly at day 35 and 42 of culture, particularly during the later stages of differentiation. Alizarin red S-positive nodules were first observed on day 14 and then increased in number during the entire culture period. Osteocalcin and VEGF were first detected in the culture medium on day 14, and their levels increased thereafter in a time-dependent manner. These results suggest that VEGF secretion from cultured human periosteal-derived cells increases along with mineralization process of the extracellular matrix. The level of VEGF secretion from periosteal-derived cells might depend on the extent of osteoblastic differentiation.
본 연구는 ipriflavone(isopropoxysioflavone)의 투여가 백서 두개관세포의 증식과 골조직 개조에 미치는 영향을 알아보고자 시도되었다. 태령 20-21일째의 백서 두개관세포를 분리 배양한 후, $10^{-9}M$부터 $10^{-5}M$까지 농도의 ipriflavone을 투여하고 1일째와 3일째에 MTT분석을 시행하여 흡광도를 평가한 결과, 모든 농도에서 백서 두개관세포의 증식을 보이지 않았다. 한편 골조직 개조에 미치는 영향을 알아보기 위하여 14일째에 alizarin red 염색을 시행하여, 형성된 석회화 결절 면적을 측정하였을 때, $10^{-8}M,\;10^{-7}M,\;10^-6}M$농도를 투여한 경우 석회화 결절 형성이 유의하게 증가하였다 골아세포의 분화에 미치는 영향을 알아보기 위하여 ipriflavone을 투여하고 7일째와 14일째에 추출한 RNA를 역전사 중합효소 연쇄반응(RT-PCR)을 시켜 bone sialoprotein(BSP), type I collagen(COL I) osteocalcin (OCN) 유전자 발현을 관찰한 결과 BSP와 COL I 유전자는 배양 7일째 높은 발현을 보였고, OCN 유전자는 배양 14일째 높은 발현을 보였다. 이상의 연구결과 ipriflavone이 백서 두개관세포에서 석회화를 촉진시키고 골아세포의 분화에 관여하는 BSP, COL I 및 OCN 유전자 발현을 증가시켜 골조직의 개조를 빠르게 할 수 있음을 시사하였다.
LOX는 결합조직의 세포외기질에 존재하여 교원섬유와 탄성섬유의 교차결합을 촉진시키는 핵심적인 효소이다. 뼈세포에서 세포분화 및 세포이동과 관련된 LOX의 역할은 보고되었으나, 상아모세포 분화에서 LOX에 대한 직접적인 효과는 거의 알려지지 않았다. 이 연구에서는 인간치수세포에서 상아모세의 분화과정 동안 LOX의 역할에 대해 실험하였다. 치수세포에 저 혈청 분화유도 배지를 처리하여 0, 1, 3, 7, 14일 동안 배양하였다. 세포성장은 세포계수, 분화와 관련된 유전자들은 RT-PCR, 광물화는 alrizarin red S 염색으로 각각 실험하였다. LOX 유전자 발현은 RT-PCR로 측정하였고, LOX 효소활성은 고감도 형광분석을 시행하였다. 1. 세포성장은 저 혈청 분화유도 배지에서 시간 의존적으로 증가하였다. 2. 분화표지자인 ALP, OPN, OCN, DMP1, DSPP는 시간 의존적으로 발현되었으나 초기, 중기, 말기에 대한 차별화는 이루어지지 않았다. 3. 광물화는 3일부터 관찰되기 시작하여 14일까지 증가되었다. 4. LOX와 LOXL은 mRNA 발현이 7일까지 증가되었고, 14일에서 큰 감소를 보였다. 하지만, LOXL3와 LOXL4 mRNA는 낮은 발현을 보였고, LOXL2 mRNA는 발현되지않았다. 5. Collagen type I mRNA는 7일까지 증가하다가 14일에서 의미있게 감소를 보였고, collagen type IV는 낮은 mRNA 발현을 보였다. 6. LOX 효소 활성은 분화유도기간 중 7일에서 가장 많은 발현을 보였고, 교원질 1형 기질이 교원질 IV형 기질보다 두드러진 발현을 보였다. 이상의 결과를 종합해 볼 때, LOX isoform의 발현과 LOX 효소 활성은 인간치수세포에서 상아모세포 분화과정 동안 중요한 조절자로 사료된다.
