STAT6 and PARP Family Members in the Development of T Cell-dependent Allergic Inflammation

  • Allergic inflammation requires the orchestration of altered gene expression in the target tissue and in the infiltrating immune cells. The transcription factor STAT6 is critical in activating cytokine gene expression and cytokine signaling both in the immune cells and in target tissue cells including airway epithelia, keratinocytes and esophageal epithelial cells. STAT6 is activated by the cytokines IL-4 and IL-13 to mediate the pathogenesis of allergic disorders such as asthma, atopic dermatitis, food allergy and eosinophilic esophagitis (EoE). In this review, we summarize the role of STAT6 in allergic diseases, its interaction with the co-factor PARP14 and the molecular mechanisms by which STAT6 and PARP14 regulate gene transcription.

Electron microscopic studies on Flavobacterium branchiophila in experimentally induced gill disease of rainbow trout (세균성(細菌性) 아가미병(病)에 실험적(實驗的)으로 감염(感染)된 무지개송어에 있어서 Flavobacterium branchiophila에 대한 전자현미경학적(電子顯微鏡學的) 연구(硏究))

  • Gill epithelia of normal rainbow trout fingerlings and abnormal ones suffering bacterial gill disease by experimental infection were examined by transmitting electron microscopy (TEM) and scanning electron microscopy (SEM). TEM observations revealed that Flavobacterium branchiophila consisted of slender rods measuring 0.5 by 5 to $8{\mu}m$, and they had which were long, thin, flexible filaments measuring approximately 4 nm by $1{\mu}m$, and packed together to organize into bundles. Morphological alterations of the diseased epithelia started at hypertrophy of the lamellar epithelium. F branchiophila attached to the gill surface of infected fish through pili with a regular distance, and did not invade into gill tissue. In SEM observations, normal surface ultrastructure of epithelial cell in the outermost layer were characterized by a typical labyrinth-like structure branching and anastomosing microridges on the cell surface. Hyperplastic lesions in experimentally infected gill were most serious at near the tips. Each filament exhibited a club-like, and fusion between the filaments was sometimes observed at their tips. On the surface of gill filaments, thread-like bacterial cells attached and were entangled. The bacterial cells almost covered the surface. After immersion in 5 % NaCl, the cell of F branchiophila, however, appeared to be indeterminate shape.

A Histopathological Study on the Intestine of Mice and Rats Experimentally Infected by Fibricola seoulensis (Fibricola seoulensis에 실험 감염된 마우스 및 흰쥐 소장의 병리조직학적 연구)

  • A histopathological study was carried out on the duodenum of mice and rats experimentally infected by F. seoulensis. Each mouse was infected with 500 metacercariae and killed after 1, 2, 3 days, 1 and 2 weeks from infection. Each rat was given 1, 000 metacercariae and was examined after 1, 2 and 4 weeks from infection. The duodenal tissue sections of mice and rats were stained with hematoxylin eosin, and PAS stained for the rats of 1 week group. The pathological findings are summarized as below. 1. There were no differences in mucosal findings between the mice and the rats, and between the location of duodenum, 1 and 5 em distal to the pylorus. 2. Each worm embraced a villus exclusively with its foliate fore body which was inserted into the intervillous spaces. The fluke pinched villous epithelia using its oral and ventral suckers. The tribocytic organ destroyed the villous epithelia deeply up to the stroma after 3 days from infection. 3. Apparent villous changes were observed in the mice after 3 days from infection. Villous changes were shortening, widening, blunting or fusion. The villous stroma showed edema, microscopic hemorrhage, capillary congestion, dilatation of lymphatics and inflammatory cell infiltration. The cells were lymphocytes, plasma cells, eosinophils and giant cells. Rarely submucosal and trans:nural inflammation was encountered.

Regional Difference in Distribution of Glycoconjugates in Mouse Epididymis (생쥐 부정소 부위별 당쇄 분포의 차이)

  • To characterize the difference in glycoconjugates of mouse epididymis, lectin labeling of the tissue section was conducted using Ulex europaeus agglutinin I(UEA I), succinylated wheat germ agglutinin(sWGA), and Griffonia simplicifolia lectin-I(GSL-I). UEA I which binds to outer $\alpha$-L-fucose residue that is a terminal sugar of the side chain branched from oligosaccharide chain gave the labeling in the proximal caput epithelia exclusively. Lumen was commonly labeled in all of the organ. It suggested that the glycoconjugates bearing outer $\alpha$ -L-fucose residue were largely expressed in the initial segments ot epididymis and subjected to secretion. GSL-I which binds to terminal $\alpha$ -D-galactosyl residue of glycoconjugates gave the labeling in the cytoplasm of clear cells and basal cells, and cilia in corpus and cauda regions but not in the caput region. There was no vast difference in labeling pattern by sWGA which binds to N-acetyl-glucosamine residue among the epididymal regions. Clear cells in corpus and cauda epithelia showed more intense labeling by sWGA compared to principal cells, suggesting the functional specialization of this type of cells. The labeling intensities of luminal content by UEA I and sWGA decreased in cauda region compared to corpus region suggesting the presence of enzymatic activities responsible for processing the $\alpha$-L-fucose and N-acetyl-glucosamine residues from secreted glycoconjugates. In summary, the difference in glycoconjugates bearing the $\alpha$-L-fucose, $\alpha$-D-galactose, and N-acetyl-glucosamine residues according to the type of epithelial cells and epididymal segments suggests functional specialization and different roles of each segment in the processing of sperm surface antigens during the epididymal transit.

