Mancozeb is a manganese and zinc-containing fungicide that belongs to the ethylene bisdithiocarbamate group and produces ethylene thiourea (ETU) after biotransformation or environmental degradation, which has toxicological hazard owing to its known antithyroid properties. Although mancozeb leads to negative changes in fertility capacity, the effects of ETU are less known. Therefore, this study examined the alteration of fertilization competence in boar spermatozoa exposed to ETU. The sperm motility, motion kinematics, viability, acrosome integrity, chromatin stability, and intracellular reactive oxygen species (ROS) production of sperm subjected to various ETU concentrations (10, 50, 100, and 200 µM) were evaluated after two different incubation times (30 min and 2 hrs). In addition, the relative mRNA expression of the sperm functional proteins was analyzed after exposure to ETU. A dose-dependent motility reduction was observed in sperm exposed to ETU during both incubation periods compared to the controls. The motion kinematics were reduced significantly in sperm incubated with ETU. Higher percentages of viable sperm were observed in the controls, while such viability was decreased significantly in sperm with 10 - 200 µM ETU. The acrosome integrity was particularly damaged on sperm incubated with 10 - 200 µM ETU for 30 min. Higher intracellular ROS levels were produced in sperm exposed to 200 µM ETU. In addition, lower relative levels of AKAP3, AKAP4, ODF2, and ZPBP2 expression were observed in sperm exposed to ETU compared to the controls. Mancozeb and ETU could adversely affect the reproductive functions of mammals. Hence, the effects of ETU on the reproductive system should be examined further.
The physiological characteristics of cultures of very high cell mass (e.g. 10g cell mass/L), termed“ultrahigh cell density cultures”is reviewed. A close relationship was found between the length of the optical path (OP) in flat-plate reactors and the optimal cell density of the culture as well as its areal (g m$\^$-2/ day$\^$-1/) productivity. Cell-growth inhibition (GI) unfolds as culture density surpasses a certain threshold. If it is constantly relieved, a 1.0cm OP reactor could produce ca. 50% more than reactors with longer OP, e.g. 5 or 10cm. This unique effect, discovered by Hu et al. [3], is explained in terms of the relationships between the frequency of the light-dark cycle (L-D cycle), cells undergo in their travel between the light and dark volumes in the reactor, and the turnover time of the photosynthetic center (PC). In long OP reactors (5cm and above) the L-D cycle time may be orders of magnitude longer than the PC turnover time, resulting in a light regime in which the cells are exposed along the L-D cycle, to long, wasteful dark periods. In contrast, in reactors with an OP of ca. 1.0 cm, the L-D cycle frequency approaches the PC turnover time resulting in a significant reduction of the wasteful dark exposure time, thereby inducing a surge in photosynthetic efficiency. Presently, the major difficulty in mass cultivation of ultrahigh-density culture (UHDC) concerns cell growth inhibition in the culture, the exact nature of which is awaiting detailed investigation.
Postantibiotic effects (PAE) refer to suppression of the bacterial growth following limited periods of exposure to an antibiotic and subsequent to the removal of the antibiotic agent. Fusobacterium nucleatum and Porphyromonas gingivalis are Gram-negative anaerobic bacteria associated with several periodontal diseases. In this study, postantibiotic effects (PAE), postantibiotic sub-MIC effect (PA SME) and sub-MIC effect (SME) of antibiotics on F. nucleatum ATCC 25586 and P. gingivalis W50 were investigated. The PAE was induced by 10X the MIC of antibiotic and antibiotic was eliminated by washing. The PA SMEs were studied by addition of 0.1, 0.2 and 0.3X MICs during the postantibiotic phase of the bacteria, and the SMEs were studied by exposition of the bacteria to antibiotic at the sub-MICs only. Amoxicillin, doxycycline and tetracycline induced PAE for F. nucleatum ATCC 25586 and P. gingivalis W50. But metronidazole and penicillin induced PAE for only F. nucleatum ATCC 25586. Metronidazole and doxycycline induced PA SME and SME for both species of anaerobic bacteria used in this study. The PA SME values for both strains were substantially longer than the SME values. The present study showed the existence of PAE, PA SME and SME for various antibiotics against F. nucleatum ATCC 25586 and P. gingivalis W50.