Background : Mesenchymal stem cells (MSCs) are present in most of the tissue matrix, taking part in their regeneration when injury or damage occurs. The aim of this study was to investigate the presence of cells with pluripotential characteristics in human subchondral bone and the capacity of these cells to differentiate to osteoblast. Methods : Human subchondral bone were digested with collagenase. Isolated cells were cultured with a-MEM, 15% FBS, 10-8M dexamethasone and 50 ng/mL ascoric acid. Cells from 0 day(isolated cells), 7 day (first subculture) and 14 days (third subculture) were used to carry out phenotypic characterization experiments flowcytometry analysis with 11 monoclonal antibodies) and osteogenic differentiation experiments. Osteogenic differentiation of cells was assessment by quantification of bone extracellular matrix components by following analysis: alkaline phosphatase(ALP) stains to detect ALP activity, RT-PCR and western blot to detect osteocalcin (OCN), osteopontin (OPN) and type I collagen(Col I), and Alizarin red stains to detect calcium deposition. Results : Flowcytometry analyses showed that in our population more than 98% of cells were positive for MSC markers: SH-2(CD105, 99%), CD29 (95%), CD73 (95%). Cells were negative for hematopoietic markers (CD11b, CD34, and CD45). Furthermore, cells showed positive stain to multipotent markers such as CDl17 (c-kit) (15.1%), and CD166 (74.9%), and cell adhesion molecules such as CD54 (78.1%) and CD106 (63.5%). The osteogenic specific marker analyses showed that the culture of these cells for 7 and 14 days stimulates ALP, OCN, OPN and Col I synthesis by RT-PCR and Western blot analysis. Also, after 14 days in the culture of MSCs induces mineralization by Arizarin red stain. Conclusion : In this work, we demonstrated a new and efficient method for osteoblastic differentiation of human subchondral bone stem cells. As MSCs takes part in reparative processes of adult tissues, these cells could play an important role in osteogenesis.

Regenerative orthopedics needs significant devices to transplant human stem cells into damaged tissue and encourage automatic growth into replacements suitable for the human skeleton. Soft biomaterials have similarities in mechanical, structural and architectural properties to natural extracellular matrix (ECM), but often lack essential ECM molecules and signals. Here we engineer mineralized polysaccharide beads to transform MSCs into osteogenic cells and osteoid tissue for transplantation. Bone morphogenic proteins (BMP-2) and indispensable ECM proteins both directed differentiation inside alginate beads. Laminin and collagen IV basement membrane matrix proteins fixed and organized MSCs onto the alginate matrix, and BMP-2 drove differentiation, osteoid tissue self-assembly, and small-scale mineralization. Augmentation of alginate is necessary, and we showed that a few rationally selected small proteins from the basement membrane (BM) compartment of the ECM were sufficient to up-regulate cell expression of Runx-2 and osteocalcin for osteoid formation, resulting in Alizarin red-positive mineral nodules. More significantly, nested BMP-2 and BM beads added to a non-union skull defect, self-generated osteoid expressing osteopontin (OPN) and osteocalcin (OCN) in a chain along the defect, at only four weeks, establishing a framework for complete regeneration expected in 6 and 12 weeks. Alginate beads are beneficial surgical devices for transplanting therapeutic cells in programmed (by the ECM components and alginate-chitosan properties) reaction environments ideal for promoting bone tissue.
목적: 다양한 크기의 마이크로그루브가 형성된 티타늄 표면에 열산화 처리를 한 복합 표면의 표면특성을 규명하고, 인간치주인대세포 배양 시 표면에 따른 다양한 세포행동들간 차이와 상관관계를 분석하고자 하였다. 재료 및 방법: Grade II 티타늄 디스크를 시편으로 제작하였다. 포토리소그라피를 이용하여 티타늄 시편의 마이크로그루브 크기를 폭/깊이 $0/0{{\mu}m}$, $15/3.5{{\mu}m}$, $30/10{{\mu}m}$, $60/10{{\mu}m}$로 각각 형성하였다. 평활한 티타늄 표면인 대조군(ST)을 제외한 모든 실험군(ST/TO, Gr15-TO, Gr30-TO, Gr60-TO)에 $700^{\circ}C$에서 3시간동안 열산화 처리하고, 주사현미경 사진을 사용하여 표면특성을 평가하였다. 인간치주인대세포를 배양한 후 BrdU (Bromdeoxyuridine) 실험, 알칼리성 인산가수분해효소 활성 실험, 세포외 칼슘 침착 실험을 통해 세포접착, 세포분화 및 골광화를 평가하였다. 통계분석으로는 일요인분산분석과 피어슨상관관계분석(SPSS version 17.0)을 사용하였다. 결과: 열산화를 동반한 마이크로그루브가 형성된 실험군(Gr15-TO, Gr30-TO, Gr60-TO)들은 평활한 대조군(ST)과 단순 열산화 처리 실험군(ST-TO)에 비하여 BrdU 실험, 알칼리성 인산가수분해효소 활성 실험, 세포 외 칼슘 침착 실험 모두에서 유의하게 증가된 활성도를 나타내었다. 특히, Gr60-TO군은 대조군 및 Gr15-TO, Gr30-TO, Gr60-TO 군 등에 비해 가장 증진된 세포접착 및 골아세포분화/골광화를 나타냈다. 결론: 본 연구의 한계 내에서, 열산화 처리 및 마이크로그루브 복합 티타늄 표면은 골아세포분화에 효과적 방법임이 확인되었다. 본 연구에서 규명 된 적정한 마이크로그루브 크기와 열산화 처리 조건은 마이크로그루브-열산화 복합 표면 티타늄 임플란트 개발의 기초 확립에 기여할 수 있을 것이다.
