Presence of Cryptosporidium spp. and Giardia duodenalis in Drinking Water Samples in the North of Portugal

  • Cryptosporidium and Giardia are 2 protozoan parasites responsible for waterborne diseases outbreaks worldwide. In order to assess the prevalence of these protozoans in drinking water samples in the northern part of Portugal and the risk of human infection, we have established a long term program aiming at pinpointing the sources of surface water, drinking water, and environmental contamination, working with the water-supply industry. Total 43 sources of drinking water samples were selected, and a total of 167 samples were analyzed using the Method 1623. Sensitivity assays regarding the genetic characterization by PCR and sequencing of the genes, 18S SSU rRNA, for Cryptosporidium spp. and $\beta$, -giardin for G. duodenalis were set in the laboratory. According to the defined criteria, molecular analysis was performed over 4 samples. Environmental stages of the protozoa were detected in 25.7% (43 out of 167) of the water samples, 8.4% (14 out of 167) with cysts of Giardia, 10.2% (17 out of 167) with oocysts of Cryptosporidium and 7.2% (12 out of 167) for both species. The mean concentrations were 0.1-12.7 oocysts of Cryptosporidium spp. per 10 L and 0.1-108.3 cysts of Giardia duodenalis per 10 L. Our results suggest that the efficiency in drinking water plants must be ameliorated in their efficiency in reducing the levels of contamination. We suggest the implementation of systematic monitoring programs for both protozoa. To authors' knowledge, this is the first report evaluating the concentration of environmental stages of Cryptosporidium and Giardia in drinking water samples in the northern part of Portugal.

대청호의 수질과 남조류 독소 Anatoxin-A 농도의 관계 (Water Quality and Cyanobacterial Anatoxin-a Concentration in Daechung Reservoir)

  • 남조류에 의해 생성되는 anatoxin-a의 양과 환경요인과의 관계를 알아보기 위해 대청호에서 2001년 6월부터 2001년 11월까지 조류 및 물 시료를 채취하였다. 환경요인 중 물리적 요인은 현장에서 측정하였고, 생물${\cdot}$화학적 요인은 실험실에서 측정하였다. 조류 및 물 시료에 존재하는 anatoxin-a의 양은 fluorescence detector를 이용하여 HPLC로 측정하였고, 조류 시료와 물 시료의 경우 각각$0.61-8.68\;{\mu}g/g\;dry wt$,$0.01-0.08\;{\mu}g/L$로 측정되었다. 음용수의 안전을 고려한 anatoxin-a의 권고기준 농도는 $1\;{\mu}g/L$로 제안되고 있다. 조류세포와 물 시료에서 anatoxin-a농도가 가장 높게 검출된 시기는 7월이었다. 독소 생성에 중요한 요인을 확인하기 위해 환경요인과 anatoxin-a농도와의상호관계를 살펴보았다. 조류 시료내 anatoxin-a농도는 nitrate,총질소와 총인 비율, 총용존질소(P<0.05)및 총입자성질소와 총입자성인 비율(P<0.05)과 높은 상관관계를 나타내었고, 물 시료내anatoxin-a 농도는 수온, 전기전도도(P<0.01), 수소이온농도, phycocyanin, phycocyanin과 엽록소 a비율 (P<0.01)과 높은 상관관계를 나타내었다.

부산지역 음용수의 세균학적 특성 (Bacteriological Characteristics of Drinking Water in Pusan Area)

  • One hundred and eighty-seven water samples were collected from 23 of spring water, 2 of ground water, 1 of tap water in Pusan area and 3 of natural mineral waters. Total coliform group, fecal coliform, viable cell count and microflora were investigated to evaluate water quality of drinking water. The results were as follows: range and geometric mean value of total coliform and fecal coliform MPN's of spring water were 0~1,500/100 ml, 85/100 ml and 0~460/100 ml, 24/100 ml but coliform group was not detected in the samples of tap water and natural mineral water. Viable cell count of spring water, ground water and tap water were lower as 100 cell than the criteria for drinking water but that of natural mineral water was higher as 6.5X 10$^2$~7.4X 10$^3$ /ml. Predominant speces among the 219 strains isolated from the samples were 19.6% Aeromonas spp., 19.2% Enterobacteriaceae, 16% Acinetobacter spp. Especially, spring water and vessels were contaminated by Hafnia spp. and Providencia Spp, inhabitant of the oral cavity.

