Antioxidant and Antimicrobial Effects of Extract with Water and Ethanol of Oriental Melon (Cucumis melo L. var makuwa Makino) (참외 (Cucumis melo L. var makuwa Makino)의 물과 에탄올 추출물의 항산화 및 항균효과)

  • The biological activities of water and ethanol extracts from different fruit parts, such as peel, flesh, and placenta of oriental melon were investigated. The total phenolic concentration of water extract was the highest such as 151.64 ${\mu}g/g$ in the peel, also that of ethanol extract was 224.77 ${\mu}g/g$ in the peel, respectively. The total flavonoid content in the water and ethanol extracts were high such as 45.53 ${\mu}g/g$ and 67.16 ${\mu}g/g$ of peel, respectively. In the physiological activities, DPPH in the water and ethanol extracts were high such as 25.0% and 83.3% of peel in 1% concentration. Extract of peel was higher than those of flesh and placenta. ABTS in the water extracts was 79.2% of peel, 57.6% of flesh and 74.0% of placenta in 1% concentration. Ethanol extracts was 99.9% of peel, 52.1% of flesh and 41.2% of placenta in 1% concentration. In addition, xanthine oxidase inhibitory activity and ${\alpha}$-Glucosidase inhibition activity of the peel of water and ethanol extracts appeared to be higher than those of placenta and flesh. This study showed that the antioxidant and ${\alpha}$-Glucosidase inhibition activity of peel extracts were higher than those of placenta and flesh. Also, the antimicrobial effect of ethanol extract from different fruit parts was shown only on Streptococcus agalactiae.

Effects of Dietary Supplementation of Bacteriophage on Growth Performance, Nutrient Digestibility, Blood Profiles, Carcass Characteristics and Fecal Microflora in Broilers (육계 사료 내 Bacteriophage의 첨가가 생산성, 영양소 소화율, 혈액 특성, 도체 특성 및 분내 미생물 조성에 미치는 영향)

  • This experiment was conducted to investigate the effects of bacteriophage SE supplementation on growth performance, nutrient digestibility, blood profiles, visceral organ weight, meat quality and excreta microflora in broilers. A total of 340 1-d-old ROSS 308 broilers (mixed gender) with an initial average body weight (BW) of $41.71{\pm}0.16$ g were randomly allotted to 4 treatments with 5 replicate pens per treatment and 17 broilers per pen for 31 days. Dietary treatments were: 1) CON, control diet, 2) SE05, CON+0.05% bacteriophage, SE 3) SE10, CON+0.10% bacteriophage SE, and 4) SE15, CON+0.15% bacteriophage SE. During d 15 to 31, broilers fed SE15 diet had a higher (P<0.05) body weight gain than broilers fed CON diet. Overall, body weight gain in SE10 and SE15 was greater (P<0.05) than that in CON. Apparent total tract nutrient digestibility and blood characteristics did not differ (P>0.05) among treatments. The water holding capacity was increased (P<0.05) in SE15 compared with CON. Other meat quality in terms of pH value, breast muscle color ($L^*$, $a^*$, $b^*$) and drip loss were unaffected by dietary supplementation with bacteriophage SE. The visceral weight of bursa of Fabricius was increased (P<0.05) in broilers fed the bacteriophage SE incorporated diets compared with those fed the CON diet. No difference (P>0.05) was observed in visceral weight of liver, spleen, breast muscle, abdominal fat, gizzard and excreta concentrations of Lactobacillus, Clostridium perfringens, Escherichia coli, and Salmonella. In conclusion, dietary supplementation with 0.10 and 0.15% bacteriophage SE could improve the growth performance, breast muscle water holding capacity and bursa of Fabricius visceral weight in broilers.

Among them, compounds of benzylamine and p-toludine showed an effective antimibrobial activity againt Bacillus subtilis and Pseudomonas ovalis.

  • Thirty-three Mannich bases of 2,2'-methylene bis (3,4, 6-trichlorophenoxy-acetic acid) were synthesized hexachlorophene as potential antimicrobial agents and tested against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Trichophyton rubrum, Microsporum gypseum, Epidermophyton floccosum, Aspergillus niger and Aspergillus oryzae in vitro. Comparison of Quality Characteristics of Sesame leaf Cleaned with Various Electrolyzed Water during Storage (다양한 전기분해수 세정처리에 따른 깻잎의 저장중 품질특성 비교)

