자율주행자동차의 이슈 및 문제해결에 기반한 전략적 관점에서의 활성화 방안 연구 (A study of the activation from strategic perspectives based on autonomous vehicle issues and problem solving)

  • 자율주행자동차 활성화를 위한 법·제도 연구는 많았지만, 전략적 관점에서의 자율주행자동차의 활성화 방안에 대한 연구는 미흡하였다. 본 연구에서는 자율주행자동차의 이슈 및 해결방안에 대해 살펴보고, 이를 기반으로 전략적 관점에서의 활성화 방안에 대해 제안하고자 한다. 자율주행자동차의 이슈 및 문제 해결을 위해 안전성 강화를 기반으로 법·제도 표준에 대한 명확한 정립이 필요하며, 교통사고시 책임의 소재와 배상책임의 확보가 선행 되어야 한다. 전략적 관점에서 자율주행자동차의 활성화를 위해서는 자율주행단계에 따른 확산전략을 수립하고, 정부지원정책을 초기 활성화의 트리거로 활용하고, 세그멘테이션(segmentation), 타켓팅(targeting), 포지셔닝(positioning) 전략을 기초로 효과적인 마케팅믹스(4P Mix)전략이 실행되어야 한다.

Activation of Antioxidant-Response Element (ARE), Mitogen- Activated Protein Kinases (MAPKs) and Caspases by Major Green Tea Polyphenol Components during Cell Survival and Death

  • Green tea polyphenols (GTP) have been demonstrated to suppress tumorigenesis in several chemical-induced animal carcinogenesis models, and predicted as promising chemopreventive agents in human. Recent studies of GTP extracts showed the involvement of mitogen-activated protein kinases (MAPKs) in the regulation of Phase II enzymes gene expression and induction of apoptosis. In the current work we compared the biological actions of five green tea catechins: (1) induction of ARE reporter gene, (2) activation of MAP kinases, (3) cytotoxicity in human hepatoma HepG2-C8 cells, and (4) caspase activation in human cervical squamous carcinoma HeLa cells. For the induction of phase IIgene assay, (-)-epigallocatechin-3-gallate (EGCG) and (-)-epicatechin-3-gallate (ECG) potently induced antioxidant response element (ARE)-mediated luciferase activity, with induction observed at 25 $\mu\textrm{m}$with EGCG. The induction of ARE reporter gene appears to be structurally related to the 3-gallate group. Comparing the activation of MAPK by the five polyphenols, only EGCG showed potent activation of all three MAPKs (ERK, JNK and p38) in a dose- and time-dependent manner, whereas EGC activated ERK and p38. In the concentration range of 25 $\mu\textrm{m}$ to 1 mM, EGCG and ECG strongly suppressed HepG2-ARE-C8 cell-growth. To elucidate the mechanisms of green tea polyphenol-induced apoptosis, we measured the activation of an important cell death protein, caspase-3 induced by EGCG, and found that caspase-3 was activated in a dose- and time-dependent manner. Interestingly, the activation of caspase-3 was a relatively late event (peaked at 16 h), whereas activation of MAPKs was much earlier (peaked at 2 h). It is possible, that at low concentrations of EGCG, activation of MAPK leads to ARE-mediated gene expression including phase II detoxifying enzymes. Whereas at higher concentrations of EGCG, sustained activation of MAPKs such as JNK leads to apoptosis. These mechanisms are currently under investigation in our laboratory. As the most abundant catechin in GTP extract, we found that EGCG potently induced ARE-mediated gene expression, activated MAP kinase pathway, stimulated caspase-3 activity, and induced apoptosis. These mechanisms together with others, may contribute to the overall chemopreventive function of EGCG itself as well as the GTP.

