Analysis of the Genetic Diversity of Radish Germplasm through SSR Markers Derived from Chinese Cabbage (배추 SSR 마커를 이용한 무의 육성 계통 및 수집종의 유전적 다양성 분석)

  • Since the early 1980s, the National Institute of Horticultural & Herbal Sciences has been breeding and collecting diverse radish breeds to select those samples with better horticultural characteristics, to ultimately expand and develop as good radish produce. Genetic diversity is a crucial factor in crop improvement and therefore it is very important to obtain various variations through sample collection. The collected samples were compared with one another in order to assess the level of diversity among the collections, and this procedure allowed for increased application of the gathered resources and aided in determining the direction to secure further samples. Towards this end, this experiment was conducted in order to examine whether the SSR markers derived from Chinese cabbage samples could be transferred to the radish samples. Among the radish breeding lines and introduced resources, 44 lines were used as materials to analyze the genotype using 22 SSR markers selected. As a result, the analysis showed that among all the selected markers, 'cnu_m139' and 'cnu_m289' were the most useful markers for diversity evaluation. The genetic relationship of the radish genetic resources showed that the geographic origins affected the diversity. Furthermore, the different types of radish groups were also determined by the year they were bred. This result demonstrated that there are differences between the older radish breeds and the more recently developed radish breeds. Even though a relatively small number of markers were used in the analysis, it was possible to distinguish whether the radish was bred 30 years ago or in the 2000s, and that the similar physical shapes comprised a particular group, showed that the SSR markers can indeed be successfully applied to to study the diversity within radish breeding lines. Through the results of this study, it can be concluded that the SSR marker developed for the Chinese cabbage can be applied to examine the genetic diversity and analyze the relationship (genetic resource determination) of radish.

Comparative Quality Evaluation of King Oyster Mushroom as Affected by Unit Packaging Method during Simulated Export Shipment (큰느타리버섯의 모의 수출운송과정에서 소포장 적용에 따른 품질변화 비교)

  • Potential of consumer unit packaging was investigated for quality maintenance during export simulation in king oyster mushrooms (Pleurotus eryngii). Mushrooms were harvested in late May, precooled to $4^{\circ}C$ within 6 hours, and then packaged for shipping in two ways: 2 kg bulk packaging in a polyethylene (PE) bag or three types of unit packaging methods such as 400 g in polypropylene film bag (PPB), 200 g on styrofoam tray + PE shrinkage film wrapping (STW), and 200 g in polyethylene terephthalate (PET) containers (PETC). For local distribution of bulk-packaged commodity, mushrooms were sorted again and packaged into 3 consumer units in the same way as for the initial shipping packages. Simulation of refrigerated container shipping was performed in a walk-in type pilot storage at $0.5^{\circ}C$ for 5 weeks, while local marketing simulation was carried out on the shelf at $7^{\circ}C$ for 7 days. During the shipment simulation, creation of modified atmosphere (MA) was substantial in 2 kg bulk packages with low $O_2$ below 2% and high $CO_2$ over 15% whereas, in PPB and PETC unit packages, relatively higher $O_2$ concentrations were observed. On the shelf at $7^{\circ}C$, $CO_2$ concentrations rapidly increased in PPB and PETC packages despite the short marketing period. Overall marketability evaluated by off-flavor, browning, and texture rating was maintained at excellent level when 2 kg bulk packaging in PE or unit packaging in PPB and PETC were used for shipment. In contrast, establishment of MA was very slight in STW packages during shipment and local distribution resulting in poor quality after export simulation. The results suggested that shipment using adequate consumer unit packaging is more practical and economically beneficial than using bulk packaging in the export program consisting of 5-week shipment and 7-day local distribution.

Physicochemical Characteristics of Liquid Fertilizer made from Pig Manure in Korea (국내 돼지분뇨의 액비성분 특성 비교조사)

  • Physicochemical properties of liquid fertilizer samples of resource organization, which are domestically produced and distributed, are analyzed. Major contents of the research results are as follows. 1. The ratio of complete decomposition for liquid fertilizer is 49% at Public Resource Center and 33% at Liquid Fertilizer Supply Center. The combined ratio of both half-decomposed and un-decomposed liquid fertilizers is over 50% at both centers. 2. The ratio of complete decomposed liquid fertilizer, 67%, is the highest in Gangwon and Gyeonggi-do area. The ratio of un-decomposed liquid fertilizer is high in Chungbuk and Chungnam area. The sum of ratios of the half- and un-decomposed is over 60% in the areas except Gyeonnggi-do and Gangwon-do. 3. As a result of regional comparison of the physicochemical properties of liquid fertilizers, concentration variation in most of the items are large, and the degree of uniformity is found to be considerably low. In particular, concentration variation in T-N and $NH_4$-N is the most noticeable. 4. The items that physicochemically correlated to the degree of decomposition of liquid fertilizer are appeared to be T-N, $NH_4$-N, $NO_3$-N, EC, $SCOD_{Mn}$, and ORP. 5. The physicochemical average values of the liquid fertilizer estimated as "complete decomposed" are appeared to be T-N 829 mg/L,$NH_4$-N 517 mg/L, $NO_3$-N 151 mg/L, $SCOD_{Mn}$ 1,205 mg/L, EC 10.32 mS/cm, ORP -117.12 mV.

