Up-regulation of Thy-1 Promotes Invasion and Metastasis of Hepatocarcinomas

  • Increasing evidence has revealed that thy-1 was a potential stem cell marker of liver cancer, but no data have been shown on how thy-1 regulates the pathophysiology of liver cancer, such as proliferation, apoptosis, invasion and migration. We previously demonstrated that thy-1 was expressed in about 1% of hepg2 cells, thy-1+hepg2 cells, but not thy-1-, demonstrating high tumorigenesis on inoculation $0.5{\times}10^5$ cells per BACA/LA mouse after 2 months. In the present study, our results showed that higher expression of thy-1 occurs in 72% (36/50 cases) of neoplastic hepatic tissues as compared to 40% (20/50 cases) of control tissues, and the expression of thy-1 is higher in poorly differentiated liver tumors than in the well-differentiated ones. In addition, thy-1 expression was detected in 85% of blood samples from liver cancer patients, but none in normal subjects or patients with cirrhosis or hepatitis. There was a significant negative correlation between thy-1expression and E-cadherin expression (a marker of invasion and migraton), but not between thy-1 expression and AFP expression in all the liver cancer and blood samples. We further investigated the relationship between thy-1 and E-cadherin in liver cancer hepg2 cell line which was transfected with pReceiver-M29/thy-1 eukaryotic expression vector followed by aspirin treatment. Lower expression of E-cadherin but higher expressions of thy-1 were detected in hepg2 cells transfected with pReceiver-M29/thy-1. Taken together, our study suggested that thy-1 probably regulates liver cancer invasion and migration.

  • Objective: Parathyroid hormone like hormone (PTHLH), as an essential factor for bone growth, is involved in a variety of physiological processes. The aim of this study was to explore the role of PTHLH gene in the growth of antlers. Methods: The coding sequence (CDS) of PTHLH gene cDNA was obtained by cloning in sika deer (Cervus nippon), and the bioinformatics was analyzed. The quantitative real-time polymerase chain reaction (qRT-PCR) was used to analyze the differences expression of PTHLH mRNA in different tissues of the antler tip at different growth periods (early period, EP; middle period, MP; late period, LP). Results: The CDS of PTHLH gene was 534 bp in length and encoded 177 amino acids. Predictive analysis results revealed that the PTHLH protein was a hydrophilic protein without transmembrane structure, with its secondary structure consisting mainly of random coil. The PTHLH protein of sika deer had the identity of 98.31%, 96.82%, 96.05%, and 94.92% with Cervus canadensis, Bos mutus, Oryx dammah and Budorcas taxicolor, which were highly conserved among the artiodactyls. The qRT-PCR results showed that PTHLH mRNA had a unique spatio-temporal expression pattern in antlers. In the dermis, precartilage, and cartilage tissues, the expression of PTHLH mRNA was extremely significantly higher in MP than in EP, LP (p<0.01). In the mesenchyme tissue, the expression of PTHLH mRNA in MP was significantly higher than that of EP (p<0.05), but extremely significantly lower than that of LP (p<0.01). The expression of PTHLH mRNA in antler tip tissues at all growth periods had approximately the same trend, that is, from distal to basal, it was first downregulated from the dermis to the mesenchyme and then continuously up-regulated to the cartilage tissue. Conclusion: PTHLH gene may promote the rapid growth of antler mainly through its extensive regulatory effect on the antler tip tissue.

Low-area FFT Processor Structure using Common Sub-expression Sharing (Common Sub-expression Sharing을 사용한 저면적 FFT 프로세서 구조)

  • In this paper, a low-area 256-point FFT structure is proposed. For low-area implementation CSD(Canonic Signed Digit) multiplier method is chosen. Because multiplication type should be less for efficient CSD multiplier application to the FFT structure, the Radix-$4^2$ algorithm is chosen for those purposes. After, in the proposed structure, the number of multiplication type is minimized in each multiplication block, the CSD multipliers are applied for implementation of multiplication. Furthermore, in CSD multiplier implementation, cell-area is more reduced through common sub-expression sharing(CSS). The Verilog-HDL coding result shows 29.9% cell area reduction in the complex multiplication part and 12.54% cell area reduction in overall 256-point FFT structure comparison with those of the conventional structure.

Implication of Angiogenesis and Expression of VEGF in Follicular Thyroid Tumor (여포상 갑상선종양에서 신생혈관형성 및 혈관내피성장인자(VEGF)의 발현의 의의)