Interaction of Bovine Growth Hormone with Buffalo Adipose Tissue and Identification of Signaling Molecules in Its Action

  • Results on localization of growth hormone receptor (GHR), interaction of growth hormone (GH) with receptor in buffalo adipose tissue and identification of activated signaling molecules in the action of GH are presented. Bovine GH (bGH) was labeled with fluorescein or biotin. Fluorescein-labelled bGH was used for localization of GHRs in buffalo adipocytes. The receptors were present on the cell surface. The affinity of binding of GH to its receptor was determined by designing an experiment in which buffalo adipose tissue explants, biotinylated GH and streptavidin-peroxidase conjugate were employed. The affinity constant was calculated to be $2{\times}10^8M^{-1}$. The receptor density on adipose tissue was found to be 1 femto mole per mg of tissue. Signalling molecules generated in the action of GH were tentatively identified by employing Western blot and enhanced chemiluminescence techniques using anti-phosphotyrosine antibody. Based on molecular weights of proteins reactive to anti-phosphotyrosine antibody, three signaling molecules viz. insulin receptor substrate, Janus activated kinase (Jak) and mitogen activated protein were tentatively identified. These signaling molecules appeared in a time (incubation time of explants with growth hormone) dependent way. The activation of Jak2 was confirmed by employing anti-Jak2 antibody in a Western blot. The activation of Jak2 occurred during 5 min incubation of buffalo adipose tissue explants with GH and incubation for an additional period, viz. 30 min. or 60 min., resulted in a drastic reduction in activation. The results suggest that Jak2 activation is an early event in the action of GH in buffalo adipose tissue.

Neural Switching Mechanism in the late Korean-English bilinguals by Event-Related fMRI

  • Functional MRI technique was used in this study for examining the language switching mechanisms between the first language (L1) and the second language (L2). Language switching mechanism is regarded as a complex task that involves an interaction between L1 and L2. The aim of study is to find out the brain activation patterns during the phonological process of reading real English words and English words written in Korean characters in a bilingual person. Korean-English bilingual subjects were examined while they covertly read four types of words native Korean words, Korean words of a foreign origin, English words written in Korean characters, and English words. The fMRI results reveal that the left hemispheric language-related regions at the brain, such as the left inferior frontal, superior temporal, and parietal cortices, have a greater response to the presentation of English words written in Korean characters than for the other types of words, in addition, a slight difference was observed in the occipital-temporal lobe. These results suggest that a change in the brain circuitry underlying the relational processes of language switching is mainly associated with general executive processing system in the left prefrontal cortex rather than with a similarity-based processing system in the occipital-temporal lobes.

  • The advanced passive pressurized water reactor (PWR) is being constructed in China and the passive residual heat removal (PRHR) system was designed to remove the decay heat. During accident scenarios with increase of heat removal from the primary coolant system, the actuation of the PRHR will enhance the cooldown of the primary coolant system. There is a risk of power excursion during the cooldown of the primary coolant system. Therefore, it is necessary to analyze the thermal hydraulic behavior of the reactor coolant system (RCS) at this condition. The advanced passive PWR model, including major components in the RCS, is built by SCDAP/RELAP5 code. The thermal hydraulic behavior of the core is studied for two typical accident sequences with PRHR actuation to investigate the core cooling capability with conservative assumptions, a main steam line break (MSLB) event and inadvertent opening of a steam generator (SG) safety valve event. The results show that the core is ultimately shut down by the boric acid solution delivered by Core Makeup Tank (CMT) injections. The effects of CMT boric acid concentration and the activation delay time on accident consequences are analyzed for MSLB, which shows that there is no consequential damage to the fuel or reactor coolant system in the selected conditions.

현장 굴착 실험을 통한 사면붕괴 거동 연구 (A Study on behavior of Slope Failure Using Field Excavation Experiment)

  • Recently, the occurrence of landslides has been increasing over the years due to the extreme weather event. Developments of landslides monitoring technology that reduce damage caused by landslide are urgently needed. Therefore, in this study, a strain ratio sensor was developed to predict the ground behavior during the slope failure, and the change in surface ground displacement was observed as slope failed on the field model experiment. As a result, in the slope failure, the ground displacement process increases the risk of collapse as the inverse displacement approaches zero. It is closely related to the prediction of precursor. In all cases, increase in displacement and reverse speed of inverse displacement with time was observed during the slope failure, and it is very important event for monitoring collapse phenomenon of risky slopes. In the future, it can be used as disaster prevention technology to contribute in reduction of landslide damage and activation of measurement industry.

