Antibacterial Activity of Euphorbia humifusa Extracts on Food-Borne Pathogenic Bacteria (비단풀 추출물의 식중독세균에 대한 항균활성)

  • This study was performed to investigate the antimicrobial effects of Euphorbia humifusa ethanol-extract against food-borne pathogens. The growth inhibitory effects of the extract at a concentration of 250, 500, 1,000 or 2,000 mg/L on food poisoning microorganism were determined against Salmonella typhimurium, Listeria monocytogenes, Yersinia enterocolitica, Escherichia coli O157:H7 and Staphylococcus aureus. The microorganisms growth was not affected by the extract at the concentration up to 250 mg/L, but was significantly (p<0.05) inhibited by the extract at a concentration higher than 1,000 mg/L. The extract of Euphorbia humifusa had strong antimicrobial activity against all test strains at a concentration of 2,000 mg/L. The results in the present study demonstrate antimicrobial effects of Euphorbia humifusa ethanol-extract against food-borne pathogens, suggesting that Euphorbia humifusa could be an effective natural antibacterial agent in food.

Cellular Protective Effect and Component Analysis of Euphorbia humifusa Extracts (땅빈대 추출물의 세포 보호 효과 및 성분 분석에 관한 연구)

  • In this study, the cellular protective effect, antioxidative property and component analysis of Euphorbia humifusa extracts were investigated. The ethyl acetate fraction ($3.68\;{\mu}g/mL$) and aglycone fraction ($3.15\;{\mu}g/mL$) of Euphorbia humifusa extract showed prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of Euphorbia humifusa extract on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction ($0.43\;{\mu}g/mL$) and aglycone fraction ($0.35\;{\mu}g/mL$) of extract showed higher ROS scavenging activity than L-ascorbic acid ($1.50\;{\mu}g/mL$). The cellular protective effects of fractions of Euphorbia humifusa extract on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction and aglycone fraction of extract protected cellular membranes against ROS in a concentration dependent manner ($5{\sim}25\;{\mu}g/mL$), and was more effective than (+)-${\alpha}$-tocopherol, lipid peroxidation chain blocker. Aglycone fraction from Euphorbia humifusa extract showed 2 bands in TLC and 2 peaks in HPLC. In HPLC chromatogram of aglycone fraction, peak 1 and peak 2 were identified as quercetin and kaempferol, respectively. And these components are very effective as antioxidant. Thus, these results indicate that fractions of Euphorbia humifusa extracts can function as antioxidant in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. Fractions of Euphorbia humifusa extracts can be applicable to new functional cosmetics for antioxidant.

Cytotoxic Activities of Panax ginseng and Euphorbia humifusa in Human Brain Tumor Cells (인삼 비당부와 땅빈대의 뇌암세포 독성작용)

  • The effects of acid hydrolysis product of Panax ginseng and MeOH extract of Euphorbia humifusa on the growth of human brain tumor cells were evaluated using U-373 MG human astrocytoma and SK-N-MC human neuroblastoma cells as model cellular systems. These plant extracts induced cytotoxicity in both cells in a dose-dependent manner. These cytotoxic effects were significantly inhibited by GSH, an antioxidant, in both cells. BAPTA/AM, an intracellular $Ca^{2+}$ chelator, significantly blocked the cytotoxic effects of these extracts in U-373 cells, but enhanced these effects in SK-N-MC cells. These results suggest that the plant extracts may be a valuable choice for the studies on the treatment of human brain tumors.

Antioxidant Activity and Cytotoxic Effect of Extracts from Euphorbia humifusa (땅빈대 추출물의 항산화 활성 및 세포독성 효과)

  • The antioxidant activities, anti-inflammatory activity and cytotoxic effects of methanol extract from Euphorbia humifusa were evaluated in this study. Total phenolic compound contents were $68.35{\pm}0.16$ mg/g and total flavonoid compound contents were estimated as $38.74{\pm}1.26$ mg/g. EC50 values for DPPH radical scavenging activity of methanol extract was $56.26{\pm}0.66{\mu}g/mL$ and those of positive controls as ascorbic acid, ${\alpha}$-tocopherol and BHA were $8.38{\pm}0.14{\mu}g/mL$, $16.45{\pm}0.89{\mu}g/mL$ and $21.18{\pm}1.01{\mu}g/mL$ respectively. NO scavenging activity increased in depending on concentration of extract. Treatment of RAW 264.7 cells with extract caused inhibition of LPS-induced nitric oxide production. The cell viability showed that the methanol extract had cytotoxicity in the growth of breast cancer cell ($66.54{\pm}1.91%$ at $400{\mu}g/mL$ conc., $43.98{\pm}3.35%$ at $800{\mu}g/mL$ conc.). Based on the results, It was suggested that the methanol extract of Euphorbia humifusa has a potential candidate for functional cosmetic and medicine.

