4가지 한방 소재(행인, 호장근, 자초, 강황)의 화장품약리활성에 관한 연구 (Study on Cosmeceutical Activity of four Kinds of Korea Herb Medicine Materials)

  • The aim of this study was to assess the cosmeceutical activies of four kinds of Korea herb medicine extracts using in cosmetics and related industries. The cosmeceutical activities of extracts were investigated by tyrosinase inhibition, astringent, anti-bacterial and MTT assay for cell viability. In the whitening effect, PA that the highest tyrosinase inhibition activity showed 56% at 10 ppm in ethanol extract. Also water and ethanol extract of RE showed 54%, 68% at 1,000 ppm, respectively, but LE and CA showed lower effect. Astringent effect of water and ethanol extract of PA appeared over 60% at 1,000ppm concentration but other extracts showed no astringent effect. In the anti-bacterial test, water and ethanol extract of PA showed no anti-bacterial effect against all microorganisms. But water and ethanol extract of RE showed anti-bacterial effect on Staphylococcus aureus, ethanol extract of showed on Staphylococcus aureus and Propionibacterium acnes, ethanol extract of CA showed on Candida albicans. The resUlt of stability test showed that the emulsion of containing PA were very stable at various temperature and sun-light test. Viscosity and pH of emulsion did not change. From the results of human patch test to assess the safety of cosmetics containing PA there was no negative reaction on skin was found.

도라지 잎 에탄올 추출물의 주요 성분 분석 및 마우스 대식세포와 인체 폐암세포에서 항염효과 (Analysis of Major Constituents of an Ethanol Extract of Platycodon Grandiflorum Leaves and Protective Effects on Inflammation in Murine Macrophage and Human Lung Carcinoma Cells)

  • This study investigated major constituents and anti-inflammatory effects of an ethanol extract of Platycodon grandiflorum leaves. Through HPLC analysis, chlorogenic acid and luteolin-7-O-glucoside were identified as predominant constituents in the ethanol extract. Their anti-inflammatory effects were evaluated using murine macrophage (RAW 264.7 cells) and human lung carcinoma cells (NCI-H292 & A549). The ethanol extract significantly (p<0.01) inhibited the production of nitrite, interleukin-6 (IL-6), and prostaglandin E2 (PGE2) induced by lipopolysaccharide (LPS) in RAW 264.7 cells. Furthermore, the ethanol extract suppressed the expression of cyclooxygenase-2 (COX-2) and inducible NO synthase (iNOS) proteins in RAW 264.7 cells stimulated with LPS. In NCI-H292 and A549 cells, treatment with the ethanol extract significantly (p<0.05) decreased levels of pro-inflammatory cytokines IL-6 and IL-8 induced by IL-1β. The phosphorylation of ERK rather than JNK in the mitogen-activated protein kinase signaling pathway was observed to be a more important mediator in the down-regulation of pro-inflammatory cytokines in NCI-H292 cells. These findings suggest that the ethanol extract of Platycodon grandiflorum leaves containing luteolin-7-O-glucoside exhibits promising anti-inflammatory properties.

Inhibitory Effect of Corn Silk Extract on Growth of Food-Borne Bacterial Pathogens

  • Various levels of antibacterial activity have been identified for water and ethanol extracts of corn silk, particularly against Salmonella typhimurium KCTC 2515. In general, the water extract was more effective than the ethanol extract. The minimum inhibitory concentration (MIC) for the water extract was 7.5 mg/disc for S. typhimurium KCTC 2515 and B. cereus KCTC 1092, as well as for the ethanol extract against S. typhimurium KCTC 2515 and S. typhimurium KCTC 1925. However, the MICs for the water extract were lower than those for the ethanol extract against all bacteria tested, except S. typhimurium KCTC 1925 and B. cereus KCTC 1014. The growth of the tested organisms in the synthesized broth medium was inhibited with the addition of 5-fold levels of MIC. Using sterilized milk as the model food system, we found that the lag phase for these microorganisms was extended up to 3 days at $20^{\circ}C$, but was not affected at $4^{\circ}C$. These results indicate that bacterial growth was strongly inhibited by corn silk extract at $20^{\circ}C$.