The Usefulness of Visceral Fat Thickness Measured by Ultrasonography as an Abdominal Obesity Index (초음파로 측정된 내장지방두께의 복부 비만지표로서의 유용성)

  • Purpose : Abdominal obesity with visceral fat accumulation have been known to be intimately associated with the development of metabolic syndrome. Therefore, it is important to estimate the precise amount of visceral fat. Ultrasonography has been reported that it is a simple and noninvasive method for visceral fat evaluation. Purpose of this study is to evaluate the association of ultrasonographic visceral fat thickness, anthropometric indexes, and risk factor of metabolic syndrome, and to investigate the cut-off value of abdominal visceral fat thickness leading to increased risk of metabolic syndrome. Materials and methods : The subject included 200 men and 200 women who visited D healthcare center in Daejeon from January to April 2008. The subcutaneous fat thickness and visceral fat thickness were measured by ultrasonograph. As anthropometric index, we measured body mass index, waist circumference and waist/height ratio. As for the risk factor of metabolic syndrome, we measured blood pressure, high density lipoprotein cholesterol, triglyceride and fasting serum glucose. Results : VFT was significantly correlated with waist circumference, (r=0.683/M, r=0.604/F), waist to height ratio (r=0.633/M, r=0.593/F) and BMI (r=0.621/M, r=0.534/F) in both men and women. In addition it was significantly correlated with Systolic blood pressure (r=0.229/M, r=0.232/F), Diastolic blood pressure ((r=0.285/M, r=0.254/F), high density cholesterol (r=-0.254/M, r=-0.254/F), Triglyceride (r=0.475/M, r=0.411/F), and Fasting blood sugar (r=0.158/M, r=0.234/F) in both men and women. The cut-off value of visceral fat thickness leading to the increased risk of metabolic syndrome was 4.58cm (sensitivity89.2%, specificity 71.2%) in men and 3.50cm (sensitivity61.2% specificity 80.8%) in women respectively. The odds ratio of the risk of metabolic syndrome was dramatically increased with the abdominal visceral fat thickness level over 6cm in men and 5cm in women. Conclusion : The visceral fat thickness using ultrasonography was significantly correlated with anthropometric indexes and risk factors of metabolic syndrome in both men and women. The cut-off value of visceral fat thickness leading to the increased risk of metabolic syndrome was 4.58cm in men and 3.50cm in women.

  • Purpose Recently PET/CT image's attenuation correction is used CTAC(Computed Tomgraphy Attenuation Correction). it can quantitative evaluation by SUV(Standard Uptake Value). This study's purpose is to evaluate SUV and to find proper CT kernel using CTAC with applied various CT kernel to PET/CT construction. Materials and Methods Biograph mCT 64 was used for the equipment. We were performed on 20 patients who had examed at our hospital from february through March 2017. Using NEMA IEC Body Phantom, The data was reconstructed PET/CT images with CTAC appiled various CT kernel. ANOVA was used to evaluated the significant difference in the result. Results The result of measuring the radioactivity concentration of Phantom was B45F 96% and B80F 6.58% against B08F CT kernel, each respectively. the SUVmax increased to B45F 0.86% and B80F 6.54% against B08F CT kernel, In case of patient's parts data, the Lung SUVmax increased to B45F 1.6% and B80F 6.6%, Liver SUVmax increased to B45F 0.7% and B80F 4.7%, and Bone SUVmax increased to B45F 1.3% and B80F 6.2%, respectively. As for parts of patient's about Standard Deviation(SD), the Lung SD increased to B45F 4.2% and B80F 15.4%, Liver SD increased to B45F 2.1% and B80F 11%, and Bone SD increased to B45F 2.3% and B80F 14.7%, respectively. There was no significant difference discovered in three CT kernel (P >.05). Conclusion When using increased noise CT kernel for PET/CT reconstruction, It tends to change both SUVmax and SD in ROI(region of interest), Due to the increase the CT kernel number, Sharp noise increased in ROI. so SUVmax and SD were highly measured, but there was no statistically significant difference. Therefore Using CT kernel of low variation of SD occur less variation of SUV.

