Biological activity of water-soluble polysaccharides from Cedrela sinensis according to extraction methods (참죽의 추출방법에 따른 수용성 다당류의 생리기능성)

  • The biological activity of water-soluble polysaccharide (WSP) fractions extracted from Cedrela sinensis was examined in this study. Cedrela sinensis was extracted using hot water, ultrasonication, and enzymes (Viscozyme, Shearzyme) and precipitated using ethanol to produce crude polysaccharides. The yield (3.51%) and total polysaccharide content (28.03 g/100 g) of WSP extracted using Shearzyme (WSPs) were highest compared to other extracts. The antioxidant activity of WSP extracted using hot water was highest and had the lowest $IC_{50}$ values in DPPH, ABTS, hydroxyl radical scavenging activity, reducing power, and superoxide dismutase-like activity. Tyrosinase inhibitory activity increased as the concentration increased. All extracts showed higher retardation effects on glucose and bile acid compared to the control; particularly, WSPs showed a similar glucose retardation effect to carboxymethyl cellulose. This study suggests that WSP from C. sinensis can be used as a functional food material.

Preparation and Characterization of Phytochemical-Rich Extract from Sasa quelpaertensis Leaf (식물화합물 다량 함유 제주조릿대 잎 추출물의 제조와 특성)

  • Sasa species leaves have been used in traditional medicine for their anti-inflammatory, antipyretic, and diuretic properties. Sasa quelpaertensis Nakai is a small bamboo grass that grows only on Mt. Halla on Jeju Island, Republic of Korea. This small bamboo grass has recently been the focus of much attention due to its potential biomass as well as its beneficial health effects. In this study, to promote the efficient utilization of the S. quelpaertensis leaf, we established a simple preparation method for phytochemical-rich extract (PRE) by comparing phytochemical contents and biological activities according to extraction methods. high performance liquid chromatography (HPLC) analysis revealed that the contents of two major phytochemicals such as, tricin (5.35 mg/g) and p-coumaric acid (44.10 mg/g) contained in PRE were higher than those in fresh hot water extract (SQH, p-coumaric 23.39 mg/g, tricin 0.18 mg/g) and ethanol extract (SQE, p-coumaric 10.8 mg/g, tricin 0.38 mg/g). The antioxidant activities [1,1-diphenyl-2-picrylhydrazy (DPPH) radical scavenging activity, 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging activity, nitric oxide (NO) scavenging activity, and xanthine oxidase inhibitory activity] of PRE were higher than those of SQH and SQE. PRE effectively inhibited NO production in LSP- stimulation RAW 264.7 cells, and the growth of human promyelocytic leukemia (HL-60) cells. These results suggest that PRE has a potential as a promising antioxidant and anti-inflammatory agent.

Phenolics Content and Browning Capacity during the White Winemaking (백포도주 양조중 페놀류의 함량과 갈변도)

  • Juices of Golden Queen, Neo-Muscat and Seibell-9110 were prepared with the addition of $SO_2(100ppm)$ in must and juice, of $SO_2(100ppm)$ and pectinase (40mg/l) in must, respectively and then 300mg/1 of Polyclar AT were added on different white wines from grape juices. Phenolics content in grape juices and white wines were determined. and also changes of browning capacity of white wines during the storage periods at $45^{\circ}C$ were investigated by accelerated method. Extraction of total phenol into grape juices was increased significantly in $SO_2-pectinase$ addition lot and content of total phenol of Seibell-9110 juice was two or three times higher than the others. Yield of grape juices was highest in $SO_2-pectinase$ addition lot. Ethanol content of white wines were 11.4-12.0 v/v% , and total acid, fusel oil and methanol content of Neo-Muscat white wine was lower than that of the other white wines. Total phenol content of white wines was 25-50% lower than that of grape juices and 50-80% lower in colour. Browning capacity of white wines was decreased by 25-40% for 18 days at $45^{\circ}C$ with addition of Polyclar AT, but Seibell-9110 white wine prepared from $SO_2-pectinase$ addition lot was not available commercially because of severe browning.

Antioxidative Activity and Chemical Characteristics of Leaf and Fruit Extracts from Thuja orientalis (측백나무 잎.열매 추출물의 이화학적 특성 및 항산화 효과)

