Disruption of the metC Gene Affects Methionine Biosynthesis in Pectobacterium carotovorum subsp. carotovorum Pcc21 and Reduces Soft-Rot Disease

  • Plant pathogenic Pectobacterium species cause severe soft rot/blackleg diseases in many economically important crops worldwide. Pectobacterium utilizes plant cell wall degrading enzymes (PCWDEs) as the main virulence determinants for its pathogenicity. In this study, we screened a random mutant, M29 is a transposon insertion mutation in the metC gene encoding cystathionine β-lyase that catalyzes cystathionine to homocysteine at the penultimate step in methionine biosynthesis. M29 became a methionine auxotroph and resulted in growth defects in methionine-limited conditions. Impaired growth was restored with exogenous methionine or homocysteine rather than cystathionine. The mutant exhibited reduced soft rot symptoms in Chinese cabbages and potato tubers, maintaining activities of PCWDEs and swimming motility. The mutant was unable to proliferate in both Chinese cabbages and potato tubers. The reduced virulence was partially restored by a complemented strain or 100 µM of methionine, whereas it was fully restored by the extremely high concentration (1 mM). Our transcriptomic analysis showed that genes involved in methionine biosynthesis or transporter were downregulated in the mutant. Our results demonstrate that MetC is important for methionine biosynthesis and transporter and influences its virulence through Pcc21 multiplication in plant hosts.

  • Greater yang disease(太陽病) is a syndrome induced by peripheral obstruction. One of them is "wind stroke(中風)" with the obstruction in lymphatic system. The other obstruction appearing on circulatory system is called "cold damage(傷寒)." Kyejitang(桂枝湯) is the formula prescribed for greater yang wind stroke pattern(太陽中風證) which is caused by peripheral lymphatic duct obstruction. Ramulus Cinamoni acts as a vasodilator and Radix Paeoniae relieves the abdominal tension. They make blood move to the internal organ and this can remove the retention of peripheral lymphatic system. Covering the patient with a blanket and getting him/her to have hot and thin rice gruel causes slight Diaphoresis, contributing to relieving the retention of lymphatic system. Disharmony between nutrient and defense(營衛不和) means that pressure becomes different between lymphatic system and vascular system. Kyejitang(桂枝湯) is called releasing muscles formula(解肌劑) because it can resolve such pressure difference. Diaphoresis is not a means to eliminate pathogenic qi(邪氣) from the body. That is the syndrome proving that the body fluid has moved around when disordered fluid distribution is corrected. Therefore, diaphoresis should be induced weakly all the time. If diaphoresis is induced excessively, body fluid will move more than desired and then illness cannot be cured. In Sanghanlun(傷寒論), dispersing drugs aim at addressing the retention in the exterior field, but it actually applies to the entire exterior and interior to make body fluid move. Therefore, diaphoresis does not just act on exterior field, and freeing the stool does not only apply to interior field. Distribution of body fluid changed by pathogenic qi(邪氣) influences the whole body because the human body has a closed circulatory system. Sanghanlun(傷寒論) has included treatments for pathogenic disease. However, its value should not be limited to pathogenic disease. It is because controlling blood flow by sending body fluid to the place a doctor wants is certainly worth using for treatment of non-exogenous disease or chronic illnesses.

Month and Season of Birth as a Risk Factor for Alzheimer's Disease: A Nationwide Nested Case-control Study

  • Objectives: Season of birth, an exogenous indicator of early life environment, has been related to higher risk of adverse psychiatric outcomes but the findings for Alzheimer's disease (AD) have been inconsistent. We investigated whether the month or season of birth are associated with AD. Methods: A nationwide nested case-control study including all community-dwellers with clinically verified AD diagnosed in 2005 to 2012 (n=70 719) and up to four age- sex- and region of residence-matched controls (n=282 862) residing in Finland. Associations between month and season of birth and AD were studied with conditional logistic regression. Results: Month of birth was not associated with AD (p=0.09). No strong associations were observed with season (p=0.13), although in comparison to winter births (December-February) summer births (June-August) were associated with higher odds of AD (odds ratio, 1.03; 95% confidence interval, 1.00 to 1.05). However, the absolute difference in prevalence in winter births was only 0.5% (prevalence of those born in winter were 31.7% and 32.2% for cases and controls, respectively). Conclusions: Although our findings do not support the hypothesis that season of birth is related to AD/dementia risk, they do not invalidate the developmental origins of health and disease hypothesis in late-life cognition. It is possible that season does not adequately capture the early life circumstances, or that other (postnatal) risk factors such as lifestyle or socioeconomic factors overrule the impact of prenatal and perinatal factors.

