The purpose of this study was to investigate the effects of dietary Indongcho(L. japonica Thunb) powder on blood glucose, serum lipid levels and antioxidative enzymes in normal and streptozotocin(STZ)-induced diabetic rats. Four groups of rats(3-week-old inbred Sprague-Dawley male rats) were normal rats fed control diet(NC), diabetic rats fed control diet(DC), normal rats fed Indongcho powder diet(NI), and diabetic rats fed Indongcho powder diet(DI). Diabetes was induced by single injection of streptozotocin(60mg/kg B.W., i.p.). The animals were fed ad libium each of the experimental diet for 5 weeks. Food and water intakes were determined everyday. Blood glucose and serum total cholesterol levels were determined every week. After 5 weeks the animals were sacrificed and activities of antioxidant enzymes and lipid peroxidation products were determined in their liver and kidney homogenates. We also determined serum concentrations of total lipid(TL), total cholesterol(TC), triglycerides(TG) and HDL-cholesterol(HDL-C). Blood sugar and water intake were higher in diabetic group(DC and DI group) than normal group(NC and NI group) and were not significantly decreased by dietary Indongcho intake. Body weight gain and FER(feed efficiency ratio) were reduced by STZ treatment. But, Final body weight was recovered by Indongcho-contained diet. LHR(LDL-cholesterol/HDL-cholesterol) of the DI g re up was significantly lower than the other experimental groups(NC, NI and DC groups). The hepatic glucose 6-phosphatase(G6Pase) activity of the groups fed Indongcho diet(NI and DI group) was lower than the groups fed control diet(NC and DC group) and the G6Pase activity of NI group was recovered to the normal levels(p<0.05). However, The glutathione peroxidase(GPx) and glutathione reductase(GR) activities in liver and G6Pase activity in kidney were not statistically different between the control and diabetic control groups. Renal GST activity of the DI group was recovered by Indongcho intake. In conclusion, these results confirm oxidative stress in the liver and kidney of rats with STZ diabetes and antioxidative effect of Indongcho.
용액 상에서 페놀류에 대해 95%의 분해특성을 지니는 동식물세포유래의 효소(horseradish peroxidase, HRP, EC 1.11.1.7)를 다공성 탄소 전극에 고정화시키고 이를 전기화학 반응기에 도입하여 전극반응에 의해 연속적으로 발생되는 과산화수소를 이용하여 페놀의 분해를 수행하였다. FT-IR 분석을 통해 다공성탄소전극 표면에 HRP의 아민기와 펩티드 결합을 위한 카르복실기가 생성되었음을 확인하였고 가교제(coupling agent)로 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride(EDC)를 이용하여 공유결합으로 고정화시켰다. 또한 HRP를 활성화 시켜 페놀을 처리하기 위해 전해질로 사용된 인산염 완충용액의 농도($10{\sim}200$ mM)와 pH($5.0{\sim}8.0$), 외부 산소 주입량($10{\sim}50$ mL/min)및 potentiostat/galvanosta에 의한 외부 공급전압($-0.2{\sim}-0.8$ volt, vs. Ag/AgCl)의 조건을 달리하며 RVC 전극 표면에서 발생되는 과산화수소 농도 및 전류효율을 고려하여 최적 자체 발생조건을 결정하였다. HRP가 고정화된 RVC 전극은 초기 고정화된 HRP 활성에 대해 4주 동안 89%의 상대적 효소 활성도(relatively enzymatic activity)를 지니는 안정한 전극임을 확인하였으며 실험실 스케일의 연속식 전기화학 반응기에 도입되어 최적 과산화수소의 발생조건에서 86%의 분해 효율을 보였다.
돈 태반의 화장품소재로서의 응용가능성을 조사하기 위해 알칼리 가수분해, 효소처리 및 산처리 가수분해 방법을 이용하여 추출물을 얻었다. 돈 태반 추출물의 중금속함량을 측정한 결과 납, 비소, 수은 등이 매우 낮아서 화장품소재로서 적절하였다. 화장품소재의 안전성시험(MTT assay) 결과 돈 태반 추출물은 모두 $50{\mu}g/ml$의 농도에서 80% 이상의 세포생존율을 보여주어서 독성이 비교적 낮은 것으로 나타났다. DPPH free radical scavenging assay를 사용한 항산화시험 결과, $50{\mu}g/ml$의 농도에서 pH9로 알칼리 처리한 경우가 항산화율이 63%로 가장 높았다. Tyrosinase 활성저해시험을 사용한 미백효과 측정 결과 $50{\mu}g/ml$의 농도에서 알칼리처리한 추출물의 효과가 30% 정도의 tyrosinase 억제효과를 보여주어서 가장 우수하였지만, 대조군인 arbutin이나 비타민 C에 비해 그 효과가 다소 낮았다. Elastase 활성억제시험을 이용한 주름개선 효과 측정결과 $50{\mu}g/ml$의 농도에서 5가지의 처리법에서 모두 20~30%의 elastase 저해활성을 보여주어서, 대조군보다 우수하였으며, 특히 효소처리한 추출물의 효과가 가장 우수하였다. 돈 태반 추출물 1%를 함유한 스킨제형을 제조하여 온도, 시간에 따른 안정성 시험결과, 안정성 역시 매우 우수하였다. 따라서 돈 태반 추출물은 주름개선 기능성 화장품소재로서의 가능성이 높음을 알 수 있었다.
