에탄올을 투여한 흰쥐 노조직의 항산화효소계 활성에 미치는 단백질과 섬유소의 영향 (Effects of Protein and Fiber on Antioxidant Enzyme Activites of Brain in Ethanol-Treated Rats)

  • This study was to investigate the effect of dietary protein and fiber on the antioxidant enzyme activities of brain in acute or chronic ethanol-treated rats. Male Sprague-Dawley rats were fed on diets containing two levels of protein(7%, 20%) with two levels of fiber (5%, 10%) Rats were administered 40%(v/v) ethanol(5g/kg body weight)orally 90min before decaptiation in acute ethanol-treated groups and 25%(v/v) ethanol(5g/kg body weight) once a day for 5 weeks in chronic ethanol treated-groups. The rats were sacrificed after 5 weeks of feeding periods. Superoxide dismutase and gluthathione S-transferase activities were lower in chronic ethanol-treated groups than acute ethanol-treated groups whereas catalase and glutathuone peroxidase activities were significantly increased by chronic ethanol treatment. Low protein supplement accelerated to change of their activities however dietary fiber levels did not affect antioxidant enzyme activities. Chronic ethanol treatment and/or low protein supplement results in increasing the brain lipid peroxide content but in lowering glutathione level. (Korean J Nutrition 33(6) ; 613~618, 2000)

제비콩(Dolichos lablab L.) 잎 추출물이 쥐의 혈청효소활성에 미치는 영향 (Effect of Jebikong(Dolichos lablab L.) Leaves Extract on Enzyme Activities in Mice Blood)

  • The effect of Dolichos lablab leave extract on enzyme activities in mice blood was investigated in this study to clarify the new useful application of the Plant leaves. There were not significant differences in the enzyme activities in mice blood among treatment fed with the leave extract and non fed control. The feeding treatment with the extract showed a tendency to activity compared to the nun fed control. The activities of glutamate oxaloacetate transaminase, glutamate pyruvate transaminase and lactate dehydrogenase in mice blood at the treatment fed with lead and the leave extract were significantly low compared to their activities at the non fed treatment with the extract, respectively. The choline esterase activity was high at the leave extract feeding. The cadmium dietary treatment showed the same result as the lead treatment. In conclusion, the physiological function of the Jebikong leave was significantly in creased when the mice was stressed by the hear metal intake. Therefore, the plant leave extract would consider the reduction of heart metal effect.

하변토양의 미생물체외효소활성에 미치는 칩입성 식물의 영향 (Influences of Invasive Plant on Extracellular Enzyme Activities in Riparian Ecosystems)

  • 하변에 침입하는 외래종 식물은 하변 토양의 생태구조 및 기능에 영향을 미칠 수 있기 때문에 관심의 대상이다. 이에 본 연구에서는 1년간 국내 하변 총 네 지점에서 유기물질 분해율을 대표할 수 있는 미생물 체외효소 활성(${\beta}$-glucosidase, N-acetylglucosaminidase, phosphatase, arylsulfatase)을 외래종 유무에 따라 측정하였다. 하변 침입종인 환삼덩굴과 가시박 생장구 및 외래종 제거구의 토양을 분석한 결과 일부 침입성 덩굴식물 실험구에서 효소활성이 높았으나 계절 및 효소별로 그 특성이 달라 어떤 경향을 밝혀낼 수는 없었다. 허나, 교란이 발생한 하변 생태계에서는 침입성 덩굴 식물이 토양의 유기물질 분해를 가속화시키는 것으로 판단된다.

판크레아틴의 규격 표준화 연구 (Standardization of Pancreatin)

  • Pancreatin is a enzyme mixture breaking down carbohydrates, proteins and lipids. Most pancreatin used in Korea is imported from foreign countries. However, guideline of each country for pancreatin produced from each country is different. Therefore, guideline for pancreatin imported from several countries, such as Europe, Japan and America, it is standardized to control its quality. Assay of enzyme activity for pancreatin in KP is similar to tat in JP, but it is significantly different from those in FP ad in USP. We measured pancreatin digestive activities of 17 commercial products. Activity assay of digestive enzymes, starch- and lipid-digestive enzymes, for pancreatin by KP method (including JP) was difficult compared to those by FIP ad USP methods. Particularly, activity assays of starch- and lipid-digestive enzymes by KP method were mistakable, ad varied in diluted samples than those by FIP. However, activity assay of protein-digestive enzyme by KP method was similar to that by FIP. Starch-digestive enzyme activities of 17 commercial pancreatins by KP method were lower 0.079-fold compared to those by FIP method. Their protein-digestive enzyme activities by KP method were higher 75.7-fold than those by FIP method. Their lipid-digestive enzyme activities by KP method were lower 0.234-fold compared to those by FIP method.