溫水수도꼭지와 온수에서 Legionella pneumophila의 分離同定 (Isolation and Identification of Legionella pneumophila from Hot Water Spouts and Hot Water)

  • A study was carried out to find distribution of Legionella spp. in hot water supplying systems of buildings in Seoul. Water samples were taken from the hot water taps in three research institute buildings during the period from the 2nd to 27th February, 1987. And all of the three buildings were supplied with hot water from the same central hot water tank. GVPC BCYEa and blood agar were used for the isolation of Legionella pneurnophila and slide agglutination test (SAT) was performed to identify their strain types. Main findings were as follows 1. 44 samples taken from hot tap-water revealed 20% of positive culture and 44 samples taken from hot water spouts revealed 11% of positive culture. 2. The 78% of positive samples taken from hot water was obtained from 25.1$\circ$C~45.0$\circ$C temperature range. 3. Only, Legionella pneumophila serogroup 3 was identified and most of them has been than 2$\mu$m in length. 4. The densities of the Legionella pneurnophila isolated from hot water samples were within the range of $1.0\times 10^2$ to $1.1\times 10^3$ CFU/l and their densities isolated from hot water spouts were either 1 CFU/plate or 2 CFU/plate.

1,4-Dioxane의 분석방법 연구 (A Study on the Research of Analytical Method for 1,4-Dioxane in Water)

  • The study was carried out to develop and standardize the analytical method of 1,4-dioxane in water. The National Institute of Environmental Research in Korea has monitored for 1,4-dioxane in water since 1998 and it has been detected in environmental water occasionally. But the environmental pollution and analytical method in water is not well known over the world. The trace analytical method at low-${\mu}/L$ level is prerequisite in order to evaluate the residue of 1,4-dioxane in water. Evaluation of the method was demonstrated by comparing and analyzing with liquid-liquid extraction, solid-phase extraction and purge & trap technique. As a result of experiment by standard solutions and raw samples, the precision and accuracy for three methods were obtained within error rate of about 10%. Therefore, three methods were standardized as official monitoring method in Korea. Also, a convenient and simple liquid-liquid extraction method for the analysis of 1,4-dioxane in water was developed by combined with gas chromatography/mass spectrometry and applied to the water samples in Korea

세포배양 생화학적 기법에 의한 수환경오염 평가 (Cell Culture Microbioassay for the Water Pollution Monitoring)

  • So far, investigation of environmental pollution has been achieved in field study. This remains the most exhaustive approach, current dimensions of environmental researches and their inherent complexity require that relatively inexpensive and simple laboratory procedures are developed to make possible the screening of large numbers of sites and samples. At this point. microbioassay has been high-lighted. The purpose of this study is to evaluate the water pollution using microbioassay. Two microbioassay methods were optimized and validated for the sensitive and quantitative determination of total toxic effects in the water. EROD(Ethoxyresorufin-O-deethylase) microbioassay was focused to detect PARs, PCBs and dioxinlike components in the water and E-screen assay to xenoestrogens. The EROD microbioassay was executed in rat hepatoma cell line, H4IIE and E-screen assay in MCF7-BUS cell line. Kumho river was selected for this study. 5ι of river water was extracted using combined solid-phase extraction in static adsorption mode with soxhlet extraction. Pollutants adsorbed to the XAD-4 resin were recovered by elution with ethyl acetate and methylene chloride (1 : 9). Toxic effects of extracts were determined by EROD-microbioassay and E-screen assay. EROD activities of water samples were 7.24-72.24 ng/ι MEQ. The estrogenic effect of various water samples is quantitatively evaluated by EEQ. The EEQ of samples range from 0.05 to 6.07 ng-EEQ/ι. These results suggested that Kumho river was highly polluted with organic toxic chemicals.

ICP/AES에 의한 먹는물의 무기원소 분석 (Elemental Analysis of Drinking Water with ICP/AES)

  • Inductively coupled plasma atomic emission spectrophotometer (ICP/AES) is a versatile modern instruments for the multi-element analysis, but quantitative analysis using ICP/AES with normal pneumatic nebulizer is not applicable for the measurement of elemental concentrations in water down to the drinkining water standard level except a few elements because of poor detection limits. However, the detection limit can be lowered enough to measure drinking water standard, if ultrasonic nebulizer and/or hydride vapor generator is attached. This method is tested with groundwater samples from Tajeon area. It is confirmed that the elemental concentrations in these samples are within the limit of drinking water standard for the most elements. However, uranium concentration is very high in some samples compared with the concentrations suggested by Environmental Protection Agency of U.S.A. There is no standard concentration level to this element in Korea and it should be prepared immediately.