Identification of the active components inhibiting the expression of matrix metallopeptidase-9 by TNFα in ethyl acetate extract of Euphorbia humifusa Willd

  • Euphorbia humifusa Willd (EuH), called Ttang-Bin-Dae in Korea, is a traditional medicinal plant widely used for its anti-inflammatory and antiviral activity. Ethyl acetate (EA) extracts of EuH (EA/EuH) inhibit invasion and metastasis by inhibiting tumor necrosis factor TNFá-induced matrix metalloproteinases (MMP)-9 expression in human breast cancer cells. However, the bioactive components of EA/EuH mediating the inhibition of MMP-9 expression have not been identified. In the present study, three bioactive constituents of EA/EuH were isolated using high-performance liquid chromatography. Nuclear magnetic resonance spectroscopy revealed isoquercetin, avicularin, and astragalin as the bioactive compounds responsible for preventing TNFα-induced MMP-9 mRNA expression in breast cancer cells. These findings suggest that isoquercetin, avicularin, and astragalin could be used as valuable anti-metastatic agents against metastatic cancers.

Antioxidant activities and physiological properties of Euphorbia humifusa extracts prepared using different solvents (추출용매에 따른 비단풀 추출물의 항산화 및 생리활성)

  • This study was conducted to investigate the antioxidant activities and physiological properties of Euphorbia humifusa extracts prepared using three different solvents (water, ethanol, and methanol). The highest total polyphenol content (293.25 mg/100 g) and total flavonoid content (21.05 mg/100 g) were observed in the methanol extract. The content of substances related to proanthocyanidin were highest in the water extract (8.42 mg/100 g), followed by methanol (5.70 mg/100 g) and ethanol (5.39 mg/100 g) extracts. The DPPH and ABTS radical scavenging activities of the methanol extract were 91.72% and 85.83%, respectively, at 50 mg% concentrations, which were higher than those of the other extracts. The extract reducing power decreased in the following order: ethanol > methanol > water. The methanol extract had relatively high antioxidant activity. The ${\alpha}-glucosidase$ and xanthine oxidase inhibitory activities of the methanol extract at a concentration of 10 mg% were somewhat higher than the other extracts. HMG-CoA reductase inhibitory activity in the water extract was slightly higher than in the methanol and water extracts. These results indicated that Euphorbia humifusa extracts were a high-value food ingredient due to their antioxidant activities and nutritional value.

Cytotaxonomic study of Korean Euphorbia L. (Euphorbiaceae) (한국산 대극속(Euphorbia L., Euphorbiaceae)의 세포분류학적 연구)

  • Somatic chromosomes about 13 taxa of Korean Euphorbia L. was investigated to estimate its taxonomic significance. Somatic chromosome numbers of treated taxa were 2n= 12, 20, 22, 28, 40, 42, 56, therefore basic chromosome numbers of those were x=6, 7, 10, 11. The chromosome numbers of E. pallasii Turcz. (2n=20), E. hylonoma Hand.-Mazz (2n=20.), E. fauriei H. L$\acute{e}$v. & Vaniot ex H. L$\acute{e}$v (2n=28) and E. jolkini Boiss. (2n=28) were determined for the first time in this study. The chromosome numbers of four taxa were same as previous ones; E. sieboldiana Moor. & Decne. (2n=20), E. ebracteolata Hayata (2n=20), E. humifusa Willd. ex Schlecht. (2n=22). But those of six taxa were different; E. esula L (2n= 16, 20, 60, 64 vs 2n=20), E. helioscopia L. (2n=12, 42 vs 2n=42), E. lucorum Rupr. (2n=28, 40 vs 2n=56), E. pekinensis Rupr. in Maxim. (2n=24 vs 2n=28, 56), E. maculata L. (2n=28, 42 vs 2n=12), E. supina Raf. (n=7 vs 2n=40). E. ebracteolata, E. pallasii and E. hylonoma were distingushcd from the other taxa by the chromosome numbers, size and satellites, E. maculata, E. humifusa, E. supina had the different basic and somatic chromosome numbers in spite of the similar morphological. anatomical and palynological chracters. The chromosomal character of Korean Euphorbia was supported the Ma and Hu's systems, and as above results, it was found to be a good character in delimiting above sections and estimating relationships for some species.

Antioxidative Effect of Domestic Plants (국내 유용식물의 항산화 효과)

  • This study was carried out to investigate the antioxidative activities of domestic plants grown in the area of Kangwon-do province. Through the examination of methanol extracts from 15 plants for radical scavenging effects using DPPH method, the extracts from Pinus densiflora Sieb. et Zucc. And Euphorbia humifusa Willd. Showed strong antioxidative activity. Because of its highest antioxidative activity among 15 domestic plants, radical scavenging effects of four different extract compartments from leaves of P.densiflora, n-Hexane, EtOAc, BuOH and $H_2O$ extracts. Were examined by DPPH method. Antioxidative activities of EtOAc and BuOH extracts were similar or even higher than that of natural (tocopherol) or synthetic antioBfdants (BHA) , suggesting that major fraction for the antioxidative activity of P. densiflora was the EtOAc and BuOH extract compartments.
