Effect of Dietary Supplementation of Acanthopanax senticosus and Eucommiaceae on the Expression of Lipogenic, Myogenic and Antioxidant Enzyme Genes in Broiler Chickens (육계에서 가시오갈피와 두충의 첨가 급여가 항산화 효소, 지방 및 근육 관련 유전자 발현에 미치는 영향)

  • This study was carried out to investigate the effect of dietary supplementation of Acanthopanax (A) senticosus and Eucommiaceae on the expression of lipogenic, myogenic and oxidative stress genes in broiler chickens. Birds were subjected (assigned) to one of the following 5 dietary treatments: control (CON), A. senticosus 0.5% (T1), 1.0% (T2), Eucommiaceae 0.5% (T3) and 1% (T4). Each treatment was replicated 8 times with 4 birds per replication, housed in 4 birds per cage. Birds were arranged according to randomized block design. Feeding trial was conducted from day 4 to 35th day of age. Liver and muscle tissues were collected for analysis. Broilers subjected to 1% A. senticosus had higher feed conversion ratio than the other treated birds whereas no significant differences were found in body weight, weight gain and feed intake. The gene expression levels of fatty acid synthase were not different among the treatments while the transcription factor $PPAR{\gamma}$ was highly expressed in Eucommiaceae but not in control and A. senticosus. The gene expression levels of myogenin were high in both A. senticosus and Eucommiaceae compared to control group. MyoD also showed high expression in treated groups furthermore, Eucommiaceae stimulated the expression of MyoD more than that of A. senticosus. The antioxidant gene expressions (SOD, CAT, SOD, GPX) generally were not much different among the treatments, however, SOD and GPX were stimulated in broilers consumed 1% Eucommiaceae diet. The result of this experiment showed that dietary supplementation of A. senticosus and Eucommiaceae in broiler may improve the antioxidant defence system through SOD and GPX without affect of growth performance in broilers.

Standardization and Seasonal Variation of Quercetin Glycoside in Eucommiae Folium (두충엽 함유 퀘르세틴배당체의 함량 표준화 및 계절적 변화)

  • The radix and folium of Eucommia ulmoides(Eucommiaceae) has been used for backache, atrophy of the leg and knee, enuresis, hypertension. In order to evaluate to quality of folium, we have been isolated a main compound as a standard compound, which was elucidated $quercetin-3-O-{\beta}-D-xylopyranosyl(l{\rightarrow}2)-{\beta}-D-glucopyranoside$ through it's physicochemical data and IR, FAB-Mass, $^{13}C-NMR$ and $^{1}H-NMR$ analysis. It was analyzed by HPLC system using 17% $CH_3CN$ as a solvent system. The amount of $quercetin-3-O-{\beta}-D-xylopyranosyl(l{\rightarrow}2)-{\beta}-D-glucopyranoside$ from Eucommiae folium was in the range of $0.056{\pm}0.022$ mg(n=7) and also from the results of analysis through seasonal variation$(June{\sim}October)$ September and October have been evaluated to be very high in it's content.

  • The pharmacological activities on water extracts of Cortex, Ramulus and Folium from the Eucommia ulmoides Oliver were studied, and the following results were observed. The vasodilative action on the rabbit ear blood vessel was recognized in the Cortex, Ramulus and Folium. The spontaneous motility and the contraction induced by Ach., $BaCl_2$ and histamine in the isolated ileum of mice were suppressed and inhibited in the Ramulus only. On the contrary, the analgesic and anti-inflammatory activities were noted in the Cortex and Folium. Furthermore, the hypotensive effect was recognized in the Folium. The diuretic, bile secretory and anti-fatigue effects were shown in the Cortex and Folium.

  • Geniposide, an iridoid glucoside, was isolated from the cortex of Eucommia ulmoides by repealed silica gel chromography and identified by its spectral data. The quantitative determination of geniposide with HPLC has been developed and this method provided a tool for standardization of the commercial Eucommiae Cortex.

Inhibitory effects of the stem bark extract of Eucommia ulmoides on the proliferation of human tumor cell lines

  • A bioassay-guided fractionation of the stem bark extract of Eucommia ulmoides Oliver (Eucommiaceae) led to the isolation of three iridoid constituents, genipin (1), geniposide (3), geniposidic acid (4) as well as (${\pm}$)-guaiacylglycerol (2) and fatty acid mixtures as active ingredients of the extract responsible for the antitumoral property. The EtOAc soluble part and BuOH soluble part of the extract demonstrated a significant inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT-15 (colon) in vitro, whereas the remaining water soluble part exhibited a poor inhibition.   • The anti-diabetic effects of the methanol extracts of Cortex, Ramulus and Folium part of Eucommia ulmoides Oliver were investigated by the experimental diabetic model in rats. The increased blood glucose level in the diabetic rats induced by alloxan and streptozotocin was significantly lowered and a marked increase in body weight was shown with the treatments of the extracts of Cortex and Folium. The blood insulin level lowered by streptozotocin pretreatment was significantly recovered with the treatment of the extract of Folium. In glucose tolerance test, the extracts of Cortex and Folium displayed a significant inhibitory effect on the glucose level. The extracts of Cortex and Folium also caused a marked inhibition in increased blood pressure in the diabetic rats induced by alloxan.