  • Tumor growth and metastasis depends on angiogenesis. Vascular endothelial growth factor (VEGF) is a potent mitogen for vascular endothelial cells in vitro and promotes neoangiogenesis in vivo. Objective: Follicular thyroid cancers(FTC) are a vascular tumor and traditionally metastasize via blood vessels. Likely other cancers, angiogenesis may playa important role in FTC. We, therefore, investigated the expression of VEGF and microvascular density by immunohistochemistry in FTC and follicular adenoma(FA). Materials and Methods: Findings of immunohistochemical stainings for VEGF and CD31 were measured by grading scale from +1 to 4+(strongest) and by counting the stained microvessels in 14 FTCs and 14 FAs. Results: 1) Expression of VEGF. a) FTCs have stronger expression than FAs in areas of tumor adjacent to capsule($mean{\pm}SD\:\;3.2{\pm}0.9\;vs\;2.0{\pm}0.9$, p<0.01) and in central area($2.3{\pm}0.7\;vs\;1.3{\pm}0.6$, p<0.01). b) The VEGF expression of capsular area in FTCs are higher than that of central area(p<0.05). 2) Microvascular density by CD31. a) FTCs have more microvessels than FAs in areas of adjacent to capsule($78.9{\pm}27.3\;vs\;38.7{\pm}15.6$, p<0.01) and in central area($75.5{\pm}23.3\;vs\;27.8{\pm}10.7$, p<0.01). b) In FTCs, the number of microvessels of capsular area are more than that of central tumor area, but not significant statistically(p>0.05). Conclusion: The higher expression of VEGF and microvascular density in FTC suggests angiogenesis plays an important role in progression of FTC.

  • The heterotrimeric G protein subunits (G ) are region-specifically expressed in brain such as hypothalamus and pituitary gland in abundant, suggesting that is may be associated with “stress-axis”. This study was designed to examine the effect of stress on the region-specific expression of various G subunits in rat brain. The localization of mRNAs encoding seven of G and striking region-specific patterns of expression were observed in 12 different regions of both non-stressed and stressed rat brain; (1) frontal cortex area, (2) cerebral cortex area, (3) striatum, (4) hippocampus area, (5) thalamus, (6) brain stem, (7) cerebellum area, (8) hypothalamus, (9) septum, (10) amygdala, (11) preoptic area, and (12) pituitary gland.   • Acupuncture has been used as a clinical treatment in Oriental medicine for various diseases. In the present study was carried out to investigate the effects of acupuncture and electrical stimulation on the change neuronal nitric oxide synthase(nNOS) immunoreactive cells in the periaqueductal gray(PAG) area of the male SD rats. Enhanced expression of nNOS was detected in the dorsolateral-PAG(DL-PAG) area of rat with stress by fixed body, and acupuncture and needle electrode electrical stimulation groups at Hapgok like acupoint decreased the stress-induced enhancement in the expression of nNOS. The present results demonstrate that acupuncture and needle electrode electrical stimulation is effective in the modulation of expression of nNOS in the DL-PAG area under stress conditions.

  • Formula for the area of a trapezoid is an educational material that can handle algebraic and geometric perspectives simultaneously. In this note, we will make up the expression equivalent algebraically to the formula for the area of a trapezoid, and deal with the conversion of a geometric point of view, in algebraic terms of translating and interpreting the expression geometrically. As a result, the geometric conversion model, the first algebraic model, the second algebraic model are obtained. Therefore, this problem is a good material to understand the advantages and disadvantages of the algebraic and geometric perspectives and to improve the mathematical insight through complementary activity. In addition, these activities can be used as material for enrichment and gifted education, because it helps cultivate a rich perspective on diverse and creative thinking and mathematical concepts.

Low-area Pipeline FFT Structure in OFDM System Using Common Sub-expression Sharing and CORDIC (Common sub-expression sharing과 CORDIC을 이용한 OFDM 시스템의 저면적 파이프라인 FFT 구조)

  • An efficient pipeline MDC Radix-4 FFT structure is proposed in this paper. Every stages in pipeline FFT structure consists of delay' commutator and butterfly. Proposed butterflies in front and rear stages utilize CORDIC and Common Sub-expression Sharing(CSS) techniques, respectively. It is shown that proposed butterfly structure can reduce the number of adders through sharing common patterns of CSD type coefficients. The Verilog-HDL modeling and Synopsys logic synthesis results that the proposed structure show 48.2% cell area reduction in the complex multiplication part and 22.1% cell area reduction in overall 256-point FFT structure comparison with those of the conventional structures. Consequently, the proposed FFT structure can be efficiently used in various OFDM systems.

Recognizing Human Facial Expressions and Gesture from Image Sequence (연속 영상에서의 얼굴표정 및 제스처 인식)

  • In this paper, we present an algorithm of real time facial expression and gesture recognition for image sequence on the gray level. A mixture algorithm of a template matching and knowledge based geometrical consideration of a face were adapted to locate the face area in input image. And optical flow method applied on the area to recognize facial expressions. Also, we suggest hand area detection algorithm form a background image by analyzing entropy in an image. With modified hand area detection algorithm, it was possible to recognize hand gestures from it. As a results, the experiments showed that the suggested algorithm was good at recognizing one's facial expression and hand gesture by detecting a dominant motion area on images without getting any limits from the background image.

  • Generally, feature area detection methods are widely used for face expression recognition by detecting the feature areas of human eyes, eyebrows and mouth. In this paper, we proposed a face expression recognition method using the histograms of the face, eyes and mouth for many applications including robot technology. The experimental results show that the proposed method has a new type of face expression recognition capability compared to conventional methods.