Antioxidant and ACE Inhibitory Activities of Soybean Hydrolysates: Effect of Enzyme and Degree of Hydrolysis

  • Native soy protein isolate (SPI) was hydrolyzed with 4 different proteolytic enzymes, including bromelain, papain, Neutrase, and Flavourzyme. SPI hydrolysates with the degree of hydrolysis (DH) in range of 6 to 15% were prepared by each enzyme. The angiotensin 1 converting enzyme (ACE) inhibitory and the antioxidant activities of the SPI hydrolysates, such as superoxide dismutase-like activity and inhibition of the linoleic acid autoxidation, were evaluated. Overall, as the DH increased, all evaluated bioactivities of the SPI hydrolysates significantly increased. The significantly highest ACE inhibitory and antioxidant activities were found in hydrolysates made with papain and bromelain, respectively. SPI hydrolysates by Flavourzyme showed the significantly lowest activity in all tested bioactivities. The results suggested that ACE inhibitory and antioxidant activities of SPI hydrolysates were determined by the DH and by the enzyme used.

The Response of Nitrogen Deposition to Methane Oxidation Availability and Microbial Enzyme Activities in Forest Soils

  • Forest soils are often nitrogen-limited, and nitrogen input to forest soils can cause substantial changes in the structure and functions of a soil ecosystem. To determine the effects of nitrogen input on methane oxidation and the microbial enzyme activities, manipulation experiments were conducted using nitrogen addition to soil samples from Mt. Jumbong. Our findings suggested that the addition of nitrogen to the soil system of Mt. Jumbong did not affect the microbial enzyme activities. Conversely, the addition of nitrogen affected the rate of methane oxidation. Inorganic nitrogen in soils can inhibit methane oxidation via several mechanisms, such as substrate competition, toxic effects, and competition with other microbes, but the inhibitory effects are not always the same. In this research, seasonal changes were found to produce different inhibitory factors, and these different responses may be caused from differences in the methantrophic bacteria community structure.

Effects of Psoralen and Angelicin on Hepatic Drug-Metabolizing Enzyme Activities

  • The effects of psoralen and angelicin on hepatic microsomal drug-metabolizing enzyme (DME) activities were investigated to elucidate the mode of the interaction of furanocoumarins with DME system. A single administration (30 mg/kg,i. p.) of both coumarins to mice cased a significant prolonagation of hexobarbital-induced hypnosis as well as an increase in strychnine toxicity. The inhibitory potencies of both coumarins as measured by rat hepatic microsomal aminopyrine N-demethylase and hexobarbital hydroxylase activities in vitro were considerably weaker than those of other furanocoumarins which possess a side chain moiety. Both coumarins were found to have significant inducing effects of DME system, with repeated treatments of them. The activities of an angular coumarin were stronger than those of a linear coumarin.

Myricetin과 Taruine의 병용 투여가 B16F10 세포의 항산화 효소계에 미치는 영향 (Effect of Myricetin Combined with Taurine on Antioxidant Enzyme System in B16F10 Cell)

  • The antioxidant enzyme (AOE) system plays an important role in the defense against oxidative stress damage. To determine whether myricetin or myricetin/taurine can exert antioxidative effects not only by modulating the AOE system directly but also by scavenging free radical, we investigated the influence of the myricetin and taurine on cell viability ROS level, activities of different antioxidant enzyme, and the expression of different antioxidant enzyme. As results, the cell viability showed inhibition of the proliferation with treatment of 'myricetin' or 'myricetin with taruine', respectively, with dose-dependent manner. Compared to control, the treatment of 'myricetin' decreased activities and gene expressions of superoxide dismutase (SOD), and glutathione peroxidase (GPx). However, combined treatment of 'myricetin with taurine' increased activities and gene expressions of the SOD, GPx, and catalase (CAT). In addition, the combined treatment of 'myricetin with taurine' somewhat decreased ROS levels, compared to the treatment of 'myricetin'. In conclusion, our study provides that the combined treatment of different antioxidants can enhance antioxidant effects.

대추 성숙중의 세포벽 성분과 세포벽 분해효소의 활성 변화 (Changes in the Cell Wall Components and Cell Wall-Degrading Enzyme Activities of Jujube Fruits during Maturation)

  • This paper was investigated the changes of the cell wall components, enzyme activities during ripening of jujuba fruits for elucidating the softening metabolism of jujuba fruits. Firmness were decreased during ripening. Moisture content did not show any notable cahanges until ripening stage but they decreased a little In overripe jujuba fruits. Polygalacturonase activities were not detected at nature green stage and $\beta$-galactosidase activities were until turning stage. But polygalacturonase activities in ripening and overripening were 51.31 and 100.72 units/100g-fr, wt. respectively. $\beta$-galactosidase activities were 16.05 and 182.55units/100g-fr. wt. in the same stages. The content of water-soluble protein was increased in overripening. Stage the contents of cell wall and alcohol-insoluble material were. decraesed during maturation, but water-soluble material was increased. The pectin and alkali-soluble hemicellulose were increased until ripening stage, but decreased in overripe jujube fruits. The total pectin and insoluble pectin during ripening, but decreased in overripe jujuba fruits.

Enzyme Activities of the Fruit Body of Ramaria botrytis DGUM 29001

  • The fruit body of Ramaria botrytis DGUM 29001 was used to determine enzyme activities of fruit body. The specific activity of laccase was the highest(6.5 unit/mg$\cdot$protein) and that of $\alpha$-amylase and xylanase was relatively high. However, little or no enzyme activity of $\beta$-glucosidase, CMCase, exo-$\beta$-1,4-glucanase, chitinase, lipase and protease was found.