신(新) H/Device를 이용한 자연수의 수소동위원소비 측정 (Introduction to a New Sample Preparation Apparatus (H/Device) for Measurement of Hydrogen Isotope Composition of Natural Water)

  • In the hydrologic and hydrochemical studies of natural waters, oxygen and hydrogen isotope compositions of waters are very important to elucidate the origin and circulation pattern of water in the hydrologic system. The hydrogen isotope analysis of waters usually has been undertaken through the reduction of water to form hydrogen gas using pure metals (in general, zinc and uranium). In 1996, a new apparatus (H/Device) was developed to prepare the water samples (by the reduction with Cr metal) without some intrinsic problems that may yield incorrect and/or inaccurate data, and was installed at 1997 in the Center for Mineral Resources Research (CMR) in Korea University. However, the optimistic conditions of preparation and analysis of samples has not been established. In this paper, we introduce the efficiency of H/Device to obtain accurate hydrogen isotope values of water, and discuss both the optimum conditions including the effective reduction time and the probable mixing (memory) effect between successive samples. We obtained large amounts of a laboratory working standard (KUW; Korea University Water) with the average ${\delta}D_{SMOW}$ value of $-42.1{\pm}1.0$$(1{\sigma})$.

Nested PCR-RFLP 및 DNA Sequencing을 이용한 환경시료에서의 크립토스포리디움 동정 (Identification of Cryptosporidium in Environmental Sample using Nested PCR-RFLP and DNA Sequencing)

  • In order to identify various Cryptosporidium species in environment, nested PCR-RFLP and DNA sequencing method were used. The sensitivity of nested PCR-RFLP based on 18s rRNA gene was shown to 1 oocyst. Therefore, we applied nested PCR-RFLP method to environmental samples. As a result, only 4 samples out of 8 samples confirmed as Cryptosporidium parvum by standard method of Cryptosporidium were identified as Cryptosporidium parvum by nested PCR-RFLP and DNA sequencing method. The rest of 4 samples among 8 samples were identified as Cryptosporidium muris, Cryptosporidium bailey. Therefore, in addition to standard method of Cryptosporidium, supplementary verification through nested PCR-RFLP and DNA sequencing should be needed to give more accurate information about risk of Cryptosporidium.

전북 지역 일부 수원지의 물에서 불소, 칼슘 및 마그네슘 함량에 관한 조사 연구 (A Survey on the Contents of Fluoride, Calcium, and Magnesium of Reservoir Water on a Stream in the Jeon-buk Area of Korea)

  • This study was performed to investigate the levels of fluoride (F), calcium (Ca), and magnesium (Mg) in water samples taken from five reservoirs or direct sources on a stream used for agricultural or domestic water in the Iksan and Wanjoo areas, Jeon-buk, Korea, and to find a possible link between Ca or Mg and fluoride in water sources in theses areas. The samples were collected by the recommendation methods of the World Health Organization and analyzed by the recommendations of the Japanese Standard Methods. Statistical analyses were performed by the analysis of variance and correlation analysis. The F levels in water samples wire 0.80~2.53 ppm. In four sampling sites the water fluoride levels exceeded 1 ppm. which if the recommended level for fluorosis/caries control. The Ca levels in water samples were 6.82~12.98 ppm, and the Mg levels were 0.30~1.97 ppm, which are lower compared with the natural levels of water sources previously reported by other investigators. This study showed a positive correlation between Ca and Mg (r= 0.8779. p<0.01) and a negative correlation between F and Ca (r=-0.6974, p<0.05) and also between F and Mg (r=-0.5581) in the water samples. However, the study did not find remarkable relationships in fluoride levels between sampling sites. These results support the fact that there were epidemics of dental fluorosis in this area. The lack of significant positive correlations in fluoride levels between sampling sites suggests that there may be some pathways for the transfer of the metal to the water through other environmental media besides the water course. Long-term epidemiological studies are needed on the relationship between high F together with low Ca and Mg levels in the water, and total human health in this community. There should alto be a long-term monitoring of the water quality in this area.