Korean ginseng products have been fumigated with ethylene oxide (EO) for sterilization and prolongation of storage periods. However, there had been controversies indicating that the consumption of food treated with EO might cause harmful effects in human. Since, in Korea the use of EO gas for food treatment was banned in 1991. Since then, irradiation technique has been developed as an alternative. This study was carried out to investigate the effects of irradiated ginseng on fertility, and reproductive and developmental toxicity. Either EO gas fumigated or gamma-irradiated ginseng was administered to male rats by oral gavage for 63 days during the premating period. Female rats were administered from 14 days before mating to day 20 of gestation or to day 21 of lactation. The exposure amount of irradiation used was 5, 10 and 30 kGy, respectively. There were no treatment related changes of darns in clinical signs, and parturition. No treatment related changes in food consumption, body/organ weights, male/female reproductive and fertility performances were observed. F1 fetuses showed no external abnormality. Reflex/sensory junctions, physical/behavioral development, and reproductive performance of F1 rats were not adversary affected. The results of this study show that gamma-irradiated ginseng, up to 30 kGy, has no adverse effects on the fertility, reproduction and development in Wistar rats.
This review summarizes the cause of allergy and control by lactic acid bacteria. Atopic diseases such as asthma, rhinitis, eczema and food allergy have increased in most industrialized countries of the world during the last 20 years. The reasons for this increase are not clear and different hypotheses have been assessed including increased exposure to sensitizing allergens or decreased stimulation of the immune system during critical periods of development. Probiotic bacteria, which beneficially affect the host by improving its microbial balance, may mediate anti-allergenic effects by immune stimulation. Although more clinical evidences are required, the possible role of specific LAB strains in the prevention of allergic diseases has become more evident. since the role of functional food is important for prevention, it is expected for the more anti-allergy fermented milk products to be on the market.
In this paper, the potential ground motion in terms of the peak ground accelerations(PGAs) due to long-distance Sumatra earthquakes is investigated for Singapore, following the probabilistic seismic hazard assessment a, pp.oach. The case investigated differs from a conventional one, in that few attenuation equations for long-distance major earthquakes are readily available. The attenuation relationships developed for other regions of the world are thus reviewed. It is found that the existing attenuation equations, when extrapolated to distant major earthquakes, tend to underestimate the PGAs. By comparing with the PGAs recorded over long distances at stations of the Japanese Meteorological Agency for major earthquakes in Japan, an attenuation equation is chosen for this study. With the chosen attenuation equation, the probability of PGAs exceeding selected levels for various exposure periods of time is then computed. The results show that at Singapore there is a 10% probability in 50 years for the PGA at rock sites to exceed 1.1% g. In view of the results and the associated uncertainties, a base shear coefficient of 1.5% is being recommended as the tentative seismic loading in Singapore. The tentative seismic loading reflects the design value of the notional horizontal load, equal to 1.5% of the characteristic building weigh as specified in the BS code, which usualy governs the design of most buildings in Singapors.
Regular radiographic examination has been considered as an essential diagnostic method for the osseointegrated dental implantation. This study investigated the marginal bone loss through the measurement on the periapical radiograph around 46 endosseous dental implants. Marginal bone loss have been observed for 5 years after delivery of prostheses. The results were as follows : 1. According to the experimental periods, average marginal bone loss in total implant was 1.821mm in the first year, 1.833mm in the second, 1.892mm at third, 1.910mm at forth and 1.957mm at fifth after delivery of prostheses(P<0.05). 2. The average bone loss was 1.832mm in the maxillae and 1.819mm in the mandible in three years but there was no significant difference between the upper and lower jaw(P>0.05). 3. The average bone loss was 1.824mm in males and 1.818mm in females in five years but there was no significant difference according to the sex(P>0.05). These results indicate that the most of the alveolar bone loss occur within the first year after delivery of dental prostheses resulting in the exposure of polished neck positions, and the bone level stabilizes at first thread portion of the implant fixtures.