Two-Ribbon Filament Eruption on 29 September 2013

  • We have presented a classic two-ribbon filament eruption occurred in the east side of NOAA active region 11850 at 21:00 UT on 29 September 2013. Interestingly, this filament eruption was not accompanied by any flares and just there was a slight brightening in X-rays, C1.2, associated with the eruption. An accompanying huge CME was appeared at 22:12 UT in the LASCO C2 field of view and it propagates into the interplanetary space with a speed of about 440 km/s. And the related solar proton event (S2) started at 05:05 UT and peaked at 20:05 UT on 30 September 2013. The CME arrival was recorded by the ACE spacecraft around 01:30 UT on 2 October 2013. Around the CME arrival time, the solar-wind speed reached at about 640 km/s and IMF Bz showed southward component (-27 nT). Finally, the filament eruption and the CME cause geomagnetic storm (G2) at 03:00 UT on 2 October 2013. We described the detailed evolution of the filament eruption and its related phenomena such as CME, proton event, geomegnetic storm and so on. In addition, we will discuss about the activation mechanism of the filament eruption without flares.

An Extensive Analysis of High-density Electroencephalogram during Semantic Decision of Visually Presented Words

  • The purpose of this study was to investigate the spatiotemporal cortical activation pattern and functional connectivity during visual perception of words. 61 channel recordings of electroencephalogram were obtained from 15 subjects while they were judging the meaning of Korean, English, and Chinese words with concrete meanings. We examined event-related potentials (ERP) and applied independent component analysis (ICA) to find and separate simultaneously activated neural sources. Spectral analysis was also performed to investigate the gamma-band activity (GBA, 30-50 Hz) which is known to reflect feature binding. Five significant ERP components were identified and left hemispheric dominance was observed for most sites. Meaningful differences of amplitudes and latencies among languages were observed. It seemed that familiarity with each language and orthographic characteristics affected the characteristics of ERP components. ICA helped confirm several prominent sources corresponding to some ERP components. The results of spectral and time-frequency analyses showed distinct GBAs at prefrontal, frontal, and temporal sites. The GBAs at prefrontal and temporal sites were significantly correlated with the LPC amplitude and response time. The differences in spatiotemporal patterns of GBA among languages were not prominent compared to the inter-individual differences. The gamma-band coherence revealed short-range connectivity within frontal region and long-range connectivity between frontal, posterior, and temporal sites.

  • 기능적 자기공명영상기법(functional MRI: fMRI)을 통해서 인간의 뇌에 대한 연구가 지난 수년 동안 활발하게 진행되어 왔다. 이러한 기능적 자기공명영상기법의 파라다임에는 Block-diagram 방법과 최근에 개발되어 이용되는 Event-related 방법이 있다. Block-diagram 방법은 여러 블록으로 구성하여 피험자들에게 자극을 인가하여 뇌의 반응신호들을 통해 뇌의 기능을 설명해 주는 데, 이러한 뇌의 기능이 얼굴인식과 관련하여 여러 조건을 포함한 자극인 경우 뇌의 인지 시스템을 설명을 해 주는 데 한계가 있다. 따라서 Event-related 방법을 이용하여 얼굴인식에 관여하는 뇌의 인지시스템에 관련된 부분을 설명하고, 또한 그 영역에서 인종구별과 같은 의식적으로는 관찰되지 않지만 무의식적으로 관찰되는 뇌의 생리적인 현상을 보여주고자 한다. 본 연구는 Event-related 방법에 대한 이해와 얼굴인식에 적용되는 파라다임 알고리즘을 개발하여 피험자들을 통해 해당조건들은 (유명한국인, 무명한국인, 유명 백인 미국인, 무명 백인미국인) 각 Event로 구성하여 한 블록인 Event-related 파라다임으로 변경하여 얼굴을 인식하는데 주로 담당하는 영역인, Ventral occipital-temporal cortex 내의 fusiform face area(FFA)를 확인하고 그 영역에서 의식적으로 반응하는 뇌의 활동과 무의식적으로 반응하는 뇌의 활동을 생리학적 또는 심리학적으로 해석하는데 용이하도록 여러 파라미터들 (즉 Event 간격 , Event의 수, Null Event, TR 간격 등)을 최적화하였다. 자기공명영상기법을 이용한 이러한 뇌 기능 연구는 의학적인 응용연구로서 신경 과학분야와 정신의학적인 측면에서 널리 이용될 것으로 보이며 더 나아가 심리학적측면의 기초 연구로서 발전의 토대가 될 것으로 사료된다.다. 불가능하여 차나무 재배지 부적합한 것으로 나타났다.35\%$일 경우 알칼리-실리카 반응에 의한 팽창을 가장 효과적으로 방지할 수 있는 것으로 판단된다.는 accessory 개발과 보급이 중요하다.에 대하여 500MU 조사 후 방사선량의 측정결과 Field size의 영향은 거의 존재하지 않았으며, beam off 후 $1{\sim}2$분 뒤 측정 시작 시점에서 대략 $1{\mu}Sv/hr$를 나타냈으며, 반감기는 약 $3{\sim}4$분인 것으로 측정되었다. Primus 15MV의 경우 S병원에서 측정을 수행하였으며, $25{\times}25cm^2$ Field size에 대하여 500MU 조사 뒤 방사선량을 측정한 결과, beam off후 $1{\sim}2$분 뒤 측정시점에서 대략 $3.26{\mu}Sv/hr$를 나타냈으며 10MV X선보다 대략 3.3배 큰 값을 나타내었다. 결과 : 일일 치료환자가 $20{\sim}50$명이고, 환자 1인당 Wedge filter의 교체작업이 $1{\sim}2$회일 때 10MV의 경우 연간선량이 $0.08{\sim}0.4mSv$로 평가되었으며, 15MV의 경우 $0.27{\sim}1.36mSv$로 평가되어 작업종사자의 연간 허용선량인 20mSv에 비해 안전한 것으로 평가되었다.서 정상조직이 적게 조사되었다. 결과 : 기존의 ICRU계획은 그 효과 및 안전성이 입증되었음에도 불구하고 CT를 이용한 CTV계획 등을 적용 한다면 잔류종양이 적은 경우 정상조직에 대한 조사를 줄이면서 잔류종양에 목표선량을 조사할 수 있을 것이다. 다만 잔류종양이 큰 경우는 정상조직에 대한 조사선량을 줄이기 위한 효과적 치료계획에