  • This study was investigated to make sure the usage of prescriptions in which Perilla frutescens is used as a main herbal medicine in Donguibogam. Through the investigation on main treatment, pathology, etiology and nature of disease, dosage in the relevant prescriptions, The consequences are as follows; 1. The Perilla frutescens is used in 15 treatment fields which contain the cough, cold chapter ect. 2. The Perilla frutescens is used as a main heral medicine in 30 natures of disease such as cough-relative disease, an asthma, an external wind and cold, etc. 3. The Perilla frutescens is used for various pathology in the exogenous etiological factor, endogenous etiological factor, exo-endogeneous etiological factor and non-exo-endogeneous etiological factor. 4. The Perilla frutescens is used in a range of $1.2g{\sim}10g$ in relevant prescription. The main dosage is 4g. 5. The Perilla frutescens is used with various crude herbs in accordance with the pathogeny. The Perilla frutescens has been used to reduce heat from the lungs, to relieve asthma, to promote qi's movement and to soothe fetal movement etc. According to the results, 1 suggest to use the Perilla frutescens in a various pathogenic fields. The Perilla frutescens is able to remove not only pathogenic heat from the cough-relative disease, but also pathogenic asthma, an external winds and colds, etc.

A PAS-Containing Histidine Kinase is Required for Conidiation, Appressorium Formation, and Disease Development in the Rice Blast Fungus, Magnaporthe oryzae

  • Rice blast disease, caused by the ascomycete fungus Magnaporthe oryzae, is one of the most important diseases in rice production. PAS (period circadian protein, aryl hydrocarbon receptor nuclear translocator protein, single-minded protein) domains are known to be involved in signal transduction pathways, but their functional roles have not been well studied in fungi. In this study, targeted gene deletion was carried out to investigate the functional roles of the PAS-containing gene MoPAS1 (MGG_02665) in M. oryzae. The deletion mutant ΔMopas1 exhibited easily wettable mycelia, reduced conidiation, and defects in appressorium formation and disease development compared to the wild type and complemented transformant. Exogenous cAMP restored appressorium formation in ΔMopas1, but the shape of the restored appressorium was irregular, indicating that MoPAS1 is involved in sensing the hydrophobic surface. To examine the expression and localization of MoPAS1 in M. oryzae during appressorium development and plant infection, we constructed a MoPAS1:GFP fusion construct. MoPAS1:GFP was observed in conidia and germ tubes at 0 and 2 h post-infection (hpi) on hydrophobic cover slips. By 8 hpi, most of the GFP signal was observed in the appressoria. During invasive growth in host cells, MoPAS1:GFP was found to be fully expressed in not only the appressoria but also invasive hyphae, suggesting that MoPAS may contribute to disease development in host cells. These results expand our knowledge of the roles of PAS-containing regulatory genes in the plant-pathogenic fungus M. oryzae.

  • Pediatrics in Geumwon(金元) age had fanned peculiar children's etiology, Byeonggihak(病機學) and Byeonjeungnon(辦證論) curative system grounded on medical theory in Geumwonsadaega(金元四大家). Yuwanso(劉完素), who was front-running man in that age, used so many times 'Yanggyeoksan(凉隔散)' or 'Bangpungtongseongsan(防風通聖散)' which have Hanryang(寒凉) disposition for curing children's disease in view of pathology in Yeoldahanso(熱多寒少), which is his special academic thought, after then Judangye(朱丹溪) succeed Yuwanso(劉完素)'s this view of pathology, Yang-eumjahyeol(養陰滋血) developed the one and made it a standing rule to cure children. Also Idongwon(李東垣) grasped that the main cause of disease is starvation because of war. As a result of it, he insisted on theory, internal cause's outbreak of person's illness, Naesangbiwi(內傷脾胃), Baekbyeong-yusaeng(百病由牛). In this view of this principle, he assorted and used following medicine 'Seungma(升麻), Siho(柴胡), Hwanggi' as 'Seungbalyanggi(升發陽氣)' method to cure children, and concentrated on children's spleen and stomach cure with this way. Jangjongjeong(張從政) applied 'Purgation therapy[攻下法]', Hantoha(汗吐下) centered on the theory of exogenous factors[外因說] , which explains that children's disease is Sagichimseup(邪氣侵襲), too. Like this, in Geumwon(金元) age, they had studied very seriously about main cause of children' s disease in two part, an internal cause and an external cause. Also, in pathological part they had approached academically 'Hwayeolchibyeong(火熱致病)' and 'Gigiseunggangsiljo(氣機乘降失調)' from diverse angles.

Ganglioside GT1b increases hyaluronic acid synthase 2 via PI3K activation with TLR2 dependence in orbital fibroblasts from thyroid eye disease patients

  • Thyroid eye disease (TED) is a complex autoimmune disease with a spectrum of signs. we previously reported that trisialoganglioside (GT)1b is significantly overexpressed in the orbital tissue of TED patients, and that exogenous GT1b strongly induced HA synthesis in orbital fibroblasts. However, the signaling pathway in GT1b-induced hyaluronic acid synthase (HAS) expression in orbital fibroblasts from TED patients have rarely been investigated. Here, we demonstrated that GT1b induced phosphorylation of Akt/mTOR in a dose-dependent manner in orbital fibroblasts from TED patients. Both co-treatment with a specific inhibitor for PI3K and siRNA knockdown of TLR2 attenuated GT1b-induced Akt phosphorylation. GT1b significantly induced HAS2 expression at both the transcriptional and translational level, which was suppressed by specific inhibitors of PI3K or Akt/mTOR, and by siRNA knockdown of TLR2. In conclusion, GT1b induced HAS2 in orbital fibroblasts from TED patients via activation of the PI3K-related signaling pathway, dependent on TLR2.

