저장김치의 Leuconostoc mesenteroides IFO 12060 및 Nisin첨가에 의한 Escherichia coli의 소장 (Fluctuation of Escherichia coli on the storage of Kimchi treated with Leuconostoc mesenteroides IFO 12060 and Nisin)

  • 수출용김치의 유통형태를 모델화하여 김치를 4$^{\circ}C$에서 7일간 저장한 후 $25^{\circ}C$로 저장온도를 변화시켜 주며 E. coli와 젖산균에 대한 Leuconostoc mesenteroides와 nisin의 첨가효과를 검토한 결과, 김치가 발효숙성 되어감에 따라 4$^{\circ}C$에서는 E. coli가 쉽게 사멸하지 않았으나 $25^{\circ}C$에서는 E. coli가 현저히 감소하다가 과숙김치에서는 다시 나타나며 후기 E. coli 재출현은 Leu. mesenteroides나 nisin의 첨가에 의해서 효과적으로 억제할 수 있음을 제시하여 주었다.

Effects of essential oil (blended and single essential oils) on anti-biofilm formation of Salmonella and Escherichia coli

  • Background: Biofilms were the third-dimensional structure in the solid surface of bacteria. Bacterial biofilms were difficult to control by host defenses and antibiotic therapies. Escherichia coli (E. coli) and Salmonella were popular pathogenic bacteria that live in human and animal intestines. Essential oils are aromatic oily liquids from plant materials and well known for their antibacterial activities. Method: This study was conducted to determine effect of essential oil on anti-biological biofilm formation of E. coli and Salmonella strains in in vitro experiment. Two kinds of bacterial strains were separated from 0.2 g pig feces. Bacterial strains were distributed in 24 plates per treatment and each plates as a replication. The sample was coated with a Bacterial biofilm formation was. Result: Photographic result, Escherichia coli (E. coli) and Salmonella bacteria colony surface were thick smooth surface in control. However, colony surface in blended and single essential oil treatment has shown crack surface layer compared with colony surfaces in control. Conclusion: In conclusion, this study could confirm that essential oils have some interesting effect on anti-biofilm formation of E. coli and Salmonella strains from pig feces.

  • 방향족 에폭사이드 기질에 대한 입체선택적 가수분해능이 우수한 Rhodotorula glutinis의 epoxide hydrolase (EH)를 codon usage를 고려한 Escherichia coli 균주에서 고효율로 발현할 수 있었다. 효모인 R. glutinis와 박테리아인 E. coli에서의 codon usage 선호도를 분석하고 그 차이를 고려하여 E. coli 에서 잘 사용되지 않는 rare codon에 대한 tRNA유전자정보가 들어 있는 pRARE plasmid를 함유한 E. coli 균주인 Rosetta(DE3)pLysS를 숙주세포로 사용하였다. R. glutinis EH를 발현시킨 재조합 E. coli를 생촉매로 사용하여 라세믹 styrene oxide 혼합물과 반응시켰을 때, (R)-styrene oxide에 대한 입체선택적 가수분해활성이 wild type R. glutinis 대비 매우 향상됨을 관찰할 수 있었다. 또한 라세믹 기질로부터 입체적으로 고순도인 99% ee 값을 갖는 광학적으로 순수한 (S)-styrene oxide를 얻을 수 있었다.

$Al_2O_3$ 비드를 가지는 방전관의 전계 및 대장균 제거 특성 (Electric Field Simulation and Removal Characteristics of Escherichia coli for Discharge Tube with $Al_2O_3$ Bead)

  • 본 연구에서는 구형 $Al_2O_3$를 갖는 방전관에 대한 전기분포의 시뮬레이션과 대장균 제거 특성을 조사하였다. 구형 $Al_2O_3$를 갖는 방전관을 이용한 대장균의 제거 특성을 실험한 결과 인가전압이 증가하면 전계도 증가를 하게 되는데 대장균의 제거 특성은 인가전압 즉 전계강도에 관계가 있음을 알 수 있었다. 그리고 방전관의 시험수 통과량이 증가하면 단위 시간당 전계영역의 통과횟수가 증가하기 때문에 대장균의 제거율이 증가하는 경향을 보였다. 또한 구형 $Al_2O_3$의 직경이 증가하면 유전체의 유전분극이 높아져 전계가 증가하기 때문에 전반적인 대장균 제거시간이 줄어드는 것을 알 수 있었고 구형 유전체를 갖는 방전관의 대장균 제거율은 유전체가 없는 경우보다 훨씬 높게 나타났다.

자몽 종자 추출물의 항균성 (Antimicrobial Activity of Grapefruit Seed Extract)

  • 자몽 종자 추출물의 항균성을 조사하기 위하여 그람 양성균인 Bacillus cereus, Bacillus subtilis, Listeria monocytogenes와 그람 음성균인 Escherichia coli, Salmonella enteritidis, Serratia marcescem를 대상으로 하여 최소 저해 농도, 생육 저해 활성, 콜로니 형성 저해 활성을 실험하였다. 그람 양성균에 대한 자몽 종자 추출물의 최소 저해 농도는 12.5 ppm정도로 상대적으로 낮았으며, 그람음성균의 최소저해 농도는 50ppm 이상으로 상대적으로 높았다. 균주의 증식에 대한 생육 저해 활성을 조사한 결과 Bacillus cereus는 1ppm 이하, Bacillus subtilis는 6.25 ppm, Listeria monocytogenes는 1ppm 이하, Escherichia coli는 6.25 ppm, Salmonella enteritidis는 25 ppm Serratia marcescem는 25pp부터 생육이 저해되었다. 그람 양성균의 콜로니 형성 저해활성은 Bacillus cereus는 93.9%, Bacillus subtilis는 94.0%, Listeria monocytogenes는 99.9%, 그람 음성균의 콜로니 형성 저해 활성은 Escherichia coli는 4.4%, Salmonella enteritidis는 82.7%, Serratia marcesem는 86.4%로, 그람 양성균에 대해서는 높은 저해 활성을 보였으며, 그람 음성균에 대해서는 비교적 낮은 저해 활성을 보였다.