RAW 264.7 대식세포에서 마늘 유래 황 함유 화합물에 의한 요산 유도 inflammasome 활성화의 억제는 ROS 생성 차단과 연관성이 있음 (Suppression of Monosodium Urate-induced NLRP3 Inflammasome Activation by Garlic-derived Sulfur-containing Phytochemicals is Associated with Blocking ROS Generation in RAW 264.7 Macrophages)

  • 만성 염증성 관절염 질환인 통풍은 고요산혈증을 특징으로 하며 NLRP3 inflammasome의 활성화에 따른 IL-1β와 같은 염증성 cytokine 방출과 연관된 MSU에 대한 염증 반응에 의해 유발될 수 있다. 마늘에 함유된 황 함유 화합물은 다양한 질병에 대한 잠재적인 유익한 약리학적 효능을 가지지만, NLRP3 inflammasome 활성화와 연관된 통풍 억제에 대한 효능은 현재까지 입증되지 않았다. 본 연구에서는 대표적인 마늘 유래 황화합물인 DADS와 DATS가 MSU에 의한 NLRP3 inflammasome 활성을 억제할 수 있는지를 조사하였다. 본 연구의 결과에 의하면, 비세포 독성 조건에서 DADS와 DATS는 LPS가 전처리된 RAW 264.7 대식세포에서 MSU에 대한 반응으로 증가된 NO의 생성과 IL-1β 유리를 유의적으로 차단하였다. DADS와 DATS는 또한 증가된 NLRP3, ASC, caspase-1 p20 및 IL-1β의 발현을 감소시켰으며, 이는 MSU로 유도된 LRP3 inflammasome 활성화가 DADS와 DATS에 의해 억제되었음을 의미한다. 아울러, DADS와 DATS는 NLRP3 inflammasome 활성화에 상위 신호로 작용하는 산화적 스트레스를 차단했으며 ROS 생성을 제거한다는 사실에서 입증되었다. 결론적으로, 본 연구의 결과는 DADS와 DATS가 ROS/NLRP3 경로를 억제하여 inflammasome 활성화를 차단함으로서 NLRP3 의존성 통풍성 관절염 치료를 위한 잠재력이 우수함을 의미한다.