Resistance Induction by Salicylic Acid Formulation in Cassava Plant against Fusarium solani

  • Fusarium root rot caused by the soil-borne fungus Fusarium solani is one of the most important fungal diseases of cassava in Thailand, resulting in high yield losses of more than 80%. This study aimed to investigate if the exogenous application of salicylic acid formulations (Zacha) can induce resistance in cassava against Fusarium root rot and observe the biochemical changes in induced cassava leaf tissues through synchrotron radiation based on Fourier-transform infrared (SR-FTIR) microspectroscopy. We demonstrated that the application of Zacha11 prototype formulations could induce resistance against Fusarium root rot in cassava. The in vitro experimental results showed that Zacha11 prototype formulations inhibited the growth of F. solani at approximately 34.83%. Furthermore, a significant reduction in the disease severity of Fusarium root rot disease at 60 days after challenge inoculation was observed in cassava plants treated with Zacha11 at a concentration of 500 ppm (9.0%). Population densities of F. solani were determined at 7 days after inoculation. Treatment of the Zacha11 at a concentration of 500 ppm resulted in reduced populations compared with the distilled water control and differences among treatment means at each assay date. Moreover, the SR-FTIR spectral changes of Zacha11-treated epidermal tissues of leaves had higher integral areas of lipids, lignins, and pectins (1,770-1,700/cm), amide I (1,700-1,600/cm), amide II (1,600-1,500/cm), hemicellulose, lignin (1,300-1,200/cm), and cellulose (1,155/cm). Therefore, alteration in defensive carbohydrates, lipids, and proteins contributed to generate barriers against Fusarium invasion in cassava roots, leading to lower the root rot disease severity.

  • Objectives The purpose of this study is to understand the formation of Jogakdaehwang-tang(JD-T)'s patholoy and the compositional change of it in Sasang Constitutional Medicine(SCM). Methods The pathologic condition in which JD-T was used and the change of its composition was studied in SCM Results and Conclusions 1. Jogak and Daehwang in "Dongmuyoogo Haedong" were presented with Ungdam because of the common purpose flowing the energy of lung. Therapeutic method for exogenous Ungibyeong in "Dongyisusebowon sangchobongwon" was subdivided into Hangwul's treated by Ungdam, and the heat pattern featuring contagious cold and constipation, treated by Jogakhwanggeumdaehwang-tang(JHD-T). 2. JD-T in "Dongyisusebowon Gabogubon" was used for Yangdok and Onbyeong which are exogenous disease causing by heat. The composition of JD-T has been completed as it included Mahwang, Gilgyeong, and Seungma derived from prescriptions relevant with Yangdok. 3. JD-T in "Dongyisusebowon Sinchukbon(Sinchukbon)" was included in pathology of Ganyeol, and it substituted Galgeunhaegi-tang(GH-T) for Onbyeong, not for Yangdok, so Galgeun and Seungma became the main drug in JD-T. Galgeunseung-gitang(GS-T) was created for treating heat pattern owing to Daehwang, its main drug, and moreover Jogak's effect is too strong. Therefore, Mahwang, Hwanggeum and Gilgyeong was excluded from the composition of JD-T. 4. The origins of JD-T could be Yiseonggugo-hwan, Daesiho-tang, Jowiseungchung-tang, Heukno-hwan, and GH-T. The change of JD-T went for them throughout JHD-T to JD-T. JD-T in "Sinchukbon" consists of Galgeun and Daehwang which directly gets rid of severe heat from small intestine, and Jogak and Seungma which help to eliminate mild heat existing from small intestine throughout nose to flesh.

A Case of Exogenous Lipoid Pneumonia after Ingestion of Squalene in Patients with Achalasia (식도이완불능증 환자에서 스쿠알렌 복용 후 발생한 지방성 폐렴 1예)

  • Exogenous lipoid pneumonia (ELP) is a chronic inflammatory reaction of the lungs resulting from the aspiration of vegetable, animal or mineral oils. Squalene, is a derivative of shark liver oil that is taken as a traditional remedy in some Asian countries, and is used widely also in cosmetics. Similar to the symptoms in most cases of oil aspiration, the symptoms of squalene-induced lipoid pneumonia are either absent or nonspecific. Hence, the disease is generally detected incidentally. Although many cases with predisposing factors have been reported, ELP with achalasia is quite rare. We report a 47-year old woman with achalasia who developed ELP after ingesting squalene. The patient was treated successfully by supportive care and surgical treatment of the achalasia.