  • This study was carried out to investigate the cleaning effect of sesame leaf, the sterilization effect and physicochemical properties, treated with various electrolyzed water. Initial physicochemical properties could be kept more than 1 month in electrolyzed oxidizing water(EW-1) of diaphragm type and 15 days in electrolyzed water(EW-2 and EW-3) of non-diaphragm system, there was no significant difference by storage temperature. 4 kinds of microorganism (initial total counts, $10^7\~10^9$ CFU/mL) were sterilized within $0.5\~1$ minutes by electrolyzed water. In fresh sesame leaves, total viable cell count and coliform group in the treatment of electolyzed water were decreased to about $2\~3$ log scale comparing non-treated ones. Especially Bacillus cereus was not detected until 13th day when treated with EW-l. Decaying ratio of sesame leaf appears on day 6 of storage in the untreated but the treatments of electrolyzed water has no sign until day 10 of storage. Change in color difference(${\Delta}E$) during storage was observed the treatments of electrolyzed low-alkaline water(EW-2) and electrolyzed neutral water(EW-3) were very desirable at the level $1\~2$ after day 13 of storage comparative to the untreated Change of Chlorophyll content was biggest decreased to 6.8 $mg\%$ on the untreated and decreased least to 8.35 $mg\%$ on EW-3 treated group on 13th day from initial value of $9.0\~10.3\;mg\%$ The overall sensory evaluation appeared most acceptable in the treatments of EW-2 and EW-3.

Adaptive Response to ionizing Radiation Induced by Low Doses of Gamma Rays in Human Lymphoblastoid Cell Lines (인체임파양세포에서 저선량의 감마선에 의해서 유도되는 적응 반응)

  • When cells are exposed to low doses of a mutagenic or clastogenic agents. they often become less sensitive to the effects of a higher dose administered subsequently. Such adaptive responses were first described in Escherichia coli and mammalian cells to low doses of an alkylating agent. Since most of the studies have been carried out with human lymphocytes, it is urgently necessary to study this effect in different cellular systems. Its relation with inherent cellular radiosensitivity and underlying mechanism also remain to be answered. In this study, adaptive response by 1 cGy of gamma rays was investigated in three human lymphoblastoid cell lines which were derived from ataxia telangiectasia homozygote, ataxia telangiectasia heterozygote, and normal individual. Experiments were carried out by delivering 1 cGy followed by 50 cGy of gamma radiation and chromatid breaks were scored as an endpoint. The results indicate that prior exposure to 1 cGy of gamma rays reduces the number of chromatid breaks induced by subsequent higher dose (50 cGy), The expression of this adaptive response was similar among three cell lines despite of their different radiosensitivity. When 3-aminobenzamide, an inhibitor of poly (ADP-ribose) polymerase, was added after 50 cGy, adaptive responses were abolished in all the tested cell lines. Therefore it is suggested that the adaptive response can be observed in human lymphoblastoid cell lines, which was first documented through this study. The expression of adaptive response was similar among the cell lines regardless of their radiosensitivity. The elimination of the adaptive response by 3-aminobenzamide is consistent with the proposal that this adaptive response is the result of the induction of a certain chromosomal repair mechanism.

Binding Mode Analysis of Bacillus subtilis Obg with Ribosomal Protein L13 through Computational Docking Study

  • Introduction: GTPases known as translation factor play a vital role as ribosomal subunit assembly chaperone. The bacterial Obg proteins ($Spo{\underline{0B}}$-associated ${\underline{G}}TP$-binding protein) belong to the subfamily of P-loop GTPase proteins and now it is considered as one of the new target for antibacterial drug. The majority of bacterial Obgs have been commonly found to be associated with ribosome, implying that these proteins may play a fundamental role in ribosome assembly or maturation. In addition, one of the experimental evidences suggested that Bacillus subtilis Obg (BsObg) protein binds to the L13 ribosomal protein (BsL13) which is known to be one of the early assembly proteins of the 50S ribosomal subunit in Escherichia coli. In order to investigate binding mode between the BsObg and the BsL13, protein-protein docking simulation was carried out after generating 3D structure of the BsL13 structure using homology modeling method. Materials and Methods: Homology model structure of BsL13 was generated using the EcL13 crystal structure as a template. Protein-protein docking of BsObg protein with ribosomal protein BsL13 was performed by DOT, a macro-molecular docking software, in order to predict a reasonable binding mode. The solvated energy minimization calculation of the docked conformation was carried out to refine the structure. Results and Discussion: The possible binding conformation of BsL13 along with activated Obg fold in BsObg was predicted by computational docking study. The final structure is obtained from the solvated energy minimization. From the analysis, three important H-bond interactions between the Obg fold and the L13 were detected: Obg:Tyr27-L13:Glu32, Obg:Asn76-L13:Glu139, and Obg:Ala136-L13:Glu142. The interaction between the BsObg and BsL13 structures were also analyzed by electrostatic potential calculations to examine the interface surfaces. From the results, the key residues for hydrogen bonding and hydrophobic interaction between the two proteins were predicted. Conclusion and Prospects: In this study, we have focused on the binding mode of the BsObg protein with the ribosomal BsL13 protein. The interaction between the activated Obg and target protein was investigated with protein-protein docking calculations. The binding pattern can be further used as a base for structure-based drug design to find a novel antibacterial drug.