Phthalates and di-2-ethylhexyladipate are both widely used as industrial chemicals and exposure high levels over long periods of time can cause toxicity, estrogenic responses and endocrine disruption properties in both humans and animals. So far, their global monitoring in environmental matrices has been performed continuously. A developed method, including modified SPEED'98 (Japan Environment Agency) and USEPA was established for simple and rapid determination of phthalates and di-2-ethylhexyladipate in various matrices. This method was applied to explore the distribution levels in domestic environmental media such as water, soil and sediment. Eight phthalates (DEP(di-ethyl), DEHP(di-[2-ethyl-hexyl]), DprP (di-propyl), DBP(di-n-butyl), DPP(di-n-pentyl), DHP(di-n-hexyl), DCHP(di-cyclohexyl), BBP(butyl benzyl) and Adipate (di-2-ethylhexyl adipate) were investigated by seasonal sampling(spring, autumn) at 24 domestic sites. Phthalates and adipate were not detected in water samples and DEP, DBP, and DEHP were mainly detected in soil and sediment samples. The concentrations of DEP and DBP excluding DEHP in spring were higher in soil than those of sediment. Total concentrations of phthalates were significantly decreased in autumn for both soil and sediment.
Laying hens kept in different light and dark periods of the day at high ambient temperature of maximum $35^{\circ}C$ were challenged to $38.5{\pm}0.5^{\circ}C$ acute heat 3 hours daily for 7 consecutive days. They were found to have a significant (p<0.01) acclimatization response (rectal temperature) to heat stress during the dark period compared to those exposed to the same temperature during the light period. The blood pH was not significantly different. The partial pressure of carbon dioxide ($PCO_2$) was significantly high (p<0.01) except in day 4. Similarly the blood bicarbonate ($HCO_3$) concentration was significantly high (p<0.05) except day three and day four. Acute heat exposure in the first day increased the body temperature in both groups (Light and Dark) reaching $44^{\circ}C$, followed by gradual reduction in body temperature. The dark treated birds showed rapid reduction in body temperature ($42.88^{\circ}C$) and adaptation to high temperature during days 2-4 but that this was lost to some extent in days 6-8. However this was not obvious in the light treated birds. It is concluded that darkness reduce hyperthermia and enhance acclimatization responses during acute heat stress.
Background: This study was performed to provide a long-term bacterial seal through the formation of reparative dentin bridge, calcium silicate-based pulp capping materials have been used at sites of pulpal exposure. The aim of this study was to evaluate the mineralization-inducing potentials of calcium silicate-based pulp capping materials (ProRoot MTA [PR], Biodentine [BD], and TheraCal LC [TC]) in human dental pulp cells (HDPCs). Methods: Specimens of test materials were placed in deionized water for various incubation times to measure the pH variation and the concentration of calcium released. The morphology of HDPCs cultured on the specimens was examined using a confocal laser scanning microscope (CLSM). Alizarin red S staining and alkaline phosphatase assays were used to evaluate mineralization-inducing potentials of the capping materials. Results: BD showed the highest calcium release in all test periods, followed by PR and TC. (p<0.05). All experimental groups showed high alkalinity after 1 day, except at 14 days. BD showed the highest cell viability compared with PR and TC after 1 and 3 days, while TC showed the lowest value (p<0.05). The CLSM analysis showed that cells were well adhered and expressed actin filaments for all pulp capping materials. Mineralization by PR and BD groups was higher than that by TC group based on alizarin red S staining. BD showed significantly higher alkaline phosphatase activity than PR and TC, while TC showed the lowest value (p<0.05). Conclusion: Within the limitations of the in vitro study, BD had higher mineralization-inducing potential than PR and TC.