AIN-93G 식이조성을 기본으로 corn starch를 potato starch로 100% 대치한 고지방식이를 42일간 rat에 급여하여 비만을 유도 후, GPS군, SPS군, EZ군, H40군으로 나누어 70일 급여한 후 지질대사에 미치는 영향을 살펴보았다. 그 결과, 체중증가량은 GPS, EZ, H40군에서 차이가 없었고 식이효율 또한 유의적으로 차이가 없었다. 혈중 혈당농도, TC, LDL-cholesterol 농도는 변성 전분 섭취 군(EZ, H40군)에서 높았고, atherogenic index는 H40이 다른 군에 비교해 높게 측정되어 H40군이 비만, 동맥경화에 미치는 영향이 크게 나타났다. 간과 신장 조직의 MDA 생성량은 H40군이 혈청 MDA 생성량이 다른 군에 비해 매우 높았고, 간과 신장 조직에서는 EZ, H40군이 낮았다. Glutathione 함량은 각 조직별로 경향이 달랐으나, GP-x활성은 SPS군에서 낮았다. 본 연구에서 전분을 달리하여 첨가한 고지방 식이가 비만한 식이에 미치는 영향을 살펴본 결과, 알감자 전분을 100% 대치하여 제조한 SPS군 및 효소 처리한 변성전분(EZ)을 100% 대치하여 급여한 군에서 비만억제 효과가 나타났다. 따라서 변성전분의 심혈관계 질환 및 혈중 지질 패턴 변화에 관련된 기전 연구가 지속적으로 필요하며, 생리활성이 규명된 후 다양한 가공식품 개발이 이루어져야 할 것으로 사료된다.
현대인들에게는 다양한 증상과 징후를 나타내는 스트레스성 질환이 증가되고 있는 추세에 있다. 이러한 스트레스 시에 구강 내에는 구강건조이나 구강작열감증후군, 구강점막질환, 구취 등이 호발 된다는 것은 이미 널리 알려진 사실이다. 이때 대부분의 환자에서 공통적으로 나타나는 현상은 타액선의 기능적 및 구조적 변화에 따른 타액성분의 변화이다. 이에 저자는 스트레스가 구강 내 타액선에 어떠한 영향을 미치며, 타액선으로부터 분비되는 ${\alpha}$-amylase에 어떠한 영향을 미치는지를 알아보고자 본 실험을 시행한 결과 다음과 같은 유의성 있는 결과를 얻었다. 1. 구속 스트레스에 의하여 타액선 조직은 선포세포가 공포화되며 변성되었고, 선포간극이 이개되었으며, 도관주위조직도 변성되었다. 2. 구속 3시간 후에는 선포세포가 위축되기 시작하였고, 구속 6시간 후에는 선포 간 이개현상이 관찰되었으며, 구속 72시간 후에는 도관 주위 조직도 변성되기 시작하였고, 점차 심화되어 구속 168시간 후에는 선포와 도관 주위조직이 모두 심하게 변성되었다. 3. 면역조직화학적 검사에서 구속 3시간 후에는 amylase의 발현이 부분적으로 불규칙하게 관찰되었으며, 구속 6시간 후에는 다소 감소되었고, 구속 12시간 후에는 amylase가 부분적으로 응집되었으며, 점차 감소되고 응집되어, 구속 48시간 후에는 전 타액선에 amylase의 응집현상이 심화되었고, 구속 168시간 후에는 amylase의 발현이 매우 감소되었다. 이상의 결과를 바탕으로 정서적인 스트레스를 받으면 타액선이 변성될 수 있으며, 타액선으로부터 분비되는 중요한 소화 효소인 amylase가 변화되어 소화 장애를 일으키거나 구강건강과 관계없이 구취발생에도 영향을 미칠 수 있다고 예측할 수 있어, 향후 amylase의 분비변화에 따른 지속적인 연구가 필요할 것으로 생각된다.