  • The contents of bioactive materials (e.g. polyphenolics compounds, flavonoids, minerals, and fatty acids) and antioxidative activities (DPPH (${\alpha}$,${\alpha}$'-diphenyl-${\beta}$-picrylhydrazyl) free radical scavenging activity, peroxidation of linoleic acid and rat hepatocyte microsome, and Fe/Cu reducing power) were tested by in vitro experimental models using water, ethanol and methanol extracts of leaves (TOL) and fruits (TOF) from Thuja orientalis. Methanol extract from TOL showed the highest extraction yield (12.90%) as well as contents of polyphenolic compounds (16.02%) and flavonoids (0.25%). Major minerals were Ca, K, and Mg. Major fatty acids were palmitic and lauric acids in TOL and palmitic and decanoic acids in TOF. In oxidation of in vitro models using DPPH free radical scavenging activity, Fe/Cu reducing power, $Fe^{2+}$/ascorbate-induced linolenic acid peroxidation by ferric thiocyanate and thiobarbituric acid (TBA) methods, and autooxidation of rat hepatic microsomes membrane, anti-oxidative activities were stronger in all extracts of TOL than in those of TOF in a dose-dependent manner. From these results, methanol extract of TOL was shown to have the most potent anti-oxidative properties and the highes content of antioxidative compounds such as polyphenolic compounds and flavonoids.

Experimental Research on the Effect of Haeyeol-tang (해열탕(解熱湯)의 항염증(抗炎症), 항산화(抗酸化), 항노화(抗老化) 작용(作用)에 대한 실험적(實驗的) 연구(硏究))

  • Objectives : The purpose of this study was to examine the effect of Haeyeol-tang on infectious disease of respiratory system. Methods : Haeyeol-tang was used by cold storage after dilution with 1, 10 and $100{\mu}g/ml$ of PBS, from dried material as extracted material with extraction with 50% ethanol. Statistical significance was examined through measuring TNF-$\alpha$, RT-PCR, DPPH, nitric oxide(NO), and MTT of Raw264.7 cells activated with LPS. Results : For the toxicity experiment of Haeyeol-tang, almost no toxicity was shown on cells in the concentration ranges of 1, 10 and $100{\mu}g/ml$. For the effect of NO production, Haeyeol-tang showed significant (p<0.001) decrease of NO production, according to its concentration. For the effect on the eliminating activity of DPPH radical, Haeyeol-tang showed increasing tendency of eliminating activity of DPPH radical, according to its concentration. For the effect on revelation of NOS2 enzyme, Haeyeol-tang showed inhibiting tendency against the revelation of NOS2, according to its concentration. For the effect on the TNF-$\alpha$ production, Haeyeol-tang showed significant (p<0.001) decrease of TNF-$\alpha$ production. Conclusions : This result means that Haeyeol-tang is effective for anti-inflammation, as well as excellent effect for anti-oxidation and anti-aging.

Eficacy and Sfety Human Study of the Ethanol Extracts from Lythrum Salicaria L. and Aceriphyllum Rossii for Reducing Body Fat Percentage of Korean Adult (털부처꽃과 돌단풍 에탄올 추출물의 한국 성인 체지방량 감소에 대한 유효성 및 안전성에 관한 인체적용시험)

  • This research, as a basic preliminary study for development of functional health food, is aimed at assessing the body fat reduction effect and for application to human body for such reduction in actual clinical settings by preliminary extraction of 2 types of wild edible greens, Lythrum salicaria L. and Aceriphyllum rossii. Subjects over the age of 19 and less than 60 years old with BMI value range of $23.0kg/m^2{\sim}29.9kg/m^2$ were recruited through screening were divided into experimental group and control group, each with 25 subjects, through randomized allocation. With both patients and evaluators wearing blindfold, the experimental group was orally administered with 4 capsules of 500mg of composite preparation containing the extracts of Lythrum salicaria L. and Aceriphyllum rossii 3 times a day for a period of 8 weeks while the control group was orally administered with 4 capsules of 500mg of placebo (fake food) with the same appearance as the preparation administered to the experimental group 3 times a day for a period of 8 weeks. After having carried out evaluation on physical examinations (body weight, BMI and body fat ratio, etc.), laboratory tests (general blood test, biochemical test of blood and urine test), lipid test, the changes were analyzed. There was no significant change between the 2 groups and within the groups in BMI and body fat ratio, which are the primary effectiveness evaluation at each time. There was no significant difference between the 2 groups in serum lipid and WHR, obesity related KOQOL, KEAT-26 which are the secondary effectiveness evaluations. There was no change between the 2 groups and within the groups in vital sign, CBC, BC and urin test. These results suggest that Lythrum salicaria L. and Aceriphyllum rossii ext. showed no significant reduction in BMI, body fat ratio and serum lipid. Additional confirmative clinical application test is needed in the future.

