Identification of Total Extracellular Fibrinase from Bacillus sp. DJ Using One-or Two-Dimensional Fibrin Zymography for Proteomic Approach

  • An extracellular fibrinolytic-enzyme-producing bacterium was isolated from Doen-Jang, a Korean traditional fermented flood, and identified as Bacillus sp. DJ based on its morphology and cellular fatty acid composition. The total extracellular fibrinase (EF) from Bacillus sp. DJ was analyzed using three fibrin zymographic techniques, SDS-fibrin zymography (SDS-FZ), isoelectrofocucing-fibrin zymographs(IEF-FZ), and a two-dimensional SDS-fibrin zymographic analysis (2D SDS-FZ). As a result, the EP map of Bacillus sp. DJ was established. The results suggest that the 2D SDS-FZ method will be a useful tool for the proteomic approach for many other bacterial pretenses.

  • Ethyl caproate, one of the major flavor compounds deciding the quality of sake (Japanese wine), is produced during the brewing by the action of alcohol acyltransferase and esterases of sake yeast and koji mold. Extracellular esterases of Aspergillus oryzae required for ethyl caproate synthesis were purified partially. The enzymes had different optimum pH and affinity toward substrates. Substrate preferences and inhibition features showed the three enzymes to be B-type esterases or carboxylesterases (EC 3.1.1.1).

Accumulation of Selenium and Changes in the Activity of Inulinase and Catalase in the Cells of Kluyveromyces marxianus on Pulsed Electric Field Treatment

  • Pulsed electric field (PEF) of 1Hz, 1.5 kV, and 1ms increased the activities of catalase and inulinase over the whole range of applied Se concentrations compared with the non-treated cultures. A significant effect of selenium concentration (in the range of 5-14 ${\mu}g/ml$) on both intra- and extracellular enzyme activities was noted. At a Se concentration of 10 ${\mu}g/ml$, the activities of intra- and extracellular inulinases and extracellular catalase in the PEF-treated cultures reached the maximum of 71 U/g d.m., 46 U/g d.m., and approx. 8 U/ml, respectively. The maximum activity of intracellular catalase of approx. 6 U/ml (with and without PEF) was recorded at 5 ${\mu}g$ Se/ml. Further increasing of selenium concentration caused a decrease in the activity of the enzymes.

  • Toll-like receptor (TLR) family proteins, type I transmembrane proteins, play a central role in human innate immune response by recognizing common structural patterns in diverse molecules from bacteria, viruses and fungi. Recently four structures of the TLR and ligand complexes have been determined by high resolution x-ray crystallographic technique. In this review we summarize reported structures of TLRs and their proposed activation mechanisms. The structures demonstrate that binding of agonistic ligands to the extracellular domains of TLRs induces homo- or heterodimerization of the receptors. Dimerization of the TLR extracellular domains brings their two C-termini into close proximity. This suggests a plausible mechanism of TLR activation: ligand induces dimerization of the extracellular domains, which enforces juxtaposition of intracellular signaling domains for recruitment of intracellular adaptor proteins for signal initiation.

  • The production of an extracellular polysaccharide, EPS-R, from the marine bacterium Hahella chejuensis was investigated at various aeration rates in a batch culture. Higher aeration rate resulted in enhanced EPS production and increased the viscosity of the culture broth. At an aeration rate of 1.5 vvm, EPS-R (12.2 g/L) was obtained with a yield (Y$\_$p/s) of 0.6 from the STN medium after 72 h of cultivation. The H. chejuensis cells changed rod morphology to a short-rod form in the stationary growth phase.

Extracellular Prostaglandin $E_2$ Upregulation Effect of the Methanol Extract of Artemisia argyi

  • Since 15-hydroxyprostaglandin dehydrogenase (15-PGDH) is the key metabolic enzyme of prostaglandin $E_2$ ($PGE_2$), inhibition of 15-PGDH is supposed to facilitate various physiological functions by increasing $PGE_2$. Methanol extract of Artemisia argyi (AAME) inhibited 15-PGDH ($IC_{50}$: $13.13{\mu}g/mL$) with relatively low cytotoxicity ($IC_{50}$: $415.00{\mu}g/mL$) and elevated extracellular $PGE_2$ levels in HaCaT cells. Real-time PCR analysis showed that AAME decreased significantly mRNA expression of PG transporter (PGT) in HaCaT cells. These results indicate that AAME could be applicable to functional materials as a 15-PGDH inhibitor and PGT expression inhibitor for the upregulation of extracellular $PGE_2$ level.

  • In the detection of pathogenic microorganisms ATP-bioluminescence reaction is a fascinating method. ATP(adenosine triphosphate) is an energy source of all kinds of living organism and ATP-bioluminescence reaction uses this ATP. However, ATP exists not only in the cells but also outside the cells. Therefore ATP-bioluminescence reaction only with intracellular ATP is very important in pathogenic microorganism detection. Because of that reason we developed a microfluidic channel containing Dielectrophoretic zone which capture microorganisms and eliminating and washing extracellular ATP with ATP-degarading enzymes, adenosine phosphate deaminase and apyrase. Microorganisms are captured by pDEP force at the DEP electrode zone and only extracellular ATPs are washed and eliminated outside the zone.

  • 당근세포(Daucus carota)로 부터 체세포배 생산성 증대를 위하여 체세포 배 현탁 배양액으로부터 추출한 세포외 분비물질을 이용하여 다음과 같은 효과를 측정하였다.: 분자량의 차이, 세포외 분비 물질 추출 시기, 주입시기, 주입농도에 따른 각종 체세포 배 생산성 증감 효과. 체세포배 생산량은 최대 1,500개/ml까지 얻었으며, 10kDa 이상 분자량이 포함된 세포외 분비물질을 초기에 주입시켰을 때 생산성이 특히 증가함을 보여 주었다.

  • The Burkholderia genus consists of over 40 Gram-negative, ${\beta}$-proteobacteria species that occupy remarkably diverse ecological niches. This genus contains species pathogenic to human, animals, and plants, as well as species involved in promoting plant growth and biodegradation of pollutants. This is largely explained by the extraordinary versatility of Burkholderia, as reflected by the remarkable diversity of extracellular products released by these bacteria. We exhaustively surveyed the extracellular enzymes, siderophores, toxins, antimicrobials, and other secondary metabolites produced by the members of this very diverse genus. Available information on regulation, especially quorum sensing mechanisms, and secretion is highlighted.

  • Producing an extracellular alkaline lipase, this isolate JM123 was identified as a Pseudomonas aeruginosa strain from the results of the analyses of its morphological, biochemical and physiological properties. This strain showed the highest productivity of alkaline lipase when grown at pH 9.0 and 30C for 13-20 hours in the medium of 2% starch, 1% soytone, 0.5% peptone and 1% MgSO4.7H2O. However, this enzyme was greatly repressed when grown in the glucose containing medium. The culture broth was fractionated by the order of the ammonium sulfate precipitation, Sephadex G-200 gel filtration, DEAE-cellulose column chromatography, and Sephadex G-150 gel filtration.