Anti-Biofilm Activities of Manuka Honey against Escherichia coli O157:H7

  • Manuka honey (MH) has been shown anti-bacterial activity against several pathogenic bacteria. However, the inhibitory effect of MH on biofilm formation by Escherichia coli O157:H7 has not yet been examined. In this study, MH significantly reduced E. coli O157:H7 biofilm. Moreover, pre- and post-treatment with MH also significantly reduced E. coli O157:H7 biofilm. Cellular metabolic activities exhibited that the viability of E. coli O157:H7 biofilm cells was reduced in the presence of MH. Further, colony forming unit of MH-treated E. coli O157:H7 biofilm was significantly reduced by over 70%. Collectively, this study suggests the potential of anti-biofilm properties of MH which could be applied to control E. coli O157:H7.

Thioredoxin in the Periplasmic Space of Escherichia coli as a Physiological Electron Donor to Periplasmic Thiol Peroxidase, p20

  • We previously reported that a novel thiol peroxidase (p20) from Escherichia coli is a distinct periplasmic peroxidase that detoxifies hydroperoxides together with glutathione or thioredoxin. Until now, there was no experimental evidence for the presence of thioredoxin (Trx) in the periplasmic space. In an attempt to confirm the physiological function of p20 as a thiol peroxidase supported by Trx in the periplasmic space, we have purified a Trx activity from the periplasmic space of Escherichia coli and identified the Trx as the same protein as the cytoplasmic Trx. The presence of Trx in the periplasmic space of Escherichia coli suggests that p20 is a unique extracellular Trx-linked thiol peroxidase.

  • For isolation of the CMCase gene of the alkalophilic Bacillus sp. strain N-4 to analyze their genetic information for the multicomponents of the cellulase, Bscherichia coli K12 and plasmid DNA pBR322 was used as host-vector system. After the digestion of purified chromosomal DNA and plasmid DNA pBR322 with HindIII, these were ligated. The ligated DND were transformed into Escherichia coli, and recombinant plasmid 107 carried the gene coding for CMCase was constructed. The CMCase produced by Escherichia coli cells containing plasmid DNA pYBC107 was found in the cells as intracellular enzyme and nearly 60% of the total CMCase activity was localized in cellular fraction. Also, the optimum pH for the reaction of CMCase produced by Escherichia coli was appeared at pH .8.0 and the enzyme was stable between pH 7.0 and pH 8.0.

PCR법을 이용한 옹달샘물의 대장균군 및 대장균 검출 (Detection of Coliform and Escherichia coli in Spring Water by Polymerase Chain Reaction)

  • The polymerase chain reaction(PCR) of target lacZ and uidA genes were used to detect total coliform and Escherichia coli for determining water quality, respectively. Of 109 spring waters, coliform were detected from 38 spring waters by lacZ PCR method but 21 spring waters by culture method accepted by the Ministry of Environment for water quality monitoring. The lacz PCR method gave the results statistically equivalent to those of the culture method(kappa=0.62, McNemar=17.00). The uidA PCR method gave the same results to those of the culture method. The sensitivity and specificity of coliform and E. coli by PCR method were 100% and 80.7%, respectively. Therefore, PCR can be used for the rapid identification of Escherichia coli and coliform in potable water using uidA and lacZ.

향신료 분말의 Esdcherichia coli 와 Staphylococcus aureus 에 대한 항균작용 (Antibacterial Activity of Powdered Spice against Escherichia coli and Staphylococcus aureus)

  • Antibacterial activities of powdered spices(garlic , ginger, cinnamon and clove) against pathogenic Escherichia coli )157:H7 and Staphyloccus auresus were investigated. Spice powder was added in was exponetial phase of each bacterial culture . Growth inhibition was determined by the absorbance at 660nm and morphological changes of the cells were observed by transmission electron microscope (TEM). Ginger powder has the highest antibacterial activity, following cinnamon , clove and garlic has the least activity.Growth of Escherichia coli O157:H7 and Staphyloccus aureus were completely inhibited within 5 hours after addition of 1 % of garlic , 0.3% of ginger or cinnamon , 0.5% of clove powder on the exponential phase of the cells. Spice untreated cells of E. coli and S. aureus, the cytoplasm was entirely surrounded by rigid cell wall and cell walls formed a smooth layer well attached to the plasma membrane. In the cells of E. coli and S. aureus treated with spice powder, cell wall and plasma membrane were lysed and severely damaged. E.coli cells growth in the presence of spice powder showed plammolysis, the loss of electron dense material, the formation of extra cellular blebs and cytoplasm burst out from the cell. S .sureus cells grown in the presence of spice powder showed swell of cell wall, the loss of electron dense material , coagulation of cell cytoplasm and formation of extra cellular blebs. Severely damaged cells of S. aureus lost whole cytoplasm and left as ghost of the cell. Spice powder stimulated autolyssi and induced cell death.