Evaluation of the Wound-healing Activity of Rice Cell Extracts in Vitro (In vitro 실험을 통한 벼세포 추출물의 창상 치유 효능 평가)

  • In the present study, we evaluated the in vitro wound-healing properties of two types of rice cell extracts (RCEs; prepared using ethanol and pressurized hot water extraction methods), using human dermal fibroblasts and keratinocytes. The effects of the RCEs (at 25–100 μg/ml) on cytotoxicity and cell migration were assessed. Both RCEs were not cytotoxic to the two cell types, instead increasing their proliferation by up to 25% in a dose-dependent manner compared with the controls. Furthermore, both RCEs significantly enhanced the migratory ability of the two cell types (fibroblast, 230–450%; keratinocyte, 170–350%). Additionally, we examined the effect of the RCEs on type I collagen synthesis, which is important in the wound reconstruction process. The RCEs significantly increased collagen type I mRNA and protein levels to a degree comparable to that induced by vitamin C. These results suggest the RCEs to be candidate materials for use in promoting wound healing, through their actions of increasing cell migration and accelerating wound re-epithelialization.

Identification of a Bioactive Compound, Violacein, from Microbulbifer sp. Isolated from a Marine Sponge Hymeniacidon sinapium on the West Coast of Korea (한국 서해안에 서식하는 주황해변해면에서 분리된 해양세균 Microbulbifer sp.으로부터 생리활성물질 비올라세인의 규명)

  • Microbial secondary metabolites of marine organisms are regarded as major sources of structurally and biologically novel compounds with numerous potential uses. Sponge-microbe associations are among the most interesting sources for exploring bioactive compounds. In this study, the bacterial strain Microbulbifer sp. (127CP7-12) was isolated from the Asian marine sponge Hymeniacidon sinapium collected at an intertidal zone on the west coast of Korea. Cultured bacteria produced a violet pigment, and optimal culture conditions for violet pigment production were investigated. Maximum production of the violet pigment from the strain culture was observed under the conditions of $25^{\circ}C$, pH 6.0, and 3% NaCl. Acetone provided better extraction of the pigment from fermented broth compared with ethanol and methanol. The proposed structure of the major component in the extracted crude pigment was determined via high-performance liquid chromatography, nuclear magnetic resonance, mass spectrometry, and UV spectra analyses, which showed that the metabolite was the promising bioactive compound violacein. This study describes the examination of marine bioactive materials from microbe-engaged metabolites and the ecological implications of the sponge-microbe association in a changing ocean.

Optimization of KOH pretreatment conditions from Miscanthus using high temperature and extrusion system (고온 압출식 반응시스템을 이용한 억새 바이오매스의 KOH 전처리조건 최적화)

  • The purpose of this study is to investigate the optimum conditions of biomass pretreatment with potassium hydroxide (KOH) for efficient utilization of cellulose, hemicellulose and lignin from Miscanthus. The optimization of variables was performed by response surface methodology (RSM). The variation ranges of the parameters for the RSM were potassium hydroxide 0.2~0.8 M, reaction temperature 110~190℃ and reaction time 10~90 min. The optimum conditions of alkali pretreatment from Miscanthus were determined as follows: concentration of KOH 0.47 M, reaction temperature 134℃ and reaction time 65 min. At the optimum conditions, the yield of cellulose from the solid fraction after pretreatment was predicted to be 95% by model prediction. Finally, 66.1 ± 1.1% of cellulose were obtained by verification experiment under the optimum conditions. The order contents of solid extraction were hemicellulose 26.4 ± 0.4%, lignin 3.7 ± 0.1% and ash 0.5 ± 0.04%. The yield of ethanol concentration of 96% was obtained using separated saccharification and fermentation.

Antioxidant, Anti-wrinkle and Whitening Effect of Fermented Extracts of Hwangryunhaedoktang (황련해독탕 발효물의 항산화, 항주름 및 미백 효과)

  • Hwangryunhaedoktang (HHT) has been traditionally used as a preventive and therapeutic medicine to treat enervation and diverse chronic diseases. This study was designed to compare the antioxidant, anti-wrinkle and whitening effects of HHT extract and its fermented extract by Leuconostoc mesenteroides (FHHT). FHHT was prepared by inoculation of L. mesenteroides after the extraction procedure with 70% ethanol. HHT and FHHT was investigated via high-performance liquid chromatography (HPLC). Simultaneous qualitative analysis of two bioacitive components, berberine and palmatine. was achieved by comparing their retention times ($t_R$) and UV spectra with those of the standard components. Cell viability test results indicated that both HHT and FHHT were non-toxic. In DPPH radical scavenging ability, $SC_{50}$ values of the FHHT was $68.85{\mu}g/mL$, which is more effective than HHT. Moreover, FHHT showed higher expression in production of procollagen type I than HHT. In nontoxic concentration range, FHHT showed strong melanin production inhibitory effect in ${\alpha}-melanocyte$ stimulating hormone (${\alpha}-MSH$)-stimulated B16F10 cell ($IC_{50}=9.82{\mu}g/mL$). These results suggested that fermented extracts of hwangryunhaedoktang had considerable potential as a cosmetics ingredient with an antioxidant and anti-wrinkle and whitening effects.