유근피 추출물의 생리활성 (Physiological Activities of $Ulmus$ $pumila$ L. Extracts)

  • 본 연구는 느릅나무과(Ulmaceae)에 속하는 유근피(Ulmuspumila L.)추출물을 기능성식품에 활용하기 위하여 폴리페놀 함량, 항산화능 및 암세포 증식 억제능의 기능성에 대하여 조사하였다. 물 추출물 및 에탄올 추출물의 수율은 각각 12.7%와 12.0%였으며, polyphenol 화합물의 함량은 $623.5{\pm}2.4$ mg/100g 및 $710.5{\pm}2.1$ mg/100g이었고, 전자공여능은 농도에 비례하여 높아, 1,000 ppm에서 물추출물은 76%, 에탄올 추출물은 64%로 물 추출물이 에탄올 추출물 보다 높았다. SOD유사활성능은 전 농도에서 물 추출물이 에탄올 추출물보다 높아 1,000 ppm에서는 물 추출물이 53%, 에탄올 추출물이 38%이었고, 아질산염 소거능은 1,000 ppm에서 에탄올 추출물이 47%, 물 추출물이 43%로 에탄올 추출물이 물 추출물보다 다소 높았다. 항암성은 MDA cell에서 1,000 ppm의 경우 물 추출물은 62%, 에탄올 추출물은 42%이었으며, A549 cell에서 1,000 ppm의 경우 물 추출물은 60%, 에탄올 추출물은 45%로 물 추출물이 에탄올 추출물보다 암세포증식 억제력이 월등히 높았다.

복분자 에탄올 추출물의 식중독 관련 위해 세균에 대한 항균활성 분석 (Antimicrobial Activity of the Ethanol Extract from Rubus coreanum against Microorganisms Related with Foodborne Illness)

  • This study analyzed the antibacterial activity of a Rubus coreanum (Bokbunja) ethanol extract. The antimicrobial activity was determined by disc diffusion, minimum inhibitory concentration (MIC), and growth inhibition methods with seven kinds of bacteria related to foodborne illness (Bacillus cereus, Escherichia coli, Escherichia coli O157:H7, Listeria monocytogenes, Pseudomonas aeruginosa, Staphylococcus aureus, and Salmonella typhimurium). In the results, disc diffusions of the ethanol extract from R. coreanum (9.8-17.5 mm at $4,000{\mu}g/disc$) clearly showed the antimicrobial activity of the extract against all tested microorganisms. Rubus coreanum promoted an inhibitory effect as follows: E. coli O157:H7 > P. aeruginosa > L. monocytogenes > E. coli > S. aureus > B. cereus ${\geq}$ S. typhimurium. In the MIC test, R. coreanum showed high antimicrobial effect against L. monocytogenes at 500 ppm. Moreover, the R. coreanum ethanol extract showed strong growth inhibition against microorganisms, similar to the MIC results. These results show that a R. coreanum ethanol extract has powerful antimicrobial activity against all tested microorganisms, suggesting that R. coreanum will be useful as a potential natural preservative.

매생이 추출물의 생리 활성과 항산화 활성에 관한 연구 (A Study on Physiological Activity and Antioxidative Activity of Maesangi(Capsosiphon fulvescens) Extract)

  • Physiological activity and antioxidative activity of Maesangi(Capsosiphon fulvescens) extracts with distilled water or 95% ethanol were investigated. For the evaluation of physiological and antioxidative activities, some evaluation assay methods such as measurement of Hunter color value, chlorophyll a/b value, total phenolics, reducing power and thiobarbituric acid(TBA) value of soybean oil were used. Proximate composition and mineral contents of Masaengi were orders of crude protein>crude fiber>moisture>crude ash>crude lipid, and K>Ca>Mg>Na>P>Fe>Zn, respectively. In ethanol extract, the content of total phenolic compounds in Maesangi was determined to half times of that in Dasima(Laminaria). The reducing power of Maesangi-ehtanol extract was about 5 % of vitamin C and was lower than that of Dasima-ethanol extract. The TBA value of Maesangi-ethanol extract on soybean oil oxidation was about 47 % and 68.4 % to control in three and eight days oxidation, respectively. But TBA value difference was not observed significantly with the dosage below 5mL of ethanol extract.

냉이(Capsella bursa-pastoris) 에탄올 추출물의 유리라디칼 소거 및 항산화 활성 (Free Radical Scavenging and Antioxidative Activities by Ethanol Extract from Capsella bursa-pastoris)

  • Screening was performed on edible natural sources to examine superoxide radical scavenging activities by using the method of superoxide dismutase assay. Among 47 kinds of samples, the extract of Capsella bursa-pastoris showed a potent superoxide radical scavenging activity of 11.6 unit / mg solid and was selected for further studies. In order to select optimal extraction solvent system, Cappella bursa-pastoris was extracted with various solvents and the electron donating abilities and inhibitory effects on lipid peroxidation of linoleic acid were measured. Among them, the ethanol extract of Capsella bursa-pastoris possessed the highest level of activities. The ethanol extract of Cappella bursa-pastoris was found to have an inhibitory effect on autoxidation of soybean oil at 5$0^{\circ}C$ from peroxide value, conjugated diene value and refractive index. In the soybean oil containing 0.2% of ethanol extract, induction period was increased 2 times in comparison with control.