오존에 노출된 갈참나무 4가계의 오존 내성 차이와 생리적 지표들의 오존 민감성은 광합성 특성, 광색소 함량, MDA 함량 및 항산화효소 활성과 같은 생리적 지표들을 표준화하여 결정하였다. 오존 처리 후, 갈참나무의 잎내 탄소고정효율 감소는 4가계 모두에서 관찰되었으며, 그들의 감소율은 대조구의 -24.1%에서 -56.9% 범위를 보였다. 광색소 함량은 가계 간 및 처리 간 차이가 뚜렷하였으며, 총 엽록소 함량의 감소는 SU4 가계에서 가장 높았고 (-40.6%), US2 가계에서 가장 낮았다(-18.8%). APX 활성은 가계 간 및 처리 간 차이를 뚜렷하게 보였으며, US2 가계를 제외한 3가계에서 대조구보다 높게 나타났다. 생리적 지표들을 기준으로 한, 갈참나무 4가계의 오존 내성은 US1 > SU4 > US2 > SU1의 순으로 나타났다. 결론적으로 광합성 특성, 광색소 함량과 APX 활성은 갈참나무의 오존 스트레스 내성을 평가하는데 적절한 지표로 판단된다.
Vulva and Vaginal cancers are rare among all gynecological cancers worldwide, including Thailand, and typically affect women in later life. Persistent high risk human papillomavirus (HR-HPV) infection is one of several important causes of cancer development. In this study, we focused on HPV investigation and specific type distribution from Thai women with abnormality lesions and cancers of the vulva and Vaginal. A total of ninety paraffin-embedded samples of vulva and Vaginal abnormalities and cancer cells with histologically confirmed were collected from Thai women, who were diagnosed in 2003-2012 at the National Cancer Institute, Thailand. HPV DNA was detected and genotyped using polymerase chain reaction and enzyme immunoassay with GP5+/bio 6+ consensus specific primers and digoxigenin-labeled specific oligoprobes, respectively. The human ${\beta}$-globin gene was used as an internal control. Overall results represented that HPV frequency was 16/34 (47.1%) and 8/20 (40.0%) samples of vulva with cancer and abnormal cytology lesions, respectively, while, 3/5 (60%) and 16/33 (51.61%) samples of Vaginal cancer and abnormal cytology lesions, respectively, were HPV DNA positive. Single HPV type and multiple HPV type infection could be observed in both type of cancers and abnormal lesion samples in the different histological categorizes. HPV16 was the most frequent type in all cancers and abnormal cytology lesions, whereas HPV 18 was less frequent and could be detected as co-infection with other high risk HPV types. In addition, low risk types such as HPV 6, 11 and 70 could be detected in Vulva cancer and abnormal cytology lesion samples, whereas, all Vaginal cancer samples exhibited only high risk HPV types; HPV 16 and 31. In conclusion, from our results in this study we suggest that women with persistent high risk HPV type infection are at risk of developing vulva and Vaginal cancers and HPV 16 was observed at the highest frequent both of these, similar to the cervical cancer cases. Although the number of samples in this study was limited and might not represent the overall incidence and prevalence in Thai women, but the baseline data are of interest and suggest further study for primary cancer screening and/or developing the efficiency of prophylactic HPV vaccines in Thailand.
Objective: To evaluate the application value of serum CA19-9, CEA, CA125 and CA242 in diagnosis and prognosis of pancreatic cancer cases treated with concurrent chemotherapy. Materials and Methods: 52 patients with pancreatic cancer, 40 with benign pancreatic diseases and 40 healthy people were selected. The electrochemiluminescence immunoassay method was used for detecting levels of CA19-9, CEA and CA125, and a CanAg CA242 enzyme linked immunoassay kit for assessing the level of CA242. The Kaplan-Meier method was used for analyzing the prognostic factors of patients with pancreatic cancer. The Cox proportional hazard model was applied for analyzing the hazard ratio (HR) and 95% confidential interval (CI) for survival time of patients with pancreatic cancer. Results: The levels of serum CA19-9, CEA, CA125 and CA242 in patients with pancreatic cancer were significantly higher than those in patients with benign pancreatic diseases and healthy people (P<0.001). The sensitivity of CA19-9 was the highest among these, followed by CA242, CA125 and CEA. The specificity of CA242 is the highest, followed by CA125, CEA and CA19-9. The sensitivity and specificity of joint detection of serum CA19-9, CEA, CA125and CA242 were 90.4% and 93.8%, obviously higher than single detection of those markers in diagnosis of pancreatic cancer. The median survival time of 52 patients with pancreatic cancer was 10 months (95% CI7.389~12.611).. Patients with the increasing level of serum CA19-9, CEA, CA125, CA242 had shorter survival times (P=0.047. 0.043, 0.0041, 0.029). COX regression analysis showed that CA19-9 was an independent prognostic factor for patients with pancreatic cancer (P=0.001, 95%CI 2.591~38.243). Conclusions: The detection of serum tumor markers (CA19.9, CEA, CA125 and CA242) is conducive to the early diagnosis of pancreatic cancer and joint detection of tumor markers helps improve the diagnostic efficiency. Moreover, CA19-9 is an independent prognostic factor for patients with pancreatic cancer.