  • The morphological development of colonic epithelia in fetuses between 60-. 90-, and 120-days gestation and neonates of Korean native goat were investigated by transmission electron microscopy. In the 60-day-old fetuses, the colonic epithelial cells contained nuclei, nucleoli, mitochondria, free ribosomes, and shoo granular and agranular endoplasmic reticula. The zonula occludens, zonula adherens, desmosomes, short microvilli, and masses of glycogen granules were also obsrved. The goblet cells contained a few secretory granules. In the 90-day-old fetuses, the cell organelles of the colonic epithelial cells were better developed than those in the 60 day old fetuses. Increased number of endoplasmic reticula, digitiform intercellular junctions, mitochondria, and Golgi complexes was observed. The goblet cells contained a lot of secretory granules. In the 120-day-old fetuses, the colonic epithelial cells contained long microvilli and well developed cell organelles. The nuclear cleft and large intercellular space were also appeared. Nunerous fibroblasts were seen in the basement membrane. The number of goblet cells was further observed. In the 120 day old fetuses, all colonic epithelial cells shape simple columnar cells. In newborns, the colonic epithelial cells were covered with extensive microvilli. There were many goblet cells with a lot of secretory granules protruding into the intestinal lumen, and some goblet cells secreted their secretory granules into the lumen. In the 60-and 90-day-old fetuses, the colonic epithelial cells appeared to be either simple columnar or stratified columnar depending on areas.

Morphology and Histology of the Digestive Organ in the Sablefish, Anoplopoma fimbria (Teleostei: Anoplopomatidae) (은대구, Anoplopoma fimbria 소화기관의 형태 및 조직학적 특징)

  • The RLG (relative length of gut) is 1.52 (n=12) in the sablefish, Anoplopoma fimbria. The digestive tract has five or six pyloric caeca in the posterior region of stomach. Morphology of mucosal fold is unbranched type in the esophagus and stomach, but branched type in the intestine. The histological structure of digestive tract can be divided into mucosal layer, submucosal layer, muscular layer and serous membrane in the cross section. In the esophagus, mucosal epithelial layer is a simple, and consists of ciliated columnar epithelia and mucous cells. In the stomach, gastric gland of mucosal epithelial layer is a tubular, and is composed of chief cell, parietal cell and mucin secreting cell, which is columnar and contained secretory granules of red and blue colors in the AB-PAS (pH 2.5) reaction. In the intestine, mucosal epithelial layer is a simple, and consists of ciliated columnar epithelia and goblet cells. The submucosal layer is composed mainly of collagen fibers, and well developed in the esophagus. And the muscular layer of digestive tract is divided into longitudinal and circular muscle layer, and well developed in the stomach. The liver is composed of numerous lobular structure and bile canaliculi. Stainability of hepatocyte cytoplasm was eosinophilic, and nucleus and nucleolus showed basophilic in the H-E stain. The pancreatic tissue was scattered in the fatty tissue near the digestive tract, and acinar gland consisting of numerous exocrine cells. And cytoplasmic stainability of exocrine cell was basophilic, and contained numerous zymogen granules of eosinophilic in the H-E stain.

Localization of Dendroaspis natriuretic peptide (DNP) in the rat salivary glands (흰쥐 타액선내에서의 DENDROASPIS NATRIURETIC PEPTIDE(DNP)의 분포)

  • Dendroaspis natriuretic peptide (DNP), a fourth member of the natriuretic peptide isolated from the venom of the Dendroaspis angusticeps snake, has been reported to be present in human plasma and atrial myocardium and caused vasorelaxation and diuresis in experimental animals. However, it is uncertain whether they are present in peripheral organs other than the heart and its further physiological roles also remains to be clarified. To assess the possible physiological role of DNP in the salivary glands, I investigated the localization of DNP peptide in the rat salivary glands by immunohistochemistry and the binding sites for radiolabelled DNP in the rat salivary glands and oral mucosa using in vitro autoradiography. DNP immunoreactivity was widely distributed in the submandibular, sublingual and parotid glands, particularly in the ducts such as the intercalated and striated ducts, where atrial natriuretic peptide (ANP) was colocalized in consecutive sections, but not in acini. High density $^{125}I-DNP$ binding sites were localized in the epithelia of the tongue and hard palate, while low density binding sites for $^{125}I-DNP$ were also distributed in the submandibular, sublingual, and parotid glands. In the hard palate and tongue, the precise location of this binding was revealed on the basal and parabasal cells of the epithelia by emulsion microautoradiography. These results suggest that DNP may not only have a role in the salivary glands but also play a role in the regulation of growth in the oral epithelium, particularly in the hard palate and tongue.