임플란트와 골 사이의 결합력을 증가시키기 위하여 타이타늄 표면에 변화를 주기위한 많은 연구들이 진행되고 있다. 타이타늄의 표면 구조나 미세 지지도의 변화가 임플란트에 대한 세포의 반응에 영향을 미치며, 골아 유사세포는 표면 조도가 높은 타이타늄 표면에 더 잘 부착하며, 세포외 기질의 합성과 광물화 결정이 더 잘 일어난다고 알려져 있다. 그러나 대부분의 연구들은 마이크로 단위의 미세 지지도에 대한 연구들이고 나노 단위의 미세 지지도에 대한 연구들은 미미하다. 이에 본 연구에서는 ROS 17/2.8 cell line을 이용하여 기계적 처리만한 군을 대조군으로 하여 blasting 처리한 마이크로 단위의 미세 지지도 표면과 알칼리 처리된 나노 단위의 미세 지지도 표면에 대한 골아 유사세포의 세포 부착양상, 증식 그리고 골아 유사세포의 표식인자 발현양상 등을 상호 비교하여 골아 유사세포에 미치는 영향을 관찰하고자 하였다. SEM을 이용한 미세 지지도 관찰에서 알칼리 처리군에서는 약 200mm의 초미세 다공성의 양상을 나타내었고, blasting 처리한 군에서는 $10\;{\mu}m$ 이하의 움푹 파인 양상을 보였다. 표면조도 측정에 있어서는 blasting 처리한 군에서 기계적 처리와 알칼리 처리된 군보다 더 높은 표면 조도를 보였으며 이는 통계학적으로 유의한 차이를 나타내었다 (p<0.01). 표면결정성 분석에서는 알칼리처리 군에서 anatase와 rutile결정형이 보였으나, blasting 처리한 군과 기계적 처리 군에서는 관찰되지 않았다. 골아 유사세포 1시간 배양 후의 전자현미경 관찰에서 모든 군의 세포는 부착 및 전개 과정을 보였고, 3시간 배양에서는 모든 군의 세포가 더 많이 전개되었으나, blasting 처리한 군과 알칼리처리 군에서 세포가 다소 더 불규칙한 형태를 나타내었다. 24시간 배양에서는 모든 군의 세포에서 완전히 전개가 일어난 양상을 보였다. 1, 4, 7일간 세포배양 후 세포활성을 평가하기 위한 MTT assay에서는 모든 군에서 시간이 증가함에 따라 세포수가 증가하였으며, 1일째에 blasting 처리한 군과 알칼리처리 군에서 기계적 처리 군에 비해 세포활성도가 통계학적으로 유의한 증가를 보였다(p<0.01). 골아 유사세포 표식인자인 osteopontin, alkaline phosphatase, ${\alpha}\;1(1)$ collagen의 유전자 발현양상을 관찰해 본 결과, osteopontin, alkaline phosphatase, ${\alpha}\;1(1)$ collagen의 유전자 발현양상이 세 군 모두에서 유의한 차이는 관찰할 수 없었으나, blasting 처리한 군과 알칼리처리 군에서 기계적 처리 군에 비해 유전자 발현양상이 다소 증가하는 경향을 보였다. 결론적으로 blasting 처리한 마이크로 단위의 미세 지지도 표면과 알칼리 처리된 나노 단위의 미세 지지도 표면이 기계적 처리 군에 비해 골아 유사세포의 기능을 촉진시키나, 알칼리 처리된 나노 단위의 미세 지지도 표면은 blasting 처리한 마이크로 단위의 미세 지지도 표면이 골아 유사세포의 기능에 미치는 영향을 압도하지는 않는 것으로 사료된다.