The objective of this experiment was to determine the effects of n-3 polyunsaturated fatty acids (n-3 PUFAs)-enriched diet supplemented with different levels of $\alpha$-tocopherol on the activities of hepatic lipogenic-related enzymes and the contents of liver and plasma lipid fractions in laying Tsaiya ducks. A total of 180 30-wk-old laying Tsaiya ducks, at the beginning of peak production, were allotted into 6 treatments with 3 replicates each. Ducks were fed one of the 6 experimental diets, containing 4% tallow (control), and 4% fish oil supplemented with graded levels of $\alpha$-tocopheryl acetate ($\alpha$-tocopherol) at 0, 100, 200, 300 and 400 mg/kg, respectively, for 6 wks. Feed and water were supplied ad libitum throughout the experimental period. The results indicated that the n-3 PUFAsenriched diet supplemented with different levels of $\alpha$-tocopherol did not affect (p>0.05) egg weight, feed intake, body weight change or liver and abdominal fat weights. Egg production, egg mass and feed efficiency significantly (p<0.05) improved as dietary $\alpha$-tocopherol levels increased. The activities of hepatic lipogenic-related enzymes including acetyl-CoA carboxylase (EC 6. 2. 1. 3; ACC), glucose-6-phosphate dehydrogenase (EC 1. 1. 1. 49; G-6-PDH), ATP-citrate cleavage enzyme (EC 4. 1. 3. 8; CCE), NADP-malate dehydrogenase (EC 1.1.1.40; NADP-MDH) and fatty acid synthetase (FAS) were higher (p<0.05) in birds fed with the tallow diet than in those fed with fish oil diets and increased with increasing dietary $\alpha$-tocopherol levels. None of the dietary treatments significantly affected the contents of triglyceride and total cholesterol in the liver, or total cholesterol, phospholipid and total lipid in the plasma. However, the contents of phospholipid and total lipid in the liver, and triglyceride in the plasma increased as dietary $\alpha$-tocopherol levels increased. Increasing dietary $\alpha$-tocopherol levels decreased the non-esterified fatty acid (NEFA) content in the plasma and trended to decrease the cholesterol contents in the egg yolk. The lipid metabolism of laying Tsaiya ducks was influenced not only by the dietary fat but also by the supplementation levels of $\alpha$-tocopherol.
A total of 126 crossbred weanling pigs (average body weight of $6.3{\pm}0.3$ kg) were used to investigate the effect of chito-oligosaccharide (COS) on growth performance, nutrient digestibility, pH of gastro-intestinal tract (GI), intestinal and fecal microflora of young piglets. Pigs were allocated to three dietary treatments based on body weight and gender in a single factorial arrangement. Treatments were control (No COS), T1 (0.2% COS during starter (6-13 kg) and 0.1% COS during grower (13-30 kg) phases, and T2 (0.4% COS during starter (6-13 kg) and 0.3% COS during grower (13-30 kg) phases, respectively. Each treatment had 3 replicates and 14 pigs were raised in each pen. COS is a low molecular weight water-soluble chitosan that can be obtained from chitin of the crab shell after deacetylation with concentrated sodium hydroxide at high temperature and then further decomposition by chitosanase enzyme in the presence of ascorbic acid. For the starter and grower periods, there were no significant differences (p>0.05) in average daily gain (ADG) and feed to gain ratio among treatments. However, during the overall period (6-30 kg), T2 showed better (p<0.05) feed to gain ratio than other treatments. A digestibility study was conducted at the end of grower phase which showed improvement (p<0.05) in DM and crude fat digestibility in T2 over the control. At 25 kg body weight, 6 pigs per treatment (2 per replicate) were sacrificed to determine the effect of diets on pH and microbial count at different sections of the GI tract. The pH of the cecal contents in pigs fed 0.1% COS was higher (p<0.05) than in the other treatments. Total anaerobic bacterial number increased from cecum to rectum in all treatments. The weekly total bacterial counts showed higher (p<0.05) in feces of pigs fed COS than that of untreated pigs at the $8^{th}$ week. The number of fecal E. coli in untreated pigs at $4^{th}$ wk was 7.35 log CFU/g compared to 6.71 and 6.54 log CFU/g in 0.1 and 0.3% COS-treated pigs, respectively. Similarly, at $8^{th}$ wk, fecal clostridium spp. were lower in pigs fed 0.3% COS (5.43 log CFU/g) than in untreated pigs (6.26 log CFU/g). In conclusion, these results indicated that chito-oligosaccharide could improve feed efficiency in young pigs and inhibited the growth of harmful bacteria.

제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