Ecophysiological Responses and Subsequent Recovery of the Olive Flounder, Paralichthys olivaceus Exposed to Hypoxia and Iron II.   • The effects of iron on gill tissue and metabolic rate represented by oxygen consumption of olive flounder, Paralichthys olivaceus were determined. The effects were further studied by means of survival rate of the fish exposed to a serial concentrations of iron. The olive flounder exposed to iron concentrations over 0.93 mg/$\ell$ showed curvature and terminal clubbing of gill lamellae at 2 weeks post-exposure. In iron concentration 4.89 mg/$\ell$, gill of the fish were seriously damaged just after 2 weeks, showing hyperplasia of filament epithelia, deformation of lamella epithelia, chloride cell damage, and separation of lamella epithelial layer, Gills exposed to 9.78 mg/$\ell$ iron concentration resulted in fusion and necrosis of the lamellae after 2 weeks. Significant decreases of metabolic rate of the fish were observed after 4 weeks at iron concentration 0,93 mg/$\ell$ and after 2 weeks at iron concentrations over 4.89 mg/$\ell$. Survival rate of the olive flounder decreased significantly after 4 weeks at the iron concentration over 4.89 mg/$\ell$. These results lead us to conclude that, as far as the iron effects are concerned, its concentrations should not exceed at least more than 0.93 mg/$\ell$ in the fish farm and coastal waters for normal growth of the olive flounder.

Tributyltin chloride (TBTCl) toxicity on the survival rate and burrowing activity of the equilateral venus, Gomphina veneriformis (Bivalvia: Veneridae) (대복, Gomphina veneriformis의 생존 및 저질 잠입성에 미치는 TBTCl의 독성)

  • This study was conducted to find out biological responses of bivalves exposed to organotin compound.The results of the study confirmed that tribultyltin chloride (TBTCl) induce reduction of survival rate andburrowing activity, and histopathological feature in the foot structure of the equilateral venus, Gomphinaveneriformis. The experimental period was 36 weeks. The experimental groups consisted of a control and 3TBTCl exposure groups (0.4, 0.6, 0.8 ym TBTCl L'). The survival rate and burrowing activity were record-ed daily. For histological analysis, foot tissues were fixed in Bouin' s fluid and then stained H-E stain, AB-PAS (PH 2.5) reaction and Masson's trichrome stain after having serially sectioned the tissue by paraffinmethod at thickness of 4-6 ym. The survival rate was not significantly different between the control andexposure groups for 20 weeks, but in 0.8 Um TBTCl L', it was on the decreased ever since the exposure. Theburrowing activity was not significantly different in the exposure group compared to the control up to 12weeks, but in 0.6 and 0.8 ym TBTCl L', it measured the lowest level after 20 weeks. The foot is composedof the epidermal layer, connective tissue, and muscular layer. The epidermal layer is composed of simplecolumnar, cuboidal epithelia and mucous cells. The cilia were well developed on the apical surface ofepithelium, Circular, longitudinal and transverse muscle bundle were well developed in the muscular layer.The majority mucous cells showed blue color (542c) when it subjected to AB-PAS (PH 2.5) reaction. Nohistopathological alterations in the foot were observed up to 12 weeks. After 20 weeks of exposure to 0.8 (anTBTCl L'', the foot samples of exposed G. veneriformis showed disappearance of cilia and striated borderpartially and extension of hemolymph sinus. The mucous cell increased in the marginal of foot. At 28-weekof exposure to 0.4 ym TBTCl L', it observed weekly acid (564c), neutral (264c) and mixed mucous cell. At36-week of exposure to 0.6 ym TBTCl L', it showed fragmentation of the muscle and collagen fiber bundle,and also diappearance of cilia on epithelia and edema of epithelium in 0.8 ym TBTCl L''.

Histopathological changes of epithelium following the exposure of N-methyl-N-nitro-N-nitrosoguanidine in Manila clam, Ruditapes philippinarum (N-methyl-N-nitro-N-nitrosoguanidine 노출에 따른 바지락 (Ruditapesphilippinarum) 상피조직의 병리조직학적 반응)

  • To understand responses of bivalve tissues to various injurious agents, histopathological observation was done in the Manila clam, Ruditapes hilippinarum, during 240 hours after an intramuscular injection of N-methyl-N-nitro-N-nitrosoguanidine (MNNG). Some histopathological changes were observed with the epithelia of stomach, midgut, hindgut, digestive diverticula and gonads after injecting MNNG. The earliest degenerative changes were recognized in the epithelium of midgut 12 hours after the injection. Partial or complete, epithelial destruction was constantly accompanied by the massive infiltration of a mononuclear, necrotic cell group including hemocytes. At 144 hours later, gonads showed the necrotic desquamation of their germinal epithelia. Nearly all of the infiltrated cells within the destructive epithelium were suggested to be hemocytes, from the binding property with a hemocyte-specific lectin, Ricinus communis (RCA-1). From these results it was concluded that the epithelium of digestive system is vulnerable to parenteral MNNG and hemocytes might be deeply involved in this MNNG-induced destruction of the digestive epithelium.