뽕잎 추출물의 돌연변이 억제효과 (Antimutagenic Effect of Mulberry Leaf Extract)

  • Background: The present study was carried out to asses whether mulberry leaves (MLs) have the potential to inhibit the mutagenic effect of cigarette smoke condensates (CSCs). Methods and Results: ML powder was extracted with 70% ethanol, and a yield of 35.1% by weight was obtained. The 70% ethanol extract of ML was further extracted sequentially using diethyl ether, chloroform, butanol, dichloromethane and water. The crude 70% ethanol extract of MLs and its solvent fractions did not show any mutagenic effect when tested at concentrations up to 1 mg/plate against Salmonella typhimurium TA98. In contrast, the crude 70% ethanol extract showed an inhibitory activity against the mutagenicity of CSCs in the presence of S-9 mixture. Among the solvent fractions, the diethyl ether fraction showed the highest inhibitory activity, which increased in a dose-dependent manner, inhibiting mutagenesis by approximately 97.1% at a concentration of 1 mg/plate. Conclusions: In this study, we found that a crude 70% ethanol extract of MLs and the diethyl ether fraction themselves are potentially not mutagenic, but inhibit the mutagenic effect of CSCs.

Aucklandia lappa Causes Membrane Permeation of Candida albicans

  • Candida albicans is a major fungal pathogen in humans. In our previous study, we reported that an ethanol extract from Aucklandia lappa weakens C. albicans cell wall by inhibiting synthesis or assembly of both (1,3)-β-D-glucan polymers and chitin. In the current study, we found that the extract is involved in permeabilization of C. albicans cell membranes. While uptake of ethidium bromide (EtBr) was 3.0% in control cells, it increased to 7.4% for 30 min in the presence of the A. lappa ethanol extract at its minimal inhibitory concentration (MIC), 0.78 mg/ml, compared to uptake by heat-killed cells. Besides, leakage of DNA and proteins was observed in A. lappa-treated C. albicans cells. The increased uptake of EtBr and leakage of cellular materials suggest that A. lappa ethanol extract induced functional changes in C. albicans cell membranes. Incorporation of diphenylhexatriene (DPH) into membranes in the A. lappa-treated C. albicans cells at its MIC decreased to 84.8%, after 60 min of incubation, compared with that of the controls, indicate that there was a change in membrane dynamics. Moreover, the anticandidal effect of the A. lappa ethanol extract was enhanced at a growth temperature of 40℃ compared to that at 35℃. The above data suggest that the antifungal activity of the A. lappa ethanol extract against C. albicans is associated with synergistic action of membrane permeabilization due to changes in membrane dynamics and cell wall damage caused by reduced formation of (1,3)-β-D-glucan and chitin.

추출 방법에 따른 감길탕가미방(甘桔湯加味方)의 항염증(抗炎症), 진통(鎭痛), 해열(解熱), 항산화(抗酸化) 및 Histamine 투과 억제효과 (Anti-inflammatory Effects, Analgesic Effects, Antipyretic Effects, Anti-oxidative Effects, and Histamine Permeation Inhibition Effects of Modified Gamgil-tang per Extraction Method)

  • Objectives : Modified Gamgil-tang is a prescription commonly used for respiratory diseases. This thesis was carried out to check the treatment effects and diversity of drug formulation by comparing extraction method of ethanol and water of modified Gamgil-tang. Methods : All experiments were carried out with water and 50% ethanol extraction for comparison. In vivo experiment, hyaluronidase inhibitory effects and trypsin inhibitory effects were tested to measure the anti-inflammatory effects activity. Scavenging effects of DPPH free radical, xanthine oxidase inhibitory effects and inhibition on TBA-RS formation were experimented to measure anti-oxidative effects. With the in vivo experiment, ICR group mice and SD group rats were used as experimental animals. An anti-inflammatory effects experiment were carried out to measure the action on carrageenin-induced hind paw edema: analgesic effects were measured using writhing syndrome induced by 0.7% acetic acid in mice: antipyretic effect was measured using endotoxin, and inhibitory effects of increase vascular permeability induced by 0.5% histamine were measured. Results : For extraction of glycyrrhizin contents, ethanol extract was extracted 2 times of that of water extract. Anti-inflammatory effects showed high in ethanol extract. Anti-oxidative effects measured high in ethanol extract. No significant result was found in inhibition on TBA-RS formation. Analgesic effects were found to be similar in water and ethanol extract. Antipyretic effects were found to be stronger in water extract. Inhibitory effects of increase vascular permeability induced by 0.5% histamine showed stronger in ethanol extract. Conclusion : By measuring anti-inflammatory effects, analgesic effects, antipyretic effects, anti-oxidative effects, and histamine permeation inhibition effects both in water extract and ethanol extract after adding agents such as Mentha Herba, Gardenias Fructus, and propolis to existing Gamgil-Tang, ethanol extract was found to be more effective in anti-inflammatory effects, analgesic effects, anti-oxidative effects, and histamine permeation inhibition effects. The converse was found for antipyretic effect.