Evaluation on Spectral Analysis in ALOS-2 PALSAR-2 Stripmap-ScanSAR Interferometry (ALOS-2 Stripmap-ScanSAR 위상간섭기법에서의 스펙트럼 분석 평가)

  • It is well known that alluvial sediment located in coastal region has been easily affected by geohazard like ground subsidence, marine or meteorological disasters which threaten invaluable lives and properties. The subsidence is a sinking of the ground due to underground material movement that mostly related to soil compaction by water extraction. Thus, continuous monitoring is essential to protect possible damage from the ground subsidence in the coastal region. Radar interferometric application has been widely used to estimate surface displacement from phase information of synthetic aperture radar (SAR). Thanks to advanced SAR technique like the Small BAseline Subset (SBAS), a time-series of surface displacement could be successfully calculated with a large amount of SAR observations (>20). Because the ALOS-2 PALSAR-2 L-band observations maintain higher coherence compared with other shorter wavelength like X- or C-band, it has been regarded as one of the best resources for Earth science. However, the number of ALOS-2 PALSAR-2 observations might be not enough for the SBAS application due to its global monitoring observation scenario. Unfortunately, the number of the ALOS-2 PALSAR-2 Stripmap images in area of our interest, Busan which located in the Southeastern Korea, is only 11 which is insufficient to apply the SBAS time-series analysis. Although it is common that the radar interferometry utilizes multiple SAR images collected from same acquisition mode, it has been reported that the ALOS-2 PALSAR-2 Stripmap-ScanSAR interferometric application could be possible under specific acquisition mode. In case that we can apply the Stripmap-ScanSAR interferometry with the other 18 ScanSAR observations over Busan, an enhanced time-series surface displacement with better temporal resolution could be estimated. In this study, we evaluated feasibility of the ALOS-2 PALSAR-2 Stripmap-ScanSAR interferometric application using Gamma software considering differences of chirp bandwidth and pulse repetition frequency (PRF) between two acquisition modes. In addition, we analyzed the interferograms with respect to spectral shift of radar carrier frequency and common band filtering. Even though it shows similar level of coherence regardless of spectral shift in the radar carrier frequency, we found periodic spectral noises in azimuth direction and significant degradation of coherence in azimuth direction after common band filtering. Therefore, the characteristics of spectral bandwidth in the range and azimuth direction should be considered cautiously for the ALOS-2 PALSAR-2 Stripmap-ScanSAR interferometry.

Evaluation of Available Soil Silicon Extracting Procedures for Oriental Melon (참외 시설재배 토양에 대한 유효규산 추출방법 비교)

  • Soil testing for silicon (Si) in the upland soils has not been sufficiently investigated. The objective of this study was to identify a suitable Si extraction method for upland soils of oriental melon (Cucumis melo L.). Thirty-eight surface soil samples and matured leaf samples were collected from plastic film houses in Sungju, Gyeongbuk province. In the laboratory, six different methods were used for extracting Si from the soils. The methods included 0.5 N HCl extraction, 1 N sodium acetate buffer (PH 4.0) extraction, citric acid 1% extraction, water extraction, Tiis buffer pH 7.0 extraction, and extraction after incubation with water for 1 week. The concentration of dissolved Si in soil extracts from all methods was determined colorimetrically. With 1 N sodium acetate buffer extraction, as the available soil Si increased, the concentration ofSi in oriental melon leaf increased until around $14g\;SiO_2\;kg^{-1}$ was reached in the form of a saturation curve. Also, among the methods studied, extraction with 1 N sodium acetate buffer was the only method provided a significant linear correlation with oriental melon leaf Si content in the range of extractable soil Si lower than the level which inducing Si saturation in oriental melon leaf. These results indicate that 1 N sodium acetate buffer extraction procedure is the best soil Si test method for upland soils of oriental melon. This sodium acetate buffer extraction procedure is rapid and quite well acquainted with scientists and farmers, since the method has been used for routine paddy soil testing.

The Comparison and Distribution of Temperatures Established in Display Stands and Food Surfaces for Cold and Frozen Foods in large Discount Stores in Korea (대형할인매장에서의 냉장.냉동식품 판매대의 설정온도와 판매식품 표면온도 비교 및 온도분포 분석)