Growth inhibition of hydrotrope-combined copper against Microcystis aeruginosa and evaluation of its toxicity (Microcystis aeruginosa에 대한 hydrotrope-combined copper의 생장억제 및 독성 평가)

  • Hydrotrope-combined copper (HCC) is a copper ($Cu^{2+}$)-based algicide, which is combined with a hydrotrope that keeps copper ion in solution to improve performance. This study assessed the growth inhibition effect of HCC against Microcystis aeruginosa which is one of the most common toxic cyanobacterium in eutrophic freshwater environment. Various HCC doses, ranging from 5.5 to $550{\mu}g/L$ as $Cu^{2+}$, were applied to either BG-11 or 1/4 diluted medium with low- or high-inoculum density of M. aeruginosa. Growth inhibition was monitored based on a decrease in chlorophyll-a content in culture medium during the incubation. Results showed that HCC significantly inhibited the growth of M. aeruginosa in a dose-dependent manner. In case of 1/4 diluted BG-11 medium, HCC dose as low as $5.5{\mu}g$ $Cu^{2+}/L$ completely inhibited the production of chlorophyll-a by M. aeruginosa. It was found that HCC did not induce any significant release of microcystin-LR from M. aeruginosa. Acute toxicity of HCC was tested using Daphnia magna, and the 24-h $EC_{50}$ value was 0.30 mg/L as $Cu^{2+}$ which was much higher than the actual inhibition dose. Ames test was performed using Salmonella enterica serovar Typhimurium TA100, and HCC showed no increase in the number of revertant colonies. The result suggested that HCC does not have any mutagenic potential in the aquatic environment. In addition, no genotoxic effect of HCC was also confirmed based on the SOS ChromoTest using Escherichia coli PQ37. Therefore, HCC could be used as a relatively safe and effective pre- and post-treatment agent to control hazardous algal blooming in aquatic environments.

Clinical Characteristics of Fever without Localizing Sign in Infants Younger than 100 Days of Age in a Single Center (단일기관에서 시행한 생후 100일 미만 영아에서 발생한 국소 증상 없는 발열에 대한 임상적인 특징에 관한 연구)

  • Purpose: This study was done to define clinical characteristics of fever without localizing signs (FWLS) in infants younger than 100 days of age with a goal of providing baseline data to establish a new diagnostic paradigm in the future. Methods: We reviewed medical records of 183 patients who admitted to Daegu Catholic University Medical Center for FWLS younger than 100 days of age from January 2013 to September 2015 retrospectively. Demographic, clinical features and laboratory findings were analyzed. Patients were divided into serious bacterial infection (SBI) and non-SBI groups, and then were compared between two groups to find risk factors for SBI. Results: Among 183 patients, lumbar puncture was performed in 98.9% and CSF pleocytosis was present in 35.9%. Sterile CSF pleocytosis was found in 43% of urinary tract infection (UTI) patients. None had concomitant bacterial meningitis in patients with UTI. As final diagnosis, febrile syndrome without source (25.7%) was most common. Among SBI, UTI was most common (99%). Birth weight, ESR, and CRP were significantly higher in SBI group compared to non-SBI group. Male sex (OR 4.93, 95% CI 1.60-15.24) and pyuria (OR 18.88, 95% CI 6.76-52.76) were identified as risk factors for SBI. Presence of sibling (OR 0.30, 95% CI 0.11-0.83) was significantly lower in SBI group. Conclusions: Our results showed UTI was the most common SBI in young infants with FWLS. Though aseptic meningitis can be coexisting with UTI, lumbar puncture may not be necessary in all patients having UTI.

The water holding capacity of breast meat increased (P<0.05) in broiler fed the diets containing bacteriophage CP compared with those fed the CON diet. Other meat characteristics such as pH value, breast muscle color ($L^*$, $a^*$, $b^*$) and drip loss were unaffected by dietary supplementation of bacteriophage CP. The weight of bursa of Fabricius increased (P<0.05) in CP05 when compared with CON. No significant difference was observed (P>0.05) among treatments in visceral weight and fecal microflora concentrations of Lactobacillus spp., Clostridium perfringens, Escherichia coli and Salmonella spp. In conclusion, dietary supplementation with 0.10 and 0.15% bacteriophage CP could improve the growth performance.