  • This study surveyed and compared the temperatures established in display stands and food surfaces for cold and frozen foods in large discount stores in Korea. The temperatures established in display stands for cold food ranged with $3.5{\pm}1.8^{\circ}C$ as mean, minimum and maximum were $0^{\circ}C$ and $7^{\circ}C$. However, the surface temperatures of cold food on sale ranged with $10.7{\pm}2.9^{\circ}C$ as a mean, minimum $4.6^{\circ}C$ and maximum $18.4^{\circ}C$. Totally, the surface temperature of cold food on sale was $7.2^{\circ}C$, as a mean, higher than established in display stands for cold food in large discount stores in Korea. 53% of the surveyed cold foods were more than $10^{\circ}C$ in surface temperature and only 47% was less than $10^{\circ}C$. The differences between temperatures were lowest in fruits, salads and vegetables, but highest in milk products. On the other hand, the temperatures established in display stands for frozen food showed a range with $-20.7{\pm}1^{\circ}C$ as a mean. However, the surface temperatures of frozen food on sale showed a range with $-15.4{\pm}5^{\circ}C$ as a mean, minimum $-28^{\circ}C$ and maximum $-4.6^{\circ}C$ (included defrosting). The surface temperatures of frozen food, frozen meats, frozen processed foods and ice creams were $-13.8^{\circ}C$, $-15.9^{\circ}C$, and $-16.8^{\circ}C$, respectively. Only 32.3% of surveyed frozen foods showed less than $-18^{\circ}C$ in surface temperature. In conclusion, the temperatures established on cold and frozen food display stands were less than those of cold and frozen food surfaces on sale. There was also much variation in food surface temperatures during cold and frozen food storage and sales. Therefore, a temperature management system technology use at the distribution level for cold and frozen foods will be developed.

Utility of CoaguChek XS for Monitoring the Prothrombin Time (프로트롬빈시간 모니터링을 위한 CoaguChek XS의 유용성)

  • Background: In order to achieve a maintenance level and to prevent hemorrhagic complications, regular monitoring of the INR is mandatory for patients on oral anticoagulation therapy (OAT). A point-of-care instrument for INR monitoring is convenient for users, but the accuracy of the results has been controversial, and so this calls for exact evaluation of the point-of-care instrument that is used for INR monitoring. Methods: From Aug 2007 through Feb 2008, 85 patients on OAT among the all the patients who were admitted to Soonchunhyang University Bucheon Hospital were involved in this study. Parallel measurements of the PT INR were performed using a CoaguChek-XS and, a CA-7000 laboratory reference instrument and the results were analyzed. In addition, the patients' clinical data, including the diagnosis and the frequency and interval of the INR measurements, were also analyzed. Results: Of the 85 patients, 25 were admitted more than once to undergo INR testing and the mean interval between testing was 8.6 weeks with 39% and 38% of the tests being less than INR 2 units with using the CoaguChek-XS and the reference method, respectively. The coefficients of variation of CoaguChek-XS were 4.50 and 2.45 for the high and low INR patients, respectively. An excellent correlation was found between the two methods with a $R^2$ of 0.966 (p<0.001). Through Bland-Altman analysis, the mean INR difference between the two methods was 0.13 with the limit of agreement being -0.47~+0.72 with a 95% confidence interval. CoaguChek-XS was shown to overestimate the INR value for patients with an increasing INR, as compared to the reference method. Conclusion: CoaguChek-XS demonstrated great precision and accuracy for patients on OAT when compared to the laboratory INR results. Accordingly, the instrument should help to monitor the INR in the patients on OAT.

Present Conditions of Mental Health Care in Rural Areas: Community Mental Health Program of Public Health Center (농촌지역 정신보건관리실태: 보건소 지역사회정신보건사업)

  • Objectives: This paper introduces need and supply level of rural mental health care service and especially focuses on the evaluation for the community mental health programs of Public Health Centers(PHCs) in rural areas as the facilities for primary mental health care. Methods: We defined the need as prevalance rate and service utilization rate, for which reviewed the results of the epidemiological study of mental disorders using Korean Composite International Diagnostic Interview surveyed on a nationwide scale in 2001. Supply was appraised in terms of psychiatric beds and primary mental heath care facilities such as private psychiatric clinics, facilities for social rehabilitation, PHCs running community mental health programs. For this, we reveiwed a variety of annual reports related mental health published by Ministry of Health and Welfare. To evaluate the community mental health programs of PHCs in rural areas, we selected. randomly samples out of the 3rd community health plans including the contents of community mental health programs, which submitted by 89 rural counties and 44 cities mixed with rural areas, and used the program's guideline established by central government as a standard. Results: Prevalence rates of major psychiatric diseases such as schizophrenia, alcoholism, major depression, anxiety disorder were higher in rural area than in urban area and 8.9% of psychiatric patients in both areas stayed at homes contacted with mental health manpower more than one time during the last year. Psychiatric beds were sufficiently supplied, but urban area had less beds than rural area contrary to general health care service. Psychiatric clinics were supplied very insufficiently in rural areas and PHCs bridged the gap instead. However rural PHCs got less financial support for community mental health programs from higher positioned agencies than urban PHCs. Rural community health programs not supported hardly worked out. Conclusions: Central government should consider a special policy for rural primary mental health care, because private psychiatric clinics can't be introduced in rural areas due to demand-deficiency and the financial independence of rural counties was very vulnerable.
